Mechanism of AiTongXiao granule in the treatment of hepatocellular carcinoma based on network pharmacology and rat transplanted liver cancer model

Objective:To investigate the mechanism of action and material basis of AiTongXiao granule in the treatment of hepatocellular carcinoma(HCC)based on network pharmacology and transplanted liver cancer rat model.Methods:TCMSP database was used to screen out effective components and its corresponding potential pharmaceutical targets,and databases including Gene Cards,OMIM,Drugbank and TTD were further used to collect HCC-related drug targets.The intersecting targets were obtained by mapping the drug and disease targets.The component-targets network was constructed and visualized by Cytoscape 3.8.2 software.Protein-protein interaction(PPI)network was built by STRING online platform,and the topological relationship and core targets was analyzed and screened by using CytoNCA software.In addition,Metascape database was used to perform gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis of the core targets.At last,rat liver transplanted liver cancer model was established by using Walker-256 cell line and treated by AiTongXiao granule for 15 days.Western blot was used to further compare the expression levels of AKT,pAKT,p53,p-p53,ERK1/2 and ERK1/2 in the tumor between treatment group and the control group.Results:257 active components were obtained from AiTongXiao granule,corresponding to 294 drug targets.Meanwhile,233 of the 7993 HCC disease targets were screened out between AiTongXiao granule drug and HCC disease targets.11 core targets including AKT1,IL6,TP53,MAPK3,TNF,JUN,CASP3,MAPK1,MYC,PTGS2,MMP9 were further obtained by median screening.GO and KEGG analysis results showed that these core targets enriched to HBV,TNF and cancer related pathways.The rat transplanted liver cancer model results indicated significant down regulation for AKT,p-AKT,pERK1/2,and significant up regulation of p-p53 after AiTongXiao granule treatment(P<0.05).Conclusion:AiTongXiao granule could act to multiple cancer related pathways,and AKT,p53 and ERK1/2 were validated to be regulated by ATXF in rat model.The mechanism may be through the regulation of the above signaling pathways to exert anti-liver cancer effect. 展开更多

In order to investigate the effects of seed rate on root twining power and seedling quality of machine-transplanted super rice cultivars, Yongyou 12 and Zhongzheyou No.l, total 6 seed rates were arranged, and the growth characteris- tics, quality and root twining power of machine-transplanted rice seedlings were studied. The results showed that seed rate had great effects on root twining power and qualityof rice seedlings; with the increase of seed rate, the root twining power of rice seedlings was increased, but the quality of rice seedlings was significantly reduced; under the condition of high seed rate, rice seedlings were weak with less accumulate dry matter and nonuniform growth, and the proportion of weak seedlings was higher. When the seed rate of both rice cultivars was reduced by 10 g/tray, the average seedling height was increased by 0.5 cm, leaf age was increased by 0.1, stem base diameter was increased by 0.1 cm, and root number per plant was increased by 0.3. When the seed rate was increased by 10 g/tray, the accumulated dry matter per plant was reduced by 5.1 mg. With the increase of seed rate, the seedling quality of Zhongzheyou No.1 was decreased rapidly, and the proportion of weak seedlings was increased rapidly. However, Yongyou 12 has a larger elasticity in seed rate. When the seed rate reached to 50 g/tray, blanket seedlings of Yongy- ou 12 were nursed with uniform growth and higher quality, but the shoots were sparse with lower leaf area index. However, when the seed rate was up to 50 g/tray, blanket seedlings of Zhongzheyou No.1 were nursed with uniform growth and higher quality, as well as higher-density shoots. From a comprehensive point of view, the optimum seed rates of Yongyou 12 and Zhongzheyou No.1 were 65 and 50 g/tray, respectively. 展开更多

