The effects of 5 lactic acid bacteria(LAB)fermentation on the pasting properties of glutinous rice flour were compared,and suitable fermentation strains were selected based on the changes of viscosity,setback value,an...The effects of 5 lactic acid bacteria(LAB)fermentation on the pasting properties of glutinous rice flour were compared,and suitable fermentation strains were selected based on the changes of viscosity,setback value,and breakdown value to prepare LAB compound starter cultures.The results revealed that Latilactobacillus sakei HSD004 and Lacticaseibacillus rhamnosus HSD005 had apparent advantages in increasing the viscosity and reducing the setback and breakdown values of glutinous rice flour.In particular,the compound starter created using the two abovementioned LAB in the ratio of 3:1 had better performance than that using a single LAB in improving the pasting properties and increasing the water and oil absorption capacity of glutinous rice flour.Moreover,the gelatinization enthalpy of the fermented samples increased significantly.For frozen glutinous rice dough stored for 28 days,the viscoelasticity of frozen dough prepared by compound starter was better than that of control dough,and the freezable water content was lower than that of control dough.These results indicate that compound LAB fermentation is a promising technology in the glutinous rice-based food processing industry,which has significance for its application.展开更多

Rice callus suspension cultures(RCSC) and seed...Tissue culture techniques were used to produce large amounts of bioactive compounds with medicinal potential, overcoming space and time constraints for cancer prevention. Rice callus suspension cultures(RCSC) and seed extracts prepared from aromatic rice varieties were used to evaluate the cytotoxic impact on human colon and lung cancer cell lines, as well as a normal control cell line, using Taxol as a positive control. RCSC and seed extracts from two Indian aromatic rice varieties were applied at different concentrations to treat the cancer cell lines and normal lung fibroblasts over varying time intervals. Apoptosis was assessed in 1:5 dilutions of the A549 and HT-29 cell lines treated with RCSC for 72 h, using propidium iodide staining and flow cytometry. RCSC showed a more potent cytotoxic effect than seed extracts with minimal effect on the normal cell line, in contrast to Taxol. Confocal microscopy and flow cytometry further confirmed the apoptotic effect of RCSC. Gas chromatography-mass spectrometry-based metabolic profiling identified metabolites involved in cytotoxicity and highlighted altered pathways. RCSC is proposed as an alternative source for the development of novel anticancer drugs with reduced side effects.展开更多
Three-dimensional(3D)cell cultures have contributed to a variety of biological research fields by filling the gap between monolayers and animal models.The modern optical sectioning microscopic methods make it possible...Three-dimensional(3D)cell cultures have contributed to a variety of biological research fields by filling the gap between monolayers and animal models.The modern optical sectioning microscopic methods make it possible to probe the complexity of 3D cell cultures but are limited by the inherent opaqueness.While tissue optical clearing methods have emerged as powerful tools for investigating whole-mount tissues in 3D,they often have limitations,such as being too harsh for fragile 3D cell cultures,requiring complex handling protocols,or inducing tissue deformation with shrinkage or expansion.To address this issue,we proposed a modified optical clearing method for 3D cell cultures,called MACS-W,which is simple,highly efficient,and morphology-preserving.In our evaluation of MACS-W,we found that it exhibits excellent clearing capability in just 10 min,with minimal deformation,and helps drug evaluation on tumor spheroids.In summary,MACS-W is a fast,minimally-deformative and fluorescence compatible clearing method that has the potential to be widely used in the studies of 3D cell cultures.展开更多
Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic fac...Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic factors determining the taste of sweet potato leaves remain unclear.This study evaluated the taste and metabolic profile of sweet potato leaves cultivated in soil and hydroponic systems,revealing that hydroponic culture improved the taste of the leafy sweet potatoes.Using gas chromatography system(GC)-ToF-MS,200 metabolites were identified,encompassing most plant metabolic pathways.A comparison of good-tasting vs.poor-tasting sweet potato leaves identified 71 metabolites associated with taste quality formation.Poor-tasting leaves exhibited lower levels of amino acid metabolites and higher levels of carbohydrate and secondary metabolites.This research provides novel insights into enhancing the taste of leafy sweet potatoes.展开更多
This study aimed to evaluate the effects of dietary yeast culture(YC)on lipopolysaccharide(LPS)-induced oxidative stress,immune and inflammatory response in P.ussuriensis.The fish were randomly assigned into three gro...This study aimed to evaluate the effects of dietary yeast culture(YC)on lipopolysaccharide(LPS)-induced oxidative stress,immune and inflammatory response in P.ussuriensis.The fish were randomly assigned into three groups as the control group,LPS group and YC+LPS group.The fish in the control were fed diet with no YC supplementation and no LPS challenge,and the fish in the LPS group or YC+LPS group were fed diet supplemented with no YC or 20 g·kg^(-1)YC,and with LPS challenge,respectively.The results showed that compared with the control group,intestinal total antioxidant capacity(T-AOC)level and superoxide dismutase(SOD)activity were significantly decreased,while intestinal malondialdehyde(MDA),plasma aspartate transaminase(AST)and alanine transaminase(ALT)levels were significantly increased in the LPS group(P<0.05).Besides,lower plasma alkaline phosphatase(ALP),alternative complement pathway(ACH50)activity and the albumin(ALB)level,as well as higher lysozyme(LZM)activity,were also found in the LPS group.However,dietary 20 g·kg^(-1)YC supplementation could relieve the above LPS-induced changes in Pseudobagrus ussuriensis.Furtherly,LPS challenge could significantly up-regulate gene expression of interleukin-8(IL-8),heat shock protein(HSP70)and NF-κBp65 except for toll-like receptors 2(TLR2),while dietary 20 g·kg^(-1)YC supplementation suppressed the increased expression of NF-κBp65 and IL-8 induced by LPS in P.ussuriensis.In summary,LPS challenge could induce immune impairment,oxidative stress and hepatic damage,and the protective effect of dietary 20 g·kg^(-1)YC supplementation on LPS-induced immune impairment and oxidative stress was observed in the present study,which was associated with the enhanced levels of antioxidant enzymes and immune parameters.Also,dietary 20 g·kg^(-1)YC supplementation could suppress LPS-induced inflammatory response by down-regulating NF-κBp65,HSP70 and IL-8 gene expression.展开更多
Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma ...Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma pathobiology.Conventional cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under discussion.In vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell culture.However,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)characteristics.Therefore,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between them.Given the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy development.In this review,we first provide a general overview of 3D cell cultures compared to 2D cell cultures.We then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO methodologies.Finally,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.展开更多

