Correlation and Expression of COX-2 and P53 Protein in Basal Cell Carcinoma of Eyelid 被引量:1

The correlation between the expression of COX-2 and p53 protein in basal cell carcinoma (BCC) of eyelid and apoptosis was investigated. Specimens of BCC were collected from 40 cases (aged 28-68 y) at the Department of Pathology, Renmin Hospital of Wuhan University, and Department of Pathology, Zhongnan Hospital of Wuhan University during from 1999 to 2006. Five specimens of paracancerous tissues served as control group. Immunohistochemical staining was performed to detect the expression of COX-2 and p53 in the tissues. The average absorbance (A) and the average positive area rate of COX-2 and p53 protein were measured by image analysis. The positive area rate of COX-2 and p53 protein was analyzed by linear correlation analysis. It was found that COX-2 and p53 proteins were highly expressed in BCC of eyelid, and weakly expressed in paracancerous tissues. Image analysis revealed that the expression of COX-2 and p53 proteins in BCC of eyelid was sig- nificantly higher than that in paracancerous tissues (P〈0.01). Spearman rank correlation analysis demonstrated a positive correlation between the expression of COX-2 and p53 (r=0.113, P=0.421). It was concluded that COX-2 can increase the expression of p53 protein, therefore suppressing apoptosis. 展开更多

Objective To investigate the protective effects of hydrogen peroxide preconditioning (HPP) on the pheochromocytoma (PC12) cells treated with 1-methyl-4-phenylpyridinium (MPP^+) and to explore the potential mechanisms. Methods The viability and apoptosis of PC 12 cells were determinded by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and 4′,6′-diamidino-2-phenylindole (DAPI) staining, respectively. The expressions of 14-3-3 protein and phospholylated p38 mitogen-activated protein kinase (MAPK) were determined by Western blot. Enzyme-linked immunosorbent assay (ELISA) was used to measure the activity of extracellular signal-regulated protein kinase 1/2 (ERK1/2). Results The cell viability decreased and the number of apoptotic cells increased dramatically in MPP^+ group compared with that in Control group. HPP induced a significant increase in cell viability and a marked decrease in population of apoptotic cells of the MPP^+- treated PC 12 cells, accompanied with up-regulation of 14-3-3 protein and increase of ERK 1/2 and p38 MAPK activities. The 14-3-3 protein expression was positively correlated with the phosphorylation of ERK1/2. Furthermore, inhibition of the ERK1/2 with PD98059 abolished the 14-3-3 protein up-regulation in PC 12 cells induced by HPP. Conclusion HPP protects PC 12 cells against MPP+ toxicity by up-regulating 14-3-3 protein expression through the ERK1/2 and p38 MAPK signaling pathways. 展开更多

In 3 subtypes of ACC, expressions of 3 proteins were different. As an important agronomic trait, inclination of leaves is crucial Ior crop architecture and grain yields. 10 understand the molecular mechanism controlling rice leaf angles, one rice leaf inclination2 (1c2, three alleles) mutant was identified and functionally characterized. Compared to wild-type plants, lc2 mutants have enlarged leaf angles due to increased cell division in the adaxial epidermis of lamina joint. The LC2 gene was isolated through positional cloning, and encodes a vernalization insensitive 3-like protein. Complementary expression of LC2 reversed the enlarged leaf angles of lc2 plants, confirming its role in controlling leaf inclination. LC2 is mainly expressed in the lamina joint during leaf development, and particularly, is induced by the phytohormones abscisic acid, gibberellic acid, auxin, and brassinosteroids. LC2 is localized in the nucleus and defects of LC2 result in altered expression of cell division and hormone-responsive genes, indicating an important role of LC2 in regulating leaf inclination and mediating hormone effects. 展开更多

