Objective: To study the effect on apoptosis of CD 4 +T、CD 19 +B in spleen of BXSB mice with systemic lupus erythematosus treated with Langchuangjing Granule (LCJG, and to probe into the mechanism of the tr eatment. Methods: The apoptosis was examined by the flow cytometric analysis and immunofluorescence double-staining method. Results: Apoptosis of male BXSB mice speeds up. LCJG can restrain the excessive apoptosis of CD 4 +T and CD 19 +B cells in spleen. Conclusion: LCJG treated systemic lupus erythematosus by restraining the excessive apoptosis of T, B lymphocytes, probably restraining the release of excessive amount of apoptotic DNA fragments, so decreasing abnormal proliferation of B cells and the produce of autoantibodies. 展开更多

The thyroid, an endocrine gland located at the base of the neck, produces thyroid hormones (triiodothyronine (T3) and thyroxine (T4)). The production of these hormones is possible by iodine and other nutrients such as selenium and certain vitamins. To assess the thyroid disturbances in the mountain population of Benin, a survey was conducted in Natitingou, a mountain town located in the department of Atacora, in the northwest of Benin, on a sample of thirty (30) adults (15 men and 15 women), most of whom are educated. The results of the questionnaire revealed that 43% of the surveyed population acknowledged having knowledge on the mentioned subject and have dietary habits based on the consumption of seafood, and also legumes (20%). The examination of the serum results of the dosage of T3, T4 and TSH hormones revealed cases of thyroid disturbances in the region (36.32% in men and 44.98% in women). The analysis of a comparative table including the “VEDALAB Easy Reader+” and five (05) other readers, presents the performance, reading techniques, principles, advantages and disadvantages of each device. Pending further studies, some recommendations were made at the end of this study to the academic authorities regarding probable cases of dysthyroidism for which additional examinations are required and an awareness for the improvement of dietary habits. 展开更多

Objective:To determine the anti-breast cancer activities and the safety oral consumption of Dillenia suffruticosa root aqueous extract(DRAE)in BALB/c mice.Methods:In the anti-breast cancer study,female BALB/c mice were divided into five groups(n=12),which were(1)positive control(with breast cancer,untreated),(2)negative control(without breast cancer,untreated)and other three groups of mice with breast cancer treated with 1 000,500 and 250 mg/kg of DRAE,respectively,by oral gavage for 28 days.All mice except from the negative control group were injected into the mammary fat pad with 4T1 cells(1×1054T1 cells/0.1 m L of phosphate buffer solution).DRAE was administered orally on Day 11 after the tumor has developed.Results:The tumor volume of the 1 000 mg/kg of DRAE group reduced significantly compared to the positive control while treatment with 500 mg/kg of DRAE had significantly inhibited metastasis to the heart.In the acute toxicity study,treatment with up to5 000 mg/kg of DRAE was not toxic to the animals,indicating its safety when a large amount of this plant extract was ingested.Based on the sub-acute toxicity study,treatment of the highest dose of DRAE(1 000 mg/kg)had mild liver toxicity indicated by mild focal hemorrhage.Conclusions:DRAE possesses anti-breast cancer properties but at the same time it shows mild toxicity to the liver.The non observable adverse effect dose for DRAE is500 mg/kg. 展开更多

基于MRI征象与影像组学诊断胃癌T3与T4a分期效能的对比分析

目的探讨基于MRI征象与影像组学诊断进展期胃癌T3和T4a分期效能的对比分析。方法选取于同济大学附属东方医院庐江分院行MRI检查,经病理结果证实,共计纳入208例胃癌患者,其中T3期96例,T4a期112例。首先应用传统影像学征象判断进展期胃癌... 目的探讨基于MRI征象与影像组学诊断进展期胃癌T3和T4a分期效能的对比分析。方法选取于同济大学附属东方医院庐江分院行MRI检查,经病理结果证实,共计纳入208例胃癌患者,其中T3期96例,T4a期112例。首先应用传统影像学征象判断进展期胃癌侵犯浆膜层等征象,比较多序列MRI征象在病理证实T3和T4a期胃癌中表现的差异性;其次按7:3的比例随机分为训练组(n=145)和验证组(n=63),分别从常规T2非抑脂序列及高比值DWI序列(b=1000 s/mm^(2))图像中提取影像组学特征,构建影像组学联合模型;然后分别绘制基于传统MRI征象与影像组学联合模型工作特征(receiver operating characteristic,ROC)曲线,并计算ROC曲线下面积(area under the curve,AUC)、特异度及灵敏度,量化两种诊断方式对胃癌T3和T4a分期的诊断效能。结果传统多序列MRI征象诊断AUC:0.929(95%CI:0.887~0.970),特异度0.912,灵敏度0.916;MRI影像组学联合模型训练组诊断AUC:0.975(95%CI:0.974~0.976),特异度0.946,灵敏度0.956,验证组诊断AUC:0.971(95%CI:0.965~0.974),特异度0.946,灵敏度0.943,均具有统计学意义(P<0.05)。结论基于MRI影像组学模型诊断T3和T4a分期效能高于传统MRI征象,值得临床工作中进一步推广使用。 展开更多

