目的:研究血清抗转录中介因子1-γ(transcriptional intermediary factor 1-γ,TIF1-γ)抗体在诊断皮肌炎(dermatomyositis,DM)和临床无肌病性皮肌炎(clinically amyopathic dermatomyositis,CADM)合并恶性肿瘤中的价值。方法:2016年1月... 目的:研究血清抗转录中介因子1-γ(transcriptional intermediary factor 1-γ,TIF1-γ)抗体在诊断皮肌炎(dermatomyositis,DM)和临床无肌病性皮肌炎(clinically amyopathic dermatomyositis,CADM)合并恶性肿瘤中的价值。方法:2016年1月至2019年12月间,收集上海交通大学医学院附属瑞金医院皮肤科病房收治的223例DM和CADM患者[包括209例成人DM(107例)和CADM(102例),5例青少年DM(juvenile DM,JDM)和9例青少年JCADM(juvenile CADM,JCADM)]的临床资料。成人DM和成人CADM分别被分为恶性肿瘤组和无肿瘤组。初诊时血清用酶联免疫吸附测定法检测患者血清抗TIF1-γ抗体,分别比较成人DM和成人CADM患者中恶性肿瘤与无肿瘤患者之间抗TIF1-γ抗体的阳性率,进一步比较抗TIF1-γ抗体阳性与抗体阴性患者之间恶性肿瘤的发生率。结果:①成人DM和CADM中恶性肿瘤共42例(20.1%),占前3位的分别是鼻咽癌、乳腺癌和肺癌。JDM和JCADM患者中无一例发生恶性肿瘤。②成人DM和CADM中有74例(74/209,35.4%)血清抗TIF1-γ抗体阳性,其中DM患者抗TIF1-γ抗体阳性率33.6%(36/107),CADM患者抗TIF1-γ抗体阳性率37.3%(38/102)。5例JDM患者中,无一例抗TIF1-γ抗体阳性;9例JCADM患者中,3例抗TIF1-γ抗体阳性。③成人DM中,恶性肿瘤患者的抗TIF1-γ抗体阳性率显著高于无肿瘤患者[26/35(74.3%)比10/72(13.9%),P<0.0001];抗TIF1-γ抗体阳性患者恶性肿瘤的发生率显著高于阴性患者[26/36(72.2%)比9/71(12.7%),P<0.0001]。成人CADM中,恶性肿瘤患者与无肿瘤患者之间抗TIF1-γ抗体的阳性率差异无统计学意义[4/7比34/95(35.8%),P=0.420];抗TIF1-γ抗体阳性与抗体阴性患者之间恶性肿瘤的发生率差异无统计学意义[4/38(10.5%)比3/64(4.7%),P=0.420]。结论:抗TIF1-γ抗体阳性与成人DM患者恶性肿瘤的发生相关,而与成人CADM患者恶性肿瘤发生无关。抗TIF1-γ抗体可能是成人DM合并恶性肿瘤的血清标志物,对于抗TIF1-γ抗体阳性的成人DM患者需要加强恶性肿瘤筛查。 展开更多

