BACKGROUND Previous studies have shown that the Shi-pi-xiao-ji(SPXJ)herbal decoction formula is effective in suppressing hepatocellular carcinoma(HCC),but the underlying mechanisms are not known.Therefore,this study investigated whether the antitumor effects of the SPXJ formula in treating HCC were mediated by acetyl-coA acetyltransferase 1(ACAT1)-regulated cellular stiffness.Through a series of experiments,we concluded that SPXJ inhibits the progression of HCC by upregulating the expression level of ACAT1,lowering the level of cholesterol in the cell membrane,and altering the cellular stiffness,which provides a new idea for the research of traditional Chinese medicine against HCC.AIM To investigate the anti-tumor effects of the SPXJ formula on the malignant progression of HCC.METHODS HCC cells were cultured in vitro with SPXJ-containing serum prepared by injecting SPXJ formula into wild-type mice.The apoptotic rate and proliferative,invasive,and migratory abilities of control and SPXJ-treated HCC cells were compared.Atomic force microscopy was used to determine the cell surface morphology and the Young’s modulus values of the control and SPXJ-treated HCC cells.Plasma membrane cholesterol levels in HCC cells were detected using the Amplex Red cholesterol detection kit.ACAT1 protein levels were estimated using western blotting.RESULTS Compared with the vehicle group,SPXJ serum considerably reduced proliferation of HCC cells,increased stiffness and apoptosis of HCC cells,inhibited migration and invasion of HCC cells,decreased plasma membrane cholesterol levels,and upregulated ACAT1 protein levels.However,treatment of HCC cells with the water-soluble cholesterol promoted proliferation,migration,and invasion of HCC cells as well as decreased cell stiffness and plasma membrane cholesterol levels,but did not alter the apoptotic rate and ACAT1 protein expression levels compared with the vehicle control.CONCLUSION SPXJ formula inhibited proliferation,invasion,and migration of HCC cells by decreasing plasma membrane cholesterol levels and altering cellular stiffness through upregulation of ACAT1 protein expression. 展开更多

BACKGROUND In recent years,immune checkpoint inhibitors(ICIs)have demonstrated remarkable efficacy across diverse malignancies.Notably,in patients with advanced gastric cancer,the use of programmed death 1(PD-1)blockade has significantly prolonged overall survival,marking a pivotal advancement comparable to the impact of Herceptin over the past two decades.While the therapeutic benefits of ICIs are evident,the increasing use of immunotherapy has led to an increase in immune-related adverse events.CASE SUMMARY This article presents the case of a patient with advanced gastric cancer and chronic plaque psoriasis.Following sintilimab therapy,the patient developed severe rashes accompanied by cytokine release syndrome(CRS).Fortunately,effective management was achieved through the administration of glucocorticoid,tocilizumab,and acitretin,which resulted in favorable outcomes.CONCLUSION Glucocorticoid and tocilizumab therapy was effective in managing CRS after PD-1 blockade therapy for gastric cancer in a patient with chronic plaque psoriasis. 展开更多

BACKGROUND Fanlian Huazhuo Formula(FLHZF)has the functions of invigorating spleen and resolving phlegm,clearing heat and purging turbidity.It has been identified to have therapeutic effects on type 2 diabetes mellitus(T2DM)in clinical application.Non-alcoholic fatty liver disease(NAFLD)is frequently diagnosed in patients with T2DM.However,the therapeutic potential of FLHZF on NAFLD and the underlying mechanisms need further investigation.AIM To elucidate the effects of FLHZF on NAFLD and explore the underlying hepatoprotective mechanisms in vivo and in vitro.METHODS HepG2 cells were treated with free fatty acid for 24 hours to induce lipid accumulation cell model.Subsequently,experiments were conducted with the different concentrations of freeze-dried powder of FLHZF for 24 hours.C57BL/6 mice were fed a high-fat diet for 8-week to establish a mouse model of NAFLD,and then treated with the different concentrations of FLHZF for 10 weeks.RESULTS FLHZF had therapeutic potential against lipid accumulation and abnormal changes in biochemical indicators in vivo and in vitro.Further experiments verified that FLHZF alleviated abnormal lipid metabolism might by reducing oxidative stress,regulating the AMPKα/SREBP-1C signaling pathway,activating autophagy,and inhibiting hepatocyte apoptosis.CONCLUSION FLHZF alleviates abnormal lipid metabolism in NAFLD models by regulating reactive oxygen species,autophagy,apoptosis,and lipid synthesis signaling pathways,indicating its potential for clinical application in NAFLD. 展开更多