Low T3 vs low T3T4 euthyroid sick syndrome in septic shock patients:A prospective observational cohort study

BACKGROUND Both phases of euthyroid sick syndrome(ESS)are associated with worse prognosis in septic shock patients.Although there are still no indications for supplementation therapy,there is no evidence that both phases(initial and prolonged)are adaptive or that only prolonged is maladaptive and requires supplementation.AIM To analyze clinical,hemodynamic and laboratory differences in two groups of septic shock patients with ESS.METHODS A total of 47 septic shock patients with ESS were divided according to values of their thyroid hormones into low T3 and low T3T4 groups.The analysis included demographic data,mortality scores,intensive care unit stay,mechanical ventilation length and 28-day survival and laboratory with hemodynamics.RESULTS The Simplified Acute Physiology Score II score(P=0.029),dobutamine(P=0.003)and epinephrine requirement(P=0.000)and the incidence of renal failure and multiple organ failure(MOF)(P=0.000)were significantly higher for the low T3T4.Hypoalbuminemia(P=0.047),neutrophilia(P=0.038),lymphopenia(P=0.013)and lactatemia(P=0.013)were more pronounced on T2 for the low T3T4 group compared to the low T3 group.Diastolic blood pressure at T0(P=0.017)and T1(P=0.007),as well as mean arterial pressure at T0(P=0.037)and T2(P=0.033)was higher for the low T3 group.CONCLUSION The low T3T4 population is associated with higher frequency of renal insufficiency and MOF,with worse laboratory and hemodynamic parameters.These findings suggest potentially maladaptive changes in the chronic phase of septic shock. 展开更多

Nanopharmaceuticals containing quantum dot nanoparticles (Q-Dot NPs) for treating serious cancers such as breast cancer have made fantastic proposals. In this study, ZnO quantum dot NPs are formulated via ZnO@PVP nanopolymer as co-assistants coordinating with efficacious suitable wetting agents, PEG-binding compound, and W/O emulsifier for producing eco-friendly water-based nanodrug. Several characterization techniques containing SEM, TEM, FTIR, photoluminescence, zeta potential, and UV-Vis absorption were employed for ZnO Q-Dot NPs in nanodrug. This work aims to investigate the anti-tumor effects of such nanomedicine on the 4T1 breast cancer cell line in BALB/c mice, being elaborated through intraperitoneal, injection (IVP) and oral therapy. The impressive findings showed that ZnO nanodrug caused changes in blood factors, having the most effectiveness at 40 μg/ml concentration after two weeks of oral treatments. The significant increase in white blood cells (WBC) neutrophils and meaningful decreases in lymphocytes and especially cholesterol were powerful simultaneous impacts, successfully treating malignant breast cancer masses. In this significant animal model research for breast cancer, the sick mice recovered entirely and even had a safe space to mate. Histopathological results showed no evidence of breast tumor formation or metastasis in the group treated with nanodrug and their children. This nanomedicine has a therapeutic effect, and is ready to be applied for treating volunteer breast cancer patients. However, its prevention (inhibitory) effect can also be analyzed and added to current data in future studies. 展开更多

METHODS: Hematological changes and changes of lymphocyte subpopulations were detected in Smad3"/- mice using cell counter and flow cytometry, respectively, and compared to their littermate controls. RESULTS: The numbers of neutrophils and lymphocytes in peripheral blood were significantly increased in Smad3^-/- mice compared to littermate controls. CD19^+ expressing cells in blood and spleen, and CD8^+ T cells in thymus were all markedly decreased in Smad3^-/- mice. More important, Smad3^-/- mice had an increased population of CD4^+CD25^+ T cells in peripheral lymphoid tissues, including thymus, spleen, and lymph nodes. CONCLUSION: These observations suggest that the changes of lymphocyte subpopulations might play a role in susceptibility to inflammation of Smad3^-/- mice. 展开更多

