Zhenhui 249) with low chlorophyll b was recently discovered in the field. The mutant was mainly characterized by the decrease of the content of extrinsic antennae complex. This variation was shown in the stage when the leaves were expanding. When the leaves are at the final developmental stage, the content would approach to that of the wild type. It was discovered that only moderate amount of chlorophyll b decreased in this mutant. The photosynthetic apparatus of the mutant was rather stable in the whole life span of the leaf. The extrinsic antennae complex of the mutant might make efficient use of light and meanwhile reduce the production of O -· 2. 展开更多

目的探讨丹参酮nA(TSA)对过氧化氢(H2O2)诱导的人脐静脉内皮细胞衰老的作用及其可能机制。方法体外培养人脐静脉内皮细胞(HUVEC),H2O2处理诱导细胞衰老,TSA预处理24 h,分为对照组、模型组和TSA组。CCK-8法检测细胞活力,Western blot法... 目的探讨丹参酮nA(TSA)对过氧化氢(H2O2)诱导的人脐静脉内皮细胞衰老的作用及其可能机制。方法体外培养人脐静脉内皮细胞(HUVEC),H2O2处理诱导细胞衰老,TSA预处理24 h,分为对照组、模型组和TSA组。CCK-8法检测细胞活力,Western blot法检测细胞内细胞衰老相关蛋白(P21和P26)、沉默信息调节因子1(SIRT1)、内皮型一氧化氮合酶(eNOS)、磷酸化eNOS(p-eNOS)蛋白水平,B-半乳糖昔酶染色来评估细胞衰老;转染SIRT1 siRNA敲低SIRT1的表达,检测TSA处理后细胞衰老水平的变化。结果(10、20、40)μg/mL的TSA预处理24 h不影响细胞活力,而(80、160、320)μg/mL TSA预处理的细胞活力下降。(10、20、40)μg/mL的TSA可成剂量依赖性促进比。H2O2介导的HUVEC活力。与对照组比较,模型组0-半乳糖昔酶染色阳性率增加,而TSA组阳性率较模型组明显降低。模型组P21和P16蛋白表达量较对照组增加,而SIRT1和p-eNOS/eNOS较对照组降低;TSA组P21和P16蛋白表达量较模型组降低,SIRT1和p-eNOS/eNOS较模型组增加。转染SIRT1 siRNA的细胞内SIRT1水平明显降低,敲低SIRT1的TSA处理组0-半乳糖昔酶染色阳性率增加。结论TSA通过激活SIRTl/eNOS通路减轻H2O2诱导的血管内皮细胞衰老。 展开更多

Allium senescens,is an important economic and ecological grassland plant with drought-resistant characteristics.A TCP protein transcription factor is important in the regulation of plant development and adverse responses.However,the mechanism by which TCP transcription functions in drought resistance in Allium senescens is still not clear.Here,we obtained a total of 190,305 transcripts with 115,562 single gene clusters based on RNA-Seq sequencing of Allium senescens under drought stress.The total number of bases was 97,195,096 bp,and the average length was 841.06 bp.Furthermore,we found that there were eight genes of the TCP family that showed an upregulated expression trend under drought stress in Allium senescens.We carried out an investigation to determine the evolution and function of the AsTCP family and how they produce an effect in drought resistance.The 14 AsTCP genes were confirmed and divided into class I and class II containing CIN and CYC/TBI subfamilies,respectively.We also found that the expression of AsTCP17 was remarkably upregulated with drought treatment.Besides,the transformation of AsTCP17 in Arabidopsis revealed that the protective enzymes,namely polyphenol oxidase(POD)and superoxide dismutase(SOD),were increased by 0.4 and 0.8 times,respectively.Chlorophyll content was also increased,while the H2O2 and malondialdehyde(MDA)contents were decreased.Staining assays with 3,3′-diaminobenzidine(DAB)also suggested that the AsTCP17 downregulates reactive oxygen species(ROS)accumulation.In addition,overexpression of the AsTCP17 affected the accumulation of drought-related hormones in plants,and the synthesis of ABA.The expression of AtSVP and AtNCED3,related ABA synthesis pathway genes,indicated that the level of expression of AtSVP and AtNCED3 was obviously enhanced,with the overexpression of line 6 showing a 20.6-fold and 7.0-fold increase,respectively.Taken together,our findings systematically analyze the AsTCPs family at the transcriptome expression level in Allium senescens,and we also demonstrated that AsTCP17 protein,as a positive regulator,was involved in drought resistance of Allium senescens.In addition,our research contributes to the comprehensive understanding of the drought stress defense mechanism in herbaceous plants. 展开更多

Activation of the tumour suppressor p53 upon cellular stress can induce a number of different cellular processes?The diverse actions of these processes are critical for the protective function of p53 in preventing the development of cancer.However,it is still not fully understood which process(es)activated by p53 is/are critical for tumour suppression and how this might differ depending on the type of cells undergoing neoplastic transformstion and the nature of the drivers of oncogenesis.Moreover,it is not clear why upon activation of p53 some cells undergo cell cycle arrest and senescence whereas others die by apoptosis.Here we discuss some of the cellular processes that are crucial for p53-mediated tumour suppression and the factors that could impact cell fate upon p53 activation.Finally,we describe therapies aimed either at activating wild-type p53 or at changing the behaviour of mutant p53 to unleash tumour growth suppressive processes for therapeutic benefit in malignant disease. 展开更多
