Increased excitatory amino acid transporter 2 levels in basolateral amygdala astrocytes mediate chronic stress–induced anxiety-like behavior

The conventional perception of astrocytes as mere supportive cells within the brain has recently been called into question by empirical evidence, which has revealed their active involvement in regulating brain function and encoding behaviors associated with emotions.Specifically, astrocytes in the basolateral amygdala have been found to play a role in the modulation of anxiety-like behaviors triggered by chronic stress. Nevertheless, the precise molecular mechanisms by which basolateral amygdala astrocytes regulate chronic stress–induced anxiety-like behaviors remain to be fully elucidated. In this study, we found that in a mouse model of anxiety triggered by unpredictable chronic mild stress, the expression of excitatory amino acid transporter 2 was upregulated in the basolateral amygdala. Interestingly, our findings indicate that the targeted knockdown of excitatory amino acid transporter 2 specifically within the basolateral amygdala astrocytes was able to rescue the anxiety-like behavior in mice subjected to stress. Furthermore, we found that the overexpression of excitatory amino acid transporter 2 in the basolateral amygdala, whether achieved through intracranial administration of excitatory amino acid transporter 2agonists or through injection of excitatory amino acid transporter 2-overexpressing viruses with GfaABC1D promoters, evoked anxiety-like behavior in mice. Our single-nucleus RNA sequencing analysis further confirmed that chronic stress induced an upregulation of excitatory amino acid transporter 2 specifically in astrocytes in the basolateral amygdala. Moreover, through in vivo calcium signal recordings, we found that the frequency of calcium activity in the basolateral amygdala of mice subjected to chronic stress was higher compared with normal mice.After knocking down the expression of excitatory amino acid transporter 2 in the basolateral amygdala, the frequency of calcium activity was not significantly increased, and anxiety-like behavior was obviously mitigated. Additionally, administration of an excitatory amino acid transporter 2 inhibitor in the basolateral amygdala yielded a notable reduction in anxiety level among mice subjected to stress. These results suggest that basolateral amygdala astrocytic excitatory amino acid transporter 2 plays a role in in the regulation of unpredictable chronic mild stress-induced anxiety-like behavior by impacting the activity of local glutamatergic neurons, and targeting excitatory amino acid transporter 2 in the basolateral amygdala holds therapeutic promise for addressing anxiety disorders. 展开更多

The Janus fabrics designed for personal moisture/thermal regulation have garnered significant attention for their potential to enhance human comfort.However,the development of smart and dynamic fabrics capable of managing personal moisture/thermal comfort in response to changing external environments remains a challenge.Herein,a smart cellulose-based Janus fabric was designed to dynamically manage personal moisture/heat.The cotton fabric was grafted with N-isopropylacrylamide to construct a temperature-stimulated transport channel.Subsequently,hydrophobic ethyl cellulose and hydrophilic cellulose nanofiber were sprayed on the bottom and top sides of the fabric to obtain wettability gradient.The fabric exhibits anti-gravity directional liquid transportation from hydrophobic side to hydrophilic side,and can dynamically and continuously control the transportation time in a wide range of 3–66 s as the temperature increases from 10 to 40℃.This smart fabric can quickly dissipate heat at high temperatures,while at low temperatures,it can slow down the heat dissipation rate and prevent the human from becoming too cold.In addition,the fabric has UV shielding and photodynamic antibacterial properties through depositing graphitic carbon nitride nanosheets on the hydrophilic side.This smart fabric offers an innovative approach to maximizing personal comfort in environments with significant temperature variations. 展开更多

BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)is an aggressive lethal malignancy with limited options for treatment and a 5-year survival rate of 11%in the United States.As for other types of tumors,such as colorectal cancer,aberrant de novo lipid synthesis and reprogrammed lipid metabolism have been suggested to be associated with PDAC development and progression.AIM To identify the possible involvement of lipid metabolism in PDAC by analyzing in tumoral and non-tumoral tissues the expression level of the most relevant genes involved in the long-chain fatty acid(FA)import into cell.METHODS A gene expression analysis of FASN,CD36,SLC27A1,SLC27A2,SLC27A3,SLC27A4,SLC27A5,ACSL1,and ACSL3 was performed by qRT-PCR in 24 tumoral PDAC tissues and 11 samples from non-tumoral pancreatic tissues obtained via fine needle aspiration or via surgical resection.The genes were considered significantly dysregulated between the groups when the p value was<0.05 and the fold change(FC)was≤0.5 and≥2.RESULTS We found that three FA transporters and two long-chain acyl-CoA synthetases genes were significantly upregulated in the PDAC tissue compared to the non-tumoral tissue:SLC27A2(FC=5.66;P=0.033),SLC27A3(FC=2.68;P=0.040),SLC27A4(FC=3.13;P=0.033),ACSL1(FC=4.10;P<0.001),and ACSL3(FC=2.67;P=0.012).We further investigated any possible association between the levels of the analyzed mRNAs and the specific characteristics of the tumors,including the anatomic location,the lymph node involvement,and the presence of metastasis.A significant difference in the expression of SLC27A3(FC=3.28;P=0.040)was found comparing patients with and without lymph nodes involvement with an overexpression of this transcript in 17 patients presenting tumoral cells in the lymph nodes.CONCLUSION Despite the low number of patients analyzed,these preliminary results seem to be promising.Addressing lipid metabolism through a broad strategy could be a beneficial way to treat this malignancy.Future in vitro and in vivo studies on these genes may offer important insights into the mechanisms linking PDAC with the long-chain FA import pathway. 展开更多

In addition to the loss of motor function,~60% of patients develop pain after spinal cord injury.The cellular-molecular mechanisms are not well understood,but the data suggests that plasticity within the rostral,epicenter,and caudal penumbra of the injury site initiates a cellularmolecular interplay that acts as a rewiring mechanism leading to central neuropathic pain.Sprouting can lead to the formation of new connections triggering abnormal sensory transmission.The excitatory glutamate transporters are responsible for the reuptake of extracellular glutamate which makes them a critical target to prevent neuronal hyperexcitability and excitotoxicity.Our previous studies showed a sexually dimorphic therapeutic window for spinal cord injury after treatment with the selective estrogen receptor modulator tamoxifen.In this study,we investigated the anti-allodynic effects of tamoxifen in male and female rats with spinal cord injury.We hypothesized that tamoxifen exerts anti-allodynic effects by increasing the expression of glutamate transporters,leading to reduced hyperexcitability of the secondary neuron or by decreasing aberrant sprouting.Male and female rats received a moderate contusion to the thoracic spinal cord followed by subcutaneous slow-release treatment of tamoxifen or matrix pellets as a control(placebo).We used von Frey monofilaments and the“up-down method”to evaluate mechanical allodynia.Tamoxifen treatment decreased allodynia only in female rats with spinal cord injury revealing a sexdependent effect.The expression profile of glutamatergic transporters(excitatory amino acid transporter 1/glutamate aspartate transporter and excitatory amino acid transporter 2/glutamate transporter-1)revealed a sexual dimorphism in the rostral,epicenter,and caudal areas of the spinal cord with a pattern of expression primarily on astrocytes.Female rodents showed a significantly higher level of excitatory amino acid transporter-1 expression while male rodents showed increased excitatory amino acid transporter-2 expression compared with female rodents.Analyses of peptidergic(calcitonin gene-related peptide-α)and non-peptidergic(isolectin B4)fibers outgrowth in the dorsal horn after spinal cord injury showed an increased calcitonin gene-related peptide-α/isolectin B4 ratio in comparison with sham,suggesting increased receptive fields in the dorsal horn.Although the behavioral assay shows decreased allodynia in tamoxifen-treated female rats,this was not associated with overexpression of glutamate transporters or alterations in the dorsal horn laminae fibers at 28 days post-injury.Our findings provide new evidence of the sexually dimorphic expression of glutamate transporters in the spinal cord.The dimorphic expression revealed in this study provides a therapeutic opportunity for treating chronic pain,an area with a critical need for treatment. 展开更多

