(Td22), inhibitory to Sclerotinia minor Jagger, to grow and survive in mature wood fibre waste (WFW) compost of paper mill origin following nutrient amendment. The growth and survival of the fungus in the WFW compost was assessed by serial dilution plate count method followed by confirmation of the fungal identity using pectic enzyme analysis as described in Cruickshank and Pitt [1]. It was found in this study that the population densities of TdE2 achieved under non-sterile conditions in the WFW compost following nutrient amendment was approximately in the range of 7.0 lOgl0 CFU/g dw - 8.5 log10 CFU/g dw after 28 days, depending on pre-treatment. The efficacy of this WFW compost-grown TdE2 for protection of lettuce from attack by S. minor was also demonstrated in glasshouse trials. This study indicates that cellulosic paper mill waste compost could provide an abundant low-cost growth medium for the large-scale cultivation of fungal antagonists, improving prospects for cost-competitiveness with chemical treatments.展开更多

Additionally, four previously characterized 18-residue peptaibols neoatroviridins A-D ( 14−17 ) were also identified. The structural configurations of the newly identified peptaibols ( 1−13 ) were determined by comprehensive nuclear magnetic resonance (NMR) and high-resolution electrospray ionization tandem mass spectrometry (HR-ESI-MS/MS) data. Their absolute configurations were further determined using Marfey’s method. Notably, compounds 12 and 13 represent the first 14-residue peptaibols containing an acidic amino acid residue. In antimicrobial assessments, all 18-residue peptaibols ( 1 − 7 , 14 − 17 ) exhibited moderate inhibitory activities against Staphylococcus aureus 209P, with minimum inhibitory concentration (MIC) values ranging from 8−32 μg·mL^(−1). Moreover, compound 9 exhibited moderate inhibitory effect on Candida albicans FIM709, with a MIC value of 16 μg·mL^(−1).展开更多

Their structures were identified as harzianone (1), trichoacorenol (2), and trichodimerol (3) by extensive spectroscopic analysis. Among them, 1 was a harziane diterpene, 2 was a sesquiterpene alcohol, and 3 was a polyketide with a completely symmetric configuration. Compound 3 exhibited medium inhibitory activity with an IC50 value of 74.6 μM using a NA (H7N9)/MUNANA model.展开更多

Xy24 by using various column chromatography techniques. Their structures were determined on the basis of extensive spectroscopic(HR-ESI-MS, 1D NMR, 2D NMR and CD) analyses. Among them, 1 was the reductive product of harzianone and 2 was the Baeyer–Villiger monooxygenase catalyzed oxidation product of harzianone. Compound 1 exhibited cytotoxic activity against He La and MCF-7 cell lines with IC(50) values of 30.1 μmol/L and 30.7 μmol/L,respectively.展开更多

[Method] Using orthognnal experimental design, with the variation of sugar content after enzymatic hydrolysis measured by DNS method as the indicator, the fermentation conditions were optimized. [Result] Taking colloidal chitin as the carbon source and 2% peptone as the nitrogen source with the shaking speed of 170 r/rain, the optimum fermentation conditions of T. aureoviride for the production of chitinase were as follows : initial pH value of medium, 5.0; inoculation amount, 8% ; bottle volume, 20 ml; 6 d cultivation at 28℃. [Conclusion] The optimum conditions for the production of chitinase were confirmed, which provided basis for the utilization of T. aureoviride.展开更多

Production of antifungal substance(s) in the H921-EAE-CF began to increase up to 3 days after isolate H921 incubation. Furthermore, heat treatment (105°C or 121°C) of H921-EAE-CF did not alter its inhibitory effect on M. oryzae spore germination compared to non-heat-treated H921-EAE-CF. Blast lesion formation inhibition by H921-EAE-CF was dose-dependent. Internal transcribed spacer (ITS) region sequence analysis indicated that this isolate shared similarities with species of the genera Trichoderma. This study suggests that H921-EAE-CF contains some antifungal substances that could be promising candidates for control of rice blast disease.展开更多

The results show that the time of fungicide selection on the transformed Trichoderma sp. may have very important effect on the resistance gene transformation. By this method, the resistance gene to carbendazim was transformed into Trichoderma harzianum, a strain of T. harzianum with resistance to carbendazim was achieved. The transformant could grow on the medium containing 150μg/mL carbendazim. The resistance is stable after 10 times transfer on non-selective medium.展开更多
