Enzymatic Determination of Glucose by Optical-Fiber Sensor Sequential Injection Renewable Surface Spectrophotometry

On the basis of oxidative decoloration of bromopyrogallol red (BPR) with H2O2, catalyzed by horseradish peroxidase( HRP), and the sequential injection renewable surface technique( SI-RST), a highly sensitive optical-fiber sensor spectrophotometric method for the enzymatic determination of hydrogen peroxide was proposed. By coupling with a glucose oxidase(GOD)-catalyzed reaction, the method was used to determine glucose in human serum. The considerations in system and flow cell design, and factors that influence the determination performance are discussed. With 100μL of sample loaded and 0. 6 mg of bead trapped, the linear response range from 5.0 × 10^-8 to 5.2 × 10^-6 mol/L BPR with a detection limit(3σ) of 2. 5 ×10 ^-8 mol/L BPR, and a precision of 1.1% RSD( n = 11 ) and a throughput of a 80 samples per hour can be achieved. Under the conditions of a 8. 7 × 10^ -6 mol/L BPR substrate, 0. 04 unit/mL HRP, 600 s reaction time and a reaction temperature of 37℃, the linear response range for H2O2 was from 5.0 × 10^-8 to 7.0 × 10^-6 mol/L with a detection limit(3σ) of 1.0 × 10^-8 mol/L and a precision of 3.7% RSD ( n = 11 ). The linear response range by coupling with a GOD-catalyzed reaction was from 1.0 × 10^-7 to 1.0 × 10^-5 mol/L. The method was directly applied to determine glucose in human serum. Glucose contents obtained by the proposed procedure were compared with those obtained by using the phenol-4-AAP method, the error was found to be less than 3%. 展开更多

A gas-liquid mass transfer model based on an unsteady state film mechanism applied to a single bubble is presented. The mathematical model was solved using Laplace transform to obtain an analytical solution of concentration profile in terms of the radial position r and time t. The dynamic mass transfer flux was deduced and the influence of the bubble size was also determined. A mathematical method for deducing the average mass transfer flux directly from the Laplace transformed concentration is presented. Its accuracy is verified by comparing the numerical results with those from the indirect method. The influences of the model parameters, namely, the bubble size R, liquid film thickness δ, and the surface renewal constant s on the average mass transfer flux were investigated. The proposed model is useful for a better understanding of the mass transfer mechanism and an optimum design of gas-liquid contact equipment. 展开更多

The process of producing high viscosity polyester by transesterification polycondensation needs to adjust the operating conditions and equipment structure of pre-polycondensation kettle and final polycondensation kettle to realize process intensification.In view of this,the fluid volume function method of computational fluid dynamics numerical simulation was used to investigate the film formation and surface renewal characteristics of horizontal polycondensation kettle under different operating conditions,including viscosity,rotating speed and liquid height.The results show that the viscosity and rotating speed were positively correlated with the film area and surface renewal in the pre-polycondensation stage.However,increasing the viscosity by several orders of magnitude in the final polycondensation kettle,the larger the film area and film thickness,but the overall surface renewal of the disk decreased.Therefore,a hexagonal hole disk is designed.By comparison,it is found that the film is more uniform,the surface update frequency is higher,and the power consumption can be reduced by more than 20%. 展开更多

Air-Sea Exchange of Volatile Organic Compounds: A New Model with Microlayer Effects

The authors propose a new "three-layer" conceptual model for the air-sea exchange of organic gases, which includes a dynamic surface microlayer with photochemical and biological processes. A parameterization of this three-layer model is presented, which was used to calculate the air-sea fluxes of acetone over the Pacific Ocean. The air-sea fluxes of acetone calculated by the three-layer model are in the same direction but possess half the magnitude of the fluxes calculated by the traditional two-layer model in the absence of photochemical and biological processes. However, photochemical and biological processes impacting acetone in the microlayer can greatly vary the calculated fluxes in the three-layer model, even reversing their direction under favorable conditions. Our model may help explain the discrepancies between measured and calculated acetone fluxes in previous studies. More measurements are needed to validate our conceptual model and provide constraints on the model parameters. 展开更多