目的 探讨抗黑素瘤分化相关基因5(MDA5)抗体阳性重症皮肌炎患者抗转录中介因子1γ(TIF1γ)抗体状态与临床特点的相关性。方法 选取自2018年1月至2022年1月收治的重症皮肌炎患者62例为研究对象。检测62例重症皮肌炎患者的抗MDA5抗体和抗T... 目的 探讨抗黑素瘤分化相关基因5(MDA5)抗体阳性重症皮肌炎患者抗转录中介因子1γ(TIF1γ)抗体状态与临床特点的相关性。方法 选取自2018年1月至2022年1月收治的重症皮肌炎患者62例为研究对象。检测62例重症皮肌炎患者的抗MDA5抗体和抗TIF1γ抗体状态。分析抗MDA5抗体阳性重症皮肌炎患者抗TIF1γ抗体状态与临床特点的关系。Log-rank检验分析抗MDA5抗体水平与抗TIF1γ抗体状态对预后的影响。以抗MDA5抗体浓度的中位数为阈值,划分为弱抗MDA5抗体水平和强抗MDA5抗体水平,分析不同抗MDA5抗体水平的重症皮肌炎患者中抗TIF1γ抗体状态与临床症状的关系。结果 抗TIF1γ抗体阳性重症皮肌炎患者的铁蛋白水平、人肌酸激酶水平、人乳酸脱氢酶水平、C反应蛋白水平、红细胞沉降率均高于抗TIF1γ抗体阴性重症皮肌炎患者,差异有统计学意义(P<0.05)。在弱抗MDA5抗体水平的重症皮肌炎患者中,抗TIF1γ抗体阳性与阴性患者比较,差异无统计学意义(P>0.05);但在强抗MDA5抗体水平的重症皮肌炎患者中,抗TIF1γ抗体阳性患者显著多于抗TIF1γ抗体阴性患者,差异有统计学意义(P<0.05)。Log-rank检验结果显示,存活率从低到高依次为强抗MDA5抗体阳性+抗TIF1γ抗体阳性[60.0%(15/25)]、强抗MDA5抗体阳性+抗TIF1γ抗体阴性[66.7%(4/6)]、弱抗MDA5抗体阳性+抗TIF1γ抗体阳性[87.5%(14/16)]、弱抗MDA5抗体阳性+抗TIF1γ抗体阴性[93.3%(14/15)],差异有统计学意义(P<0.05)。重症皮肌炎患者临床表现中,抗MDA5抗体阳性伴抗TIF1γ抗体阳性患者皮肤溃疡、声嘶、快速进展性间质性肺病(RP-ILD)所占比例明显高于抗MDA5抗体阳性伴抗TIF1γ抗体阴性患者,差异有统计学意义(P<0.05)。结论 抗TIF1γ抗体在抗MDA5抗体阳性的重症皮肌炎患者中的阳性率较高,且抗MDA5抗体阳性伴抗TIF1γ抗体阳性与皮肤溃疡、声嘶、RP-ILD相关。结合抗MDA5抗体水平和抗TIF1γ抗体状态有助于评估患者预后。 展开更多

目的探讨抗转录中介因子1-γ(TIF1-γ)抗体相关皮肌炎(DM)的临床特征。方法回顾性分析137例DM患者的临床资料,随访时间≥2年。依据是否存在抗TIF1-γ抗体分为阳性组(23例)和阴性组(114例),比较两组临床特征,分析DM患者发生恶性肿瘤的影... 目的探讨抗转录中介因子1-γ(TIF1-γ)抗体相关皮肌炎(DM)的临床特征。方法回顾性分析137例DM患者的临床资料,随访时间≥2年。依据是否存在抗TIF1-γ抗体分为阳性组(23例)和阴性组(114例),比较两组临床特征,分析DM患者发生恶性肿瘤的影响因素。结果与阴性组相比,阳性组诊断时间较长,用力肺活量和一氧化碳弥散量较高,DM相关典型皮疹和恶性肿瘤发生率较高,“技工手”和间质性肺疾病发生率较低(P<0.05或P<0.01)。与阴性组相比,阳性组球蛋白、血清铁蛋白、乳酸脱氢酶、AST、ESR和IgA水平均较低(P<0.05或P<0.01)。多因素logistic回归分析结果显示,抗TIF1-γ抗体阳性[OR=4.301,95%CI(1.182~15.642)]和DM发病年龄较大[OR=1.057,95%CI(1.007~1.110)]是DM患者发生恶性肿瘤的独立危险因素(P<0.05)。结论抗TIF1-γ抗体相关DM患者恶性肿瘤发生率高,对典型皮疹的识别有利于早期诊断该病。 展开更多