The immune effect of two recombinant protein fragments of spike protein in severe acute respiratory syndrome coronavirus (SARS CoV) was investigated in Balb/c mice. Two partial spike gene fragments S1 (322-1464 bp) and S2 (2170-2814 bp) of SARS coronavirus were amplified by RT-PCR, and cloned into pET-23a prokaryotic expression vector, then transformed into competent Escherichia E.coli BL21 (DE3)(pLysS) respectively. Recombinant proteins were expressed and puri- fied by Ni2+ immobilized metal ion affinity chromatography. The purified proteins mixed with com- plete Freund adjuvant were injected into Balb/c mice three times at a two-week interval. High titer antibody was detected in the serum of immunized Balb/c mice, and mice immunized with S1 protein produced high titer IgG1, IgG2a, IgG2b and IgG3, while those immunized with S2 protein produced high titer IgG1, IgG2a, but lower titer IgG2b and IgG3. Serum IFN-γ concentration was increased significantly but the concentrations of IL-2, IL-4 and IL-10 had no significant change. And a marked increase was observed in the number of spleen CD8+ T cells. The results showed that recombinant proteins of SARS coronavirus spike protein induced hormonal and cellular immune response in Balb/c mice. 展开更多

Objective:Acupuncture is considered an important part of the alternative medical treatment of tumors.However,it is still unclear whether acupuncture has a direct effect on inhibition of tumor growth.The purpose of this study was to evaluate the effects of acupuncture treatment on tumors in BALB/c mice that were or were not administered cyclophosphamide.Methods:A 4T1 breast carcinoma mouse model for this study was established.When the mice developed 5 mm × 5 mm visible subcutaneous tumors,they were subjected to acupuncture and/or cyclophosphamide (CTX) treatment for 15 days.Results:Acupuncture and CTX treatment both reduced the size of the solid tumors.When used in conjunction,the tumor size reduction was even more obvious.Hematoxylin and eosin staining showed that acupuncture had a significant effect on induction of tumor cell necrosis and inhibition of angiogenesis.Immunohistochemistry showed that the expression of CD34 and matrix metalloproteinase-9 (MMP-9),which were related to angiogenesis,decreased after treatment with either acupuncture or CTX.However,when the two treatments were used in conjunction,they showed a synergistic effect on inhibiting the expression of CD34.However,the synergistic effect was not observed onMvMP-9 expression.Conclusion:Acupuncture plays an important role in improving the effect of chemotherapy by inhibiting angiogenesis. 展开更多

Most patients lost their vocal ability after ablative surgery for the laryngeal cancer. Total laryngectomy has a history of over 100 years, hence many surgeons are accustomed to the en bloc organ resection and the loss of ability for communication after surgery. The recent advance of surgery introduces the concept and technique of radical cure and functional preservation of the cancer-afflicted-organ at same setting. For lesions of late T categories (T3, T4), the maintenance of normal function after wide resection to ensure radicality demands a special technique to repair the crippled larynx. For this purpose a local osteomuscular flap was designed to reconstruct a functioning larynx. The surgical manoeuvre was described and the result proved to be successful. The outcome of 138 patients with T3 and T4 laryngeal carcinomas following conservation laryngectomy was analyzed. The five-year survival rate was 69% for supraglottic type and 75% for glottic type. The quality of life was much improved. 展开更多

Glucose transporter 4 (GLUT4) is responsible for insulin-stimulated glucose transporting into the insulin-sensitive fat and muscle cells. The dynamics of GLUT4 storage vesicles (GSVs) remains to be explored and it is unclear how GSVs are arranged based on their mobility. We examined this issue in 3T3-L1 cells via investigating the three-dimensional mobility of single GSV labeled with EGFP-fused GLUT4. A thin layer of cytosol right adjacent to the plasma membrane was illuminated and successively imaged at 5 Hz under a total internal reflection fluorescence microscope with a penetration depth of 136 nm. Employing single particle tracking, the three-dimensional subpixel displacement of single GSV was tracked at a spatial precision of 22 nm. Both the mean square displacement and the diffusion coefficient were calculated for each vesicle. Tracking results revealed that vesicles moved as if restricted within a cage that has a mean radius of 160 nm, suggesting the presence of some intracellular tethering matrix. By constructing the histogram of the diffusion coefficients of GSVs, we observed a smooth distribution instead of the existence of distinct groups. The result indicates that GSVs are dynamically retained in a continuous and wide range of mobility rather than into separate classes. 展开更多

