P16蛋白在宫颈癌和宫颈上皮内瘤变中的表达及临床意义

目的分析在宫颈上皮内瘤变、宫颈癌中P16蛋白表达水平与临床意义。方法收集2019年5月-2020年12月惠州市第六人民医院上皮内瘤变、宫颈癌、宫颈炎患者共142例,根据疾病类型分组,宫颈炎组21例、宫颈癌组63例、宫颈上皮内瘤变组58例,经免... 目的分析在宫颈上皮内瘤变、宫颈癌中P16蛋白表达水平与临床意义。方法收集2019年5月-2020年12月惠州市第六人民医院上皮内瘤变、宫颈癌、宫颈炎患者共142例,根据疾病类型分组,宫颈炎组21例、宫颈癌组63例、宫颈上皮内瘤变组58例,经免疫组化染色,检测所有受试者P16水平,分析P16表达水平与宫颈病变的关系。结果本组142例受试者中,P16阳性检出97例(68.31%),阴性检出45例(31.69%);宫颈炎组患者P16阳性检出率0.00%,显著低于CIN组的70.69%、宫颈癌组的88.89%,差异有统计学意义(P<0.05)。CINⅠ级患者P16阳性检出率52.94%,显著低于CINⅡ级组(68.75%)与CINⅢ级组(84.00%),差异有统计学意义(P<0.05)。随着HR-HPV载量等级升高,P16蛋白阳性表达率越高,差异有统计学意义(P<0.05)。HR-HPV载量阳性表达在宫颈炎组中占28.57%,显著低于CINⅠ级组(70.59%)、CINⅡ级组与Ⅲ级组(80.49%)、宫颈癌组(95.24%),差异有统计学意义(P<0.05)。Spearman分析,在宫颈炎、CINⅠ级组中HR-HPV载量与P16呈正相关(P<0.01);而在宫颈癌、CINⅡ级与Ⅲ级组中,HR-HPV载量与P16表达并无明显相关性(P>0.05)。结论CIN病变程度、宫颈癌与病灶组织中P16蛋白表达存在相关性,P16可作为临床诊断重要的生物学标记。 展开更多

Objective Bmi-1, a putative proto-oncogene, is a core member of the polycomb gene family, which is expressed in many human tumors. The p16 protein negatively regulated cell proliferation, whereas CD44v6 is associated with proliferation as an important protein. Additionally, CD44v6 is an important nuclear antigen closely correlated to tumor metastasis. Tlle present study aims to investigate the expression and significance of Bmi-1, p16, and CD44v6 in uterine cervical carcinoma (UCC). Methods A total of 62 UCC, 30 cervical neoplasic, and 20 normal cervical mucosal tissues were used ill the current study. The expression of Bmi-1, p16, and CD44v6 in these tissues was determined using immunohistochemical assay. The relationships among the expression of these indices, the clinicopathologic features of UCC, and the survival rate of UCC patients were also discussed. The correlation between Bmi-1 protein expression and p16 or CD44v6 protein in UCC was analyzed. Results The expression of Bmi-l, p16, and CD44v6 was significantly high in cervical carcinoma compared with that in tlle cervical neoplasia and normal colorectal mucosa (P〈0.05). The over-expression of Bmi-1 protein in UCC was apparently related to the distant metastasis (P〈0.01) and the tumor, nodes and metastasis-classification, i.e. the TNM staging, World Health Organization (P〈0.05). Nevertheless, the positive expression of p16 protein in UCC was not significantly associated with the clinicopathologic features (P〉0.05). The Kaplan-Meier survival analysis showed that the over-expression of Bmi-1 significantly decreased the survival rate of UCC patients (P〈0.05). A strong correlation indicated that there was statistical significance between the expression of Bmi-1 and CD44V6 proteins in UCC (r=0.419, P=0.001). Conclusions The over-expression of Bmi-1 and CD44v6 protein closely correlate to the tumorigenesis, metastasis, and prognosis of UCC. Bmi-I and CD44v6 may be used to predict the prognosis of cervical carcinoma. Bmi-1 may indirectly regulate the expression of CD44v6 in UCC patients. The positive expression of p16 protein is possibly associated with the tumorigenesis, but not with the metastasis or prognosis of UCC. 展开更多

Association of cyclin D1, p16 and retinoblastoma protein expressions with prognosis and metastasis of gallbladder carcinoma 被引量:4