The unique survey in December 1998 mapped the entire western boundary area of the South China Sea(SCS),which reveals the three-dimensional structure and huge volume transport of the swift and narrow winter western boundary current of the SCS(SCSwwbc) in full scale. The current is found to flow all the way from the shelf edge off Hong Kong to the Sunda Shelf with a width around 100 km and a vertical scale of about 400 m. It appears to be the strongest off the Indo-China Peninsula, where its volume transport reached over 20×10~6 m^3/s. The current is weaker upstream in the northern SCS to the west of Hong Kong. A Kuroshio loop or detached eddy intruded through the Luzon Strait is observed farther east where the SCSwwbc no more exists. The results suggest that during the survey the SCSwwbc was fed primarily by the interior recirculation of the SCS rather than by the"branching" of the Kuroshio from the Luzon Strait as indicated by surface drifters, which is likely a near-surface phenomenon and only contributes a minor part to the total transport of the SCSwwbc. A global transport model is proposed in which a multimoment constrained finite volume (MCV) scheme is applied to a Yin-Yang overset grid. The MCV scheme defines 16 degrees of freedom (DOFs) within each element to build a 2D cubic reconstruction polynomial. The time evolution equations for DOFs are derived from constraint conditions on moments of line-integrated averages (LIA), point values (PV), and values of first-order derivatives (DV). The Yin-Yang grid eliminates polar singularities and results in a quasi-uniform mesh. A limiting projection is designed to remove nonphysical oscillations around discontinuities. Our model was tested against widely used benchmarks; the competitive results reveal that the model is accurate and promising for developing general circulation models. 展开更多

In order to accurately simulate strong three-dimensional (3-D) free surface flows and sediment transport, the fully 3- D non-hydrostatic pressure models are developed based on the incompressible Navier-Stokes equations and convection-diffusion equation of sediment concentration with the mixing triangle and quadrilateral grids. The governing equations are discretized with the unstructured finite volume method in order to provide conservation properties of mass and momentum, and flexibility with practical application. It is shown that it is first-order accurate on nonuniform plane two-dimensional (2-D) grids and second-order accurate on uniform plane grids. A third-order approximation of the vertical velocity at the top-layer is applied. In such a way, free surface zero stress boundary condition is satisfied maturely, and very few vertical layers are needed to give an accurate solution even for complex discontinuous flow and short wave simulation. The model is applied to four examples to simulate strong 3-D free surface flows and sediment transport where non-hydrostatic pressures have a considerable effect on the velocity field. The newly developed model is verified against analytical solutions with an excellent agreement. 展开更多

Northward infl ow through the East Taiwan Channel is vital in modulation of water exchange processes off northeastern Taiwan,China.In addition to the eff ects of the Kuroshio Current and westward-propagating oceanic mesoscale eddies,the seas off eastern Taiwan,China,are frequently infl uenced by typhoons.Focusing on extrema of East Taiwan Channel volume transport(ETCVT)that appear within days of typhoon infl uence,this study investigated 124 historical cyclones including 91 typhoons that passed over the study sea area off eastern Taiwan,China.Based on 25-year HYbrid Coordinate Ocean Model(HYCOM)data,71%of short-term(within 10 d)ETCVT absolute values with>5 Sv occurred under the infl uence of typhoons crossing the study sea area,and the maximum short-term ETCVT extrema induced by typhoons were 12.5 and-10.9 Sv.The ETCVT extrema induced by typhoons showed reasonable positive correlation with typhoon wind speed.More importantly,the ETCVT extrema diff ered in response to diff erent typhoon tracks.Three types of typhoon were identifi ed based on their track and impact on ETCVT.Representative typhoon cases were examined to elucidate the specifi cs of each typical response.Based on historical best track data and HYCOM data,it was established that Type I typhoons caused the ETCVT to exhibit a negative extremum followed by a positive extremum.All Type I,II,and III typhoons tended to result in typical ETCVT fl uctuations in the surface mixing layer above the depth of 50-100 m,while Type II typhoons were more likely to induce ETCVT fl uctuations in the subsurface layer.The fi ndings of this study enhance understanding of ETCVT extrema that occur following typhoon passage,which is valuable for short-term physical-biogeochemical studies both in the study region and in areas downstream owing to the large net volume transport changes induced by typhoons. 展开更多

