目的探讨山楂制剂调脂与肠道菌群的相关性。方法将100只SPF级载脂蛋白E基因敲除小鼠(C57BL/6J Apoe^(-/-)小鼠)随机分为5组,每组20只。分为空白对照组、动脉粥样硬化(atherosclerosis,AS)模型组、AS加抗生素对照组及AS加山楂加抗生素组...目的探讨山楂制剂调脂与肠道菌群的相关性。方法将100只SPF级载脂蛋白E基因敲除小鼠(C57BL/6J Apoe^(-/-)小鼠)随机分为5组,每组20只。分为空白对照组、动脉粥样硬化(atherosclerosis,AS)模型组、AS加抗生素对照组及AS加山楂加抗生素组、AS加山楂组。检测指标:检测小鼠血脂水平,包括低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C);运用RT-PCR技术定量检测腹腔巨噬细胞清道夫受体A1(scavenger receptor class A type 1,SR-A1)、CD36mRNA的表达水平;运用Western-blot方法检测CD36,SR-A1的蛋白表达。结果比较空白对照组、AS模型组、AS加抗生素对照组及AS加山楂加抗生素组、AS加山楂组的血脂水平、CD36和SR-A1的mRNA水平和蛋白水平变化,结果表明:(1)通过与空白对照组比较,AS加山楂组有显著降脂效果;(2)通过与AS模型组比较,AS加抗生素对照组和AS加山楂加抗生素组及AS加山楂组均有降脂效果;(3)AS加山楂加抗生素组降脂水平高于AS加抗生素对照组;(4)AS加山楂降脂水平高于AS加山楂加抗生素组。结论山楂具有改善高胆碱饮食喂养造成动脉粥样硬化模型小鼠血脂的作用,山楂制剂调脂与肠道菌群相关。展开更多