AIM: To investigate the role of cyclin D1, p16 and retinoblastoma in cancerous process of gallbladder carcinomas and to assess the relation between cyclin D1, p16, Rb and the biological characteristics of gallbladder carcinoma. METHODS: Forty-one gallbladder carcinoma, 7 gallbladder adenoma and 14 chronic cholecystitis specimens were immunohistochemically and histopathologically investigated for the relation of cyclin D1, p16 and Rb with Nevin staging and pathologic grading. RESULTS: The expression rates of abnormal cyclin Dl in gallbladder carcinoma (68.3%)and gallbladder adenoma (57.1%) were significantly higher than those in chronic cholecystitis (7.1%) (P<0.05). No significant difference was found both among the pathological grades G1, G2 and G3 and among Nevin stagings S1-S2, S3 and S4-S5 of gallbladder carcinoma. The positive rates of p16 (48.8%) and Rb (58.5%) in gallbladder carcinoma were significantly lower compared to those in adenoma (100.0%) and cholecystitis (100.0%) (P<0.05). The positive rates of p16 and Rb in Nevin stagings S1-S2 (80.0% and 90.0%) and S3 (46.2% and 61.5%) gallbladder carcinomas were significantly higher than those in S4-S5(33.3% and 38.8%) (P<0.05), and those in pathologic grades G1(54.5% and 81.8%) and G2 (50.0% and 62.5%) gallbladder carcinoma were significantly higher than those in G3 (28.6% and 35.7%) (P<0.05). The protein expression of p16 and Rb had a negative-correlation in gallbladder carcinoma (r= -0.2993, P<0.05), and this negative-correlation was correlated with Nevin staging (P<0.05). Moreover, the protein expression of p16 and cyclin Dl had a negative-correlation in gallbladder carcinoma (r = -0.9417, P<0.05). CONCLUSION: Cyclin Dl may play a role in the early stage of gallbladder carcinoma. Mutation of p16 and Rb genes might be correlated with progression of gallbladder carcinoma. Analysis of p16 and Rb can estimate the prognosis of gallbladder carcinoma. Expression of p16 and Rb may be correlated with Nevin staging and pathologic grading in gallbladder carcinoma. 展开更多

Objective: To investigate the expression of cyclooxygenase-2 (COX-2) and p16 proteins in non-Hodgkin's lymphomas (NHL) and their relationship with the genesis and progress of it. Methods: The expression of COX-2 and p16 protein were studied in the lymph nodes tissue from 60 NHL patients and 10 control patients with non-malignant diseases by flow cytometry. Results: Positive rate of COX-2 protein expression in NHL tissues (63.3%, 38/60) was higher than that in normal lymphaden tissues (0, 0/10). The difference was significant between the two groups (P 〈 0.01). Expression of COX-2 protein was related with the clinical stage of NHL. In stage Ⅰ + Ⅱ patients, it was significantly lower (35.0% ± 54.6%) than that in stage Ⅲ + Ⅳ patients (84.6% ±87.5%) (P 〈 0.01). In different sex, age, tumor malignant degree, IPI grade, extranodal involvement and B symptoms groups, the differences of COX-2 expression were not statistically significant (P 〉 0.05). Positive rate of p16 protein expression (41.7%, 25/60) in NHL' was statistically lower than that in normal lymphomas (100%, 10/10) (P 〈 0.01). Expression of p16 protein was related to malignant degree of NHL. The positive rates of p16 protein in low malignant degree tissues (64.7%, 11/17) was higher than that in high malignant degree tissues (14.3%, 2/14) (P 〈 0.05). Positive rates of p16 protein of NHL tissues in different sex, age, IPI grade, extranodal involvement, clinical stages and B symptoms were not statistically significant (P 〉 0.05). The p16 protein expression in COX-2 positive patients was 47.4% (18/38), and in negative patients it was 31.8% (7/22). There was no statistically difference between them (P 〉 0.05). Correlation analysis revealed there was no correlation between expression of COX-2 and p16 protein. Conclusion: Both COX-2 and p16 protein may all have relationship with the genesis and progress of NHL. The expression of COX-2 protein in NHL may be a poor prognostic indicator. COX-2 and p16 protein probably have different mechanisms in the genesis and progress of NHL. Their relationship is firstly put forward in this article and needed further studying. 展开更多