In recent years,cancer immunotherapy has introduced novel treatments,such as monoclonal antibodies,which have facilitated targeted therapies against tumor cells.Programmed death-1(PD-1)is an immune checkpoint expressed in T cells that regulates the immune system’s activity to prevent over-activation and tissue damage caused by inflammation.However,PD-1 is also expressed in tumor cells and functions as an immune evasion mechanism,making it a therapeutic target to enhance the immune response and eliminate tumor cells.Consequently,immune checkpoint inhibitors(ICIs)have emerged as an option for certain tumor types.Nevertheless,blocking immune checkpoints can lead to immune-related adverse events(irAEs),such as psoriasis and cytokine release syndrome(CRS),as exemp-lified in the clinical case presented by Zhou et al involving a patient with adva-nced gastric cancer who received sintilimab,a monoclonal antibody targeting PD-1.Subsequently,the patient experienced exacerbation of psoriasis and CRS.The objective of this editorial article is to elucidate potential immunologic mechanisms that may contribute to the development of CRS and psoriasis in patients receiving ICIs.It is crucial to acknowledge that while ICIs offer superior safety and efficacy compared to conventional therapies,they can also manifest irAEs affecting the skin,gastrointestinal tract,or respiratory system.In severe cases,these irAEs can lead to life-threatening complications such as circulatory shock or multiorgan failure.Consequently,it is recommended that patients receiving ICIs undergo regular monitoring to identify and manage these adverse events effectively. 展开更多

目的:探索D-松醇对银屑病皮损大鼠Th1/Th2平衡影响的机制研究。方法:构建咪喹莫特(IMQ)大鼠模型,随机分为对照组、IMQ组、D-松醇低剂量组、D-松醇中剂量组、D-松醇高剂量组和阳性对照组,每组10只。造模成功后大鼠给与D-松醇低、中、高... 目的:探索D-松醇对银屑病皮损大鼠Th1/Th2平衡影响的机制研究。方法:构建咪喹莫特(IMQ)大鼠模型,随机分为对照组、IMQ组、D-松醇低剂量组、D-松醇中剂量组、D-松醇高剂量组和阳性对照组,每组10只。造模成功后大鼠给与D-松醇低、中、高剂量组(50mg/kg、100mg/kg、150mg/kg)和阳性对照组(1mg/kg,甲氨蝶呤)灌胃治疗,对照组和IMQ组给予相同剂量的生理盐水,每天一次,连续7d。在给药第8天对各组大鼠银屑病皮损症状和指数(MPASI)评价;HE染色观察银屑病皮损病理变化;ELISA检测血清中干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白介素-4(IL-4)、白介素-10(IL-10)水平;以流式细胞术检测辅助性T细胞1(Th1)/辅助性T细胞2(Th2)比值;Western blot检测T盒子转录因子(T-bet)、GATA结合蛋白3(GATA-3)、缺刻基因1(Notch1)、发状分裂相关增强子1(Hes1)蛋白表达。结果:与对照组比较,IMQ组大鼠皮肤出现银屑病样病变如红斑、过度角质化、炎性细胞浸润等,IFN-γ、TNF-α、Th1细胞、T-bet、Notch1、Hes1水平显著升高(P<0.05),IL-4、IL-10、Th2细胞、GATA-3水平显著降低,Th1/Th2细胞比例失衡(P<0.05);经D-松醇治疗后,大鼠银屑病样病变减轻或消失,皮损指数下降,IFN-γ、TNF-α、Th1细胞、T-bet、Notch1、Hes1水平下降(P<0.05),IL-4、IL-10、Th2细胞、GATA-3水平升高,Th1/Th2比例有所恢复(P<0.05)。结论:D-松醇可能通过抑制Notch1/Hes1信号通路调节Th1/Th2平衡,降低炎性因子产生,减轻IMQ大鼠银屑病皮损。 展开更多

