BACKGROUND: Cerebral infarction is poorly treated due to neuronal necrosis and secondary pathophysiological changes; for example, free radical production and inflammatory reactions. OBJECTIVE: To detect the levels of high-sensitivity C-reactive protein (hs-CRP), interleukin-6 (IL-6), and tumor necrosis factor- a (TNF- α ) in elderly males with cerebral infarction. DESIGN: Non-randomized current control study. SETTING: Cadre Medical Department, Guizhou Provincial People's Hospital. PARTICIPANTS: Forty elderly males (65-89 years old) with cerebral infarction were selected from Cadre Medical Department, Guizhou Provincial People's Hospital from February 2004 to December 2006. All patients met the diagnostic criteria of cerebral infarction modified at the 4th National Cerebrovascular Disease Academic Meeting, and were diagnosed on the basis of CT or MRI tests. Furthermore, 35 elderly male inpatients (65-87 years old) without cerebral infarction were selected as the control group. Included subjects provided confirmed consent and did not have heart disease, diabetes mellitus, lipid disorder, acute trauma, infection, rheumatism, or other inflammatory diseases. The study was approved by the local ethics committee. There were no significant differences in age, blood pressure, and lipid levels between the cerebral infarction group and the control group (P 〉 0.05), and this suggested that the baseline data of both groups were comparable. METHODS: Fasting venous blood was drawn from cerebral infarction patients 24 hours after cerebral infarction attack and from control subjects 24 hours after hospitalization. A latex-enhanced immunoturbidimetric assay and an enzyme-linked immunosorbent assay were used to detect the levels of hs-CRP, IL-6, and TNF- α in the serum. MAIN OUTCOME MEASURES: The levels of hs-CRP, 1L-6, and TNF- α in the serum in both groups. RESULTS: Forty cerebral infarction patients and thirty-five control subjects were included in the final analysis without any loss. Levels of hs-CRP, IL-6, and TNF-α in the cerebral infarction group were significantly higher than those in the control group (P 〈 0.01 ). CONCLUSION: Levels of serum inflammatory reactive factors are increased in elderly males with cerebral infarction. 展开更多

AIM:To study the effects of Helicobacter pylori(H. pylori)tumor necrosis factor-α(TNF)inducing protein (Tip-α)on cytokine expression and its mechanism. METHODS:We cloned Tip-αfrom the H.pylori strain 26695,transformed Escherichia coli with an expression plasmid,and then confirmed the expression product by Western blotting.Using different concentrations of Tip-αthat affected SGC7901 and GES-1 cells at different times,we assessed cytokine levels using enzyme-linked immunosorbent assay.We blocked SGC7901 cells with pyrrolidine dithiocarbamate(PDTC),a specific inhibitor of nuclear factorκB(NF-κB).We then detected interleukin(IL)-1βand TNF-αlevels in SGC7901 cells. RESULTS:Western blot analysis using an anti-Tip-α antibody revealed a 23-kDa protein,which indicated that recombinant Tip-αprotein was recombined successfully.The levels of IL-1β,IL-8 and TNF-αwere sig-nificantly higher following Tip-αinterference,whether GES-1 cells or SGC-7901 cells were used(P<0.05).However,the levels of cytokines(including IL-1β,IL-8 and TNF-α)secreted by SGC-7901 cells were greater than those secreted by GES-1 cells following treatment with Tip-αat the same concentration and for the same duration(P<0.05).After blocking NF-κB with PDTC, the cells(GES-1 cells and SGC-7901 cells)underwent interference with Tip-α.We found that IL-1βand TNF-αlevels were significantly decreased compared to cells that only underwent Tip-αinterference(P<0.05). It may induce sleep, fatigue, and alertness loss at work, and endanger public safety. It’s unclear that cytokines of train driver would be influenced by their job. The research considers the hypothesis that stressful professions, such as train driver, influence the body’s immune system through the long-time and high-pressure working, and change production of neuro-immune factors. Using enzyme linked immunosorbent assay (ELISA), several neuro-immune factors were assayed among train drivers (N = 82) and health blood donors (N = 80) enrolled in the Yunnan Collaborative Innovation Center for Public Health and Disease Control. The concentrations of angiotensin, C-reactive protein (CRP), interleukin-8 (IL-8), and tumor necrosis factor-alpha (TNF-α) were determined. Kruskal-Wallis test and Dunn’s multiple comparisons test were performed for overall comparison between groups and for pairwise comparison, respectively. Statistical significance level was set at P < 0.05. The profession of train driving was not associated with significant increases or decreases in the systemic levels of inflammatory (CRP, IL-8, and TNF-α), but it was associated with the high expression of angiotensin in vivo. These findings suggest that the job of train driving may not be associated with significant alterations in systemic immune condition, but arouse the level of angiotensin. 展开更多

