Objective:To improve the quality of post-thawing Boer buck semen for artificial insemination by adding green tea extract chitosan nanoparticles to skimmed egg yolk diluent,and the proper thawing temperature.Methods:The ejaculate of Boer buck was added to skimmed egg yolk diluent without(the control group)and with adding 1μg of chitosan nanoparticles of green tea extract per mL of diluent(the treatment group).Then,the diluted semen was filled in French mini straws containing 60×106 live sperm per straw,frozen in a standard protocol,and stored as frozen semen at−196℃for a week.Six replicates from each group were diluted for 30 s at 37℃or 39℃sterile water to evaluate the semen quality.Results:Post-thawing(at 37℃or 39℃)of live sperm,progressive motility,and plasma membrane integrity were lower compared to those of the pre-freezing stage(P<0.05).Thawing at 37℃resulted in no significant difference in live sperm,progressive motility,and plasma membrane between the control group and the treatment group(P>0.05).The live sperm,progressive motility,and plasma membrane of the treatment group in the pre-freezing stage,and post-thawed at 39℃were higher compared to those of the control group(P<0.05).There was no significant difference in malondialdehyde(MDA)concentration,DNA fragmentation,and catalase concentration of thawing at 37℃compared to those of 39℃in the same group.The MDA concentration and DNA fragmentation in thawing at 37℃and 39℃of the treatment group were significantly lower than those of the control group(P<0.05).However,the catalase concentration in thawing at 37℃and 39℃of the treatment group was not significantly different than the control group(P>0.05).Conclusions:Higher quality post-thawing Boer buck semen is achieved by adding 1μg/mL of chitosan nanoparticles of green tea extract to the skimmed egg yolk diluent and thawing at 39℃. 展开更多

Amyloid-beta-induced neuronal cell death contributes to cognitive decline in Alzheimer’s disease.Citri Reticulatae Semen has diverse beneficial effects on neurodegenerative diseases,including Parkinson’s and Huntington’s diseases,however,the effect of Citri Reticulatae Semen on Alzheimer’s disease remains unelucidated.In the current study,the anti-apoptotic and autophagic roles of Citri Reticulatae Semen extract on amyloid-beta-induced apoptosis in PC12 cells were first investigated.Citri Reticulatae Semen extract protected PC12 cells from amyloid-beta-induced apoptosis by attenuating the Bax/Bcl-2 ratio via activation of autophagy.In addition,Citri Reticulatae Semen extract was confirmed to bind amyloid-beta as revealed by biolayer interferometry in vitro,and suppress amyloid-beta-induced pathology such as paralysis,in a transgenic Caenorhabditis elegans in vivo model.Moreover,genetically defective Caenorhabditis elegans further confirmed that the neuroprotective effect of Citri Reticulatae Semen extract was autophagy-dependent.Most importantly,Citri Reticulatae Semen extract was confirmed to improve cognitive impairment,neuronal injury and amyloid-beta burden in 3×Tg Alzheimer’s disease mice.As revealed by both in vitro and in vivo models,these results suggest that Citri Reticulatae Semen extract is a potential natural therapeutic agent for Alzheimer’s disease via its neuroprotective autophagic effects. 展开更多

Exploring the relationship between ambient sulfur dioxide and semen quality parameters:A systematic review and meta-analysis

