A 12-year field experiment was conducted to investigate the effect of different tillage methods and fertil- ization systems on microbial biomass C, N and P of a gray fluvo-aguic soil in rice-based cropping system. Five fertilization treatments were designed under conventional tillage (CT) or no tillage (NT) system: no fertilizer (CK); chemical fertilizer only (CF); combining chemical fertilizer with pig manure (PM); combining chemical fertilizer with crop straw (CS) and fallow (F). The results showed that biomass C, N and P were enriched in the surface layer of no-tilled soil, whereas they distributed relatively evenly in the tilled soil, which might result from enrichment of crop residue, organic manure and mineral fertilizer, and surficial development of root systems under NT. Under the cultivation system, NT had slightly greater biomass C, N and P at 0~5 cm depth, significantly less biomass C, N and P at 5~15 cm depth, less microbial biomass C, N and equivalent biomass P at 15 ~30 cm depth as compared to CT, indicating that tillage was beneficial for the multiplica tion of organisms in the plowed layer of soil. Under the fallow system, biomass C, N and P in the surface layer were significantly greater for NT than CT while their differences between the two tillage methods were negligible in the deeper layers. In the surface layer, biomass C, N and P in the soils amended with organic manure combined with mineral fertilizers were significantly greater than those of the treatments only with mineral fertilizers and the control. Soils without fertilizer had the least biomass nutrient contents among the five fertilization treatments. Obviously, the long-term application of organic manure could maintain the higher activity of microorganisms in soils. The amounts of biomass C, N and P in the fallowed soils varied with the tillage methods; they were much greater under NT than under CT, especially in the surface layer, suggesting that the frequent plowing could decrease the content of organic matter in the surface layer of the fallowed soil. 展开更多

Phosphorus (P) risk indices are commonly used in the USA to estimate the field-scale risk of agricultural P runoff. Because the Ohio P Risk Index is increasingly being used to judge farmer performance, it is important to evaluate weighting/scoring of all P Index parameters to ensure Ohio farmers are credited for practices that reduce P runoff risk and not unduly penalized for things not demonstrably related to runoff risk. A sensitivity analysis provides information as to how sensitive the P Index score is to changes in inputs. The objectives were to determine 1) which inputs are most highly associated with P Index scores and 2) the relative impact of each input variable on resultant P Index scores. The current approach uses simulations across 6134 Ohio point locations and five crop management scenarios (CMSs), representing increasing soil disturbance. The CMSs range from all no-till, which is being promoted in Ohio, rotational tillage, which is a common practice in Ohio to full tillage to represent an extreme practice. Results showed that P Index scores were best explained by soil test P (31.9%) followed by connectivity to water (29.7%), soil erosion (13.4%), fertilizer application amount (11.3%), runoff class (9.5%), fertilizer application method (2.2%), and finally filter strip (2.0%). Ohio P Index simulations across CMSs one through five showed that >40% scored <15 points (low) while <1.5% scored >45 points (very high). Given Ohio water quality problems, the Ohio P Index needs to be stricter. The current approach is useful for Ohio P Index evaluations and revision decisions by spatially illustrating the impact of potential changes regionally and state-wide. 展开更多