The structure of current speed and the variability of volume transports of the Kuroshio in the Tokara-kaikyo and Osumi-kaikyo are discussed on the basis of data of KER in the period from 1977 to 1984. The average geostrophic transport through these two straits is estimated to be 24. 5×106 m3/s and only 1/12 of the transport is through the Osumi-kaiky5. Countercurrents on both sides of the Kuroshio trunk are observed in the Tokara-kaikyo. Calculation indicates that the average geostrophic current speed is less than the GEK current speed, systematically. On the basis of the current measurements, the northward transports through the Taiwan Strait in winter and summer are estimated to be 1. 05×106and 3. 16×106m3/s, respectively. From Chu's data (1976) the average transport of the Kuroshio flowing into the East China Sea passing through the passage east of Taiwan is about 29. 3×106m3/s. From Miita and Ogawa's data (1984) the average transport through the Tsushima-kaikyo is 3. 6×106m3/s. Thus the volume transports through the above four straits are roughly in balance, the total outflowing transport is slightly larger than the total inflowing transport. BACKGROUND Sodium-dependent glucose transporter 2 inhibitors(SGLT2i)have shown efficacy in reducing heart failure(HF)burden in a very heterogeneous groups of patients,raising doubts about some contemporary assumptions of their mechanism of action.We previously published a prospective observational study that evaluated mechanisms of action of SGLT2i in patients with type 2 diabetes who were in HF stages A and B on dual hypoglycemic therapy.Two groups of patients were included in the study:the ones receiving SGLT2i as an add-on agent to metformin and the others on dipeptidyl peptidase-4 inhibitors as an add-on to metformin due to suboptimal glycemic control.AIM To evaluate the outcomes regarding natriuretic peptide,oxidative stress,inflammation,blood pressure,heart rate,cardiac function,and body weight.METHODS The study outcomes were examined by dividing each treatment arm into two subgroups according to baseline parameters of global longitudinal strain(GLS),N-terminal pro-brain natriuretic peptide,myeloperoxidase(MPO),high-sensitivity C-reactive protein(hsCRP),and systolic and diastolic blood pressure.To evaluate the possible predictors of observed changes in the SGLT2i arm during follow-up,a rise in stroke volume index,body mass index(BMI)decrease,and lack of heart rate increase,linear regression analysis was performed.RESULTS There was a greater reduction of MPO,hsCRP,GLS,and blood pressure in the groups with higher baseline values of mentioned parameters irrespective of the therapeutic arm after 6 months of follow-up.Significant independent predictors of heart rate decrease were a reduction in early mitral inflow velocity to early diastolic mitral annular velocity at the interventricular septal annulus ratio and BMI,while the predictor of stroke volume index increase was SGLT2i therapy itself.CONCLUSION SGLT2i affect body composition,reduce cardiac load,improve diastolic/systolic function,and attenuate the sympathetic response.Glycemic control contributes to the improvement of heart function,blood pressure control,oxidative stress,and reduction in inflammation. 展开更多

With the depletion of fossil fuels and the demand for high-performance energy storage devices,solidstate lithium metal batteries have received widespread attention due to their high energy density and safety advantages.Among them,the earliest developed organic solid-state polymer electrolyte has a promising future due to its advantages such as good mechanical flexibility,but its poor ion transport performance dramatically limits its performance improvement.Therefore,single-ion conducting polymer electrolytes(SICPEs)with high lithium-ion transport number,capable of improving the concentration polarization and inhibiting the growth of lithium dendrites,have been proposed,which provide a new direction for the further development of high-performance organic polymer electrolytes.In view of this,lithium ions transport mechanisms and design principles in SICPEs are summarized and discussed in this paper.The modification principles currently used can be categorized into the following three types:enhancement of lithium salt anion-polymer interactions,weakening of lithium salt anion-cation interactions,and modulation of lithium ion-polymer interactions.In addition,the advances in single-ion conductors of conventional and novel polymer electrolytes are summarized,and several typical highperformance single-ion conductors are enumerated and analyzed in what way they improve ionic conductivity,lithium ions mobility,and the ability to inhibit lithium dendrites.Finally,the advantages and design methodology of SICPEs are summarized again and the future directions are outlined. 展开更多

