大豆拟茎点种腐病菌(Phomopsis/Diaporthe longicolla)可侵染大豆引起种腐病和茎枯病等多种病害。本研究根据该病菌基因组的密码子偏好性,优化了外源的荧光素酶基因序列,通过酵母同源重组构建了荧光素酶表达载体,利用PEG介导的原生质体... 大豆拟茎点种腐病菌(Phomopsis/Diaporthe longicolla)可侵染大豆引起种腐病和茎枯病等多种病害。本研究根据该病菌基因组的密码子偏好性,优化了外源的荧光素酶基因序列,通过酵母同源重组构建了荧光素酶表达载体,利用PEG介导的原生质体转化法获得了稳定表达荧光素酶的大豆拟茎点种腐病菌菌株PlYC2-1-Luc。结果显示:荧光素酶基因的引入不影响大豆拟茎点种腐病菌的生长速率和致病力。以PlYC2-1-Luc菌株进行示踪可直观发现:健康大豆中分离的3株内生镰孢菌能够显著抑制PlYC2-1-Luc生长,具有生防潜力;含苯醚甲环唑、精甲霜灵·咯菌腈和咯菌腈·噻霉酮的种衣剂均可抑制PlYC2-1-Luc向大豆种子扩展,表明这些药剂或其成分可用于病害防控;PlYC2-1-Luc对大豆品种Williams的致病力强于齐黄34,表明齐黄34更加抗病。该荧光素酶标记方法具有较好的有效性和稳定性,该研究为进一步开展大豆拟茎点种腐病菌的致病机理及病害防控技术研究提供了一种可视化、高效的菌株材料。 展开更多

After insufflating extended endophytes phomopsis B3 ferment diluent on seedling bed of rice, the effect of microbial inoculum on rice growth was studied. The investigation result of seedlings before transplanting seedling showed that leaf age,the number of green leaves,the number of tiller of individual plant and the weight of per 100 fresh plants in microbial inoculum treatment group increased 0.11,0.12 ,0. 03 and 2.6 g respectively compared with those of control group. 20 days after transplanting, increased tiller number in microbial inoculum treatment group was 55 500 hm^2 more than that in control group. 30 days after transplanting, tiller number in microbial inoculum treatment group was 42 000 hm^2 more than that in control group. In harvest season, the yield ,weight of I 0130 grain and seed-setting rate increased 300 kg/hm2, 0. 7 g and 0. 23% respectively compared with these in control group. The result of preliminary study demonstrated that microbial inoculum confected by endophytes phomopsis B3 could increase the number of strong seedling and yield. 展开更多

No significant genetic difference was detected among the three strains with 492 bp long ITS1-5.8S-ITS2 sequence. It was then identified through colony growth, conidia morphology, and molecular characterization. The physiological response to oxidation and osmosis stress, and virulence to Asparagus officinalis L. were analyzed. The results showed that the pathogen causing asparagus stem blight for A. officinalis L. in Jiangxi Province is Phomopsis asparagri (Sacc.) Bubák. Under pure culture conditions, the conidia were oval-shaped (α-type), with colorless single spore and single nucleus, containing 0-2 oil balls. Its vegetative growth rate was higher when cultured on 0.2 × potato dextrose agar (0.2 × PDA) medium than that on oatmeal agar (OA) medium. However, the pycnidia appeared earlier on OA medium than on 0.2 earlier PDA medium. The vegetative growth rate was depressed under oxidation (H2O2) or osmosis (NaCl) stress conditions, and totally inhibited under 7 mmol/L H2O2 or 2.4 mol/L NaCl. All the strains caused typical pathogenic symptoms to Asparagus officinalis L. at 7 days-post-inoculation (dpi) with conidia. 展开更多