Background:Mental health literacy(MHL)refers to one’s knowledge and understanding of mental health disorders and their treatments.This literacy may be influenced by cultural norms and values that shape individuals’e...Background:Mental health literacy(MHL)refers to one’s knowledge and understanding of mental health disorders and their treatments.This literacy may be influenced by cultural norms and values that shape individuals’experiences,beliefs,attitudes,and behaviors regarding mental health.This study focuses on adapting the Mental health literacy scale(MHLS)for use in the multicultural context of Israel.Objectives include validating its construct,assessing its accuracy in measuring MHL in this diverse setting and examining and comparing levels of MHL across different cultural groups.Methods:The data collection included 1057 participants,representing all the ethnic groups of the Israeli population aged 18 and over.The tools included the MHLS and a demographic questionnaire.Confirmatory factor analysis(CFA)was employed to assess the original structure of the MHLS.Results:The results revealed that after evaluating the original MHLS,five items were excluded,leading to the validation of a modified version—Israeli mental health scale(IMHLS)with four factors and 25 items.CFA and reliability analyses supported an established and robust four-factor model.Significant ethnic differences in MHLS scores were identified,with Muslim participants showing the highest familiarity with mental disorders,followed by Druze and Christian participants,while Jewish participants had the lowest familiarity.Conclusion:The study concluded that the IMHLS is a valid and reliable tool for assessing MHL in Israel’s diverse and multicultural population.The revised scale better reflects the cultural nuances of the Israeli context.The significant ethnic differences that the study revealed in IMHLS emphasize the importance of culturally sensitive mental health interventions tailored to different ethnic groups in Israel.展开更多
BACKGROUND Human mesenchymal stromal cells(MSCs)possess regenerative potential due to pluripotency and paracrine functions.However,their stemness and immunomod-ulatory capabilities are sub-optimal in conventional two-...BACKGROUND Human mesenchymal stromal cells(MSCs)possess regenerative potential due to pluripotency and paracrine functions.However,their stemness and immunomod-ulatory capabilities are sub-optimal in conventional two-dimensional(2D)culture.AIM To enhance the efficiency and therapeutic efficacy of MSCs,an in vivo-like 3D culture condition was applied.METHODS MSCs were cultured on polystyrene(2D)or in a gellan gum-based 3D system.In vitro,prostaglandin-endoperoxide synthase 2,indoleamine-2,3-dioxygenase,heme oxygenase 1,and prostaglandin E synthase gene expression was quantified by quantitative real-time polymerase chain reaction.MSCs were incubated with lipopolysaccharide(LPS)-treated mouse splenocytes,and prostaglandin E2 and tumor necrosis factor-alpha levels were measured by enzyme linked immuno-sorbent assay.In vivo,LPS was injected into the lateral ventricle of mouse brain,and MSCs were administered intravenously the next day.Animals were sacrificed and analyzed on days 2 and 6.RESULTS Gellan gum polymer-based 3D culture significantly increased expression of octamer-binding transcription factor 4 and Nanog homeobox stemness markers in human MSCs compared to 2D culture.This 3D environment also heightened expression of cyclooxygenase-2 and heme-oxygenase 1,enzymes known for immunomodulatory functions,including production of prostaglandins and heme degradation,respectively.MSCs in 3D culture secreted more prostaglandin E2 and effectively suppressed tumor necrosis factor-alpha release from LPS-stimulated splenocytes and surpassed the efficiency of MSCs cultured in 2D.In a murine neuroinflammation model,intravenous injection of 3D-cultured MSCs significantly reduced ionized calcium-binding adaptor molecule 1 and glial fibrillary acidic protein expression,mitigating chronic inflammation more effectively than 2D-cultured MSCs.CONCLUSION The microenvironment established in 3D culture serves as an in vivo mimetic,enhancing the immunomodulatory function of MSCs.This suggests that engineered MSCs hold significant promise a potent tool for cell therapy.展开更多