AIM:To determine the expression and clinical significance of transcriptional intermediary factor 1 gamma (TIF1γ),Smad4 and transforming growth factor-beta (TGFβR) across a spectrum representing colorectal cancer (CRC) development.METHODS:Tissue microarrays were prepared from archival paraffin embedded tissue,including 51 colorectal carcinomas,25 tubular adenomas (TA) and 26 HPs,each with matched normal colonic epithelium.Immunohistochemistry was performed using antibodies against TIF1γ,Smad4 and TGFβ RⅡ.The levels of expression were scored semi-quantitatively (score 0-3 or loss and retention for Smad4).RESULTS:Overexpression of TIF1γ was detected in 5/26 (19%) HP;however,it was seen in a significantly higher proportion of neoplasms,15/25 (60%) TAs and 24/51 (47%) CRCs (P<0.05).Normal colonic mucosa,HP,and TAs showed strong Smad4 expression,while its expression was absent in 22/51 (43%) CRCs.Over-expression of TGFβ RⅡ was more commonly seen in neoplasms,13/25 (52%) TAs and 29/51 (57%) CRCs compared to 9/26 (35%) HP (P<0.05).Furthermore,there was a correlation between TIF1γ overexpression and Smad4 loss in CRC (Kendall tau rank correlation value=0.35,P<0.05).The levels of TIF1γ overexpression were significantly higher in stage Ⅲ than in stage Ⅰ and Ⅱ CRC (P<0.05).CONCLUSION:The findings suggest that over-expression of TIF1γ occurs in early stages of colorectal carcinogenesis,is inversely related with Smad4 loss,and may be a prognostic indicator for poor outcome. 展开更多

AIM To establish a model to enrich and characterize stemlike cells from murine normal liver and hepatocellular carcinoma(HCC) cell lines and to further investigate stem-like cell association with epithelial-to-mesenchymal transition(EMT).METHODS In this study,we utilized a stem cell conditioned serumfree medium to enrich stem-like cells from mouse HCC and normal liver cell lines,Hepa 1-6 and AML12,respectively.We isolated the 3-dimensional spheres and assessed their stemness characteristics by evaluating theRNA levels of stemness genes and a cell surface stem cell marker by quantitative reverse transcriptase-PCR(q RTPCR).Next,we examined the relationship between stem cells and EMT using q RT-PCR.RESULTS Three-dimensional spheres were enriched by culturing murine HCC and normal hepatocyte cell lines in stem cell conditioned serum-free medium supplemented with epidermal growth factor,basic fibroblast growth factor and heparin sulfate.The 3-dimensional spheres had enhanced stemness markers such as Klf4 and Bmi1 and hepatic cancer stem cell(CSC) marker Cd44 compared to parental cells grown as adherent cultures.We report that epithelial markers E-cadherin and ZO-1 were downregulated,while mesenchymal markers Vimentin and Fibronectin were upregulated in 3-dimensional spheres.The 3-dimensional spheres also exhibited changes in expression of Snai,Zeb and Twist family of EMT transcription factors.CONCLUSION Our novel method successfully enriched stem-like cells which possessed an EMT phenotype.The isolation and characterization of murine hepatic CSCs could establish a precise target for the development of more effective therapies for HCC. 展开更多

Evidence has accumulated to suggest an important role of ethanol and/or its metabolites in the pathogenesis of alcohol-related liver disease. In this review, the fibrogenic effects of ethanol and its metabolites on hepatic stellate cells (HSCs) are discussed. In brief, ethanol interferes with retinoid metabolism and its signaling, induces the release of fibrogenic cytokines such as transforming growth factor β-1 (TGFβ-1) from HSCs, up-regulates the gene expression of collagen I and enhances type I collagen protein production by HSCs. Ethanol further perpetuates an activated HSC phenotype through extracellular matrix remodeling. The underlying pathophysiologic mechanisms by which ethanol exerts these pro-fibrogenic effects on HSCs are reviewed. 展开更多

Transforming growth factor-β1(TGF-β1)acts as a tumor promoter in advanced prostate cancer(PCa).We speculated that microRNAs(miRNAs)that are inhibited by TGF-β1 might exert anti-tumor effects.To assess this,we identified several miRNAs downregulated by TGF-β1 in PCa cell lines and selected miR-3691-3p for detailed analysis as a candidate anti-oncogene miRNA.miR-3691-3p was expressed at significantly lower levels in human PCa tissue compared with paired benign prostatic hyperplasia tissue,and its expression level correlated inversely with aggressive clinical pathological features.Overexpression of miR-3691-3p in PCa cell lines inhibited proliferation,migration,and invasion,and promoted apoptosis.The miR-3691-3p target genes E2F transcription factor 3(E2F3)and PR domain containing 1,with ZNF domain(PRDM1)were upregulated in miR-3691-3p-overexpressing PCa cells,and silencing of E2F3 or PRDM1 suppressed PCa cell proliferation,migration,and invasion.Treatment of mice bearing PCa xenografts with a miR-3691-3p agomir inhibited tumor growth and promoted tumor cell apoptosis.Consistent with the negative regulation of E2F3 and PRDM1 by miR-3691-3p,both proteins were overexpressed in clinical PCa specimens compared with noncancerous prostate tissue.Our results indicate that TGF-β1-regulated miR-3691-3p acts as an anti-oncogene in PCa by downregulating E2F3 and PRDM1.These results provide novel insights into the mechanisms by which TGF-β1 contributes to the progression of PCa. 展开更多