目的探究补肺健脾方(Bufei Jianpi formula,BJF)通过调控IRS-1/PI3K信号轴对COPD大鼠骨骼肌线粒体损伤的影响。方法将60只SPF级SD大鼠随机分为空白(Control)组、COPD稳定期模型(Model)组、氨茶碱(Am)组、补肺健脾方(BJF)组、吡格列酮(P... 目的探究补肺健脾方(Bufei Jianpi formula,BJF)通过调控IRS-1/PI3K信号轴对COPD大鼠骨骼肌线粒体损伤的影响。方法将60只SPF级SD大鼠随机分为空白(Control)组、COPD稳定期模型(Model)组、氨茶碱(Am)组、补肺健脾方(BJF)组、吡格列酮(PIO)组以及补肺健脾方+吡格列酮(BJF+PIO)组,10只/组。采用烟熏加鼻腔滴菌(肺炎克雷伯杆菌)的方法建立COPD稳定期大鼠模型,自第9周开始给药至20周结束后取材,每周给予大鼠体重测量。分别对肺组织和骨骼肌组织进行常规切片与HE染色,并于光镜下观察其相应的病理学改变。分别于第0、8、20周采用非束缚全身体积描记系统观察大鼠肺功能,包括VT、PEF、EF50。采用qPCR技术检测大鼠骨骼肌组织中IRS-1、PI3K、PGC-1α以及Leptin mRNA的表达。采用Western blot技术检测大鼠骨骼肌组织中IRS-1、PI3K、AKT、p-AKT、PGC-1α、TFAM和Leptin蛋白的表达。结果光镜观察显示与Control组比,Model组肺病理可见肺泡间质以及肺支气管存有大量的炎性细胞浸润,部分肺泡壁出现断裂并融合形成气腔、纤维网被破坏等;与Model组比,用药治疗后各组肺泡壁的断裂以及纤维网的破坏均得到改善,支气管中炎性细胞浸润减轻,其中以BJF组与Am组尤为明显。用药治疗后各组骨骼肌病理与Model组比,可不同程度改善肌纤维之间排列间隙、萎缩与断裂,肌细胞胞质染色不均一等,其中以BJF组疗效较为显著。与Control组比,Model组PEF、VT和EF50第8周起显著降低(P<0.01),BJF组、BJF+PIO组和Am组可以显著提高PEF、EF50(P<0.01)。与Control组比,Model组中IRS-1、PGC-1α和PI3K mRNA与蛋白表达水平显著降低(P<0.05,P<0.01),Leptin mRNA与蛋白表达水平显著增高(P<0.01);与Model组比,BJF组IRS-1、PGC-1α、PI3K mRNA与蛋白表达水平显著增高(P<0.05,P<0.01);PIO组IRS-1 mRNA表达水平显著增高(P<0.01);BJF+PIO组PGC-1αmRNA水平显著增高(P<0.01),IRS-1、PI3K mRNA与蛋白水平显著升高(P<0.05,P<0.01);Am组中PI3K mRNA与蛋白表达水平显著增高(P<0.01);4个用药组中Leptin mRNA的表达水平均显著降低(P<0.01),除Am组外,其余3个用药组Leptin蛋白表达显著降低(P<0.01);与Control组比,Model组股四头肌组织中TFAM、p-AKT蛋白表达有明显的下降趋势,各治疗组的TFAM、p-AKT蛋白表达均有升高趋势,但无显著性差异(P>0.05)。结论补肺健脾方可通过调控IRS-1/PI3K信号轴,改善骨骼肌线粒体的损伤,同时提高PGC-1α与线粒体转录因子TFAM的表达,增强线粒体的生物合成,从而减轻肺与骨骼肌组织的病理性损伤。 展开更多