The experiment included four nitrogen solution concentration levels, with ...This study investigated the effects of chlorine-based fertilizers under varying nitrogen solution concentrations in a soilless culture system. The experiment included four nitrogen solution concentration levels, with nitrogen concentrations of 6 mmol/L (C1), 12 mmol/L (C2), 18 mmol/L (C3), and 24 mmol/L (C4). Each nutrient concentration level was further divided into four chloride ion treatments (R1, R2, R3, and R4), where 100%, 60%, 33%, and 0% of the NH4+ and K+ ions were derived from NH4Cl and KCl, respectively. The length, surface area and volume of root were significantly higher by 25.3%~136.9%, 40.1%~173.1%, 27.9%~178.0%, respectively, in the R4 treatment than in the R1 and R2 treatments at flowering stage. The aboveground biomass and yield in the R4 treatment were significantly higher, by approximately 15.6%~43.5% and 16.6%~28.6%, respectively, than in the R1, R2, and R3 treatments at the picking stage. The C3 and C4 treatments significantly decreased biomass and yield by 31.9%~50.2% and 20.7%~50.5%, respectively, compared to the C1 and C2 treatments at the picking stage. Besides, the higher nutrient solution increased the incidence of blossom-end rot. In conclusion, high concentrations of chloride ions in nutrient solutions, especially when the Cl− concentration exceeded 10 mmol/L, have been shown to inhibit tomato growth in soilless culture systems. Therefore, replacing sulfur-based fertilizers by chlorine-based fertilizers is not recommended for tomato production under the studied conditions.展开更多
The artemisinin accumulation in the hairy root cultures of Artemisia annua L. was enhanced via a treatment of three fungal elicitors separately ( Verticillium dahliae Kleb., Rhizopus stolonifer (Ehrenb. ex ...The artemisinin accumulation in the hairy root cultures of Artemisia annua L. was enhanced via a treatment of three fungal elicitors separately ( Verticillium dahliae Kleb., Rhizopus stolonifer (Ehrenb. ex Fr.) Vuill and Colletotrichum dematium (Pers.) Grove). Among these three elicitors, V. dahliae had the highest inducing efficiency, but none of them manifests any noticeable effects on the cell growth of the hairy root cultures. The artemisinin content of the hairy root cultures treated with V. dahliae elicitor was 1.12 mg/g DW, which was 45% higher than the control (0.77 mg/g DW). The results showed that elicitation was dependent on the elicitor concentration, the incubation period and the physiological stage at which the hairy root cultures were treated. In addition, the authors found that for V. dahliae , the optimum concentration was 0.4 mg carbohydrate per millilitre medium, the strongest response of A. annua hairy root cultures to the elicitation was at the late exponential growth stage, and the highest artemisinin content of the hairy root cultures was on the 4th day post treatment.展开更多