Methods: Immunohistochemical technique (SABC) was used to detect p16 and C-fos protein expression in 41 cases of osteosarcoma. Results: The positive rates of p16 and C-fos protein expression were 51.2% and 82.9% respectively. Their expression was not correlated to pathological subtype, but correlated to clinic grade, and the latter was associated with tumor metastasis. There was a negative correlation between p16 and C-fos protein expression. Conclusion: The alteration of p16 and C-fos protein expression may be related to the tumorigenesis and development of osteosarcoma, and C-fos proteins may take part in osteosarcoma metastasis. These data will offer useful helpness to determine the prognosis of osteosarcoma. 展开更多

Objective: To investigate the expression of p16, cyclin D1 and Rb protein in gastric carcinoma and premalignant lesions including dysplastic gastric mucosa and intestinal metaplasia gastric mucosa. Methods: Using SP immunohistochemical methods, the expression of pl6, cyclin D1 and Rb proteins was detected in 10 specimens of normal gastric mucosa, 15 specimens of dysplastic gastric mucosa, 15 specimens of intestinal metaplasia gastric mucosa, 30 specimens of gastric carcinoma. The clinical characteristics of the 30 patients with gastric carcinoma were analysed to explore the relationship between the parameter detected and biological action of gastric cancer. Results: Expression of p16 protein was detected in 90% of normal gastric mucosa, 86.67% of dysplastic gastric mucosa, 86.67% of intestinal metaplasia gastric mucosa, 36.67% of gastric carcinoma. The positive rate of p16 protein expression in gastric carcinoma is significantly lower than that in normal gastric mucosa and gastric premalignant lesions mucosa (P<0.01). Expression of cyclin D1 protein was detected in 10% of normal gastric mucosa, 20% of dysplastic gastric mucosa, 20% of intestinal metaplasia gastric mucosa, 53.33% of gastric carcinoma. The positive rate of cyclin D1, protein expression in gastric carcinoma is significantly higher than that in normal gastric mucosa and gastric premalignant lesions mucosa (P<0.05). Expression of Rb protein was detected in 90% of normal gastric mucosa, 80% of dysplastic gastric mucosa, 80% of intestinal metaplasia gastric mucosa, 50% of gastric carcinoma. The positive rate of Rb protein expression in gastric carcinoma is significantly lower than that in normal gastric mucosa (P<0.05). The expression of p16, cyclin D1 gene were associated with the degree of differentiation of gastric carcinoma, lymphnodes metastasis and distant metastasis. Conclusion: p16, Cyclin D1 and Rb gene play important role in gastric carcinoma genesis. The expression of p16, cyclin D1 and Rb gene have some value to the diagnosis at earlier stage of gastric cancer. Detection of expression of p16, cyclin D1 gene would be helpful to judge the prognosis of gastric cancer. 展开更多

Methods:Thirty-seven glioma specimens were classified as astrocytoma(25 cases,including 7 fibrillary cases;6 protoplasmic cases;12 anaplastic cases),and glioblastoma(12 cases,including 4 GBM cases).Ten normal brain tissues were taken as controls.The expression of cyclin D1,p16 and pRB were detected by immunohistochemical method.Cellular proliferation was assessed by Ki-67 label index(Ki-67 LI).Cellular apoptosis was detected by TUNEL and apoptotic indices(AI)was calculated. Results:The alterations of three proteins were cyclin D1 overexpression(28/37,75.7%),p16 and pRB deletion(20/37,54.1 % and 12/37,32.4%),which were closely related to tumor types,particularly in malignant glioma.Ki-67 LI and AI were higher when pRB pathway was abnormal.Apoptosis was minor in astrocytic tumors(astrocytomas,0.010±0.002;glioblastomas,0.057±0.016). Results: There was abnormal expression of p16 protein in CA and cancerized CA, mainly major protein expression. The p16 protein expresseed in different locations in different cases was as follows: In basal layer cells in normal cuits; in spinous layer, granular layer and stratum corneum layer cells in CA;in keratin pearl peripheral and spinous layer cells in cancerized CA. Conclusion.. There was major expression of p16 protein in CA and cancerized CA, and these protein of the two groups might not naturally be the same. 目的探讨人乳头状瘤病毒感染与乳腺癌的关系以及与P16蛋白表达的相关性,为患者治疗提供临床参考。方法选取2011年1月-2013年1月医院收治的150例乳腺肿瘤患者作为本研究对象,分析人乳头状瘤病毒感染与乳腺癌的关系以及与P16蛋白表达的相... 目的探讨人乳头状瘤病毒感染与乳腺癌的关系以及与P16蛋白表达的相关性,为患者治疗提供临床参考。方法选取2011年1月-2013年1月医院收治的150例乳腺肿瘤患者作为本研究对象,分析人乳头状瘤病毒感染与乳腺癌的关系以及与P16蛋白表达的相关性;使用SPSS18.0软件对数据进行分析。结果 150例患者进行HPV16/18-DNA检测显示,癌组织与正常乳腺组织HPV16/18-DNA阳性率比较差异有统计学意义(P<0.05);癌旁组织与正常乳腺组织HPV16/18-DNA阳性率比较差异有统计学意义(P<0.05);肿瘤最大直径以及患者是否绝经与HPV16/18在乳腺癌组织中的表达无相关性,存在腋窝淋巴结转移的患者HPV16/18在乳腺癌组织中的表达与不存在腋窝淋巴结转移的患者比较差异有统计学意义(P<0.05);HPV16/18-DNA与P16表达显示呈负相关性,并且解脲脲支原体、沙眼衣原体、假丝酵母菌属、人支原体、荫道毛滴虫均为患者HPV感染的相关因素(P<0.05)。结论人乳头状瘤病毒在乳腺癌患者组织内得到高度表达,对乳腺癌的诊断治疗具有病因学的意义,并且人乳头状瘤病毒与P16蛋白的表达为负相关,两者对于乳腺癌患者的病情相互影响。 展开更多

