目的优化肉豆蔻总木脂素的纯化工艺并对其抗炎活性进行研究。方法采用大孔吸附树脂法富集纯化肉豆蔻总木脂素,以吸附率、解吸率及总木脂素含量为指标,考察树脂型号、洗脱剂浓度、上样量及洗脱剂用量等相关参数;采用二甲苯致小鼠耳肿胀模...目的优化肉豆蔻总木脂素的纯化工艺并对其抗炎活性进行研究。方法采用大孔吸附树脂法富集纯化肉豆蔻总木脂素,以吸附率、解吸率及总木脂素含量为指标,考察树脂型号、洗脱剂浓度、上样量及洗脱剂用量等相关参数;采用二甲苯致小鼠耳肿胀模型,通过测定肿胀度、抑制率考察其抗炎活性。结果总木脂素的纯化采用AB-8型树脂,取粗提物干膏1 g溶解后作为上样液,加入40 m L(含树脂16 g)预处理的树脂,静态吸附12 h,上柱,以2 BV/h流速,先用30%乙醇洗脱至无色,再用70%乙醇300 m L洗脱,收集70%乙醇洗脱液,纯度可达45.8%。肉豆蔻总木脂素高、中、低剂量组的肿胀度与模型组比较,差异均有统计学意义(P<0.05)。结论总木脂素纯化工艺稳定、重复性好,肉豆蔻总木脂素具有明显的抗炎活性。展开更多