Fu keeps a high taxol_producing level after successive subcultures for more than eight years. In this study, eight taxanes were isolated from the suspension cell cultures of this cell line. Based on NMR and MS analyses, and comparison with literature data and standards, their structures were determined to be 2α,5α,10β_triacetoxy_14β_propionyloxy_4(20),11_taxadiene (1), 2α,5α,10β_triacetoxy_14β_(2′_methyl)_butyryloxy_4(20),11_taxadiene (2), 2α,5α,10β_14β_tetra_acetoxy_4 (20),11_taxadiene (3, taxuyunnanine C), 2α,5α,10β_triacetoxy_14β_(2′_methyl_3′_hydroxy)_butyryloxy_4(20),11_taxadiene (4, yunnanxane) and its 3′_epimer (5), baccatin Ⅳ (6), baccatin Ⅲ (7) and taxol (8), respectively. Among those compounds, 3, 5, 6 and 7 were reported to be isolated from the suspension cell cultures of T. yunnanensis for the first time. TLC and HPLC analyses indicated that the chemical constituents of the culture solution were similar to those of cultured cells. Moreover, the highest taxol content of this cell line reached 0.3% and the cell line could be applied for a large_scale culture.展开更多

Ethyl acetate extract of the cultures showed inhibitory activities against Penicillium avellaneum UC-4376 which was sensitive to maytansinoids. Exhaustive isolation of natural products from a large scale of suspension cell cultures did not yield maytansine instead of affording nine compounds including one novel triterpenoid, named 2, 3-diacetoxyl maytenusone (1), and eight known ones including squalene (2), beta-sitosterol (3), 2', 3', 4-triacetyl-sitoindoside I (4), salaspermic acid (5), maytenonic acid (6), 2alpha-hydroxy-maytenonic acid (7), 6, 11,12-trihydroxy-8, 11, 13-abietrien-7-one (8) and 11, 12-dihydroxy-8, 11, 13-abietatrien-7-one (9) elucidated on the basis of 1D and 2D NMR data. The H-1-NMR and C-13-NMR assignments were made for 1, 5, 6 and 7, while the C-13-NMR assignments for 5 and 6 were revised. The chemical results suggested that the suspension cell cultures of M. hookeri did not produce maytansinoids under the reported experiment conditions.展开更多

Local vernacular dwellings can be classified in the following 3 ways on the basis of their own regional features:First, classifying by the origin and development history:stilt building, courtyard and non-courtyard building, western-style building; Second, by building materials:bamboo-wood building, stone building, adobe building, earth-stone building, brick-wood building, building of mixed materials; Third, by architectural form:stilt building, courtyard building, non-courtyard building. Stilt wooden building in northern Guangxi is a representative of traditional vernacular dwellings in the study area. By giving mature consideration to inheritance and development of cultures of traditional vernacular dwellings, this study proposes that proper renovation plans should be made according to actual conditions of the planning site; form renovation of vernacular dwellings should be taken as the new trend of renovation; traditional cultures should be further explored, demonstrated and protected. 文摘The effects of 5 lactic acid bacteria(LAB)fermentation on the pasting properties of glutinous rice flour were compared,and suitable fermentation strains were selected based on the changes of viscosity,setback value,and breakdown value to prepare LAB compound starter cultures.The results revealed that Latilactobacillus sakei HSD004 and Lacticaseibacillus rhamnosus HSD005 had apparent advantages in increasing the viscosity and reducing the setback and breakdown values of glutinous rice flour.In particular,the compound starter created using the two abovementioned LAB in the ratio of 3:1 had better performance than that using a single LAB in improving the pasting properties and increasing the water and oil absorption capacity of glutinous rice flour.Moreover,the gelatinization enthalpy of the fermented samples increased significantly.For frozen glutinous rice dough stored for 28 days,the viscoelasticity of frozen dough prepared by compound starter was better than that of control dough,and the freezable water content was lower than that of control dough.These results indicate that compound LAB fermentation is a promising technology in the glutinous rice-based food processing industry,which has significance for its application.
文摘This paper explores the pivotal role of Matteo Ricci(1583-1610)in promoting Sino-Western cultural exchange during his mission in late Ming China,focusing on the period from his arrival in 1583 until his death in 1610.Ricci’s unique approach of cultural accommodation,scientific knowledge dissemination,and religious dialogue positioned him as a bridge between two vastly different civilizations.This study analyzes Ricci’s methods of engagement with Chinese scholars,his introduction of Western science,and the impact of his religious and philosophical exchanges.Ricci’s strategy of adopting Chinese customs and intellectual traditions allowed him to foster trust and dialogue,making him one of the most successful missionaries in China.The paper further examines how Ricci’s translations of scientific works and his attempts to harmonize Christianity with Confucian values significantly influenced Chinese intellectual circles.By introducing advanced European knowledge in astronomy,mathematics,and geography,Ricci not only facilitated scientific exchange but also enhanced the Jesuits’reputation among Chinese scholars.Ricci’s enduring legacy is examined in the context of early global interactions,emphasizing his influence on subsequent Jesuit missions and the broader history of intercultural dialogue between East and West,highlighting the transformative power of empathy,respect,and intellectual curiosity in cross-cultural engagements.

