短链全氟化合物PFBA和PFBS对多齿新米虾急性毒性与氧化应激的研究

近年来,短链全氟化合物(PFASs)作为长链PFASs替代品,被广泛应用于工业生产和商业产品中。短链PFASs具有持久性、迁移性和潜在毒性对环境造成不可忽视的影响。为探究短链全氟化合物对水生生物的毒性机制,本文研究了全氟丁酸(PFBA)与全氟... 近年来,短链全氟化合物(PFASs)作为长链PFASs替代品,被广泛应用于工业生产和商业产品中。短链PFASs具有持久性、迁移性和潜在毒性对环境造成不可忽视的影响。为探究短链全氟化合物对水生生物的毒性机制,本文研究了全氟丁酸(PFBA)与全氟丁烷磺酸(PFBS)这2种典型替代品对多齿新米虾(Neocaridina denticulata)体内过氧化氢酶(CAT)活性、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、乙酰胆碱酯酶(AChE)活性的影响。结果表明:PFBA和PFBS对多齿新米虾96 h-LC 50分别为7.866 mg·L^(-1)和0.713 g·L^(-1),PFBA比PFBS急性毒性更大,PFBS为低毒物质;CAT、SOD、AChE活性和MDA含量与PFBA和PFBS的暴露浓度均存在剂量-效应关系。MDA含量随PFBS暴露浓度增大持续显著增加,而CAT、SOD和AChE的酶活性随暴露浓度增大均呈现先增加后减小的倒“U”型曲线关系,PFBA也呈现倒“U”型关系。研究表明PFBA与PFBS不同程度地干扰多齿新米虾体内酶活性变化,中低浓度暴露对机体产生一系列抗氧化损伤反应,高浓度暴露会造成机体出现氧化应激反应,最终导致死亡。该研究为短链全氟化合物对水生生物的毒性作用机制研究提供了科学依据。 展开更多

Per-and polyfluorinated alkyl substances(PFASs) are commonly used in industrial processes and daily life products.Because they are persistent, they accumulate in the environment, wildlife and humans.Although many studies have focused on two of the most representative PFASs, PFOS and PFOA, the potential toxicity of short-chain PFASs has not yet been given sufficient attention.We used a battery of assays to evaluate the toxicity of several four-carbon and six-carbon perfluorinated sulfonates and carboxyl acids(PFBS,PFHxS, PFBA and PFHxA), with a human mesenchymal stem cell(hMSC) system.Our results demonstrate significant cyto-and potential developmental toxicity for all the compounds analyzed, with shared but also distinct mechanisms of toxicity.Moreover, the effects of PFBS and PFHxS were stronger than those of PFBA and PFHxA, but occurred at higher doses compared to PFOS or PFOA. 展开更多

Egg hatching of the soil collembolan Folsomia candida and the effects of per-and polyfluoroalkyl substances were investigated.New and effective laboratory methods for egg hatching studies with soil collembolans were established.Per-and polyfluoroalkyl compounds(PFASs)have been used industrially worldwide and are persistent organic pollutants in many soils.Twenty eggs laid by synchronized adults of the collembolan Folsomia candida were added to each Petri dish containing compressed soil substrate mixed with perfluorooctanoic acid(PFOA),heptafluorobutyric acid(PFBA),or 6:2 chlorinated polyfluoroalkyl ether sulfonic acid(F-53B),and after 25 d of exposure the number hatched declined on average by 6.9%−49.7%,10.3%−24.1%,and 3.4%−18.6%,respectively.PFASs delayed the peak of hatching by one day,and at different concentrations reduced the number of eggs hatched during the peak by 16.7%−30%and 23.3%−43.2%in PFOA and PFBA treatments,respectively.In the presence of F-53B the number of eggs hatched declined by 73.3%but the number of individuals increased by 29.3%at higher concentrations.The characteristics of egg hatching were stable and sensitive to PFASs,and may be suitable for use as indicators in the screening of contaminated soils for environmental risk assessment. 展开更多