BACKGROUND: Non-operative therapy takes an impor- tant position in comprehensive therapy of liver cancer. De- spite some effects by using ethanol, acetic acid and heat sa- line for intra-tumor injection in the treatment of liver canc- er, it is difficult to attain a complete cure but bring about injury to the liver to some extent. Hence, searching for other drugs for the local treatment of liver tumor is an im- portant option. This study was designed to set up rat mo- dels of transplanted liver cancer, intra-tumor injection of Kang-Lai-Te (KLT), and negative control (saline) and positive control (ethanol). The effect of intra-tumor injec- tion of KLT in treating transplanted hepatoma in rats and its advantages and disadvantages were assessed and the pos- sibility of its use in treating patients with liver cancer was evaluated. METHODS: Forty rats were divided into 4 groups ( G1, G2, G3 and G4, 10 rats in each group). Different drugs were injected into their implanted hepatoma (G1 with 0.2 ml saline as control, G2 with 10 mg KLT, G3 with 20 mg KLT, G4 with 0.2 ml ethanol). After 3 and 8 days, the hepa- toma volume (HV), the serum levels of albumin, alanine aminotransferase(ALT), aspartate aminotransferase alkaline phosphatase( ALP) and creatinine, as well as the expression of proliferation cell nuclear antigen (PCNA) in hepatoma were detected. RESULTS: After 3 days, the HVs were smaller in G3 and G4 than in G1 (P <0.05), the serum levels of albumin were higher in G2 and G3 than in Gl and G4 (P <0.05), the se- rum levels of ALT and AST were lower in G2 and G3 than in G4 (P<0.05), the serum levels of ALP was lower in G2 and G3 than in Gl and G4 (P <0. 05), the PCNA labeling indexes (PCNA LI) were lower in G2 and G3 than in Gl and GA (P <0.05). After 8 days, the HVs were smaller in G2, G3 and G4 than in Gl (P <0.05), and the differences of HVs among G2, G3 and G4 were not significant. The serum levels of ALP were lower in G1, G2 and G3 than in G4 (P <0.05), and the PCNA LI were lower in G3 than in Gl andG4 (P<0.05). CONCLUSION: Intra-tumor injection of KLT into implan- ted hepatoma is evidently effective, but it is less effective than ethanol. The effect of KLT on liver function is markedly lower than that of ethanol. 展开更多

Objective:To study the regulating effect of HSP70 inhibitor(PES) combined with cisplatin on cervical cancer proliferation in vitro and transplanted tumor growth.Methods:Cervical cancer Hela cell lines were cultured and divided into control group,cisplatin group,PES group and cisplatin+PES group that were treated with serum-free DMEM,cisplatin with final concentration of 10 μmol/L,PES 20 μmol/L and cisplatin 10 μmol/L combined with PES with 20 μmol/L,respectively;animal models with cervical cancer xenografts were established and divided into control group,cisplatin group,PES group and cisplatin+PES group who received intra-tumor injection of normal saline,10 μmol/L cisplatin,20 μmol/L PES as well as 10 μmol/L cisplatin+20 μmol/L PES,respectively.Cell proliferation activity,transplanted tumor volume and mitochondria apoptosis molecule expression were detected.Results:Cell viability value and Bcl-2 mRNA expression in cells of cisplatin group,PES group and cisplatin+PES group were significantly lower than those of control group while Bax,Caspase-3 and Caspase-9 mRNA expression in cells were significantly higher than those of control group;transplanted tumor volume and the Bcl-2 mRNA expression in transplanted tumor tissue of cisplatin group,PES group and cisplatin+PES group were significantly lower than those of control group while Bax,Caspase-3 and Caspase-9 m RNA expression in transplanted tumor tissue were significantly higher than those of control group.Conclusions:HSP70 inhibitor combined with cislatin can inhibit cervical cancer cell proliferation in vitro and transplanted tumor growth through mitochondrial apoptosis pathway. 展开更多

With an equivalent yield, dry seeded flooded rice (DSR) has been considered as a replacement for traditional transplanted flooded rice (TFR). However, the differences in leaf and root senescence during grain filling between DSR and TFR were seldom identified. In this study, the root length, root tip number and leaf senescence of rice varieties Huanghuazhan and Yangliangyou 6 during ripening were compared between DSR and TFR. Results showed that top three leaves in DSR had the characteristics of relatively lower SPAD value, lower N content and premature leaf senescence. In addition, both the total root length and total root tip number of DSR were significantly lower than those of TFR. In conclusion, premature and quick leaf senescence was related with inadequate root length and root tip number during ripening, which might result from the deficiency of nitrogen supply in DSR. Techniques on improving leaf nitrogen status and delaying the leaf senescence during grain-filling in DSR should be developed in future researches. 展开更多