[Objective]The study was to evaluate the indoor toxicity and field efficacy of five Strobilurins fungicides including pyraclostrobin 250 g/L EC,azoxystrobin 250 g/L SC,kresoxim-methyl 50%WG,picoxystrobin 22.5%SC and trifloxystrobin 50%WG against Phomopsis asparagi(Sacc.)Bubak,and to screen out effective fungicides.[Method]The toxicity was tested by mycelial growth rate method.The field trials were carried out by routine spraying method.[Result]The indoor toxicities successively were pyraclostrobin>kresoxim-methyl>picoxystrobin>trifloxystrobin>azoxystrobin.The EC50 values were 6.1029,52.5591,83.8257,129.8616 and 252.1214 mg/L,respectively,all significantly higher than that of benzimidazole fungicide carbendazim(753.3650 mg/L).The field efficacies successively were pyraclostrobin>kresoxim-methyl≥picoxystrobin≥azoxystrobin≥trifloxystrobin.The control effects were 81.20%,77.95%,74.80%,71.69%and 68.54%,respectively,all significantly higher than that of benzimidazole fungicide carbendazim(48.27%).[Conclusion]The five Strobilurins fungicides all have good toxicity and field efficacy against P.asparagi. 展开更多

The export oriented agricultural and horticultural crops depends on the export of residue free produce and has created a great potential and demand for the incorporation of biopesticides in crop protection. PGPR with wide scope for commercialization includes Pseudomonas putida. Pseudomonas putida was effective in reducing die back caused by a fungus Botryodiplodia theobromae and phomopsis or arm death, caused by a fungus, Phomopsis viticola of grapevine in vitro and in vivo. Pseudomonas putida showed optimum Siderophore pseudobactin production at 72 h, and growth peak at 120 h. glycerol as a source of carbon and yeast as a source of nitrogen Pseudomonas putida was formulated using polysaccharide dextran starch, which led to the elongation of conservation and the effectiveness of the biofungicide. Pseudomonas putida was very effective as biocontrol agents to reduce the die back and phomopsis diseases on grapevine. The obtained results showed that both bacterial treatments significantly increased the growth parameters as well as fruits dry weights and yield of grapevine. 展开更多

【目的】Lithocarols为新型的多异戊烯基二苯甲酮类化合物,具有良好的抗肿瘤活性。对lithocarols生物合成基因litI进行克隆和表达纯化,并对该基因的启动子进行功能鉴定,为lithocarols的生物合成及转录调控奠定分子生物学基础。【方法】... 【目的】Lithocarols为新型的多异戊烯基二苯甲酮类化合物,具有良好的抗肿瘤活性。对lithocarols生物合成基因litI进行克隆和表达纯化,并对该基因的启动子进行功能鉴定,为lithocarols的生物合成及转录调控奠定分子生物学基础。【方法】将litI基因扩增后进行原核表达,采用镍亲和层析初步纯化目的蛋白LitI,利用生物信息学方法分析该蛋白的性质和结构。同时扩增litI基因启动子片段,构建荧光素酶表达系统,分析启动子的转录活性,利用PlantCARE启动子分析网站对litI基因启动子的功能组件进行预测。【结果】LitI蛋白为亲水性蛋白,其相对分子量为51 kD,二级结构包括51.32%的α-螺旋、8.99%的延伸链、3.95%的β-转角以及35.75%无规则卷曲。litI基因启动子具有较强的转录活性且在大肠杆菌中具有启动氨苄青霉素抗性基因表达的功能,其功能组件包含TATA box和CAAT box。【结论】通过异源表达获得了LitI蛋白,分析了其性质和结构,并鉴定了具有较强转录活性的litI基因启动子片段。 展开更多

