Subsequently, the se...To analyze the genetic differentiation of the host- and geo-populations of Liriomyza sativae Blanchard, the β-tubulin gene of 5 host-populations and 6 geo-populations of L. sativae was sequenced. Subsequently, the sequences were analyzed by biosoftware DNAStar and MEGA, and the phylogenetic trees constructed. The results obtained by the two softwares were similar, that is, the sequences of β-tubulin gene were more than 98% homologous, among the host- and geo-populations of L. sativae, with only 8 variable sites, but no insertions and deletions were detected. It seems that differentiations in β-tubulin gene among the host- and geo-populations of L. sativae are related to the hobby to hosts and the geographical distributions, respectively.展开更多

Zhenjiang based on morphological characteristics and phylogenetic analyses of ribosomal sequences. This microsporidium causes silkworm pebrine, although its original host and mode of transmission are unknown. To better understand its distribution and transmission mode, it is essential to have species specific molecular markers. Towards this goal, we characterized the alpha tubulin gene from Endoreticulatus sp. Zhenjiang in this study. The full-length alpha-tubulin cDNA from Endoreticulatus sp. Zhenjiang was cloned and sequenced (GenBank ID: KJ784483) using the rapid amplification of cDNA ends (RACE) protocol. The alpha-tubulin cDNA is 1382 bp long with an open reading frame spanning 1320 bp and consisting of a short 20-bp 5’-untranslated region (5’-UTR) and a 42-bp 3’-UTR with a stop codon and a poly (A) tail. This alpha-tubulin cDNA encodes a deduced polypeptide with 439 amino acids, including a complete tubulin domain and a tubulin C domain. This protein has an estimated isoelectric point of 5.1 and a predicted molecular weight of 48.6 kDa.展开更多

Research on gene expression and regulation during zebrafish embryonic development plays an important role in revealing the molecular mechanism of morphogenesis in vertebrate. Tubulin locates the cytoplasmic determinants in eggs, organizes the formation of basic cytoskeleton and controls various cell movements. There are several types of tubulin, each of which has specific expression regions and functions. In this study, the full_length cDNA of a Xenopus β 2 tubulin was used as template to generate Dig_labeled antisense RNA which was used as a probe to carry out in situ hybridization on whole mount zebrafish embryos. Under the experimental conditions, the tubulin transcript was first detected in a group of cells in embryos at the midblastula stage. With the development of the embryo, this transcript was gradually restricted to the nervous system. These results demonstrate that the expression of this β 2 tubulin gene in zebrafish embryos is neural specific.展开更多

And the microtubule, as a carrier of transported materials, of axonal architecture plays an important role. It has also been found that the content and transported velocity of tubulin in the regeneration nerve increase significantly owing to the requirements of axonal reconstruction.
