[目的]研究分析昆明市经性途径感染HIV-1的人群中HIV gag基因亚型流行情况,为科学客观地分析昆明市HIV流行传播模式的变化提供参考。[方法]以2005-2007年间所发现的经性途径感染HIV-1的昆明市本地感染者为研究对象。取得他们口头知情同意后,利用问卷调查表进行“一对一”的当面调查,并采集血样,提取血液细胞DNA,对HIV前病毒的gag基因进行套式PCR扩增、纯化,测序,所得序列经过质量评价、校对、整理后,利用MEGA3.1软件构建系统进化树。同时通过使用洛斯阿拉莫斯HIV核酸序列库和美国NCBI提供的在线分析工具,对所获得序列进行在线比对,根据在线比对结果与所构建的系统进化树来判定序列所属HIV基因亚型。[结果]40名研究对象所感染HIV-1gag基因被成功扩增、测序、判定基因亚型。其中2人所感染HIV为CRF01-AE重组型,7人为CRF07-BC重组型.21人为CRF08-BC重组型。研究对象年龄在18~64岁之间;女性24人,男性16人;已婚者占80.0%;文化程度以初中为主,占60%;农民所占比例达57.5%。[结论]2005-2007年间所发现的经性途径感染HIV-1的昆明市本地感染者中,重组型毒株占据了绝对优势,流行情况更加复杂。 展开更多

The male and female reproductive tracts are complex microenvironments that have diverse functional demands. The immune system in the reproductive tract has the demanding task of providing a protective environment for a fetal allograft while simultaneously conferring protection against potential pathogens. As such, it has evolved a unique set of adaptations, primarily under the influence of sex hormones, which make it distinct from other mucosal sites. Here, we discuss the various components of the immune system that are present in both the male and female reproductive tracts, including innate soluble factors and cells and humoral and cell-mediated adaptive immunity under homeostatic conditions. We review the evidence showing unique phenotypic and functional characteristics of immune cells and responses in the male and female reproductive tracts that exhibit compartmentalization from systemic immunity and discuss how these features are influenced by sex hormones. We also examine the interactions among the reproductive tract, sex hormones and immune responses following HIV-1 infection. An improved understanding of the unique characteristics of the male and female reproductive tracts will provide insights into improving clinical treatments of the immunological causes of infertility and the design of prophylactic interventions for the prevention of sexually transmitted infections. 展开更多