Objective:To investigate the relationship between ambient sulfur dioxide(SO2)exposure and semen quality parameters.Methods:A systematic literature search was conducted to identify relevant studies investigating the association between SO2 exposure and semen quality parameters.This search encompassed the timeframe from January 2000 to May 2023 and included electronic databases such as Web of Science,Google Scholar,PubMed,Cochrane,and Scopus.Pooled effect estimates with 95%confidence intervals(CI)were calculated using percent changes(PC).The meta-analysis included seven studies with 6711 participants and 15087 semen samples.Results:The results revealed a significant negative association between ambient SO2 exposure and certain semen quality parameters.In particular,SO2 exposure was associated with a significant decrease in progressive motility(PC=0.032;95%CI:-0.063 to-0.001;P=0.044)and sperm concentration(PC=-0.020;95%CI:-0.036 to-0.005;P=0.012).However,no statistically significant associations were observed for total sperm count(PC=-0.038;95%CI:-0.079 to 0.003;P=0.070),seminal fluid volume(PC=-0.009;95%CI:-0.048 to-0.030;P=0.662)and sperm motility(PC=-0.17;95%CI:-0.363 to 0.022;P=0.830).In addition,the results of the subgroup analysis revealed specific variables that were associated with the decrease in relevant sperm parameters.Conclusions:This systematic review and meta-analysis provides compelling evidence supporting a consistent negative association between exposure to ambient SO2 and semen quality parameters. 展开更多

Objective:To investigate the effect of Moringa oleifera leaf extract on semen quality parameters in Teressa goat buck.Methods:A total of 25 semen samples from six bucks were selected for the study.Spermatozoa of 150×10^(6) were incubated in 0,300,500 and 700μg of leaf extract as groupⅠ,Ⅱ,Ⅲ and Ⅳ,respectively.Liquid stored semen samples were analysed for motility,viability,total sperm abnormalities,plasma membrane,acrosomal and nuclear integrities,seminal plasma intracellular enzymes[aspartate aminotransferase(AST),alanine aminotransferase(ALT)and lactate dehydrogenase(LDH)]and total antioxidant capacity(TAC)and spermatozoa malondialdehyde(MDA)and cholesterol efflux in comparison with the control group(groupⅠ)for up to 72 h.Results:Moringa oleifera leaf extract(500μg/150×10^(6) spermatozoa)-treated semen had significantly higher motility,viability,plasma membrane,acrosomal and nuclear integrities,and TAC(P<0.05),and had significantly lower total sperm abnormalities,AST,ALT,LDH,MDA production and cholesterol efflux compared to those in other Moringa oleifera leaf extract treated(300 or 700μg/150×10^(6) spermatozoa)and control groups at different hours of liquid storage(P<0.05).Semen quality parameters and TAC showed an increasing trend and total sperm abnormalities,MDA production,leakage of intra-cellular enzymes and cholesterol efflux showed a decreasing trend from group Ⅰto group Ⅲand then an opposite trend from group Ⅲ to group Ⅳ at different hours of liquid storage.Conclusions:Moringa oleifera leaf extract 500μg/150×10^(6) spermatozoa can be used for liquid semen preservation in Teressa goat at Andaman and Nicobar Islands. 展开更多

The Effects of Age and Ejaculatory Abstinence on Semen Quality and Reproductive Hormones in Africa and the Middle East

The aim of this study was to retrospectively evaluate the effects of male age and ejaculatory abstinence on semen parameters and reproductive hormones among men residing in Africa and the Middle East. A total of 70,142 semen analysis results were analysed and grouped according to the age intervals (16 - 20, 21 - 30, 31 - 40, 41 - 50, 51 - 60, >60) and ejaculatory abstinence (<2 days, 2 - 5 days and >5 days). Semen parameters i.e. volume, concentration, progressive motility, total progressively motile count, morphology, total normal sperm count, DNA fragmentation, viability, sORP, normed sORP were specifically evaluated. Additionally, for each age interval, reproductive hormones i.e. estradiol, luteinizing hormone, follicle stimulating hormone, testosterone and prolactin were evaluated. Semen volume, total progressively motile count, sperm morphology and total normal sperm count constantly decrease significantly after the age of 30 years. Sperm concentration started declining significantly after the age of 50 years. There was a constant age-  related increase in number of spermatozoa with damaged DNA. sORP constantly increased up to 60 years. Furthermore, constantage-related decreases in FSH, serum testosterone and prolact in were observed from patients aged between 16 years and 60 years. Semen volume, sperm concentration, progressive motility and normal morphology were significantly higher in patients having > 5 days of abstinence. Patients having > 5 days of abstinence had the lowest normed sORP. Male age significantly affects sperm parameters and reproductive hormones in fertile and infertile men residing in Africa and the Middle East. Prolonged abstinence days provides better semen quality. 展开更多