炎调方通过PD-1/PD-L1通路抑制脓毒症大鼠T淋巴细胞衰竭的实验研究

目的研究炎调方对脓毒症大鼠T淋巴细胞及T淋巴细胞表面表面程序性死亡蛋白(PD-1)、程序性死亡蛋白配体(PD-L1)表达的影响。方法SD大鼠(雄性)分为正常组、假手术组、模型组和炎调方组。盲肠结扎穿孔(CLP)制备脓毒症大鼠模型,于造模后12... 目的研究炎调方对脓毒症大鼠T淋巴细胞及T淋巴细胞表面表面程序性死亡蛋白(PD-1)、程序性死亡蛋白配体(PD-L1)表达的影响。方法SD大鼠(雄性)分为正常组、假手术组、模型组和炎调方组。盲肠结扎穿孔(CLP)制备脓毒症大鼠模型,于造模后12、24、48、72 h采集外周血处死大鼠(每个时间点各5只大鼠),采用流式细胞分析法测定CD3^(+)、CD4^(+)、CD8^(+)比例;双抗体夹心酶联免疫吸附法(ELISA)测定CD4^(+)及CD8^(+)表面PD-1、PD-L1表达水平。结果白介素-6(IL-6)在脓毒症模型组12 h分泌达到高峰后逐渐下降,降钙素原(PCT)在模型组24 h达到高峰后逐渐下降;炎调方组在12 h降低IL-6的水平(P<0.05),24 h时明显降低IL-6、PCT的水平(P<0.01)。模型组CD3^(+)、CD4^(+)、CD8^(+)比例早期分泌开始减少,48 h分泌达最低点(P<0.01);炎调方组在48 h时可升高CD3^(+)、CD4^(+)、CD8^(+)比例,提高CD4^(+)/CD8^(+)的比值而具体调节T细胞活化和增殖的功能。模型组CD4^(+)、CD8^(+)表面PD-1、PD-L1表达水平早期24 h即表达增多(P<0.01),72 h表达最多;炎调方组24 h时即可抑制PD-1、PD-L1的表达水平而具体改善T细胞衰竭的作用。CD4^(+)的比例与CD4^(+)表面PD-1、PD-L1的表达呈负相关(P<0.01);CD8^(+)比例与CD8^(+)表面PD-1、PD-L1的表达呈负相关(P<0.01)。结论脓毒症早期48 h内即出现了T细胞活化和增殖的功能衰竭引起的T细胞免疫抑制。炎调方能在脓毒症早期改善T细胞活化和增殖的功能,其机制可能与抑制PD-1/P-L1通路有关。 展开更多