目的研究液基薄层细胞学(TCT)、荧光定量聚合酶链反应技术(FQPCR)、P16蛋白联合检测在宫颈癌早期诊断中的价值,为宫颈癌的早期防治提供参考。方法对2012年10月-2013年10月1 786例宫颈异常女性进行TCT、FQPCR检查,监测人乳头状瘤病毒(HPV... 目的研究液基薄层细胞学(TCT)、荧光定量聚合酶链反应技术(FQPCR)、P16蛋白联合检测在宫颈癌早期诊断中的价值,为宫颈癌的早期防治提供参考。方法对2012年10月-2013年10月1 786例宫颈异常女性进行TCT、FQPCR检查,监测人乳头状瘤病毒(HPV)感染情况,检测P16蛋白表达,采用SPSS 17.0进行统计分析。结果 1 786例患者中检测HPV阳性198例,阳性率为11.09%;其中高危型HPV占38.38%、低危型占61.62%;P16蛋白随着CINⅡ级、CINⅢ级、SCC变化表达率逐渐升高,比较差异有统计学意义(P<0.05)。结论宫颈异常患者HPV阳性检出率高,P16蛋白表达率随着宫颈病变的加重而提升,FQPCR、TCT、P16联合检查有助于宫颈癌及其癌前病变的筛查。 展开更多

This study aimed at ass... Background Bmi-1 gene determines the proliferative capacity of normal and leukemia stem cells. Expression of Bmi-1 has been found in all types of myeloid leukemia cells in both humans and mice. This study aimed at assessing the effect of antisense Bmi-1 expression on K562 cells proliferation and p16 protein (p16) expression. Methods A transcriptional repressor, Bmi-1 cDNA was cloned by reverse transcriptase polymerase chain reaction (RT-PCR) of its mRNA from K562 cells. A plasmid expressing antisense Bmi-1 mRNA was then constructed by reverse design of PCR primers and cloned to the plasmid pLNCX2; G418 was added to the medium after the plasmid was successfully introduced in K562 cells by lipofectin-mediated DNA transfection. The effects of the antisense expression on the proliferation of K562 cells were analyzed by using microcuhure tetrazolium and colony forming. Cell cycle was analyzed by using flow cytometry. The p16 expression of K562 cells was observed by immunoflorescence histochemical stain. Results K562 cells transfected with antisense Bmi-1 plasmid grew significantly slower than that of controls (the parental K562 and cells transfected with empty plasmid). The colony forming ability of antisense Bmi-1 plasmid transfected ceils decreased significantly (P 〈0. O1 ) compared with controls. The p16 expression of cells transfected with antisense Bmi-1 was upgraded more apparently than that of controls. Conclusion The antisense Bmi-1 gene can inhibit the growth of K562 cell and upgrade expression of p16 in K562 cells. 展开更多
