【目的】旨在建立Rutgers番茹品种的遗传转化体系。【方法】以易感线虫番茹品种Rutgers为实验材料,采用农杆菌介导法,对无菌苗的苗龄、筛选培养基中Hyg的浓度大小、不定芽诱导培养基的激素配比和生根培养基4个方面进行优化,从而建立Rut... 【目的】旨在建立Rutgers番茹品种的遗传转化体系。【方法】以易感线虫番茹品种Rutgers为实验材料,采用农杆菌介导法,对无菌苗的苗龄、筛选培养基中Hyg的浓度大小、不定芽诱导培养基的激素配比和生根培养基4个方面进行优化,从而建立Rutgem品种的遗传转化体系。【结果】选择9~11 d的无菌苗的叶片分化愈伤组织的效果最好,筛选培养基中Hyg的浓度为10 mg/L时可以有效的筛选转基因的抗性芽,愈伤组织诱导不定芽培养基中最佳激素配比为6-BA2mg/L+IAA0.1 mg/Lo以1/2MS添加不同浹度IBA(0.05、0.1、0.15、0.20 mg/L)进行生根比较,再生芽在1/2 MS+IBA 0.1 mg/L生根培养基上生根最好。【结论】首次建立了Rutgers番茹品种的遗传转化体系,为番茄的抗根结线虫转基因育种打下基础。 展开更多