Networked manufacturing produces some problems while bringing enterprise infin ite commercial chance. Compared with traditional manufacturing, stealing intelle ctual achievements and encroaching the related property rights not only becomes easier and quicker, but also leaves no traces when enterprises transfer informat ion through network. It′s well known that competition among corporations in the future is the on e of intellectual property in the final analysis. Therefore, recently, the statu s of intellectual property protection is promoted grandly. In the following parts, several proposals on protecting intellectual property ar e put forward in detail. We can protect the intellectual property by the ways of strengthening the consci ousness of protection. We can protect the intellectual property by perfecting the law. We will know which behaviors on the network are legitimate and which one are illegitimate. As a result, it can decrease and resolve the conflicts partially. Besides, the article puts forward that the conflicts can be resolved through a m edi-institution, an authoritative governmental organization. Moreover, we should strengthen the education on ethics. However, the overhead caused by proof of compliance on authorization and the absence of certifying authorities may negate the strong and objective security advantages of policy-based approach, whilst vagueness, complexity and inaccurate characterization caused by reputation evolution may eliminate the quantitative and flexible advantages of reputation-based approach. We propose an adaptive trust management framework, which combines the merit of policy proof and reputation evolution such that authorization is aware of not only the strong and objective security traits, but also the calculability and the availability security traits. Premature Rupture of Membranes (PROM) with the resulting prematurity remains a major public health issue in the Democratic Republic of Congo (DRC). This study aimed to assess expectant management of PPROM before 34 weeks at the university hospital of Kinshasa. We conducted a retrospective analysis of expectantly managed PROM before 34 weeks between January 2008 and December 2018. Maternal and fetal outcomes were collected, and all data were analyzed using the SPSS 23.0 software. Of the 113 patients included in the study, 2.6% were diagnosed with PROM before 34 weeks. We observed prolongation of the pregnancy duration;the median latency period was eight days, and the average gestational age at delivery of 32.85 ± 2.5 weeks. Chorioamnionitis (23%), severe oligoamnios (7%), and acute fetal distress (4%) were complications observed during the latency period. In the postpartum period, endometritis (6.2%), neonatal jaundice (39.8%), anemia (25.7%), ulcerative necrotizing enterocolitis (6.2%), cerebromeningeal hemorrhage (5.3%), and acute respiratory distress syndrome (4.4%) were complications observed. The risk of infection during the latency period was significantly associated with irregular (P = 0.045) or lack (P = 0.006) antenatal care (ANC) attendances and C-Reactive Protein (CRP) results 6 (P = 0.013). The risk of neonatal death was significantly associated to infection during the latency period (P = 0.011), irregular (P = 0.009) or lack of ANC (P = 0.000) attendances, Birth weight g (P = 0.039) as well as Gestational age at birth between 28 to 30 Weeks (S) (P = 0.021). These findings report first-time pregnancy outcomes related to the management of PPROM before 34 weeks in our setting. We found that the conservative attitude adopted allowed the prolongation of pregnancies, reducing the risks associated with prematurity. Nevertheless, attendance in good quality ANC could reduce the frequency of PROM and related adverse outcomes. 展开更多

目的 观察冰水泳浴对痛风大鼠模型病理变化的影响并探究其中P2X7R的调控机制。方法 雄性SD大鼠分为正常(NORM)组和实验组,实验组包括痛风对照(GC)组、冰水泳浴(IWS)组和亮蓝G(BBG,P2X7R拮抗剂)组。实验组大鼠通过尿酸酶抑制法联合Coderr... 目的 观察冰水泳浴对痛风大鼠模型病理变化的影响并探究其中P2X7R的调控机制。方法 雄性SD大鼠分为正常(NORM)组和实验组,实验组包括痛风对照(GC)组、冰水泳浴(IWS)组和亮蓝G(BBG,P2X7R拮抗剂)组。实验组大鼠通过尿酸酶抑制法联合Coderre法塑造高尿酸血症合并痛风性关节炎模型。冰水泳浴组大鼠在Coderre法造模后的0 h和12 h,分别于深约0.5 m的冰水混合物中进行5 min的耐力游泳,BBG组则在造模后立即腹腔注射BBG溶液1次。公式计算踝关节肿胀指数;尿酸酶法检测大鼠血清尿酸水平;ELISA法检测血清IL-1β、IL-6和TNF-α含量;HE染色观察踝关节和滑膜组织的病理变化;Western blot和免疫组化分别检测滑膜组织中P2X7R和NLRP3蛋白表达。结果 与正常组相比,实验组大鼠血清尿酸水平及踝关节肿胀指数均显著升高(P<0.05或P<0.01),且滑膜组织均存在不同程度的炎性浸润。与痛风对照组相比,冰水泳浴组踝关节肿胀指数在12 h显著升高(P<0.05),血清IL-1β、IL-6和TNF-α含量(P<0.01)及滑膜组织中P2X7R、NLRP3蛋白量均有显著表达(P<0.05);病理结果显示软骨表面破损,滑膜组织出现严重增生与糜烂,并伴有大量炎性细胞聚集。BBG组大鼠滑膜组织的P2X7R、NLRP3蛋白表达与病理损伤较痛风对照组无显著变化(P>0.05),而血清IL-1β、IL-6和TNF-α的表达均有显著改善(P<0.01)。结论 冰水泳浴模拟的寒冷刺激和剧烈运动可加重痛风性关节炎的病理损伤,其机制可能与关节局部P2X7R的高表达有关。 展开更多