目的研究1株红树植物木果楝(Xylocarpus granatum Koenig)来源内生真菌Phomopsis sp.MGF222的次级代谢产物及其生物活性。方法利用硅胶柱层析、Sephadex LH-20凝胶柱层析、半制备高效液相色谱等色谱分离方法,对该菌发酵产物的乙酸乙酯... 目的研究1株红树植物木果楝(Xylocarpus granatum Koenig)来源内生真菌Phomopsis sp.MGF222的次级代谢产物及其生物活性。方法利用硅胶柱层析、Sephadex LH-20凝胶柱层析、半制备高效液相色谱等色谱分离方法,对该菌发酵产物的乙酸乙酯浸膏的次级代谢产物进行分离纯化;综合利用核磁共振波谱(NMR)、质谱(MS)等波谱解析方法以及与文献数据对照,鉴定化合物的结构;并对化合物进行抗菌和α-葡萄糖苷酶抑制活性评价。结果从内生真菌Phomopsis sp.MGF222中分离鉴定出9个化合物(1~9),结构分别为12-epicitreoisocoumarinol(1)、5-hydroxy-7methoxy-4methylphthalide(2)、5-hydroxy-7-methoxy-4,6-dimethylphthalide(3)、(2-(2'-hydroxypropyl)-5-methyl-7-hydroxychromone(4)、tyrosol(5)、2-hydroxy-benzaldehyde(6)、5-hydroxymethyl-pyrrole-2-carbaldehyde(7)、(2S,2'R,3R,4E,8E,3'E)-2-(2'-hydroxy-3'-octadecenoylamino)-9-methyl-4,8-octadecadiene-l,3-diol(8)、(22E,24S)-5,8-epidioxy-24-methyl-cholesta-6,9(11),22-trien-3-ol(9);其中化合物9对白色念珠菌Canidia albicans显示抗菌活性;化合物2、5和8具有α-葡萄糖苷酶抑制活性。结论发现1个具有抗菌活性和3个具有α-葡萄糖苷酶抑制活性的化合物,具有重要的研究和应用价值。 展开更多

METHOD: Isolation and purification were performed through silica gel column chromatography, g... AIM: To study the chemical constituents of the solid culture of the endophyte Phomopsis sp. IFB-E060 in Vatica mangachapoi. METHOD: Isolation and purification were performed through silica gel column chromatography, gel filtration over Sephadex LH-20, ODS column chromatography, and HPLC. Structures of the isolated compounds were elucidated by a combination of spectroscopic analyses(UV, CD, IR, MS, 1D, and 2D NMR). The cytotoxicity of the isolates was evaluated in vitro by the MTT method against the human hepatocarcinoma cell line SMMC-7721. RESULTS: Five compounds were isolated from the solid culture of the endophyte Phomopsis sp. IFB-E060 and their structures were identified as 18-methoxy cytochalasin J(1), cytochalasin H(2),(22E, 24S)-cerevisterol(3), ergosterol(4), and nicotinic acid(5). Compound 1 had an inhibition rate of 24.4% at 10 μg·mL-1 and 2 had an IC50 value of 15.0 μg·mL-1, while a positive control 5-fluorouracil had an inhibition rate of 28.7% at 10 μg·mL-1. CONCLUSION: 18-Methoxy cytochalasin J(1), produced by endophytic Phomopsis sp. IFB-E060, is a new cytochalasin with weak cytotoxicity to the human hepatocarcinoma cell line SMMC-7721. 展开更多

Lithocarpinols A(1) and B(2), a pair of tenellone diastereoisomers with novel fused skeleton were isolated from the deep-sea derived fungus Phomopsis lithocarpus FS508. Their structures were elucidated by comprehensive spectroscopic analyses, X-ray diffraction and quantum molecular calculation. Their plausible biogenetic pathway featured an intriguing carbonyl-ene cyclization. Lithocarpinol A exhibited moderate inhibitory effect against HepG-2 and A549 tumor cell lines with IC_(50) values of 9.4 μmol/L and10.9 μmol/L,respectively. 展开更多

Three new chromone derivatives, phomochromenones A-C (1-3), and one known chaetocyclinone B (4) were obtained from the cultures ofPhomopsis sp. HNY29-2B isolated from the mangrove Acanthus ilicifolius Linn., which was collected from the South China Sea. Their structures were determined by the analysis of 1D NMR and 2D NMR as well as mass spectroscopic data. The absolute configurations of I and 2 were assigned by quantum chemi- cal calculations of the electronic circular dichroism (ECD) spectra. Compound 3 is the third example of alkaloids possessing the unique chromeno [3,2-c] pyridine nucleus. In the bioactivity assay, compound 4 showed cytotoxicity against human prostate cancer cell lines (PC-3 and DU145 cells) with the IC50 values of 8.13 and 3.59 μmol/L, respectively. 展开更多