Objective:To select the appropriate concentrations of methanolic pomegranate extract supplemented in rams’semen extender for obtaining the best-cryopreserved semen quality.Methods:Tris-based semen extender was supplemented with 0.0,0.40,0.48,and 0.56 mg/mL pomegranate peel methanolic extract to extend semen collected from five native rams twice weekly for two months(n=80).Pooled(n=16)post-thaw semen characteristics were determined.Thawed seminal plasma of all supplemented and control groups were used to measure malondialdehyde(MDA),nitric oxide(NO),glutathione peroxidase(GPx),superoxide dismutase(SOD),ascorbic acid,zinc,copper,total cholesterol,low-density lipoproteins(LDL),lactate dehydrogenase(LDH),and alkaline phosphatase(ALP).Results:The supplementation of Tris-based semen extender with 0.48 mg/mL semen extender resulted in the highest post-thaw sperm total motility(P<0.001),sperm progressive motility(P<0.001),live sperm(P<0.001),sperm plasma membrane integrity(P<0.001),acrosome integrity(P<0.001),SOD(P<0.05),zinc(P<0.001),total cholesterol(P<0.001),and LDL(P<0.001)with the lowest percentage of abnormal sperm morphology(P<0.001),the lowest lipid peroxidation(MDA,P<0.01),ascorbic acid(P>0.05),and LDH(P>0.05).Conclusions:Pomegranate peel methanolic extract 0.48 mg/mL supplemented to Tris-based semen extender of rams is the best enrichment in preserving the sperm post-thaw characteristics via improving biochemical profiles and antioxidant capacity. 展开更多

Objective:To investigate the association between age and semen parameters among male partners of subfertile couples.Methods:This retrospective study analyzed the semen of 1523 infertile men aged 26 to 50 years.Data were extracted from GarbhaGudi IVF Centre database from January 2019 to September 2020.The basic semen parameters were interpreted according to the WHO manual 2021,6th edition.Semen parameters in different age groups were compared.Results:Total and progressive motile sperms were significantly higher in the age group of 26-30 years compared to other age groups(P<0.05).Normal sperm count was significantly higher in the age group of 26-30 years compared to the age groups of 41-45 years and>46 years(P=0.001).However,sperm head defects,neck and midpiece defects,tail defects,and cytoplasmic droplets showed statistically insignificant difference in all the age groups(P>0.05).Semen viscosity showed no statistical difference in all the age groups compared to the reference age group of 26 to 30 years.Conclusions:Higher age can lead to a significant decrease in normal sperms and motility in subfertile men.Hence,male partner age should be considered as one of the major determining factors for reproductive outcomes. 展开更多

Objective:To optimize the concentration of PDC-109 protein in semen for higher cryopreservability and fertility by sequestration or neutralization of PDC-109 by its antibodies(anti-PDC-109 antibodies)in bubaline species.Methods:PDC-109 protein was purified by applying two-step chromatography procedures.Purified protein was injected in rabbits to raise antibodies.These raised anti-PDC-109 antibodies were used in neutralization or sequestration of PDC-109 in in-vitro model.Ejaculates were collected from buffaloes and splited for four groups.Group 1 received egg yolk Tris glycerol extender,without anti-PDC-109 antibodies,while group 2 to 4 received anti-PDC-109 antibodies 266μg/mL,80μg/mL,and 26μg/mL in Tris-fructose-citrate buffer,respectively.Semen quality parameters viz.,forward progressive motility,viability,total morphological abnormality,acrosomal integrity,plasma membrane integrity,cryoinjury and in-vitro zona binding index were evaluated.Results:Semen quality parameters of neat semen were within the normal range of bubaline species.Sperm motility,livability,acrosomal integrity,plasma membrane integrity,and cholesterol content of sperm were decreased and total sperm abnormality was increased significantly in post-thaw semen compared to those in pre-freeze and fresh semen(P<0.05).Semen in group 2 had higher sperm motility,livability,acrosomal integrity,plasma membrane integrity,and cholesterol content of sperm and lower total sperm abnormality significantly compared to those in group 1,3 and 4 at pre-freeze and post-thaw stages(P<0.05).Conclusions:Sequestration or neutralization of PDC-109 by its antibodies significantly improves pre-freeze,and post-thaw semen quality parameters and in-vitro zona binding index with simultaneously reducing cryoinjury or cryodamage in the sperm of bubaline species. 展开更多