目的观察苓桂气化方对射血分数保留心力衰竭(heart failure with preserved ejection fraction,HFpEF)早期模型大鼠心脏舒张功能的干预效应,基于核苷酸结合寡聚化结构域样受体3(NOD-like receptor thermal protein domain associated pr... 目的观察苓桂气化方对射血分数保留心力衰竭(heart failure with preserved ejection fraction,HFpEF)早期模型大鼠心脏舒张功能的干预效应,基于核苷酸结合寡聚化结构域样受体3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)/胱氨酸天冬氨酸特异性蛋白酶-1(cysteine-containing aspartate-specific proteases-1,Caspase-1)/消皮素D(Gasdermin D,GSDMD)通路探讨其潜在的分子作用机制。方法40只自发性高血压大鼠高盐高脂高糖饮食联合腹腔注射链脲佐菌素溶液建立HFpEF早期大鼠模型,分为HFpEF组、恩格列净组(1.8 mg/kg)、苓桂气化方低剂量组(4.96 g/kg)、苓桂气化方高剂量组(9.92 g/kg),予相应药物灌胃;正常血压组、空白组予等剂量生理盐水灌胃,连续干预9周。采用超声心动图检测大鼠左心房内径(left atrial diameter,LAD)、舒张早期二尖瓣血流速度(left ventricular early diastolic filling peak velocity,E)与舒张晚期二尖瓣血流速度(atrial systolic left ventricular filling peak velocity,A)比值、左室射血分数(left ventricular ejection fraction,LVEF);酶联免疫吸附测定法检测血清心房钠尿肽(atrial natriuretic peptide,ANP)含量;苏木精—伊红染色观察心肌组织病理变化;蛋白质印迹(Western Blot,WB)法检测NLRP3、Caspase-1、GSDMD和白介素1β(interleukin-1β,IL-1β)的表达。结果与正常血压组相比,HFpEF组LAD和E/A增加(P<0.05),LVEF无明显变化(P>0.05);血清ANP含量升高(P<0.05);病理观察显示心肌炎症浸润;WB结果示NLRP3、Caspase-1、GSDMD和IL-1β蛋白含量升高(P<0.05),表明建模成功。与HFpEF组比,恩格列净组和苓桂气化方低、高剂量组的LAD和E/A减小(P<0.05),血清ANP含量降低(P<0.05),病理观察显示心肌炎症浸润减轻,WB结果示恩格列净组NLRP3、Caspase-1、GSDMD、IL-1β蛋白含量降低(P<0.05),苓桂气化方低剂量组NLRP3、IL-1β蛋白含量降低(P<0.05),苓桂气化方高剂量组NLRP3、GSDMD、IL-1β蛋白含量降低(P<0.05),其余蛋白含量有下降趋势。结论苓桂气化方能改善HFpEF早期模型大鼠心脏舒张功能,其机制可能与调控NLRP3/Caspase-1/GSDMD信号通路、减轻心肌炎症浸润、抑制心房重构相关。 展开更多

While the Bushen Yizhi Formula can treat Alzheimer’s disease(AD),the yet to be ascertained specific mechanism of action was explored in this work.Methods:Different concentrations of the Bushen Yizhi Formula and amyloid-beta peptide(Aβ)were used to treat rat pheochromocytoma cells(P12)and human neuroblastoma cells(SH-SY5Y).Cell morphological changes were observed to determine the in vitro cell damage.Cell Counting Kit(CCK)-8 assay and flow cytometry were employed to identify cell viability and apoptosis/cell cycle,respectively.Western blotting and immunohistochemistry were employed to measure the expressions of endoplasmic reticulum stress(ERS)-related proteins(GRP78 and CHOP),p-IRE1α,IRE1α,ASK1,p-JNK,JNK,Bax,Bcl-2,XBP-1,and Bim.Fura 2-acetoxymethyl ester(Fura-2/AM)was used to determine the intracellular calcium(Ca^(2+))concentration.Also,an AD model was constructed by injecting Aβinto the CA1 area of the hippocampus in Sprague Dawley rats.AD model rats were gavaged with different concentrations of Bushen Yizhi Formula for 14 consecutive days.The Morris water maze experiment was conducted to test the learning and memory of rats.Hematoxylin&Eosin(H&E)and Terminal-deoxynucleotidyl Transferase(TdT)-mediated dUTP Nick-End Labeling(TUNEL)staining were done to determine histopathological changes in the brain.Results:Bushen Yizhi Formula relieved the Aβ-induced effects including cell injury,decreased viability,increased apoptosis,G0/G1 phase cell cycle arrest,upregulation of GRP78,CHOP,p-IRE1α,p-JNK,Bax,XBP-1 and Bim,as well as down-regulation of Bcl-2.These results were also seen with IRE1αsilencing.While Aβsuppressed the learning and memory abilities of rats,the Bushen Yizhi Formula alleviated these effects of Aβ.Brain nerve cell injury induced by Aβcould also be treated with Bushen Yizhi Formula.Conclusion:Bushen Yizhi Formula could influence ERS through the IRE1αsignaling pathway to achieve its therapeutic effects on AD. 展开更多

