Observation on the Effect of Salvia Miltiorrhiza Injection in Treating Traumatic Hyphema and the Opportune Time for Its Application 被引量:1

Objective: To observe the effect of Salvia miltiorrhiza Injection (SMI) in treating traumatic hyphema (TH) and the opportune time for its application. Methods: A retrospective study was conducted in 174 patients with TH (all with a single eye wounded), of whom 92 patients were treated with dicynone and 82 with SMI, and their status of recurrent or aggravated hemorrhage within 5 days after trauma were analyzed. Further, a prospective study was conducted in 76 TH patients (all with a single eye wounded), who were treated with dicynone though they had hyphema 5 days after treatment. After the dicynone medication was discontinued, 39 of them were treated with Sl, but 37 were not. The status of hyphema absorption was observed. Results: The retrospective study showed that the number of cases with recurrent or aggravated bleeding in the SMI-treatment group was much more than that in the dicynone- treated group (Z=-2.531, P=0.011). On the other hand, the prospective study showed that the status of hyphema absorption among the SMI-treated patients was significantly better than in those untreated with SMI (Z=-2.642, P=0.008). Conclusion: SMI shows an effect of promoting the absorption of TH, and SMI treatment is worthy of being spread in clinical practice. For safety considerations, it is suitable to apply the treatment 5 days after trauma. 展开更多

Objective: To explore the effect of Composite Salviae miltiorrhizae Injection (CSMI) on radiosensitivity of leukemia cells. Methods:Semisolid agar culture and flow cytometry assay were performed for studying the change in radiosensitivity of HL60 cell line and fresh human leukemia cells after exposing to CSMI.Results: The D0 (the inverse of the slope of the survival curve) and SF2 (survival fraction at 2 Gy) of HL60 cell line were decreased from 1. 53, 0. 34 to 0. 93, 0. 12, respectively and apopotosis rates after radiation were raised significantly by CSMI. Furthermore, the concentration of CSMI and the time of mixed culture with CSMI before irradiation had positive relation to the effects mentioned above. Compared with control group, CSMI could increase the radiation-induced apoptosis of fresh leukemia cells (5. 89 ± 2. 91 vs 12. 05 ± 3. 06). BACKGROUND: Studies have shown that both salviae miltiorrhizae and ligustrazine can promote protein expression of nerve growth factor (NGF) and regeneration of peripheral nerve. OBJECTIVE: To verify the effect of salviae miltiorrhizae and ligustrazine hydrochloride injection on axonal regeneration and NGF protein expression in a rat model of sciatic nerve injury. DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at the Laboratory of Traditional Chinese Medicine and the Institute of Bioengineering of Jinan University from July to December 2008. MATERIALS: Salviae miltiorrhizae and ligustrazine hydrochloride injection (containing 20 mg salviae miJtiorrhizae and 100 mg ligustrazine per 100 mL injection) was provided by Guizhou Baite Pharmaceutical, China; salviae miltiorrhizae and ligustrazine decoctions (containing 1 g raw drug per 1 mL decoction) were provided by Guangzhou Baiyunshan Factory for Traditional Chinese Medicine, China; rabbit-anti-rat NGF monoclonal antibody was provided by Beijing Biosynthesis Biotechnology, China. METHODS: A total of 80 healthy, male, Sprague Dawley rats were used to establish a sciatic nerve injury model via neurotomy, and were then randomly assigned to 4 groups: salviae miltiorrhizae and ligustrazine hydrochloride injection group (intraperitoneal injection of 35 mL/kg per day salviae miltiorrhizae and ligustrazine hydrochloride injection), saIviae miltiorrhizae group (intragastric peffusion of 2 mL salviae miltiorrhizae), ligustrazine group (intragastric peffusion of 2 mL ligustrazine), and model group (intraperitoneal injection of 35 mL/kg per day saline), with 20 rats in each group. Thereafter, rats in each group were then divided into 4 subgroups according to varying time points of 1, 2, 4, and 8 weeks post-surgery, with 5 rats in each subgroup. MAIN OUTCOME MEASURES: Axons were quantified using chromotrope 2R-brilliant green and silver staining combined with image analysis to calculate the axonal regeneration rate; NGF expression was detected using immunohistochemistry and Western blot analysis; toe interspace was measured by behavior at 4 and 8 weeks. RESULTS: With increasing time after sciatic nerve expression, and toe interspace gradually increased njury, the axonal regeneration rate, NGF protein At 4 and 8 weeks post-surgery, axonal regeneration rate and NGF protein expression were significantly increased in the injured tissue of the salviae miltiorrhizae and ligustrazine hydrochloride injection, salviae miltiorrhizae, and ligustrazine groups, compared with the model group (P 〈 0.05 or P 〈 0.01), and toe interspace was remarkably enlarged (P 〈 0.05 or P 〈 0.01), especially in the salviae miltiorrhizae and ligustrazine hydrochloride injection group. CONCLUSION: Salviae miltiorrhizae and ligustrazine hydrochloride injection promoted axonal regeneration and NGF protein expression in the injured sciatic nerve, and also enhanced neurofunctional recovery. Objective: To investigate the effects of Salvia miltiorrhiza and Ligustrazine Injection(SML) on proliferation and apoptosis of human hepatic stellate cell LX-2 and the expression of N-myc downstreamregulated gene 2(NDRG2, a tumor suppressor gene). Methods: HSCs from the LX-2 cell line were cultured in vitro. The proliferative state of different initial LX-2 cell numbers was measured using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) colorimetric assay. LX-2 cells were plated in 96-well plates at an approximate density of 2.50×10;cells/mL and cultured for 24 h followed by the application of different concentrations of SML(1, 2, 4 and 8 μL/mL). Cell proliferation was measured using the MTT assay at 24 and 48 h. Apoptosis was detected by flow cytometry at 24 h. LX-2 cells were treated with different concentrations of SML and extracted with protein lysis buffer. The levels of NDRG2 and β-catenin were measured by Western blot. Results: With the exception of the 1 and 2 μL/mL concentrations, 4 and 8 μL/mL SML inhibited cell proliferation in a concentration-dependent manner at 24 and 48 h(P<0.05). With the exception of the 1 and 2 μL/mL concentrations, the NDRG2 expression level was greatly increased in a concentration-dependent manner. However, the level of β-catenin was unaffected. Conclusion: SML inhibit LX-2 cell proliferation in a concentration-dependent manner, and the mechanism may be associated with NDRG2 over-expression. 展开更多