Objective:To measure seasonal effects on semen quality profiles,seminal biochemical and oxidative stress attributes in fresh and liquid stored semen in monsoon and dry seasons.Methods:A total of 10 Teressa bucks(3-4 years)were selected from breeding farm,ICAR-Central Island Agricultural Research Institute,Port Blair,Andaman and Nicobar Islands,India.Semen samples(n=25 per season)were collected through artificial vagina method and preserved at refrigerated temperature(5℃)for 48 h using Tris citrate glucose based extender.We detected semen quality parameters[volume,mass activity,pH,sperm concentration,total motility,viability,total sperm abnormality,and plasma membrane,acrosomal and nuclear integrities],biochemical profiles[aspartate amino transferase(AST),alanine amino transferase(ALT)and total cholesterol],and oxidative stress markers[total antioxidant capacity(TAC)and malondialdehyde(MDA)]during monsoon and dry seasons.Results:Semen quality parameters significantly differed between seasons(P<0.05)and among storage periods(P<0.05).Volume,pH,mass activity,motility,viability,acrosomal,plasma membrane and nuclear integrities,and TAC were significantly higher(P<0.05).Sperm concentration,sperm abnormalities,MDA,AST,ALT and total cholesterol were significantly lower in fresh semen of monsoon than dry season(P<0.05).Motility,viability,acrosomal,plasma membrane and nuclear integrities,and TAC were significantly decreased(P<0.05)while sperm abnormality,AST,ALT,total cholesterol and MDA were significantly increased as liquid semen storage period advanced(P<0.05).Conclusions:Monsoon season has higher beneficial effects on semen quality profiles and liquid stored semen remained usable for upto 48 h.Good quality ejaculates with higher TAC and lower MDA can be cryopreserved and will be used for artificial insemination. 展开更多

Aim: To assess whether exposure to computers harms the semen quality of healthy young men. Methods: A total of 178 subjects were recruited from two maternity and children healthcare centers in Shanghai, 91 with a history of exposure to computers (i.e., exposure for 20 h or more per week in the last 2 years) and 87 persons to act as control (no or little exposure to computers). Data on the history of exposure to computers and other characteristics were obtained by means of a structured questionnaire interview. Semen samples were collected by masturbation in the place where the semen samples were analyzed. Results: No differences in the distribution of the semen parameters (semen volume, sperm density, percentage of progressive sperm, sperm viability and percentage of normal form sperm) were found between the exposed group and the control group. Exposure to computers was not found to be a risk factor for inferior semen quality after adjusting for potential confounders, including abstinence days, testicle size, occupation, history of exposure to toxic substances. Vitamin D levels have been linked to various health outcomes including reproductive disorders. The purpose of this study was to explore the association between serum vitamin D level (25-hydroxy-vitamin D, or 250HD) and semen and hormonal parameters. This is a cross-sectional study that included 170 healthy men recruited for the study of spermatogenesis from the general population. Men completed general and reproductive health questionnaires, and donated blood and semen samples. The main measures were hormonal (total and free testosterone, sex hormone-binding globulin, estradiol, follicle-stimulating hormone and luteinizing hormone) and semen parameters, adjusted (n= 147) for age, body mass index (BMI), season, alcohol intake and smoking, in relation to categories of vitamin D levels, determined apriori. The mean age of the study population was 29.0±8.5 years and mean BMI was 24.3±3.2 kg m-2. The mean 250HD was 34.1± 15.06 ng m1-1. BMI showed a negative association with 250HD. Sperm concentration, sperm progressive motility, sperm morphology, and total progressively motile sperm count were lower in men with ‘250HD ≥ 50 ng ml-1' when compared to men with‘20 ng ml-1 ≤ 250H D〈 50 ng ml-1,. Total sperm count and total progressive motile sperm count were lower in men with ‘250HD〈20 ng ml-1' when compared to men with‘20 ng ml-1≤250HD〈50 ng ml-1'. The adjusted means of various hormonal parameters did not show statistical difference in the different categories of 250HD. In conclusion, serum vitamin D levels at high and low levels can be negatively associated with semen parameters. 展开更多

