Differential Gene Expression Between Cross-Fertilized and Self-Fertilized Kernels During the Early Stages of Seed Development in Wheat

In order to understand molecular basis of cross-fertilized kernel advantage and heterosis, improved differential display of mRNA was used in this study to analyze alterations in gene expression between cross-fertilized and self-fertilized kernels at 2, 4, 6, 8, 10 and 12 days after pollination (DAP) by using 3 wheat hybrids with different level of heterosis. Four patterns of differential expression were observed: (i) bands observed in cross-fertilized kernels but not in self-fertilized kernels (BCnS); (ii) bands occurring in only self-fertilized kernels but not in cross-fertilized kernels (BSnC); (iii) cDNA over-expressed in cross-fertilized kernels compared to self-fertilized kernels (OEC); (iv) cDNA under-expressed in cross-fertilized kernels compared to self-fertilized kernels (UEC). Further analysis showed that BCnS is positively correlated with heterosis, but BSnC is negatively correlated with heterosis. Four differentially expressed cDNA fragments were verified by reverse-northern blot and sequence homology search in GenBank showed that one of them was new sequences; the other exhibited higher similarity to NBS-LRR type resistance protein, 1,6-bisphosphatase and photosystem Ⅱ chlorophyll a-binding protein psbB, respectively, which indicated diverse pathways may be involved in heterosis formation.

Analysis on Factors Influencing Sustainable Development of Self-study Examination of Agricultural Science Disciplines

Based on existing problems in the course of development of self-study examination of agricultural science disciplines, this paper analyzes logical conditions for its sustainable development. Major logical conditions include requirement for free and comprehensive development of individuals; requirement for social institutional evolution and change; requirement for fairness and justice of education and development; as well as requirement for better-established organization system and higher social reputation. Finally, it presents basic measures for sustainable development of self-study examination of agricultural science disciplines, including carrying on reasonable core elements of self-study examination system, adjusting structure of discipline, improving social assistance system, and expanding service space.

Development of a Dietary Habit Self-Management Skills Scale for Post-Gastrectomy Cancer Patients in Japan

Purpose: The purpose of the present study was to develop a Dietary Habit Self-Management Skill (DHSMS) scale, and to test the reliability and validity of this scale. Methods: A questionnaire survey was then conducted using a 69-item DHSMS scale (second draft) for which content validity had been confirmed. The participants were post-gastrectomy cancer patients aged from 40 to under 80 years who had undergone initial gastrectomy with an interval of from 1 month since discharge to <3 years post-operative. Results: The scale is a second-order factor model with “Dietary habit self-management skills” as a higher-order factor, four factors—”Skill to form partnerships with other important people”, “Skill to prevent or cope with post-gastrectomy disorder”, “Skill to grasp issues accompanying post-gastrectomy disorder”, and “Self-efficacy”—and 27 items. The construct validity of the scale was confirmed. Cronbach’s alpha coefficient of the DHSMS scale was 0.915. The criterion- related validity of the DHSMS scale was confirmed based on the correlations between overall DHSMS scale scores and “Number of post-gastrectomy disorder symptoms experienced”, the score for the subscale “Skill to grasp issues accompanying post-gastrectomy disorder” and “Number of post-gastrectomy disorder symptoms experienced”, and the subscale “Self-efficacy” and the SF-8. Discussion: By using patients’ self-administered DHSMS scale scores, medical professionals are able to assess the level of a patient’s self-management skills and the status of their dietary habits as a result of post-gastrectomy disorder.

Na^+/HCO_3^- cotransporter is expressed on β and α cells during rat pancreatic development

AIM To determine the expression and localization of the electrogenic Na^+/HCO_3^- cotransporter(NBC1) in rat pancreas during development. METHODS The rat pancreas from postnatal and embryos removed from the uterus of pregnant rats that had been sacrificed by CO2 asphyxiation were used. Rat pancreas from embryonic day(E) 15.5 and E18.5 rat embryos was isolated under a stereomicroscope. Rat pancreas from postnatal(P) days 0, 7, 14, 21 and adult was directly isolated by the unaided eye. The RT-PCR analysis of the NBC1 specific region on rat pancreastissues from different developmental stages. The two antibodies which target the NBC1 common COOHterminal region and NH2-terminal region detected a clear band of about 145 k Da in the Western blot analysis. The localization of NBC1 was examined by immuno-fluorescence detection. RESULTS The results revealed the first peak of NBC1 expression at E18.5 and the second peak at P14. Meanwhile, the low NBC1 expression occurred at P7 and adult stages. Our results demonstrated, for the first time, the presence of NBC1 in the plasma membrane of β and α cells, as well as in the basolateral membrane of acinar cells of the rat pancreas at different stages of development. CONCLUSION The data strongly suggests that NBC1 is diversely expressed in the pancreas at different developmental stages, where it may exert its functions in pancreatic development especially islet cell growth through HCO_3^- transport and pH regulation.

Attention-Deficit/Hyperactivity Disorder in Adults with High-Functioning Pervasive Developmental Disorders in Japan

Aims: This study was designed to verify the proportion of Japanese adults with pervasive developmental disorder (PDD) who met the diagnostic criteria (other than E) for attention-deficit/hyperactivity disorder (ADHD) in the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Text Revision (DSM-IV-TR). Furthermore, we examined to what extent adults with PDD think that they exhibit ADHD symptoms. Methods: We developed an original Japanese self-report questionnaire to determine the presence or absence of 18 symptoms from the diagnostic criteria for ADHD in the DSM-IV-TR. We administered the questionnaire to 64 adults with high-functioning PDD (45 men and 19 women) and 21 adults with ADHD (10 men and 11 women), aged 18 to 59 years, with a full-scale intelligence quotient ≥75. Target patients were evaluated for ADHD by their psychiatrists. Results: Twenty-nine (45.3%) adults with PDD also had ADHD. The percentage of these adults who had over six perceived inattention symptoms from the DSM-IV-TR was 96.6%. The percentage of these adults who had over six perceived hyperactivity-impulsivity symptoms was 65.5%. Thirty-five (55.6%) adults with PDD responded that they were aware of having ADHD symptoms at the level of the relevant diagnostic criteria. Conclusions: The present study is the first to examine the frequency of objective and perceived ADHD symptoms in adults with PDD in Japan. Our results show that both objective and perceived ADHD symptoms frequently appear in a large number of adults with PDD. This suggests that it is necessary to attend to concomitant ADHD symptoms in the medical care of adults with PDD.