目的:探讨环状RNA天冬酰胺β-羟化酶(CircASPH)靶向miR-28-5p/胰岛素样生长因子1受体(IGF-1R)轴对多囊卵巢综合征(PCOS)卵巢颗粒细胞增殖、迁移和侵袭的影响。方法:使用人卵巢颗粒细胞KGN、COV434为研究对象,通过双荧光素酶报告基因实... 目的:探讨环状RNA天冬酰胺β-羟化酶(CircASPH)靶向miR-28-5p/胰岛素样生长因子1受体(IGF-1R)轴对多囊卵巢综合征(PCOS)卵巢颗粒细胞增殖、迁移和侵袭的影响。方法:使用人卵巢颗粒细胞KGN、COV434为研究对象,通过双荧光素酶报告基因实验、下拉实验证实CircASPH、miR-28-5p、IGF-1R三者间的靶向关系。将KGN、COV434细胞分为si-NC组、si-ASPH组、si-ASPH+anti-NC组、si-ASPH+anti-miR-28-5p组,采用实时荧光定量PCR(qRT-PCR)检测CircASPH、miR-28-5p和IGF-1R mRNA表达水平,并用四甲基偶氮唑盐(MTT)法、5-乙炔基-2′脱氧尿嘧啶核苷(Edu)染色和迁移小室实验分别检测细胞增殖、迁移和侵袭行为;采用蛋白印迹实验检测增殖细胞核抗原、基质金属蛋白酶-2(MMP-2)、波形蛋白、N-钙黏蛋白、E-钙黏蛋白、IGF-1R蛋白表达。结果:生物信息学分析和双荧光素酶报告基因实验显示,CircASPH、IGF-1R与miR-28-5p有靶向结合位点。与si-NC组比较,si-ASPH组CircASPH表达水平、OD 490值、Edu阳性细胞率、细胞迁移与侵袭数、MMP-2、波形蛋白、N-钙黏蛋白降低,miR-28-5p表达水平和E-钙黏蛋白表达升高,差异均有统计学意义(P<0.05)。与si-ASPH+anti-NC组比较,si-ASPH+anti-miR-28-5p组miR-28-5p表达水平和E-钙黏蛋白降低,OD 490值、Edu阳性细胞率、细胞迁移与侵袭数、MMP-2、波形蛋白、N-钙黏蛋白升高,差异均有统计学意义(P<0.05)。结论:在KGN、COV434细胞中,抑制CircASPH表达可通过调节miR-28-5p/IGF-1R轴,抑制卵巢颗粒细胞增殖、迁移、侵袭及上皮间质转化,可能成为治疗PCOS的一种新靶点。 展开更多

目的:探讨异鼠李素(ISO)对口腔鳞状细胞癌(OSCC)细胞生物学特性的影响,以及在该过程中对miR-454-3p/PIK3R1轴的调控机制。方法:MTT法检测ISO对TSCC1细胞的毒性,随后将TSCC1细胞分为OSCC组、ISO组、ISO+miR-NC组、ISO+miR-454-3p mimic组... 目的:探讨异鼠李素(ISO)对口腔鳞状细胞癌(OSCC)细胞生物学特性的影响,以及在该过程中对miR-454-3p/PIK3R1轴的调控机制。方法:MTT法检测ISO对TSCC1细胞的毒性,随后将TSCC1细胞分为OSCC组、ISO组、ISO+miR-NC组、ISO+miR-454-3p mimic组。MTT法检测各组细胞的存活率;克隆形成实验检测各组细胞增殖情况;流式细胞术检测各组细胞凋亡;Transwell实验检测各组细胞迁移和侵袭能力;RT-qPCR法检测各组细胞中miR-454-3p和PIK3R1的mRNA水平;Western blotting法检测各组细胞中PIK3R1蛋白的表达;双荧光素酶实验验证miR-454-3p与PIK3R1的靶向关系;建立裸鼠移植瘤模型,将小鼠分为模型组、ISO组、ISO组+agomir-NC组,ISO组+miR-454-3p agomir组,每组5只,干预15 d后检测各组小鼠的肿瘤质量和体积;RT-qPCR法检测肿瘤组织中miR-454-3p和PIK3R1的mRNA水平。结果:体外实验发现,与OSCC组比较,ISO组和ISO+miR-NC组细胞存活率、迁移和侵袭细胞数量、miR-454-3p表达水平均降低(P<0.05),细胞凋亡率、PIK3R1 mRNA和蛋白表达水平均升高(P<0.05);miR-454-3p mimic回补实验逆转了ISO对OSCC的抑癌作用及PIK3R1的表达水平(P<0.05),同时双荧光素酶报告基因实验验证了miR-454-3p与PIK3R1之间存在靶向关系;体内实验发现,与模型组比较,ISO组小鼠移植瘤的质量和体积均减小(P<0.05),miR-454-3p表达水平、PIK3R1 mRNA表达水平以及miR-454-3p agomir的回补实验结果均与体外实验保持一致。结论:ISO能够通过调节miR-454-3p/PIK3R1轴抑制OSCC细胞的增殖、迁移和侵袭,促进细胞凋亡。 展开更多