The annual mean volume and heat transport sketches through the inter-basin passages and transoceanic sections have been constructed based on 1400-year spin up results of the MOM4p 1. The spin up starts from a state of rest, driven by the monthly climatological mean force from the NOAAWorld Ocean Atlas (1994). The volume transport sketch reveals the northward transport throughout the Pacific and southward transport at all latitudes in the Atlantic. The annual mean strength of the Pacific-Arctic-Atlantic through flow is 0.63x106 m3/s in the Bering Strait. The majority of the northward volume transport in the southern Pacific turns into the Indonesian through flow (ITF) and joins the Indian Ocean equatorial current, which subse- quently flows out southward from the Mozambique Channel, with its majority superimposed on the Ant- arctic Circumpolar Current (ACC). This anti-cyclonic circulation around Australia has a strength of 11 x 106 ms /s according to the model-produced result. The atmospheric fresh water transport, known as P-E^R (pre- cipitation minus evaporation plus runoff), constructs a complement to the horizontal volume transport of the ocean. The annual mean heat transport sketch exhibits a northward heat transport in the Atlantic and poleward heat transport in the global ocean. The surface heat flux acts as a complement to the horizontal heat transport of the ocean. The climatological volume transports describe the most important features through the inter-basin passages and in the associated basins, including: the positive P-E+R in the Arctic substantially strengthening the East Greenland Current in summer; semiannual variability of the volume transport in the Drake Passage and the southern Atlantic-Indian Ocean passage; and annual transport vari- ability of the ITF intensifying in the boreal summer. The climatological heat transports show heat storage in July and heat deficit in January in the Arctic; heat storage in January and heat deficit in July in the Antarctic circumpolar current regime (ACCR); and intensified heat transport of the iTF in July. The volume transport of the ITF is synchronous with the volume transport through the southern Indo-Pacific sections, but the year-long southward heat transport of the ITF is out of phase with the heat transport through the equatorial Pacific, which is northward before May and southward after May. This clarifies the majority of the ITF origi- natinR from the southern Pacific Ocean. 展开更多

Background Promoting the synchronization of glucose and amino acid release in the digestive tract of pigs could effectively improve dietary nitrogen utilization.The rational allocation of dietary starch sources and the exploration of appropriate dietary glucose release kinetics may promote the dynamic balance of dietary glucose and amino acid supplies.However,research on the effects of diets with different glucose release kinetic profiles on amino acid absorption and portal amino acid appearance in piglets is limited.This study aimed to investigate the effects of the kinetic pattern of dietary glucose release on nitrogen utilization,the portal amino acid profile,and nutrient transporter expression in intestinal enterocytes in piglets.Methods Sixty-four barrows(15.00±1.12 kg)were randomly allotted to 4 groups and fed diets formulated with starch from corn,corn/barley,corn/sorghum,or corn/cassava combinations(diets were coded A,B,C,or D respectively).Protein retention,the concentrations of portal amino acid and glucose,and the relative expression of amino acid and glucose transporter m RNAs were investigated.In vitro digestion was used to compare the dietary glucose release profiles.Results Four piglet diets with different glucose release kinetics were constructed by adjusting starch sources.The in vivo appearance dynamics of portal glucose were consistent with those of in vitro dietary glucose release kinetics.Total nitrogen excretion was reduced in the piglets in group B,while apparent nitrogen digestibility and nitrogen retention increased(P<0.05).Regardless of the time(2 h or 4 h after morning feeding),the portal total free amino acids content and contents of some individual amino acids(Thr,Glu,Gly,Ala,and Ile)of the piglets in group B were significantly higher than those in groups A,C,and D(P<0.05).Cluster analysis showed that different glucose release kinetic patterns resulted in different portal amino acid patterns in piglets,which decreased gradually with the extension of feeding time.The portal His/Phe,Pro/Glu,Leu/Val,Lys/Met,Tyr/Ile and Ala/Gly appeared higher similarity among the diet treatments.In the anterior jejunum,the glucose transporter SGLT1 was significantly positively correlated with the amino acid transporters B0AT1,EAAC1,and CAT1.Conclusions Rational allocation of starch resources could regulate dietary glucose release kinetics.In the present study,group B(corn/barley)diet exhibited a better glucose release kinetic pattern than the other groups,which could affect the portal amino acid contents and patterns by regulating the expression of amino acid transporters in the small intestine,thereby promoting nitrogen deposition in the body,and improving the utilization efficiency of dietary nitrogen. 展开更多