目的探讨温阳扶正方联合PD-1免疫治疗影响阳虚型肝癌微环境的临床疗效。方法选取复旦大学附属中山医院青浦分院2021年10月至2023年7月的阳虚型原发性肝癌行PD-1免疫治疗的住院患者80例为研究对象,按照随机数字表法将其分为对照组(予PD-... 目的探讨温阳扶正方联合PD-1免疫治疗影响阳虚型肝癌微环境的临床疗效。方法选取复旦大学附属中山医院青浦分院2021年10月至2023年7月的阳虚型原发性肝癌行PD-1免疫治疗的住院患者80例为研究对象,按照随机数字表法将其分为对照组(予PD-1免疫治疗)和中药组(予PD-1免疫治疗联合用温阳扶正方治疗),对两组患者的中医疗效、中医证候积分、主症评分、行为能力状况、免疫功能指标及血清中标志物水平进行对比。结果中药组的总有效率为72.50%,明显高于对照组的50.00%(P<0.05);两组患者治疗后中医证候积分明显低于治疗前(P<0.05),且中药组治疗后中医证候积分明显低于对照组(P<0.05);两组患者治疗后主症右胁胀痛、畏寒、乏力、纳差、便溏、舌质淡或淡暗、苔白腻评分明显低于治疗前(P<0.05),且中药组治疗后主症右胁胀痛、畏寒、乏力、纳差、便溏、舌质淡或淡暗、苔白腻评分明显低于对照组(P<0.05);两组患者治疗后Karnofsky得分明显高于治疗前(P<0.05),且中药组治疗后Karnofsky得分明显高于对照组(P<0.05);两组患者治疗后的CD4^(+)T细胞、CD4^(+)/CD8^(+)明显高于治疗前(P<0.05),且中药组治疗后CD4^(+)T细胞、CD4^(+)/CD8^(+)明显高于对照组(P<0.05);两组患者治疗后血清中的bFGF、VEGF、TGF-β1、AFP水平明显低于治疗前(P<0.05),且中药组治疗后bFGF、VEGF、TGF-β1、AFP水平明显低于对照组(P<0.05)。结论温阳扶正方联合PD-1免疫治疗阳虚型肝癌疗效显著,有利于改善免疫功能和抑制肿瘤标志物的释放,具有良好的参考、推广及借鉴价值。 展开更多

目的探究通痹活络方联合丁苯酞治疗对急性缺血性脑梗死(AICI)患者脑血管储备能力及血清单核细胞趋化蛋白-1(MCP-1)、基质金属蛋白酶-9(MMP-9)的影响。方法按照随机数字表法将2021年1月—2023年1月天津中医药研究院附属医院脑病一科收治... 目的探究通痹活络方联合丁苯酞治疗对急性缺血性脑梗死(AICI)患者脑血管储备能力及血清单核细胞趋化蛋白-1(MCP-1)、基质金属蛋白酶-9(MMP-9)的影响。方法按照随机数字表法将2021年1月—2023年1月天津中医药研究院附属医院脑病一科收治的200例AICI患者分为观察组和对照组,每组100例。对照组患者于发病入院的24 h内给予丁苯酞注射治疗,观察组在对照组基础上给予通痹活络方治疗。比较两组患者中医证候积分、脑血管储备功能(CVR)、大脑中动脉(MCV)血流速度、血流灌注指标(PI)、血液流变学指标(血浆黏度、全血高切黏度、红细胞比积水平)、炎症因子指标MCP-1、MMP-9及临床疗效。结果治疗后观察组总有效为95.00%,高于对照组90.00%(χ^(2)=1.802,P=0.179);治疗后口舌歪斜、感官障碍、半身不遂等主症和次症中医证候积分明显降低,且观察组低于对照组(P<0.05);治疗后两组MCV血流速度、CVR均提高,且观察组高于对照组,治疗后PI降低,且观察组低于对照组(P<0.05);治疗后两组患者血浆黏度、全血高切黏度均明显降低,且观察组低于对照组;而治疗后两组患者红细胞比积明显升高,且观察组高于对照组;治疗后两组MCP-1、MMP-9水平均明显降低,且观察组低于对照组(P<0.05);两组患者治疗后观察组总不良反应率为5.00%,低于对照组11.00%(χ^(2)=3.907,P=0.048)。结论通痹活络方联合丁苯酞治疗AICI患者效果确切,能够有效改善患者中医证候积分,提高脑血管储备能力,降低血液黏度和MCP-1、MMP-9水平,值得临床推广应用。 展开更多