Objective To develop a rapid multi-residue assay for detecting 16 demanded by the European Union (EU). Methods A recombinant penicillin-binding protein (PBP) 2x* from Streptococcus pneumoniae R6 was expressed in vitro and six β-1actams were conjugated to HRP by four methods. A rapid multi-residue assay for β-1actams was established with PBP2x* and HRP-conjugate. Results PBP2x* was expressed and purified successfully and the ideal HRP-conjugate was identified. The multi-residue assay was developed. After optimization, penicillin G, ampicillin, amoxicillin, cloxacillin, dicloxacillin, oxacillin, nafcillin, cephalexin, ceftiofur, cefalonium, cefquinome, cefazolin, cefoperazone, cephacetrile, and cephapirin can be detected at levels below MRL in milk with simple pretreatment. Conclusion This assay developed can detect all 16 β-1actams demanded by the European Union (EU). The whole procedure takes only 45 min and can detect 42 samples and the standards with duplicate analysis. 展开更多

Benzene is an established leukotoxin and leukemogen in humans. We have previously re- ported that exposure of workers to benzene and to benzene metabolite hydroquinone in cultured cells induced DNA-dependent protein kinase catalytic subunit (DNA-PKcs) to mediate the cellular response to DNA double strand break (DSB) caused by DNA-damaging metabolites. In this study, we used a new, small molecule, a selective inhibitor of DNA-PKcs, 2-(morpholin-4-yl)-benzo[h]chomen-4-one (NU7026), as a probe to analyze the molecular events and pathways in hydroquinone-induced DNA DSB repair and apoptosis. Inhibition of DNA-PKcs by NU7026 markedly potentiated the apoptotic and growth inhibitory effects of hydroquinone in proerythroid leukemic K562 cells in a dose-dependent manner. Treatment with NU7026 did not alter the production of reactive oxygen species and oxidative stress by hydroquinone but repressed the protein level of DNA-PKcs and blocked the induction of the kinase mRNA and protein expression by hydroquinone. Moreover, hydroquinone increased the phos- phorylation of Akt to activate Akt, whereas co-treatment with NU7026 prevented the activation of Akt by hydroquinone. Lastly, hydroquinone and NU7026 exhibited synergistic effects on promoting apop- tosis by increasing the protein levels of pro-apoptotic proteins Bax and caspase-3 but decreasing the protein expression of anti-apoptotic protein Bcl-2. Taken together, the findings reveal a central role of DNA-PKcs in hydroquinone-induced hematotoxicity in which it coordinates DNA DSB repair, cell cycle progression, and apoptosis to regulate the response to hydroquinone-induced DNA damage. 展开更多

To detect retrospectively the phenotype and stability of the E-protein gene in Japanese Encephalitis (JE) virus strain SA14-14-2 used in the live-attenuated JE vaccine prepears, the viral titer was titrated by plaque formation in BHK-21 cell cultures, and the neuro-virulence of viruses was assayed in mice with body weight of 12-14 g by intracerebral inoculation. Meanwhile, the total RNA of virus gene was extracted and amplified by RT-PCR with the designed primers, and then it was purified and cloned to the expression vector pGEM-T. The recombinant plasmid was purified and sequenced. It was found that the loss of viral titer of vaccines stored in -20℃ for longer than 10 years was less than 0.5 Lg PFU/ml. No mice inoculated intracerebrally showed signs of illness or even death. The size of plagues of the vaccine virus remained to be small, and the E genes of primary virus seed SA14-14-2 and the vaccines prepared at different years (1987-2001) were unchanged, in- cluding the 8 critical amino acid sites which were different from the parent wild virus strain SA14 and the related neuro-virulence. These results indicate that the genotypic and biological characteristics of the attenuated JE virus strain SA14-14-2 and its vaccines sion noted. prepared are quite stable without any reversion noted. 展开更多