目的探讨血清三碘甲状腺素(T3)、甲状腺素(T4)、促甲状腺素(H-TSH)、游离甲状腺素(FT4)、游离三碘甲状腺原氨酸(FT3)在甲状腺功能亢进(简称甲亢)与甲状腺功能减退(简称甲减)诊断中的应用价值。方法选择我院2017年1月-2018年6月门诊收治... 目的探讨血清三碘甲状腺素(T3)、甲状腺素(T4)、促甲状腺素(H-TSH)、游离甲状腺素(FT4)、游离三碘甲状腺原氨酸(FT3)在甲状腺功能亢进(简称甲亢)与甲状腺功能减退(简称甲减)诊断中的应用价值。方法选择我院2017年1月-2018年6月门诊收治且确诊的甲亢患者56例(甲亢组)、甲减患者46例(甲减组)进行研究,同时将同期接待的健康体检者56例作为对照组。纳入对象均以化学发光法测定血清T3、T4、H-TSH、FT3、FT4指标,记录三组前述指标并比较,同时对甲亢组、甲减组前述各指标诊断符合率进行比较,比较软件均为统计学SPSS23.0。结果甲亢组血清T3、T4、FT3、FT4均显著高于对照组,而H-TSH显著低于对照组,两组差异有统计学意义(P<0.05);甲减组血清T3、T4、FT3、FT4均显著低于对照组,而H-TSH显著高于对照组,两组差异有统计学意义(P<0.05)。血清T3、T4、H-TSH、FT3、FT4指标诊断甲亢与甲减符合率均达到70%以上,其中甲亢组FT3与H-TSH诊断符合率最高,而甲减组H-TSH诊断符合率最高。结论血清T3、T4、H-TSH、FT3、FT4指标在甲亢患者与甲减患者中有明显的改变,与正常人群相比差异显著,其中甲亢患者中FT3与H-TSH、T3诊断符合率高,而甲减中H-TSH、FT4、T4诊断符合率高。 展开更多

夏枯草颗粒联合泼尼松治疗亚急性甲状腺炎疗效及对血清T3、T4及TSH水平的影响 被引量:12

目的探讨夏枯草颗粒联合泼尼松治疗亚急性甲状腺炎的疗效及对血清T3、T4及TSH水平影响。方法选取医院收治的96例疗亚急性甲状腺炎患者,随机分为两组各48例。对照组患者给予醋酸泼尼松片,夏枯草组在对照组基础上口服夏枯草颗粒,两组患者... 目的探讨夏枯草颗粒联合泼尼松治疗亚急性甲状腺炎的疗效及对血清T3、T4及TSH水平影响。方法选取医院收治的96例疗亚急性甲状腺炎患者,随机分为两组各48例。对照组患者给予醋酸泼尼松片,夏枯草组在对照组基础上口服夏枯草颗粒,两组患者均持续治疗2个月。对比两组患者的临床疗效、临床症状消失时间、中医证候积分、T3、T4、TSH水平、ESR和CRP水平及TPOAb、TGAb水平。结果治疗后,夏枯草组总有效率较高(P<0.05);夏枯草组临床症状消失时间显著低于对照组(P<0.05);两组体征评分显著降低(P<0.05);并且夏枯草组降低较多(P<0.05);两组患者T3、T4水平显著降低,TSH水平显著升高(P<0.05);并且夏枯草组改善较多(P<0.05);两组ESR和CRP水平显著降低(P<0.05);并且夏枯草组降低程度较大(P<0.05);两组患者治疗前TPOAb、TGAb水平差异无统计学意义(P>0.05),两组患者治疗后TPOAb、TGAb水平明显降低(P<0.05),且夏枯草组TPOAb、TGAb水平低于对照组(P<0.05)。结论采用夏枯草颗粒联合泼尼松治疗亚急性甲状腺炎具有较好的治疗效果,能够改善血清T3、T4及TSH水平,安全性较高,值得在临床上推广应用。 展开更多