文摘Tissue culture techniques were used to produce large amounts of bioactive compounds with medicinal potential, overcoming space and time constraints for cancer prevention. Rice callus suspension cultures(RCSC) and seed extracts prepared from aromatic rice varieties were used to evaluate the cytotoxic impact on human colon and lung cancer cell lines, as well as a normal control cell line, using Taxol as a positive control. RCSC and seed extracts from two Indian aromatic rice varieties were applied at different concentrations to treat the cancer cell lines and normal lung fibroblasts over varying time intervals. Apoptosis was assessed in 1:5 dilutions of the A549 and HT-29 cell lines treated with RCSC for 72 h, using propidium iodide staining and flow cytometry. RCSC showed a more potent cytotoxic effect than seed extracts with minimal effect on the normal cell line, in contrast to Taxol. Confocal microscopy and flow cytometry further confirmed the apoptotic effect of RCSC. Gas chromatography-mass spectrometry-based metabolic profiling identified metabolites involved in cytotoxicity and highlighted altered pathways. RCSC is proposed as an alternative source for the development of novel anticancer drugs with reduced side effects.

文摘Three-dimensional(3D)cell cultures have contributed to a variety of biological research fields by filling the gap between monolayers and animal models.The modern optical sectioning microscopic methods make it possible to probe the complexity of 3D cell cultures but are limited by the inherent opaqueness.While tissue optical clearing methods have emerged as powerful tools for investigating whole-mount tissues in 3D,they often have limitations,such as being too harsh for fragile 3D cell cultures,requiring complex handling protocols,or inducing tissue deformation with shrinkage or expansion.To address this issue,we proposed a modified optical clearing method for 3D cell cultures,called MACS-W,which is simple,highly efficient,and morphology-preserving.In our evaluation of MACS-W,we found that it exhibits excellent clearing capability in just 10 min,with minimal deformation,and helps drug evaluation on tumor spheroids.In summary,MACS-W is a fast,minimally-deformative and fluorescence compatible clearing method that has the potential to be widely used in the studies of 3D cell cultures.

文摘Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic factors determining the taste of sweet potato leaves remain unclear.This study evaluated the taste and metabolic profile of sweet potato leaves cultivated in soil and hydroponic systems,revealing that hydroponic culture improved the taste of the leafy sweet potatoes.Using gas chromatography system(GC)-ToF-MS,200 metabolites were identified,encompassing most plant metabolic pathways.A comparison of good-tasting vs.poor-tasting sweet potato leaves identified 71 metabolites associated with taste quality formation.Poor-tasting leaves exhibited lower levels of amino acid metabolites and higher levels of carbohydrate and secondary metabolites.This research provides novel insights into enhancing the taste of leafy sweet potatoes.

