Phospho-specific antibodies have become powerful tools to study these processes. Cardiac myosin binding protein-C (cMyBP-C) is one of the proteins that make up the contractile apparatus of cardiomyocytes. Phosphorylation of cMyBP-C is essential for normal cardiac function, since dephosphorylation of this protein leads to its degradation and has been associated with cardiomyopathy. One of the upstream kinases, which phosphorylate cMyBP-C, is protein kinase D (PKD). While studying the role of PKD in cMyBP-C phosphorylation, we tried to analyze phosphorylation of PKD with a phospho-specific PKD-Ser744/748 antibody. Contrary to the expected 115 kDa, a signal was found for a 150-kDa protein. By MALDI-TOF mass spectrometry, we identified this protein to be cMyBP-C. These data were confirmed by immunostaining using the p-PKD-Ser744/748 antibody, which displayed a striated pattern similar to the one observed for a regular cMyBP-C antibody. To our knowledge there are no antibodies commercially available for phosphorylated cMyBP-C. Thus, the p-PKD-Ser744/748 antibody can accelerate research into the role of cMyBP-C phosphorylation in cardiomyocytes.展开更多

Our previous studies showed that tuina regulates relevant regions and indices of the spinal dorsal horn using the Dian, Bo, and Rou method in Yinmen(BL37), Yanglingquan(GB34), and Weizhong(BL40). Treatment prevents muscle atrophy, protects spinal cord neurons, and promotes sciatic nerve repair. The mechanisms of action of tuina for treating peripheral nerve injury remain poorly understood. This study established rat models of sciatic nerve injury using the crushing method. Rats received Chinese tuina in accordance with the principle of "Three Methods and Three Points," once daily for 20 days. Tuina intervention reduced paw withdrawal latency and improved wet weight of the gastrocnemius muscle, as well as promoting morphological recovery of sciatic nerve fibers, Schwann cells, and axons. The protein expression levels of phospho-p38 mitogen-activated protein kinase, tumor necrosis factor-α, and interleukin-1β also decreased. These findings indicate that "Three Methods and Three Points" promoted morphological recovery and improved behavior of rats with peripheral nerve injury.展开更多

文摘目的探讨小鼠皮肤损伤愈合过程中损伤区磷脂酰肌醇-3-激酶(phosphatidylinositol-3-kinase,PI3K)、磷酸化磷脂酰肌醇-3-激酶(phospho-PI3K,p-PI3K)、蛋白激酶B(protein kinase B,PKB,又称Akt)和磷酸化蛋白激酶B(phospho-Akt,p-Akt)的表达及其随时间的变化规律。方法应用免疫组织化学染色、Western印迹法和real-time PCR技术检测小鼠皮肤损伤不同时间段PI3K、Akt、p-PI3K及p-Akt的变化情况。结果免疫组织化学染色显示,皮肤损伤后,PI3K、p-Akt阳性染色出现在单核细胞和成纤维细胞,峰值出现在组织修复期。Western印迹法显示,各个时间段均有PI3K、p-PI3K、Akt、p-Akt的阳性条带,其中p-PI3K、p-Akt表达的峰值出现在炎症期和增生期,PI3K、Akt表达的高峰出现在组织重建期。real-time PCR结果显示,PI3K m RNA的峰值出现在炎症期和组织重建期,Akt m RNA的峰值出现在组织重建期。结论在皮肤损伤愈合过程中,PI3K、Akt、p-PI3K及p-Akt的表达变化呈现不同的规律性并有较好的时间依赖性,PI3K/Akt通路的表达规律可以用于法医学损伤时间推断。
