Least squares projection twin support vector machine(LSPTSVM)has faster computing speed than classical least squares support vector machine(LSSVM).However,LSPTSVM is sensitive to outliers and its solution lacks sparsi...Least squares projection twin support vector machine(LSPTSVM)has faster computing speed than classical least squares support vector machine(LSSVM).However,LSPTSVM is sensitive to outliers and its solution lacks sparsity.Therefore,it is difficult for LSPTSVM to process large-scale datasets with outliers.In this paper,we propose a robust LSPTSVM model(called R-LSPTSVM)by applying truncated least squares loss function.The robustness of R-LSPTSVM is proved from a weighted perspective.Furthermore,we obtain the sparse solution of R-LSPTSVM by using the pivoting Cholesky factorization method in primal space.Finally,the sparse R-LSPTSVM algorithm(SR-LSPTSVM)is proposed.Experimental results show that SR-LSPTSVM is insensitive to outliers and can deal with large-scale datasets fastly.展开更多

In this work, two chemometrics methods are applied for the modeling and prediction of electrophoretic mobilities of some organic and inorganic compounds. The successive projection algorithm, feature selection (SPA) ...In this work, two chemometrics methods are applied for the modeling and prediction of electrophoretic mobilities of some organic and inorganic compounds. The successive projection algorithm, feature selection (SPA) strategy, is used as the descriptor selection and model development method. Then, the support vector machine (SVM) and multiple linear regression (MLR) model are utilized to construct the non-linear and linear quantitative structure-property relationship models. The results obtained using the SVM model are compared with those obtained using MLR reveal that the SVM model is of much better predictive value than the MLR one. The root-mean-square errors for the training set and the test set for the SVM model were 0.1911 and 0.2569, respectively, while by the MLR model, they were 0.4908 and 0.6494, respectively. The results show that the SVM model drastically enhances the ability of prediction in QSPR studies and is superior to the MLR model.展开更多
Background Assessment of perfusion in the early period of acute renal failure (ARF) is important,and can not be made by conventional ultrasound.The aim of this study was to prospectively test in a rabbit ARF model w...Background Assessment of perfusion in the early period of acute renal failure (ARF) is important,and can not be made by conventional ultrasound.The aim of this study was to prospectively test in a rabbit ARF model whether real-time contrast-enhanced ultrasound (CEUS) can quantitatively evaluate the hemodynamic changes of renal cortex in the early period.Methods The model of ARF was induced in 30 healthy New Zealand white rabbits (2.5-3.0 kg),by intramuscular injection of 50% glycerin solution (12 ml/kg).CEUS were performed on right kidneys before,6 and 24 hours after glycerin injection.CEUS quantitative indexes were measured in renal cortex using QLAB software.Comparisons between different stages were performed using paired t test.The sensitivity of CEUS in diagnosing ARF was compared with blood serum urea nitrogen (BUN),serum creatinine (SCr) level and color Doppler flow imaging (CDFI).Results Among quantitative indexes,time to peak (TTP) and area under curve (AUC) increased significantly from (5.86±2.57) seconds and (124.4±46.7) dB.s before glycerin injection to (7.66±2.05) seconds and (288.1±64.9) dB.s 6 hours after injection (P 〈0.05).Slope rate of ascending curve (A) and descending curve (α) decreased slightly from (3.00±1.22) dB/s and (0.19±0.15) 1/s to (2.80±1.45) dB.s and (0.09±0.02) 1/s (P 〈0.05).Twenty-four hours later,only AUC increased significantly from (124.4±46.7) dB.s to (466.2±52.2) dB.s (P〈0.05).Conclusion Quantitative indexes of CEUS might be useful in predicting the hemodynamic changes of renal cortex in the early six hours of ARF model.展开更多

Methods: The changes of the cerebral cortical temperature d...Objective: To explore the possibility of using cortical infrared thermography (CIT) and temperature in studying the principle of acupuncture and moxibustion. Methods: The changes of the cerebral cortical temperature during electro-acupuncture (EA) measured by CIT in twenty cats were observed. By opening the skull before experiments, the cortex of animals was revealed. The distribution of the conical temperature of the awakened animals was studied by AGA infrared thermovision, and the thermograms were analysed by computer. Results: (1) The thermograms of the profile of the cortex could be displayed clearly, the temperature was higher in the ante and lower in post-cortex than that in other regions. (2) No tendency of conical temperature ascending within 30 minutes in cats without EA. (3 ) After EA, the temperature of cortex generally increased by 0.3 to 1.7 degrees centigrade, and 5 min after EA, the increment of temperature was the highest, 10 min after EA, the temperature increment was higher than before EA, but lower than 5 min after EA. The conical temperature recovered 10 min after stopping the stimulation. (4) The temperature increment was more obvious in the regions of ant. ectosylvian, ant. supra-Sylvian and ant. Sylvian than that in other regions. No matter where the site of EA was, every group had its own special region of higher temperature increment. Conclusion: The temperature increased in the cortex were mainly in the somatosensory regions. The extent and position of temperature response of cortex after EA could be displayed with CIT directly by picture. The display of the temperature response could be taken as the indication of the nervous activity. CIT could be used to conduct the studies of the acupuncture and the brain.展开更多
Objective To investigate the reversal of aberrant splicing of β thalassemia allele (IVS 2 654 C→T, β 654 ) by antisense RNA in vitro and in vivo.Methods The vector expressing antisense RNA which targeted a...Objective To investigate the reversal of aberrant splicing of β thalassemia allele (IVS 2 654 C→T, β 654 ) by antisense RNA in vitro and in vivo.Methods The vector expressing antisense RNA which targeted against the aberrant splice sites of β 654 pre mRNA was constructed in pcDNA3, and then used to repair the defective splicing of the mutant pre mRNA in an in vitro transcription and splicing system, as well as in HeLa β 654 cells and cultured β 654 erythroid cells by lipid mediated DNA transfection method. The effect of the antisense RNA was identified by RT PCR mediated mRNA quantitative assay as well as globin chain micro biosynthesis.Results The antisense RNA decreased the aberrant splicing product and restored the correct splicing pattern in vitro and in vivo efficiently. In the in vitro transcription and splicing system, the level of normally spliced mRNA [β/(β+β *)] increased from 0.25 to 0.60. In cultured HeLa β 654 cells, the level of β/(β+β *) increased from 0.07 to 0.43 on the 15th day after transfection. In cultured β 654 erythroid cells, the level of mRNA [β/(β+β *)] increased from 0.19 to 0.58 on the 8th day after transfection in β 654 /β 654 erythroid cells, from 0.02 to 0.38 in β 654 /β 41 42 erythroid cells, and from 0.45 to 0.83 in β 654 /βA erythroid cells, respectively. Correspondingly, the ratios of globin chain (β/α) biosynthesis increased from 0.16 to 0.52 on the 8th day after transfection in β 654 /β 654 erythroid cells, 0.05 to 0.36 in β 654 /β 41 42 erythroid cells, and 0.42 to 0.81 in β 654 /β A erythroid cells, respectively. The splicing pattern did not show significant changes as compared to the untreated, as well as to the control antisense fragment.Conclusions Antisense RNA transcribed from the expression vector described here could efficiently suppress the aberrant splicing pattern of β 654 mutant mRNA and restore the correct splicing pathway in vitro and in vivo, leading to the improvement of globin chain biosynthesis in thalassemic cells. Our antisense strategy provides an alternative approach to the gene therapy of β thalassemia.展开更多