Objective:To investigate the effects of Salvia Miltiorrhiza Liguspyragine Hydrochloride and Glucose Injection(参芎葡萄糖注射液,SLGI) on the expression of platelet membrane receptors proteinase-activated receptor-1(PAR1) and proteinase-activated receptor-4(PAR4) in end-stage renal disease(ESRD) patients on chronic haemodialysis(HD).Methods:Eighty-six ESRD patients on HD(treated group) were treated with SLGI,7 days as one therapeutic course,for two successive courses.The previous therapies were unchanged.Flow cytometry was used to assess the expression of platelet PAR1 and PAR4 in the patients,and turbidity method was used to determine the platelet maximum aggregation rate(MAR).Meanwhile,renal function was measured.The final data were compared with those before treatment and with those in the normal control group(54 healthy subjects).Results:Compared with the normal control group,the expressions of PAR1 and PAR4 and platelet MAR in ESRD patients on HD was significantly higher before treatment(P=0.001,P=0.006, and P=0.008);after treatment with SLGI,the above indices in patients were remarkably decreased(P=0.036 and P=0.046),except PAR4(P=0.067),but still higher than those in the normal control group,however,it was not statistically significant.Conclusions:(1) The overexpression of PAR1 and PAR4 might lead to increased platelet aggregation and this could be one of the reasons for the thrombotic events in ESRD patients on HD.(2) SLGI was able to down-regulate the expression of PAR1 in ESRD patients on HD,improve platelet function,and regulate platelet activation. 展开更多

The results showed that : (1) The differ-ence was not obvious in the morbidity of complications between the two groups, but the mortality (3.6% ) ofthe trial group was significantly lower tlian that ( 30. 8% ) of the control group ( P < 0. 05 ) ; ( 2 ) Hematocritwas clearly decreased from 46. 1 5. 2% to 33. 2 3. 9% in the trial one ( P < 0. 05) , but platelet andhemoglobin showed no statistical signiticance.