AIMS To study bcl-2 and P53 protein expression and inhibition of apoptosis during colorectal tumorigenesis. METHODS Expression of bcl -2 and p53 in 45 colorectal ade- nomas and 61 colorectal carcinomas was detected by immunohis- tochemical staining. RESULTS The bcl-2 and P53 protein expression was uniformly negative in normal mucosa,whereas bcl-2 and p53 positive rates were significantly higher in adenoma and carcinoma than in nor- reals(P<0.01 ).The area with strong bcl-2 expression was of- ten the area with severely dysplasia.In colorectal adenoma,ex- pression of p53 increased with the increasing size and dysplasia, in adenomas≥20 mm being higher than adenomas<10 mm(77, 8% vs 35.0%,P<0.05).p53 was relevant to differentiation and Duke's staging.A significant inverse correlation was found between bcl-2 and p53 in immunostaining in the adenomas,but not in the carcinomas.Furthermore,carcinomas with a high per- centage of bcl-2 positive cells were significantly more likely to have low rates of apoptosis. CONCLUSIONS These results suggest that bcl-2 gene appears to be an early event in colorectal tumorigenesis that can inhibit apoptosis,p53 expression plays an important role in the develop- ment and malignant change of colorectal adenoma,bcl-2 and p53 may be used as a good marker relating to cell apoptosis. 展开更多

To investigate whether the Bcl- 2 gene family is involved in m odulating mechanism of apoptosis and change of cell cycle protein induced by curcumin in acute myeloid leukemia HL - 6 0 cell line and primary acute m yelogenous leukem ic cells,the Bcl- 2 family member Mcl- 1,Bax and Bak and cell cycle proteins including P2 7kipl,P2 1wafl,cyclin D3and p Rbp- were selected and their ex- pression detected by SABC imm uno- histochem ical stain m ethod.The attitude of sub- G1 peak in DNA histogram was determined by FCM.The TU NEL positive cell percentage was identified by term inal deoxynucleotidyl transferase (Td T ) - m ediated Biotin d U NP end labeling technique.It was found that when HL - 6 0 cells were treated with 2 5μm ol/ L curcumin for 2 4 h,the expression level of Mcl- 1was down- regulated,but that of Bax and Bak up- regulated time- dependently.There was significant difference in the expression level of Mcl- 1,Bax and Bak between the curcumin- treated groups and control group(P<0 .0 5 - 0 .0 1) .At the sam e time,curcumin had no effect on progress of cell cycle in prim aty acute m yelogenous leukemia at newly diagnosis,but could in- crease the peak of Sub- G1 (P<0 .0 5 ) ,and down- regulate the expression of Mcl- 1and up- regulate the expression of Bax and Bak with the difference being statistically significant.The expression of P2 7kipl,P2 1wafl and p Rbp- were elevated and thatof cyclin D3decreased in the presence of curcum in. 目的:探究穴位刺激联合COX-2抑制剂对乳腺癌骨转移大鼠镇痛、NK细胞活性及PK2蛋白的作用机制。方法:选取50只SPF级SD雌性大鼠,随机数字表法分为正常(A)组、模型(B)组、穴位刺激(C)组、COX-2抑制剂(D)组与穴位刺激联合COX-2抑制剂(E)组,... 目的:探究穴位刺激联合COX-2抑制剂对乳腺癌骨转移大鼠镇痛、NK细胞活性及PK2蛋白的作用机制。方法:选取50只SPF级SD雌性大鼠,随机数字表法分为正常(A)组、模型(B)组、穴位刺激(C)组、COX-2抑制剂(D)组与穴位刺激联合COX-2抑制剂(E)组,每组10只,对B、C、D与E组采用胫骨内注射乳腺癌细胞建立乳腺癌骨转移模型,A组不建立该模型,建模成功后,对C组给予穴位刺激治疗,对D组给予灌胃25 mg/kg的塞来昔布,对E组给予穴位刺激联合塞来昔布治疗,A组、B组同期给予灌胃同体积生理盐水,观察并记录大鼠疼痛情况,双能线骨密度仪检测大鼠骨密度及骨矿物质含量,HE染色法检测骨组织病理形态,免疫印迹法检测NK细胞活性,免疫组化法检测PK2蛋白表达。结果:与A组比较,B组50%PWT、TWL显著降低,差异具有统计学意义(P<0.05),双侧后肢负重差值显著升高,差异具有统计学意义(P<0.05),与B组比较,C组、D组与E组50%PWT、TWL明显升高,差异具有统计学意义(P<0.05),双侧后肢负重差值显著降低,差异具有统计学意义(P<0.05),且D组与C组比较差异无统计学意义(P>0.05),E组比D组变化明显,差异具有统计学意义(P<0.05);A组大鼠骨组织形态完整,骨小梁排列整齐,骨皮质连续,无坏死区域,骨髓腔内充满深染的血细胞,B组骨组织形态遭到破坏,骨皮质出现连续性中断,并可见大量肿瘤细胞浸润,瘤细胞形态不规则,且排列紊乱,细胞核大,异型性明显,与B组比较,C组、D组与E组病理形态明显改善;与A组比较,B组BMD、BMC显著降低,差异具有统计学意义(P<0.05),与B组比较,C组、D组、E组BMD、BMC显著升高,差异具有统计学意义(P<0.05),且D组与C组比较差异无统计学意义(P>0.05),E组比D组升高显著,差异具有统计学意义(P<0.05);与A组比较,B组大鼠胫骨组织中NKp30、NKp44、NKp46蛋白表达显著降低,差异具有统计学意义(P<0.05),与B组比较,C组、D组与E组大鼠胫骨组织中NKp30、NKp44及NKp46蛋白表达显著升高,差异具有统计学意义(P<0.05),且D组与C组比较差异无统计学意义(P>0.05),E组比D组升高显著,差异具有统计学意义(P<0.05);与A组比较,B组大鼠胫骨组织中PK2蛋白表达显著降低,差异具有统计学意义(P<0.05),与B组比较,C组、D组与E组大鼠胫骨组织中PK2蛋白表达显著升高,差异具有统计学意义(P<0.05),且D组与C组比较差异无统计学意义(P>0.05),E组比D组升高显著,差异具有统计学意义(P<0.05)。结论:穴位刺激联合COX-2抑制剂对乳腺癌骨转移大鼠具有显著的镇痛作用,并可显著改善NK细胞活性,促进PK2蛋白表达。 展开更多