文摘This study aimed to evaluate the effects of dietary yeast culture(YC)on lipopolysaccharide(LPS)-induced oxidative stress,immune and inflammatory response in P.ussuriensis.The fish were randomly assigned into three groups as the control group,LPS group and YC+LPS group.The fish in the control were fed diet with no YC supplementation and no LPS challenge,and the fish in the LPS group or YC+LPS group were fed diet supplemented with no YC or 20 g·kg^(-1)YC,and with LPS challenge,respectively.The results showed that compared with the control group,intestinal total antioxidant capacity(T-AOC)level and superoxide dismutase(SOD)activity were significantly decreased,while intestinal malondialdehyde(MDA),plasma aspartate transaminase(AST)and alanine transaminase(ALT)levels were significantly increased in the LPS group(P<0.05).Besides,lower plasma alkaline phosphatase(ALP),alternative complement pathway(ACH50)activity and the albumin(ALB)level,as well as higher lysozyme(LZM)activity,were also found in the LPS group.However,dietary 20 g·kg^(-1)YC supplementation could relieve the above LPS-induced changes in Pseudobagrus ussuriensis.Furtherly,LPS challenge could significantly up-regulate gene expression of interleukin-8(IL-8),heat shock protein(HSP70)and NF-κBp65 except for toll-like receptors 2(TLR2),while dietary 20 g·kg^(-1)YC supplementation suppressed the increased expression of NF-κBp65 and IL-8 induced by LPS in P.ussuriensis.In summary,LPS challenge could induce immune impairment,oxidative stress and hepatic damage,and the protective effect of dietary 20 g·kg^(-1)YC supplementation on LPS-induced immune impairment and oxidative stress was observed in the present study,which was associated with the enhanced levels of antioxidant enzymes and immune parameters.Also,dietary 20 g·kg^(-1)YC supplementation could suppress LPS-induced inflammatory response by down-regulating NF-κBp65,HSP70 and IL-8 gene expression.
文摘Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma pathobiology.Conventional cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under discussion.In vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell culture.However,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)characteristics.Therefore,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between them.Given the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy development.In this review,we first provide a general overview of 3D cell cultures compared to 2D cell cultures.We then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO methodologies.Finally,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.

文摘Background:Mental health literacy(MHL)refers to one’s knowledge and understanding of mental health disorders and their treatments.This literacy may be influenced by cultural norms and values that shape individuals’experiences,beliefs,attitudes,and behaviors regarding mental health.This study focuses on adapting the Mental health literacy scale(MHLS)for use in the multicultural context of Israel.Objectives include validating its construct,assessing its accuracy in measuring MHL in this diverse setting and examining and comparing levels of MHL across different cultural groups.Methods:The data collection included 1057 participants,representing all the ethnic groups of the Israeli population aged 18 and over.The tools included the MHLS and a demographic questionnaire.Confirmatory factor analysis(CFA)was employed to assess the original structure of the MHLS.Results:The results revealed that after evaluating the original MHLS,five items were excluded,leading to the validation of a modified version—Israeli mental health scale(IMHLS)with four factors and 25 items.CFA and reliability analyses supported an established and robust four-factor model.Significant ethnic differences in MHLS scores were identified,with Muslim participants showing the highest familiarity with mental disorders,followed by Druze and Christian participants,while Jewish participants had the lowest familiarity.Conclusion:The study concluded that the IMHLS is a valid and reliable tool for assessing MHL in Israel’s diverse and multicultural population.The revised scale better reflects the cultural nuances of the Israeli context.The significant ethnic differences that the study revealed in IMHLS emphasize the importance of culturally sensitive mental health interventions tailored to different ethnic groups in Israel.

文摘BACKGROUND Human mesenchymal stromal cells(MSCs)possess regenerative potential due to pluripotency and paracrine functions.However,their stemness and immunomod-ulatory capabilities are sub-optimal in conventional two-dimensional(2D)culture.AIM To enhance the efficiency and therapeutic efficacy of MSCs,an in vivo-like 3D culture condition was applied.METHODS MSCs were cultured on polystyrene(2D)or in a gellan gum-based 3D system.In vitro,prostaglandin-endoperoxide synthase 2,indoleamine-2,3-dioxygenase,heme oxygenase 1,and prostaglandin E synthase gene expression was quantified by quantitative real-time polymerase chain reaction.MSCs were incubated with lipopolysaccharide(LPS)-treated mouse splenocytes,and prostaglandin E2 and tumor necrosis factor-alpha levels were measured by enzyme linked immuno-sorbent assay.In vivo,LPS was injected into the lateral ventricle of mouse brain,and MSCs were administered intravenously the next day.Animals were sacrificed and analyzed on days 2 and 6.RESULTS Gellan gum polymer-based 3D culture significantly increased expression of octamer-binding transcription factor 4 and Nanog homeobox stemness markers in human MSCs compared to 2D culture.This 3D environment also heightened expression of cyclooxygenase-2 and heme-oxygenase 1,enzymes known for immunomodulatory functions,including production of prostaglandins and heme degradation,respectively.MSCs in 3D culture secreted more prostaglandin E2 and effectively suppressed tumor necrosis factor-alpha release from LPS-stimulated splenocytes and surpassed the efficiency of MSCs cultured in 2D.In a murine neuroinflammation model,intravenous injection of 3D-cultured MSCs significantly reduced ionized calcium-binding adaptor molecule 1 and glial fibrillary acidic protein expression,mitigating chronic inflammation more effectively than 2D-cultured MSCs.CONCLUSION The microenvironment established in 3D culture serves as an in vivo mimetic,enhancing the immunomodulatory function of MSCs.This suggests that engineered MSCs hold significant promise a potent tool for cell therapy.