The World Health Organization (WHO) has extensively revised its manual for semen analysis, and seeks to provide reference limits for semen quality parameters. This raises the question of what is meant by 'normal' (excluding the use of this term to denote a Gaussian distribution). It could be taken in a purely statistical sense, using a biologically arbitrary cut-off point to denote an abnormal level, typically the extreme 5 percent of the population. Alternatively, 'normal' could be defined according to the biological concept of normality and abnormality, in terms of the point at which biological function becomes impaired. Either of these can be used in descriptive epidemiology, for example, to study trends, but in the case of fertility, both semen quality and functional fertility (time to pregnancy) are continuous variables with no clear threshold. The WHO manual uses the biological meaning of normal, in that it provides the semen parameter distributions for men who have recently fathered pregnancies that took 12 months or less to conceive. However, what is really needed is the same information the other way around: given a particular semen test result, what should be expected in terms of ability to conceive, and how long it is likely to take. In considering epidemiological research, the focus has been mainly on internal comparisons, rather than reference limits, but it would be beneficial if more attention were paid to the absolute levels and to what these mean in terms of function--in other words, if the data were better calibrated biologically. 展开更多

It is generally thought that a single ejaculate is a bad predictor of semen quality of a subject, because of significant intra-individual variation. Therefore, we investigated the degree to which the results of a first semen analysis differ from that of a second analysis among men from a how the two different semen results mirrored the overall general population in Norway. In addition, we analysed semen quality assessment. A total of 199 volunteers participated in the study and delivered two semen samples with an interval of 6 months. The semen parameters were determined according to the World Health Organization (WHO) 1999 guidelines, which were also used to determine whether semen quality was normal or abnormal. In addition, the DNA fragmentation index (DFI) was determined using the Sperm Chromatin Structure Assay. The two samples from each individual were very similar with regard to standard semen parameters and DFI (rs: 0.67-0.72), and there were no significant systematic differences between the two samples. The result of the first sample (normal/abnormal) was highly predictive of the overall conclusion based on the two samples (sperm concentration: in 93% of the cases (95% confidence interval [CI]: 89%-96%); sperm motility: in 85% of the cases (95% CI: 79%-89%); overall semen quality: in 85% of the cases (95% CI: 80%-90%). In epidemiological studies, one ejaculate is a sufficient indicator of semen quality in a group of subjects. In a clinical situation, when the question is whether the semen quality is normal or not, the first ejaculate will, in at least 85% of cases, give a correct overall conclusion. 展开更多