多模式出行信息对小汽车出行者转向P+R的影响

为倡导绿色出行理念,解决以往研究在处理重复观测数据时容易忽视的潜在相关性和个体异质性问题,针对如何利用智能手机APP提供的多模式出行信息引导小汽车出行者转向停车换乘(Park-and-Ride,P+R)模式进行了探究,同时引入广义线性混合模型... 为倡导绿色出行理念,解决以往研究在处理重复观测数据时容易忽视的潜在相关性和个体异质性问题,针对如何利用智能手机APP提供的多模式出行信息引导小汽车出行者转向停车换乘(Park-and-Ride,P+R)模式进行了探究,同时引入广义线性混合模型(Generalized Linear Mixed Model,GLMM)分析了多模式出行信息对小汽车出行者转向P+R意向的影响。首先,基于上海市路网设计意向调查问卷,整合了自驾和P+R两种出行方式的道路拥堵程度、出行时间、停车费用及地铁车厢座位情况等信息,并运用全因子设计法构建了24种不同信息水平组合的假设情景。然后,通过智能手机APP界面示意图向小汽车出行者展示这些多模式出行信息,并收集其转向P+R的意向数据。最后,运用GLMM方法处理同一个体重复决策数据中潜在的相关性和捕捉个体间的异质性。结果显示,GLMM的应用不仅解决了同一个体重复决策间的相关性,还揭示了不同个体对道路拥堵程度和地铁车厢座位情况的差异化关注;智能手机APP整合的多模式出行信息显著提升了小汽车出行者转向P+R的意愿,且这一转变占比达29.2%;高收入、长驾龄以及对P+R政策不了解的出行者转向P+R的意愿较低。研究表明,通过智能手机APP整合自驾和P+R的多模式出行信息能显著增强P+R方式的吸引力,可为提升P+R的普及率提供新思路,有效促进小汽车出行者向绿色出行方式的转变。 展开更多

目的探讨人参皂苷Rc(Ginsenoside Rc,简称Rc)对酒精性肝病的保护作用及其作用机制。方法采用50 mmol/L乙醇(EtOH)刺激小鼠肝细胞(AML-12)12 h后,采用不同浓度Rc处理24 h。Western blot方法检测AML-12细胞中P2X7r、NLRP3和IL-23蛋白表达... 目的探讨人参皂苷Rc(Ginsenoside Rc,简称Rc)对酒精性肝病的保护作用及其作用机制。方法采用50 mmol/L乙醇(EtOH)刺激小鼠肝细胞(AML-12)12 h后,采用不同浓度Rc处理24 h。Western blot方法检测AML-12细胞中P2X7r、NLRP3和IL-23蛋白表达情况;RT-qPCR测定AML-12细胞中SREBP1、Lipin-1、P2X7r、NLRP3的mRNA表达情况;通过ELISA法分析肝细胞上清液中IL-1β和IL-18水平。结果与对照组比较,EtOH能够诱导AML-12细胞脂质变性和炎症因子释放,Rc能够有效下调P2X7r、NLRP3和IL-23的蛋白或mRNA表达,抑制EtOH诱导AML-12细胞中SREBP1和Lipin-1的mRNA表达,抑制EtOH诱导AML-12细胞中IL-1β和IL-18向胞外释放(均P<0.001)。结论Rc能够抑制酒精性肝病进程,作用机制可能与抑制P2X7r-NLRP3信号通路有关。 展开更多