寻常型银屑病患者Th1、Th17和miR-150表达与病情进展的关系

目的探讨寻常型银屑病(PV)患者外周血辅助性T细胞(Th)1、Th17和miR-150表达水平与病情进展程度的关系。方法选取2021年2月至2023年4月宝鸡市人民医院收治的112例PV患者为PV组,并选取同期60名体检健康者为正常组。PV组男63例、女49例,年... 目的探讨寻常型银屑病(PV)患者外周血辅助性T细胞(Th)1、Th17和miR-150表达水平与病情进展程度的关系。方法选取2021年2月至2023年4月宝鸡市人民医院收治的112例PV患者为PV组,并选取同期60名体检健康者为正常组。PV组男63例、女49例,年龄(46.89±12.35)岁;正常组男32例、女28例,年龄(44.36±11.22)岁。根据银屑病皮损面积和严重程度指数(PASI)将PV组分为轻度组、中度组和重度组。比较PV组和正常组Th1、Th17及miR-150水平,轻度组、中度组和重度组Th1、Th17、miR-150及相关细胞因子[白细胞介素(IL)-2、IL-17、IL-22]水平,分析Th1、Th17、miR-150表达水平与PASI评分的相关性。统计学方法采用t检验、F检验、χ^(2)检验、Pearson相关性分析。结果PV组Th1和Th17水平均高于正常组[(22.76±3.21)%比(16.83±1.65)%、(3.35±0.68)%比(1.25±0.37)%],miR-150水平低于正常组[(0.80±0.09)比(0.99±0.15)],差异均有统计学意义(t=13.38、22.21、10.38,均P<0.05);轻度组Th1和Th17水平均低于中度组、高度组,miR-150水平高于中度组、高度组,差异均有统计学意义(F=85.29、19.41、92.93,均P<0.05);轻度组IL-2、IL-17和IL-22水平均低于中度组、高度组,差异均有统计学意义(F=66.03、42.79、23.34,均P<0.05);Pearson相关性分析显示,PV患者外周血Th1、Th17水平与PASI评分呈正相关(r=0.771、0.514,均P<0.05),miR-150水平与PASI评分呈负相关(r=-0.703,P<0.05)。结论PV患者外周血Th1、Th17水平升高,miR-150水平下降,三者与PV病情进展严重程度相关。 展开更多