Several animal studies indicate that mercury is a male reproductive toxicant, but human studies are few and contradictory. We examined semen characteristics and serum levels of reproductive hormones in relation to environmental exposure to mercury. Blood and semen samples were collected from 529 male partners of pregnant women living in Greenland, Poland and Ukraine between May 2002 and February 2004. The median concentration of the total content of mercury in whole blood was 9.2 ng ml-1 in Greenland (0.2- 385.8 ng ml-1), 1.0 ng m1-1 in Poland (0.2-6.4 ng ml-z) and 1.0 ng ml-I in Ukraine (0.2-4.9 ng ml-1). We found a significantly positive association between the blood levels of mercury and serum concentration of inhibin B in men from Greenland (p=0.074, 95% confidence interval (CI)=0.021 to 0.126) and in an analysis including men from all three regions (p=0.067, 95% C1=0.024 to O. 110). The association may be due to beneficial effects of polyunsaturated fatty acids (PUFAs), which are contained in seafood and fish. No significant association (P〉O.05) was found between blood concentrations of mercury and any of the other measured semen characteristics (semen volume, total sperm count, sperm concentration, morphology and motility) and reproductive hormones (free androgen index {FAI), follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone and LH xtestosterone) in any region. In conclusion, the findings do not provide evidence that environmental mercury exposure in Greenlandic and European men with median whole blood concentration up to 10 ng m1^-1 has adverse effects on biomarkers of male reproductive health. 展开更多

Pretherapy sperm cryopreservation in young men is currently included in good clinical practice guidelines for cancer patients. The aim of this paper is to outline the effects of different oncological treatments on semen quality in patients with testicular neoplasia or lymphoprol iferative disorders, based on an 8-year experience of the Cryopreservation Centre of a large public hospital. Two hundred and sixty-one patients with testicular neoplasia and 219 patients with lymphoproliferative disorders who underwent chemotherapy and/or radiotherapy and pretherapy semen cryopreservation were evaluated. Sperm and hormonal parameters (follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone, inhibin B levels) were assessed prior to and 6, 12, 18, 24 and 36 months after the end of cancer treatment. At the time of sperm collection, baseline FSH level and sperm concentration were impaired to a greater extent in patients with malignant testicular neoplasias than in patients with lymphoproliferative disorders. Toxic effects on spermatogenesis were still evident at 6 and 12 months after the end of cancer therapies, while an improvement of seminal parameters was observed after 18 months. In conclusion, an overall increase in sperm concentration was recorded about 18 months after the end of cancer treatments in the majority of patients, even if it was not possible to predict the evolution of each single case 'a priori'. For this reason, pretherapy semen cryopreservation should be considered in all young cancer patients. 展开更多

Background: Hypothermic preservation of boar semen is considered a potential method for omitting antibiotics from insemination doses, thereby contributing to the global antibiotic resistance defence strategy. The main challenges are chilling injury to spermatozoa and bacterial growth during semen storage leading to reduced fertility.Objectives: To examine chilling injury and the number and type of bacteria in boar semen stored at 5 ℃ in the absence of antibiotics, and to assess the applicability of hypothermic semen storage under field conditions.Material and methods: Boar ejaculates were extended with AndroStar~? Premium, stored at 17 ℃ with and at 5 ℃ without antibiotics and tested for functional sperm parameters by flow cytometry. Raw semen and extended samples were investigated bacteriologically. Fertility was evaluated after once-daily inseminations of 194 sows in a field study.Results: Lethal sperm damage assessed by motility and membrane integrity was low throughout storage in both experimental groups. Sublethal chilling effects based on the decrease of viable spermatozoa with low membrane fluidity were higher(P < 0.05) up until 72 h in sperm stored at 5 ℃ compared to 17 ℃ but did not differ after 144 h.After 72 h, incubation in capacitating medium for 60 min induced a similar decrease in viable sperm with high mitochondria membrane potential and low cytosolic calcium in both groups. In semen stored at 5 ℃, bacteria counts were below 103 CFU/mL and the bacteria spectrum was similar to that of raw semen. In 88% of 34 boars,cooled semen fulfilled the requirements for insemination. Fertility was high and did not differ(P > 0.05) between sow groups inseminated with semen stored antibiotic-free at 5 ℃ and semen stored at 17 ℃ with antibiotics.Conclusion: Despite subtle chilling effects and low bacterial numbers, antibiotic-free hypothermic storage of boar semen offers the possibility to reduce the use of antibiotics in pig insemination. However, strict sanitary guidelines must be maintained and further evidence of efficiency under field conditions is considered desirable. 展开更多
