Previous study have demonstrated that a compound composed of water-soluable Ganoderma lucidum polysaccharides(GLP)and Polyporus umbellatus polysaccharides(PUP)in a ratio of 3:1 named GPP enhances innate immune functio...Previous study have demonstrated that a compound composed of water-soluable Ganoderma lucidum polysaccharides(GLP)and Polyporus umbellatus polysaccharides(PUP)in a ratio of 3:1 named GPP enhances innate immune function in mice through enhancing the function of macrophage cells and activity of natural killer(NK)cells.Here in our research,we further investigated the effect of GPP on the diversity and composition of intestinal flora,and explored its effect on colitis model mice.The immunoregulatory verification experiments of GPP were conducted in both normal and DSS-induced mice model.Our research showed that GPP increased the diversity of intestinal microorganisms in mice with the extension of administration time.Daily GPP intake attenuated DSS-induced colon injury,protected the splenic lymphocyte proliferation ability,enhanced the serum hemolysin synthesis,and increased peripheral phagocytes and NK cell activity in model mice.Comparisons of the predominant gene pathways of the bacterial microbiota showed that DNA repair and recombination,base mismatch repair pathways was stronger in GPP-treatment group than in control group,indicating the possible molecular mechanisms of immune function regulation.Our study showed that GPP regulated immune function in both health and colitis model,and had a positive effect on maintaining intestinal flora homeostasis.展开更多

To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was ...To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was applied to analyze samples of medicinal materials and decoction pieces collected from different regions.Finally,ten characteristic peaks were designated in the specific chromatograms and applied to the authenticate identification of P.umbellatus samples.Nine common peaks were designated in the fingerprints,and then the similarities between 32 batches of samples were calculated.Among them,eight compounds were identified by HPLC-APCI-IT-TOF-MS^(n),four of which were identified in specific chromatograms and four in fingerprints.In the present study,we,for the first time,combined HPLC specific chromatograms and fingerprints for the species identification and quality evaluation of P.umbellatus.Collectively,our findings provided a new method for establishing a comprehensive quality standard of P.umbellatus.展开更多

In this study, three TLP genes were cloned from P. umbellatus. The full-length coding sequence of PuTLP1, PuTLP2 and PuTLP3 were 768, 759 and 561 bp long, respectively, encoding for 256, 253 and 187 amino acids. Phylogenetic trees showed that P. umbellatus PuTLP1, PuTLP2 and PuTLP3 were clustered with sequences from Gloeophyllum trabeum, Trametes versicolor and Stereum hirsutum, respectively. The expression patterns of the three TLP genes were higher in P. umbellatus with Armillaria mellea infection than in the sclerotia without A. mellea. Furthermore, over-expression of three PuTLPs were carried out in Escherichia coli BL21(DE3) strain, and high quality proteins were obtained using Ni-NTA resin that can be used for preparation of specific antibodies. These results suggest that PuTLP1, PuTLP2 and PuTLP3 in P. umbellatus may be involved in the defense response to A. mellea infections.展开更多

Methods: PPAOL was administered to treat 30 cases of infantile rotavirus diarrhea, the...Objective: To evaluate the efficacy of Poria-Polyporus anti-diarrhea oral liquor (PPAOL) in treating infantile rotavirus diarrhea. Methods: PPAOL was administered to treat 30 cases of infantile rotavirus diarrhea, the effect was observed and compared with that of Smecta. And human rotavirus RNA of patients was detected. Results: The effective rate of the PPAOL group was 83. 33 % and that of Smecta group was 90. 00 %.There was no significant difference between the effect of the two groups in respect of the duration of fever and diarrhea. Human rotavirus RNA showed positive result in 13 patients of PPAOL group and 19 in Smecta group.The duration of virus excretion in both groups was 2 ~7 days, which was much shorter than 9~ 21 days in the previous report. Conclusion: PPAOL is effective in treating infantile rotavirus diarrhea.展开更多
