This study investigated the protective effects of ampelopsin on H2O2-induced oxidative injury in porcine kidney epithelial cell line, PK-15; cells were pretreated with medium containing 0, 15, 30, and 60 ug mL-1 ampelopsin, respectively, for 1 h prior to the addition of 100 umol L-1 H2O2. After 2 h, the cell viability, intracellular superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, cellular malondialdehyde (MDA) content, and DNA strand breakage were determined. Results showed that ampelopsin pretreatment did not modify the SOD and GSH-Px inactivation but significantly decreased MDA production; except for the medium dose (30 ug mL-1), all the tested concentrations of ampelopsin did not significantly inhibit cell viability reduction and gave no influence on DNA damage. The results suggested that all the tested doses of ampelopsin pretreatment before H2O2 addition can protect PK-15 cell from H2O2-induced lipid peroxidation, which might not be mediated by the SOD and GSH-Px responsees. The appropriate dose of ampelopsin pretreatment prevents PK-15 cell from DNA damage and cell viability reduction. 展开更多

将猪细小病毒南京毒株1(N J-1)、南京毒株2(N J-2)和7909毒株接种于PK-15细胞,用免疫荧光检测方法研究了猪细小病毒增殖的基本特性与规律。在PK-15细胞中,3个毒株的增殖规律基本相似,均在感染后12 h即可检测到荧光,表明其子代病毒粒子产... 将猪细小病毒南京毒株1(N J-1)、南京毒株2(N J-2)和7909毒株接种于PK-15细胞,用免疫荧光检测方法研究了猪细小病毒增殖的基本特性与规律。在PK-15细胞中,3个毒株的增殖规律基本相似,均在感染后12 h即可检测到荧光,表明其子代病毒粒子产生,随后逐渐增多,在感染后48 h荧光几乎遍布所有细胞,随后荧光开始衰减,在84 h时病毒引起细胞大面积崩解,荧光呈现岛屿状分布。通过绘制其一步法生长曲线可知,在病毒感染后12 h,细胞培养液中的病毒TC ID50/mL为2.0左右,随后逐渐增高,感染后60 h 3种毒株的TC ID50/mL均达到最高,子代病毒粒子向细胞外释放也达到高峰,其后TC ID50逐渐下降。培养液中病毒粒子的半寿期为7 h。 展开更多

A novel agent (hence termed as P2) was isolated from pig sera in China, which contained covalently bound circular genomic DNAs of 993 nucleotides. Sequence analyses indicated that the agent was closely related to the porcine circovirus (PCV). The molecular clone of P2 was constructed subsequently and used for the following studies. Intracytoplasmic inclusions and intranuclear inclusions were only found in PK-15 cells transfected with the tandem dimer of P2 molecular DNA clone. Intracytoplasmic inclusions were round or irregular in shape and 0.1-0.4 μm in diameter, and intranuclear inclusions were electronically denser than intracytoplasmic inclusions and had two general shapes: round/small (0.1 μm in diameter) and hexagonal/large (0.5―1.4 μm in diameter). The inclusions were not membranously bound. The cells transfected with the tandem dimer of P2 molecular DNA clone were tested positive for P2 DNA at passages 5. The P2 antigen could be detected in both transfected and passaged PK-15 cells. This is the first report regarding the complete nucleotide sequence of a small DNA genome in a circovirus-like infectious agent in vitro. 展开更多