Objective Diabetic cardiomyopathy(DCM)represents a substantial risk factor for heart failure and increased mortality in individuals afflicted with diabetes mellitus(DM).DCM typically manifests as myocardial fibrosis,myocardial hypertrophy,and impaired left ventricular diastolic function.While the clinical utility of the Jianpi Qinghua(JPQH)formula has been established in treating diabetes and insulin resistance,its potential efficacy in alleviating diabetic cardiomyopathy remains uncertain.This study aims to investigate the impact and underlying molecular mechanisms of the JPQH formula(JPQHF)in ameliorating myocardial injury in nonobese diabetic rats,specifically focusing on apoptosis and inflammation.Methods Wistar rats were assigned as the normal control group(CON),while Goto-Kakizaki(GK)rats were randomly divided into three groups:DM,DM treated with the JPQHF,and DM treated with metformin(MET).Following a 4-week treatment regimen,various biochemical markers related to glucose metabolism,cardiac function,cardiac morphology,and myocardial ultrastructure in GK rats were assessed.RNA sequencing was utilized to analyze differential gene expression and identify potential therapeutic targets.In vitro experiments involved high glucose to induce apoptosis and inflammation in H9c2 cells.Cell viability was evaluated using CCK-8 assay,apoptosis was monitored via flow cytometry,and the production of inflammatory cytokines was measured using quantitative real-time PCR(qPCR)and ELISA.Protein expression levels were determined by Western blotting analysis.The investigation also incorporated the use of MAPK inhibitors to further elucidate the mechanism at both the transcriptional and protein levels.Results The JPQHF group exhibited significant reductions in interventricular septal thickness at end-systole(IVSs)and left ventricular internal diameter at end-systole and end-diastole(LVIDs and LVIDd).JPQHF effectively suppressed high glucose-induced activation of IL-1βand caspase 3 in cardiomyocytes.Furthermore,JPQHF downregulated the expression of myocardial JunB/c-Fos,which was upregulated in both diabetic rats and high glucose-treated H9c2 cells.Conclusion The JPQH formula holds promise in mitigating diabetic myocardial apoptosis and inflammation in cardiomyocytes by inhibiting JunB/c-Fos expression through suppressing the MAPK(p38 and ERK1/2)pathway. 展开更多

目的探讨双路通脑方对缺血性脑卒中大鼠神经元铁死亡的作用以及对沉默信息调节因子2同源物1(SIRT1)/核因子E2相关因子2(Nrf2)/谷胱甘肽过氧化物酶4(GPx4)信号通路的调节机制。方法采用随机数字表法选取20只大鼠作为假手术组,剩余70只大... 目的探讨双路通脑方对缺血性脑卒中大鼠神经元铁死亡的作用以及对沉默信息调节因子2同源物1(SIRT1)/核因子E2相关因子2(Nrf2)/谷胱甘肽过氧化物酶4(GPx4)信号通路的调节机制。方法采用随机数字表法选取20只大鼠作为假手术组,剩余70只大鼠均利用大脑中动脉闭塞法制备缺血性脑卒中大鼠模型,将造模成功的大鼠随机分为模型对照组、双路通脑方组、双路通脑方+SIRT1抑制剂组(双路通脑方+EX527组),每组20只。14 d后,对大鼠进行神经功能损伤评分;TTC染色检测大鼠脑梗死面积;HE染色检测大鼠脑组织病理变化;尼氏染色检测大鼠脑组织神经元数量;试剂盒检测大鼠脑组织中铁离子(Fe^(2+))、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、丙二醛(MDA)的水平;免疫组化检测大鼠脑组织中酰基-辅酶a合成酶长链家族成员4(ACSL4)、转铁蛋白受体(TFR)、铁蛋白重链多肽1(FTH1)蛋白的阳性表达;Western blotting法检测大鼠脑组织中SIRT1、Nrf2、GPx4及胱氨酸/谷氨酸转运蛋白(SLC7A11)蛋白的表达。结果与假手术组比较,模型对照组大鼠的神经功能缺损评分、脑梗死面积、Fe^(2+)和MDA含量、ACSL4、TFR蛋白表达升高(P<0.05),神经元数量、SOD和GSH含量、FTH1、SIRT1、Nrf2、GPx4及SLC7A11蛋白表达均降低(P<0.05);与模型对照组比较,双路通脑方组大鼠神经功能缺损评分、脑梗死面积、Fe^(2+)和MDA含量、ACSL4、TFR蛋白表达降低(P<0.05),神经元数量、SOD和GSH含量、FTH1、SIRT1、Nrf2、GPx4及SLC7A11蛋白表达均升高(P<0.05);采用SIRT1抑制剂进行回补实验,结果显示SIRT1抑制剂逆转了双路通脑方对神经元铁死亡的抑制作用,同时也抑制了Nrf2和GPx4的表达(P<0.05)。结论双路通脑方可能通过激活SIRT1/Nrf2/GPx4信号通路来抑制缺血性脑卒中大鼠神经元铁死亡。 展开更多