Verticillium wilt(VW),induced by the soil-borne fungus Verticillium dahliae(Vd),poses a substantial threat to a diverse array of plant species.Employing molecular breeding technology for the development of cotton varieties with heightened resistance to VW stands out as one of the most efficacious protective measures.In this study,we successfully generated two stable transgenic lines of cotton(Gossypium hirsutum L.),VdThitRNAi-1 and VdThit-RNAi-2,using host-induced gene silencing(HIGS)technology to introduce double-stranded RNA(dsRNA)targeting the thiamine transporter protein gene(VdThit).Southern blot analysis confirmed the presence of a single-copy insertion in each line.Microscopic examination showed marked reductions in the colonization and spread of Vd-mCherry in the roots of VdThit-RNAi cotton compared to wild type(WT).The corresponding disease index and fungal biomass of VdThit-RNAi-1/2 also exhibited significant reductions.Real-time quantitative PCR(qRT-PCR)analysis demonstrated a substantial inhibition of VdThit expression following prolonged inoculation of VdThit-RNAi cotton.Small RNA sequencing(sRNA-Seq)analysis revealed the generation of a substantial number of VdThit-specific siRNAs in the VdThit-RNAi transgenic lines.Additionally,the silencing of VdThit by the siVdThit produced by VdThit-RNAi-1/2 resulted in the elevated expression of multiple genes involved in the thiamine biosynthesis pathway in Vd.Under field conditions,VdThit-RNAi transgenic cotton exhibited significantly enhanced disease resistance and yield compared with WT.In summary,our findings underscore the efficacy of HIGS targeting VdThit in restraining the infection and spread of Vd in cotton,thereby potentially enabling the development of cotton breeding as a promising strategy for managing VW. 展开更多

Reactive transport modeling(RTM)is an emerging method used to address geological issues in diagenesis research.However,the extrapolation of RTM results to practical reservoir prediction is not sufficiently understood.This paper presents a case study of the Eocene Qaidam Basin that combines RTM results with petrological and mineralogical evidence.The results show that the Eocene Xiaganchaigou Formation is characterized by mixed siliciclastic-carbonate-evaporite sedimentation in a semiclosed saline lacustrine environment.Periodic evaporation and salinization processes during the syngeneticpenecontemporaneous stage gave rise to the replacive genesis of dolomites and the cyclic enrichment of dolomite in the middle-upper parts of the meter-scale depositional sequences.The successive change in mineral paragenesis from terrigenous clastics to carbonates to evaporites was reconstructed using RTM simulations.Parametric uncertainty analyses further suggest that the evaporation intensity(brine salinity)and particle size of sediments(reactive surface area)were important rate-determining factors in the dolomitization,as shown by the relatively higher reaction rates under conditions of higher brine salinity and fine-grained sediments.Combining the simulation results with measured mineralogical and reservoir physical property data indicates that the preservation of original intergranular pores and the generation of porosity via replacive dolomitization were the major formation mechanisms of the distinctive lacustrine dolomite reservoirs(widespread submicron intercrystalline micropores)in the Eocene Qaidam Basin.The results confirm that RTM can be effectively used in geological studies,can provide a better general understanding of the dolomitizing fluid-rock interactions,and can shed light on the spatiotemporal evolution of mineralogy and porosity during dolomitization and the formation of lacustrine dolomite reservoirs. 展开更多

Given the challenge of definitively discriminating between chemical and nuclear explosions using seismic methods alone,surface detection of signature noble gas radioisotopes is considered a positive identification of underground nuclear explosions(UNEs).However,the migration of signature radionuclide gases between the nuclear cavity and surface is not well understood because complex processes are involved,including the generation of complex fracture networks,reactivation of natural fractures and faults,and thermo-hydro-mechanical-chemical(THMC)coupling of radionuclide gas transport in the subsurface.In this study,we provide an experimental investigation of hydro-mechanical(HM)coupling among gas flow,stress states,rock deformation,and rock damage using a unique multi-physics triaxial direct shear rock testing system.The testing system also features redundant gas pressure and flow rate measurements,well suited for parameter uncertainty quantification.Using porous tuff and tight granite samples that are relevant to historic UNE tests,we measured the Biot effective stress coefficient,rock matrix gas permeability,and fracture gas permeability at a range of pore pressure and stress conditions.The Biot effective stress coefficient varies from 0.69 to 1 for the tuff,whose porosity averages 35.3%±0.7%,while this coefficient varies from 0.51 to 0.78 for the tight granite(porosity<1%,perhaps an underestimate).Matrix gas permeability is strongly correlated to effective stress for the granite,but not for the porous tuff.Our experiments reveal the following key engineering implications on transport of radionuclide gases post a UNE event:(1)The porous tuff shows apparent fracture dilation or compression upon stress changes,which does not necessarily change the gas permeability;(2)The granite fracture permeability shows strong stress sensitivity and is positively related to shear displacement;and(3)Hydromechanical coupling among stress states,rock damage,and gas flow appears to be stronger in tight granite than in porous tuff. 展开更多