目的探讨血清T3/T4值在诊断Graves病甲亢和亚急性甲状腺炎引起的甲状腺毒症中的临床价值。方法回顾性分析2004年8月~2008年10月在新疆医科大学第一附属医院内分泌科就诊的Graves病甲状腺功能亢进症(124例)及亚急性甲状腺炎患者(35例)... 目的探讨血清T3/T4值在诊断Graves病甲亢和亚急性甲状腺炎引起的甲状腺毒症中的临床价值。方法回顾性分析2004年8月~2008年10月在新疆医科大学第一附属医院内分泌科就诊的Graves病甲状腺功能亢进症(124例)及亚急性甲状腺炎患者(35例)的病例资料,对已确诊的患者血清T3/T4的比值进行统计学分析。结果 Graves病甲亢组及亚甲炎组患者血清T3/T4值差异具有统计学意义(P<0.01),Graves病甲亢组T3/T4值高于亚甲炎组。Graves病甲亢组血清FT3/FT4值较高,而亚甲炎组较低,其差异具有统计学意义(P<0.01)。结论在临床工作中可以通过观察血清T3/T4、FT3/FT4值来简单判断甲状腺功能亢进症的不同病因,并给予积极对症治疗。 展开更多

目的观察非气管插管下单体位单孔胸腔镜双侧交感神经T3/T4切断术治疗手足多汗症的效果。方法选取2016年1月至2019年12月入院治疗的70例手足多汗症患者作为研究对象,随机分为两组,各35例。所有患者均给予单体位单孔胸腔镜双侧交感神经T3... 目的观察非气管插管下单体位单孔胸腔镜双侧交感神经T3/T4切断术治疗手足多汗症的效果。方法选取2016年1月至2019年12月入院治疗的70例手足多汗症患者作为研究对象,随机分为两组,各35例。所有患者均给予单体位单孔胸腔镜双侧交感神经T3/T4切断术治疗,其中A组给予喉罩复合全静脉非插管麻醉,B组给予单腔气管插管全身麻醉。比较两组患者手术过程中心率、血压、血氧饱和度指标变化及围手术期结果。结果术后,两组患者双手、双足均变为干燥温暖,有效率100%,两组患者治疗效果、手术时间比较差异无统计学意义;A组麻醉起效时间、进食时间、术后下地活动时间、住院时间等均显著短于B组,术后24 h疼痛评分、术后舒适度评分、住院费用均低于B组(P<0.05)。A组并发症发生率为5.71%(2/35),B组为25.71%(9/35),两组比较差异有统计学意义(P<0.05)。T0时刻,两组HR、MBP、SpO2比较差异无统计学意义,T1~T3时刻,两组患者HR、MBP均高于T0时刻,差异有统计学意义(P<0.05),组间比较差异无统计学意义,两组不同时间点SpO2比较差异无统计学意义。结论非气管插管下单体位单孔胸腔镜双侧交感神经T3/T4切断术有助于促进手足多汗症术后康复,并发症发生率较低,值得临床推广应用。 展开更多

目的:观察食管康颗粒对反流性食管炎(RE)模型大鼠血清T3、T4、FT3、FT4含量的影响。方法:将64只Wistar大鼠,雌雄各半,随机分成4组,即正常对照组(正常组)、食管康颗粒组(食管康组)、西药治疗组(西药组)及模型组,每组16只。采用"食... 目的:观察食管康颗粒对反流性食管炎(RE)模型大鼠血清T3、T4、FT3、FT4含量的影响。方法:将64只Wistar大鼠,雌雄各半,随机分成4组,即正常对照组(正常组)、食管康颗粒组(食管康组)、西药治疗组(西药组)及模型组,每组16只。采用"食管—十二指肠端侧吻合术"制备动物模型。从术后第3天开始,正常对照组和模型组给予生理盐水灌胃,药物治疗组分别给予食管康颗粒和西药灌胃,连续7天,于第8天处死大鼠留取标本,观察食管下段黏膜大体及病理组织学变化,应用放免法测模型大鼠血清T3、T4、FT3、FT4含量。结果:模型组大鼠血清T3、T4、FT3、FT4含量较正常组明显降低,差异具有统计学意义(P<0.05);食管康组及西药组大鼠血清T3、T4、FT3、FT4含量较模型组明显升高,差异具有统计学意义(P<0.05);食管康组与西药组比较,食管康组T4、FT3、FT4含量升高更加显著,差异具有统计学意义(P<0.05);与西药组比较,食管康组T3含量升高不明显,差异无统计学意义(P>0.05)。结论:食管康颗粒能够通过升高反流性食管炎模型大鼠血清中T3、T4、FT3、FT4含量,影响能量代谢,对RE起到治疗作用。 展开更多