文摘This study investigated the effects of chlorine-based fertilizers under varying nitrogen solution concentrations in a soilless culture system. The experiment included four nitrogen solution concentration levels, with nitrogen concentrations of 6 mmol/L (C1), 12 mmol/L (C2), 18 mmol/L (C3), and 24 mmol/L (C4). Each nutrient concentration level was further divided into four chloride ion treatments (R1, R2, R3, and R4), where 100%, 60%, 33%, and 0% of the NH4+ and K+ ions were derived from NH4Cl and KCl, respectively. The length, surface area and volume of root were significantly higher by 25.3%~136.9%, 40.1%~173.1%, 27.9%~178.0%, respectively, in the R4 treatment than in the R1 and R2 treatments at flowering stage. The aboveground biomass and yield in the R4 treatment were significantly higher, by approximately 15.6%~43.5% and 16.6%~28.6%, respectively, than in the R1, R2, and R3 treatments at the picking stage. The C3 and C4 treatments significantly decreased biomass and yield by 31.9%~50.2% and 20.7%~50.5%, respectively, compared to the C1 and C2 treatments at the picking stage. Besides, the higher nutrient solution increased the incidence of blossom-end rot. In conclusion, high concentrations of chloride ions in nutrient solutions, especially when the Cl− concentration exceeded 10 mmol/L, have been shown to inhibit tomato growth in soilless culture systems. Therefore, replacing sulfur-based fertilizers by chlorine-based fertilizers is not recommended for tomato production under the studied conditions.
文摘The artemisinin accumulation in the hairy root cultures of Artemisia annua L. was enhanced via a treatment of three fungal elicitors separately ( Verticillium dahliae Kleb., Rhizopus stolonifer (Ehrenb. ex Fr.) Vuill and Colletotrichum dematium (Pers.) Grove). Among these three elicitors, V. dahliae had the highest inducing efficiency, but none of them manifests any noticeable effects on the cell growth of the hairy root cultures. The artemisinin content of the hairy root cultures treated with V. dahliae elicitor was 1.12 mg/g DW, which was 45% higher than the control (0.77 mg/g DW). The results showed that elicitation was dependent on the elicitor concentration, the incubation period and the physiological stage at which the hairy root cultures were treated. In addition, the authors found that for V. dahliae , the optimum concentration was 0.4 mg carbohydrate per millilitre medium, the strongest response of A. annua hairy root cultures to the elicitation was at the late exponential growth stage, and the highest artemisinin content of the hairy root cultures was on the 4th day post treatment.
文摘A high taxol yield cell line of Taxus yunnanensis Cheng et L. K. Fu keeps a high taxol_producing level after successive subcultures for more than eight years. In this study, eight taxanes were isolated from the suspension cell cultures of this cell line. Based on NMR and MS analyses, and comparison with literature data and standards, their structures were determined to be 2α,5α,10β_triacetoxy_14β_propionyloxy_4(20),11_taxadiene (1), 2α,5α,10β_triacetoxy_14β_(2′_methyl)_butyryloxy_4(20),11_taxadiene (2), 2α,5α,10β_14β_tetra_acetoxy_4 (20),11_taxadiene (3, taxuyunnanine C), 2α,5α,10β_triacetoxy_14β_(2′_methyl_3′_hydroxy)_butyryloxy_4(20),11_taxadiene (4, yunnanxane) and its 3′_epimer (5), baccatin Ⅳ (6), baccatin Ⅲ (7) and taxol (8), respectively. Among those compounds, 3, 5, 6 and 7 were reported to be isolated from the suspension cell cultures of T. yunnanensis for the first time. TLC and HPLC analyses indicated that the chemical constituents of the culture solution were similar to those of cultured cells. Moreover, the highest taxol content of this cell line reached 0.3% and the cell line could be applied for a large_scale culture.
文摘Suspension cell cultures of Maytenus hookeri Loos. (Celastraceae) in SH media were established from the calli induced from the leaves and young steins of M. hookeri on MS media with the supplement of 2 mg/L 2,4-D and 0.1 mg/L KIN (kinetin). Ethyl acetate extract of the cultures showed inhibitory activities against Penicillium avellaneum UC-4376 which was sensitive to maytansinoids. Exhaustive isolation of natural products from a large scale of suspension cell cultures did not yield maytansine instead of affording nine compounds including one novel triterpenoid, named 2, 3-diacetoxyl maytenusone (1), and eight known ones including squalene (2), beta-sitosterol (3), 2', 3', 4-triacetyl-sitoindoside I (4), salaspermic acid (5), maytenonic acid (6), 2alpha-hydroxy-maytenonic acid (7), 6, 11,12-trihydroxy-8, 11, 13-abietrien-7-one (8) and 11, 12-dihydroxy-8, 11, 13-abietatrien-7-one (9) elucidated on the basis of 1D and 2D NMR data. The H-1-NMR and C-13-NMR assignments were made for 1, 5, 6 and 7, while the C-13-NMR assignments for 5 and 6 were revised. The chemical results suggested that the suspension cell cultures of M. hookeri did not produce maytansinoids under the reported experiment conditions.
文摘By analyzing background and present situation of townscape renovation project in Guangxi, endeavors for inheriting and developing cultures of traditional vernacular dwellings in the implementation of this project are discussed, and characteristics of traditional vernacular dwellings in the study area are summarized. Local vernacular dwellings can be classified in the following 3 ways on the basis of their own regional features:First, classifying by the origin and development history:stilt building, courtyard and non-courtyard building, western-style building; Second, by building materials:bamboo-wood building, stone building, adobe building, earth-stone building, brick-wood building, building of mixed materials; Third, by architectural form:stilt building, courtyard building, non-courtyard building. Stilt wooden building in northern Guangxi is a representative of traditional vernacular dwellings in the study area. By giving mature consideration to inheritance and development of cultures of traditional vernacular dwellings, this study proposes that proper renovation plans should be made according to actual conditions of the planning site; form renovation of vernacular dwellings should be taken as the new trend of renovation; traditional cultures should be further explored, demonstrated and protected. These suggestions are of great instructive significance for improving the cultural taste of townscapes.