目的探讨肿瘤相关标志物与皮肌炎(dermatomyositis,DM)并发间质性肺病(interstitial lung disease,ILD)的关系,寻找临床中DM合并ILD的血清标志物,帮助判断病情,指导治疗。方法前瞻性纳入59例DM患者,其中合并ILD患者30例,另有5例合并恶... 目的探讨肿瘤相关标志物与皮肌炎(dermatomyositis,DM)并发间质性肺病(interstitial lung disease,ILD)的关系,寻找临床中DM合并ILD的血清标志物,帮助判断病情,指导治疗。方法前瞻性纳入59例DM患者,其中合并ILD患者30例,另有5例合并恶性肿瘤。测定血清转录中介因子1-γ(TIF1-γ)抗体以及肿瘤相关抗原(tumor-associated antigens,TAAs),分析肿瘤相关标志物与DM合并ILD的关系,并采用受试者工作特征(receiver operating characteristic,ROC)曲线分析单指标、多指标联合的诊断价值。结果 59例DM中合并ILD与合并肿瘤相互独立。共11例血清TIF1-γ抗体阳性。在5例合并恶性肿瘤DM患者中,4例血清TIF1-γ抗体阳性,其诊断肿瘤灵敏度为36.36%,特异度为97.92%。而在30例DM合并ILD患者中,仅1例TIF1-γ抗体阳性(P=0.002)。DM合并ILD组中血清糖类抗原125(CA125)(P=0.018)、糖类抗原153(carbohydrate antigen-125,CA153)(P=0.014)、细胞角蛋白19片段(CYFRA21-1)增高(P=0.002),CYFRA21-1诊断ILD的ROC曲线下面积(AUC)为0.745(P=0.002),当最佳诊断点为3.58ng/mL时,血清CYFRA21-1检测DM合并ILD的敏感性和特异性分别为60.71%和84%,CA125(21.5U/mL)与CYFRA21-1(3.58ng/mL)的联合诊断价值最高,AUC达0.801(P<0.001),但诊断点的敏感性和特异性未见明显提高(分别为64.29%和82.61%)。结论在DM合并ILD患者中,肿瘤相关抗原CA125、CA153、CYFRA21-1的血清水平升高提示DM合并ILD情况,尤其是CYFRA21-1,可以考虑作为DM合并ILD的较好的血清学指标。 展开更多

BACKGROUND: Phototropism is the response a plant exhibits when it is faced with a directional blue light stimulus. Though a seemingly simple differential cell elongation response within a responding tissue that results in organ curvature, phototropism is regulated through a complex set of signal perception and transduction events that move from the plasma membrane to the nucleus. In nature phototropism is one of several plant responses that have evolved to optimize photosynthesis and growth. OBJECTIVE: In the present work we will review the state of the field with respect to the molecules and mechanisms associated with phototropism in land plants. METHODS: A systematic literature search was done to identify relevant advances in the field. Though we tried to focus on literature within the past decade (1998-present), we have discussed and cited older literature where appropriate because of context or its significant impact to the present field. Several previous review articles are also cited where appropriate and readers should seek those out. RESULTS: A total of 199 articles are cited that fulfill the criteria listed above. CONCLUSIONS: Though important numerous and significant advances have been made in our understanding of the molecular, biochemical, cell biological and physiologic mechanisms underlying phototropism in land plants over the past decade, there are many remaining unanswered questions. The future is indeed bright for researchers in the field and we look forward to the next decade worth of discoveries. 展开更多