A three-dimensional reconstruction of rough fracture surfaces of hydraulically fractured rock outcrops is carried out by casting process,a large-scale experimental setup for visualizing rough fractures is built to perform proppant transport experiments.The typical characteristics of proppant transport and placement in rough fractures and its intrinsic mechanisms are investigated,and the influences of fracture inclination,fracture width and fracturing fluid viscosity on proppant transport and placement in rough fractures are analyzed.The results show that the rough fractures cause variations in the shape of the flow channel and the fluid flow pattern,resulting in the bridging buildup during proppant transport to form unfilled zone,the emergence of multiple complex flow patterns such as channeling,reverse flow and bypassing of sand-carrying fluid,and the influence on the stability of the sand dune.The proppant has a higher placement rate in inclined rough fractures,with a maximum increase of 22.16 percentage points in the experiments compared to vertical fractures,but exhibits poor stability of the sand dune.Reduced fracture width aggravates the bridging of proppant and induces higher pumping pressure.Increasing the viscosity of the fracturing fluid can weaken the proppant bridging phenomenon caused by the rough fractures. 展开更多

The elliptic azimuthal anisotropy coefficient(v_(2))of the identified particles at midrapidity(|η|<0.8)was investigated in p-Pb collisions at√s_(NN)=5.02 TeV using a multi-phase transport model(AMPT).The calculations of differential v_(2)based on the advanced flow extraction method of light flavor hadrons(pions,kaons,protons,andΛ)in small collision systems were extended to a wider transverse momentum(p_(T))range of up to 8 GeV/c for the first time.The string-melting version of the AMPT model provides a good description of the measured p_(T)-differential v_(2)of the mesons but exhibits a slight deviation from the baryon v_(2).In addition,we observed the features of mass ordering at low p_(T)and the approximate number-of-constituentquark(NCQ)scaling at intermediate p_(T).Moreover,we demonstrate that hadronic rescattering does not have a significant impact on v_(2)in p-Pb collisions for different centrality selections,whereas partonic scattering dominates in generating the elliptic anisotropy of the final particles.This study provides further insight into the origin of collective-like behavior in small collision systems and has referential value for future measurements of azimuthal anisotropy. 展开更多

As the main link of ground engineering,crude oil gathering and transportation systems require huge energy consumption and complex structures.It is necessary to establish an energy efficiency evaluation system for crude oil gathering and transportation systems and identify the energy efficiency gaps.In this paper,the energy efficiency evaluation system of the crude oil gathering and transportation system in an oilfield in western China is established.Combined with the big data analysis method,the GA-BP neural network is used to establish the energy efficiency index prediction model for crude oil gathering and transportation systems.The comprehensive energy consumption,gas consumption,power consumption,energy utilization rate,heat utilization rate,and power utilization rate of crude oil gathering and transportation systems are predicted.Considering the efficiency and unit consumption index of the crude oil gathering and transportation system,the energy efficiency evaluation system of the crude oil gathering and transportation system is established based on a game theory combined weighting method and TOPSIS evaluation method,and the subjective weight is determined by the triangular fuzzy analytic hierarchy process.The entropy weight method determines the objective weight,and the combined weight of game theory combines subjectivity with objectivity to comprehensively evaluate the comprehensive energy efficiency of crude oil gathering and transportation systems and their subsystems.Finally,the weak links in energy utilization are identified,and energy conservation and consumption reduction are improved.The above research provides technical support for the green,efficient and intelligent development of crude oil gathering and transportation systems. 展开更多