Rhodiola sachalinensis growing in the different habitats, Xiaotianchi plot (altitude 1800 m), Tree line plot (altitude 2000 m) and Tianwenfeng plot (altitude 2325 m), of Changbai Moutain (northern slop) were transplanted to Harbin Experimental Forest Farm for measuring its characteristic of gas exchange. The study results indicated that the growth state and gas exchange characteristics ofRh. sachalinensis growing in different habitats varied markedly. The plants transplanted from Tianwenfeng plot had the highest values in net photosynthesis rate (Pn) and transpiration rate (Tr), those transplanted from Tree line plot shows the second, and those transplanted rom Xiaotianchi plot had the lowest values. The variance existed in transplanted plants was the same as shown in the field. From the result it can be extrapolated that the difference of Pn and Tr existed inRh. sachalinensis transplanted from different habitats is depend not only on the environmental factors, but on the variance of physiological characteristic of plant itself. 展开更多

The number of patients reinitiating dialysis after a failed transplant increases over time and has more than doubled between the year 1988 and 2010 (an increase from 2463 to 5588). More importantly, patients returning to dialysis have been shown to have a greater thanthree-fold increase in the annual adjusted mortality rates compared with those with a functioning graft. Continuation of immunosuppression to preserve residual graft function has been implicated to be a contributing factor, seemingly due to immunosuppression-ass-ociated cardiovascular and infectious complications and malignancy risk, among others. Nonetheless, maintenance low-dose immunosuppression has been suggested to confer survival beneft in patients returning to peritoneal dialysis. Whether early vs late reinitiation of dialysis or whether transplantectomy has an impact on patient survival remains poorly defined. Consensus guidelines for the management of a failed allograft are lacking. In this article, we present a literature overview on the ideal timing of dialysis reinitiation after graft loss, the management of immunosuppression after graft failure, and the risks and benefits of transplantectomy. The authors’ perspectives on the management of this special patient population are also discussed. 展开更多

Objectives To trace and evaluate intracoronary transplanted mesenchymal stem cells(MSCs) labeled with superparamagnetic iron oxide(SPIO) by using magnetic resonance imaging(MRI) in a swine model of myocardial infarction (MI).Methods MSCs were transfected with a lentiviral vector carrying the gene encoding green fluorescent protein (GFP) and labeled in vitro with SPIO.Two weeks after MI, swine were randomized to intracoronary transplantation of dual -labeled MSCs(n = 10),MSCs-GFP(n = 10) and saline(n = 5).MRI examination was performed with a 1.5T clinical scanner at 24 hours,3 weeks and 8 weeks after cells transplantation. Signal intensity(SI) changes,cardiac function and MI size were measured using MRI.Correlation between MR findings and histomorphologic findings was also investigated. Results The labeling efficiency at a combination of 25μg Fe/ml SPIO and 0.8 pi/ml Lipofectamine 2000 reached 100%.SPIO labeling did not affect GFP fluorescence and dual-labeling did not affect cell proliferation(P】0.05). Multipotentiality was not affected especially for cardiomyocyte-like cells differentiation.Cardiac cell marker of a-MHC and actinin were positively expressed by immunofluorescence staining after induction.SI on T2 * WI decreased substantial- ly in the interventricular septum 24 hours after injection of MSCs.The intensity of hypo-intense signals appeared to increase throughout the later time points.Changes in SI at 24 hours,3 weeks and 8 weeks were 52.98%±10.74%,21.53%±5.40%and 6.23%±2.01%,respectively(P【0.01).DE-MRI demonstrated both dual-labeled MSCSs and MSCs-GFP could dramatically reduce the size of MI and improve cardiac function. Histological data revealed that prussian blue stain-positive cells were found mainly in the border zone which also showed green fluorescence but negative for macrophage marker(CD68).Gross pathologic examination revealed that engrafted MSCs dramatically reduce the extent of necrotic myocardium and promote the regeneration of new,contractile myocardium along the subendocardial surface of the MI. Conclusions MSCs could be efficiently and safely labeled with SPIO and GFP,and could be detected reproducibly and noninvasively in vivo using cardiac MRI.Intracoronary transplantation of dual-labeled MSCs could increase cardiac function and reduce the size of MI. 展开更多