文摘By studying close links between classical Chinese gardens and traditional Chinese cultures,it was stressed that spiritual characteristics pursued by classical Chinese gardens were inevitable components of their expression forms,which were also closely related to classical Chinese philosophy,social system and other cultural phenomena.Characteristics of classical Chinese gardens in terms of psychology were analyzed,and it was proposed that they had particular cultural connotations in the spiritual expression and appeal,that is,natural view of "human-nature oneness",etiquette culture of "coordinating with the nature by etiquette",seclusion view of "living in seclusion and pursuing a peaceful life".Concrete expressions of spiritual cultures in classical Chinese gardens were discussed,specifically,the expression of natural views was demonstrated as "respecting the nature,all creatures having its own course(Tao,way of evolution),adopting measures suiting local conditions,skillfully borrowing from other landscapes,simulating natural landscapes";as for the expression of seclusion view,it was demonstrated as the expression of seclusion cultures by inscriptions and steles,the expression of "seclusion" by implicit garden settings;etiquette and hierarchy of classical Chinese gardens were discussed from the perspective of private garden layout and residence construction.On this basis,spiritual world of ancient Chinese were further stu-died,mental patterns of Chinese were believed as the source of classical gardens,and what those hills,water,plants,architecture as well as their combinations expressed were spiritual pursuits of Chinese people.

文摘By studying typical "Top Eight Views" in cities like Xining,Lanzhou and Yinchuan in the upper reaches of the Yellow River,mountain and water landscapes in the study area were analyzed in terms of natural ecology;bridges,canals,ferries,pavilions,mansions,towers,temples,historical sites,production and living landscapes were analyzed in terms of humanistic ecology.On this basis,"Top Eight Views" in the upper reaches of the Yellow River were compared and the following conclusions obtained:mountain landscapes were distributed in the whole region,waterscapes unevenly valued in different cities,bridges,canals and ferries mostly located in Ningxia,temples commonly found in all cities,garden arts introduced into cities,traditional cultures carried forward by historical relics.Then the relationship between "Top Eight Views" and regional eco-cultures was analyzed,and it was proposed that humanistic and cultural connotations of these "Top Eight Views" contributed a lot to beautifying urban environment,satisfying needs of the locals' spiritual life,carrying forward local history and culture,enriching urban cultural connotations,expanding living spaces of local residents and improving integrated functions of cities."Top Eight Views" culture complied with the construction gist of urban ecological cultures,and was of great referential value for the construction of urban ecological cultures in the upper reaches of the Yellow River,and also the healthy,scientific and sustainable development of local cities.