目的:检测COX-2在食管癌中的表达,探讨其与食管癌临床病理特征及术后患者预后的关系。方法:应用SABC免疫组化染色法检测89例手术切除的食管癌组织和20例正常食管粘膜组织COX-2蛋白的表达,分析COX-2在食管癌组织的表达与患者性别、年龄... 目的:检测COX-2在食管癌中的表达,探讨其与食管癌临床病理特征及术后患者预后的关系。方法:应用SABC免疫组化染色法检测89例手术切除的食管癌组织和20例正常食管粘膜组织COX-2蛋白的表达,分析COX-2在食管癌组织的表达与患者性别、年龄、肿瘤部位、病变长度、浸润深度、区域淋巴结转移、远处转移、鳞癌分化程度的关系。并对其中81例有随访资料者,分析COX-2表达与预后的关系。结果:食管癌组COX-2呈阳性表达率为94.38%(84/89),且着色较深,正常食管组COX-2呈阳性表达率为60%(12/20),着色浅。食管癌中,随着肿瘤浸润深度的增加、鳞癌细胞分化程度的减低,COX-2的表达逐步增强(P<0.05)。COX-2的表达与其它临床病理特征,包括性别、年龄、肿瘤生长部位、肿瘤大小、有无区域淋巴结转移、有无远处转移无相关(P>0.05)。COX-2低表达组与高表达组的生存时间有显著差异,前者生存时间较后者为长(P<0.01)。结论:COX-2在食管癌中的表达高于正常食管粘膜。肿瘤浸润越深、鳞癌分化程度越低,COX-2的表达越强;COX-2的表达与食管癌患者的年龄、性别、肿瘤生长部位、肿瘤长度、有无区域淋巴结转移、有无远处转移均无关。COX-2的表达与食管癌术后患者的预后相关,COX-2呈低表达者的生存时间比COX-2高表达者长。 展开更多