Autologous submandibular gland(SMG) transplantation has been proved to ameliorate the discomforts in patients with severe keratoconjunctivitis sicca. The transplanted glands underwent a hypofunctional period and then restored secretion spontaneously.This study aims to investigate whether autonomic nerves reinnervate the grafts and contribute to the functional recovery, and further determine the origin of these nerves. Parts of the transplanted SMGs were collected from the epiphora patients, and a rabbit SMG transplantation model was established to fulfill the serial observation on the transplanted glands with time. The results showed that autonomic nerves distributed in the transplanted SMGs and parasympathetic ganglionic cells were observed in the stroma of the glands. Low-dense and unevenly distributed cholinergic axons, severe acinar atrophy and fibrosis were visible in the patients' glands 4–6 months post-transplantation, whereas the cholinergic axon density and acinar area were increased with time. The acinar area or the secretory flow rate of the transplanted glands was statistically correlated with the cholinergic axon density in the rabbit model, respectively. Meanwhile, large cholinergic nerve trunks were found to locate in the temporal fascia lower to the gland, and sympathetic plexus concomitant with the arteries was observed both in the adjacent fascia and in the stroma of the glands. In summary, the transplanted SMGs are reinnervated by autonomic nerves and the cholinergic nerves play a role in the morphological and functional restoration of the glands. Moreover, these autonomic nerves might originate from the auriculotemporal nerve and the sympathetic plexus around the supplying arteries. 展开更多

However, under the condition of mechanical transplanting, the optimal application time of tillering fertilizer is still unclear. In this study, the early rice cultivar Zhongjiazao 17 and late rice cultivar H You 518 were used as materials, and the effects of different application times of tillering fertilizer on yield and population development of double-cropping rice transplanted by machine were investigated. The tillering fertilizer was applied 7(D07), 10(D10) and 13(D13) d after the transplanting, respectively. The results showed that compared with those in the D07 treatment groups, the yield of early rice in the D10 and D13 treatment groups were reduced by 9.4% and 3.8%, respectively, and the yield of late rice in the D10 and D13 treatment groups were reduced by 4.5% and 12.6%,respectively. However, there were no significant differences in rice yield among the treatment groups. The application time of tillering fertilizer showed significant effects on grain number per panicle and seed setting rate of early rice. The grain number per panicle in the D10 treatment group was significantly lower than that in the D07 treatment group, and the seed setting rate of the D13 treatment group was significantly lower than that in the D07 treatment group(P〈0.05). For the late rice, the effect of application time of tillering fertilizer on effective tiller number was most obvious, and the effective tiller number in the D13 treatment group was significantly lower than that in the D07 treatment group(P〈0.05). Compared with those in the D07 treatment group, the effective tiller numbers, leaf area indexes and biomasses in the D10 and D13 treatment groups were all trended to be decreased. Therefore,to improve the quality of population and fulfill the high-yielding potential of double-cropping rice transplanted by machine, the tillering fertilizer should be applied as early as possible after rice seedlings turn green. 展开更多

Transplanted lung cancer models are of special interest and are widely used today. Such models are essential tools in accelerating development of new therapies for lung cancer. Four representative cultivars in Huaibei area were selected as test materials. By setting potted seedlings and carpet seedlings, the effects of different nursery meth- ods on the photosynthesis of rice at different growth stages in Huaibei area were investigated. Compared with carpet seedlings, the leaf area index of potted seedlings decreased at the maturation stage, but the dry weight of leaf shewed no significant difference.The SPAD of potted seedlings had an increasing trend after transplanting, but the SPAD increased differently according to cultivars. The net photosynthetic rate, stomatal conductance and transpiration rate of the leaves of pot seeding rice were significantly higher than those of carpet seedling rice on the 85^th d after transplanting. However, theintracellular CO2 concentration and water use efficiency of leaves had no significant differences between different 'treatments. The results indicate that the photosynthetic capacity of flag leaves of pot seedling rice is stronger in early and middle stages, but the decay rate of photosynthetic function is slightly faster than carpet seedling rice, which might be the main reason for the rice yield of potted seedlings having no remarkable difference from carpet seedlings. 展开更多

Six rice cultivars viz. PR120, PR116, Feng Ai Zan, PR115, PAU201 and Punjab Mehak 1 under the direct-seeded and transplanted conditions were used to investigate the involvement of antioxidative defence system in relation to polyamine catabolism in temporal regulation of developing grains. Activities of ascorbate peroxidase (APx), guaiacol peroxidase (GPx), catalase (CAT), superoxide dismutase (SOD), polyamine oxidases (PAO) and contents of ascorbate, a-tocopherol, proline and polyamines increased gradually until mid-milky stage and then declined towards maturity stage under both planting conditions. The transplanted condition led to higher activities of antioxidative enzymes (APx, GPx and CAT) and contents of ascorbate, a-tocopherol and proline whereas the direct-seeded condition had elevated levels of PAO and SOD activities and contents of polyamines, lipid peroxide and hydrogen peroxide. Cultivars Feng Ai Zan and PR120 exhibited superior tolerance over other cultivars by accumulating higher contents of ascorbate, a-tocopherol and proline with increasing level of PAO and SOD activities under the direct-seeded condition. However, under the transplanted condition PR116 and PAU201 showed higher activities of antioxidative enzymes with decreasing content of lipid peroxide. Therefore, we concluded that under the direct-seeded condition, enhancements of polyamines content and PAO activity enabled rice cultivars more tolerant to oxidative stress, while under the transplanted condition, antioxidative defence with decreasing of lipid peroxide content was closely associated with the protection of grains by maintaining membrane integrity during rice grain filling. The results indicated that temporal dynamics of H2O2 metabolic machinery was strongly up-regulated especially at the mid-milky stage. 展开更多

In this study, the mechanism by which Ad-p27mt inhibits the growth, invasion and metastasis of transplanted liver tumor was studied by examining the effects of Ad-27mt gene transfer on the expression of Bax, Bcl-2, VEGF and MMP-9 in the transplanted liver tumors in nude mice.The model of transplanted hepatic tumor was established in nude mice.The mice were then divided into three groups, which were injected with PBS, Ad-LacZ and Ad-p27mt and the growth of the transplanted liver tumor was observed.The expressions of P27, Bax and Bcl-2 proteins were detected by Western blotting and the expressions of VEGF and MMP-9 were immunohistochemically determined.Our result showed that the tumor size, expressions of Bax, Bcl-2 proteins, VEGF and MMP-9 were all lower than those in PBS and Ad-LacZ groups and the differences were statistically significant (P【0.05).Our study suggested that Ad-p27mt could inhibit the growth, invasion and metastasis of hepatic cancer by lowering the expressions of VEGF and MMP-9. 展开更多

Two kinds of renal scintigraphic characteristics were identified in recipients with supravesical obstruction of the graft. First, the regular type was characterized by radioactivity defect area in kidney parenchyma during early uptake period followed by ureteropelvic retention. Second, the tubular type was typified by cortical retention and attenuation in collecting system during the whole test period with a special sign of “hollow kidney”. Non obstructive dilated calyces showed similar signs as the regular type. Acute rejection reaction and tubule necrosis demonstrated obstructive time activity curves. However, the radioactivity retention appeared in cortex. It was suggested that dilated calyces and obstructive renogram might not be reliable evidence for upper urinary tract obstruction. The signs of radioactivity attenuation in kidney parenchyma during early uptake period followed by ureteropelvic retention may be more valuable for the evaluation. As for tubular obstruction, specified “hollow kidney” was the characteristic sign which is helpful for the diagnosis. 展开更多

The Influence of Different Single Radiation Dose on Delayed Growth of Transplanted Tumor in Athymic Mouse

OBJECTIVE To reveal the biological effects and effective dosage in radiotherapy model which applies high single-dose irradiation by animal experiment. METHODS We inoculated subcutaneouly human pancreatic carcinoma cell line (MIA PaCa-2) in the lateral of the right lower extremity of the athymic mouse to grow transplantation tumor. While the median diameter of transplantation tumor reached 10 mm approximately, the animals were randomly divided into 7 groups (6 animals per group) and irradiation by different dose in one fixed with consciousness for fraction (0, 2, 5, 10, 17, 25, 35 Gy). All were kept on to be bred for observation of the change in gross tumor volume, calculation of delayed growth time and delayed growth curve. RESULTS With increased dose per fraction, cutaneous reaction on the neoplasma surface of the animal, which was mainly moist yellow effusion was more and more severe. When dosage is less than 10 Gy, all animals showed similar effects, that's the delayed tumor growth was not obvious. Tumors receiving more than 10 Gy in one fraction showed very good biological effect and the delayed tumor growth was obviously related to dosage. The difference in delayed tumor growth between the 2 groups was statistically significant. The delayed tumor growth time in 10, 17, 25 Gy group was respectively 3 weeks, 6 weeks and more. CONCLUSION The biological effect of the model which applies high single-dose irradiation (more than 10 Gy in one fraction) was very good. The effect of delayed tumor growth was obviously related to the dosage after transplantation tumor was radiated. Because of its higher dose per fraction and biological effects, the model of high single-dose irradiation can get better clinical effects. 展开更多