Generalized pustular psoriasis (GPP) is the severe and rare type of psoriasis. The association between tumor necrosis factor-alpha induced protein 3 interacting protein 1 (TNIP1) gene and psoriasis was confirmed in people with multiple ethnicities. This study was to investigate the association between TNIP1 gene polymorphisms and pustular psoriasis in Chinese Hart population. Methods: Seventy-three patients with GPP, 67 patients with palmoplantar pustulosis (PPP), and 476 healthy controls were collected from Chinese Hart population. Six single nucleotide polymorphisms (SNPs) of the TNIP1 gene, namely rs3805435, rs3792798, rs3792797, rs869976, rs 17728338, and rs999011 were genotyped by using polymerase chain reaction-ligase detection reaction. Statistical analyses were performed using the PLINK 1.07 package. Allele frequencies and genotyping frequencies for six SNPs were compared by using Chi-square test, odd ratio (OR) (including 95% confidence interval) were calculated. The haplotype analysis was conducted by Haploview software. Results: The frequencies of alleles of five SNPs were significantly different between the GPP group and the control group (P ≤ 7.22 × 10^-3), especially in the GPP patients without psoriasis vulgaris (PsV). In the haplotype analysis, the most significantly different haplotype was H4: ACGAAC, with 13.1% frequency in the GPP group but only 3.4% in the control group (OR = 4.16, P = 4.459 × 10^-7). However, no significant difference in the allele frequencies was found between the PPP group and control group for each of the six SNPs (P 〉 0.05). Conclusions: Polymorphisms in TNIP1 are associated with GPP in Chinese Han population. However, no association with PPP was found. 自发性急性脑出血患者血浆sCD163/sTWEAK比值与预后的关系

目的探究自发性急性脑出血(ACH)患者血浆可溶性CD163(sCD163)/可溶性肿瘤坏死因子样凋亡弱诱导因子(sTWEAK)比值与预后的关系。方法纳入ACH患者90例作为病例组,根据格拉斯哥预后评分将病例组分为预后不良组(38例)和预后良好组(52例);另... 目的探究自发性急性脑出血(ACH)患者血浆可溶性CD163(sCD163)/可溶性肿瘤坏死因子样凋亡弱诱导因子(sTWEAK)比值与预后的关系。方法纳入ACH患者90例作为病例组,根据格拉斯哥预后评分将病例组分为预后不良组(38例)和预后良好组(52例);另选取同期体检健康者45例为对照组。酶联免疫吸附试验检测血浆sCD163、sTWEAK水平并计算sCD163/sTWEAK比值。分析血浆sCD163、sTWEAK水平及sCD163/sTWEAK比值与临床资料的相关性;Logistic回归分析ACH患者预后不良的影响因素;受试者工作特征(ROC)曲线分析sCD163/sTWEAK比值对ACH患者预后不良的预测价值。结果病例组血浆sCD163、sTWEAK水平及sCD163/sTWEAK比值均显著高于对照组;预后良好组上述指标均低于预后不良组(P<0.05)。预后良好组血肿体积、美国国立卫生研究院卒中量表(NIHSS)评分、高血压及幕下出血比例均低于预后不良组,低密度脂蛋白胆固醇(LDL-C)高于预后不良组(P<0.05)。相关性分析表明,血浆sCD163、sTWEAK水平及sCD163/sTWEAK比值与出血部位、血肿体积、NIHSS评分、白细胞计数、血小板计数、中性粒细胞/淋巴细胞比值(NLR)呈正相关(P<0.05)。Logistic回归分析显示,sCD163/sTWEAK比值、出血部位、血肿体积、NIHSS评分为ACH患者预后不良的影响因素(P<0.05)。ROC曲线结果表明,sCD163/sTWEAK比值评估ACH患者预后不良的AUC为0.850,敏感度和特异度分别为86.84%和69.23%。结论sCD163/sTWEAK比值在ACH患者血浆中水平较高,并与预后不良有关,该值对此类患者的预后有一定预测价值。 展开更多

冠心宁通过TNFAIP3-ASK1/JNK通路对动脉粥样硬化血管平滑肌细胞表型转换的调控作用

目的探讨冠心宁(GXN)调控动脉粥样硬化(AS)斑块稳定性的分子机制。方法制备含有GXN药物的血清,利用氧化低密度脂蛋白(ox-LDL)诱导血管平滑肌细胞(VSMC)构建AS体外模型,Western blot和qPCR检测VSMC表型转换情况。通过沉默肿瘤坏死因子α... 目的探讨冠心宁(GXN)调控动脉粥样硬化(AS)斑块稳定性的分子机制。方法制备含有GXN药物的血清,利用氧化低密度脂蛋白(ox-LDL)诱导血管平滑肌细胞(VSMC)构建AS体外模型,Western blot和qPCR检测VSMC表型转换情况。通过沉默肿瘤坏死因子α诱导蛋白3(TNFAIP3),检测GXN对VSMC表型转换的影响。构建凋亡信号调节激酶1(ASK1)过表达载体,并利用Lip3000转染进细胞,检测GXN是否通过凋亡信号调节激酶1/c-Jun氨基末端激酶(ASK1/JNK)通路发挥作用。结果与对照组比较,ox-LDL组的细胞表型转换,表现为I型胶原蛋白(COLIA1和COLIA2)、TNFAIP3和α-平滑肌肌动蛋白(α-SMA)的表达水平降低(均P<0.01),基质金属蛋白酶(MMP)2、MMP9、MMP13、骨桥蛋白(OPN)、ASK1磷酸化与ASK1比值(p-ASK1/ASK1)、JNK磷酸化与JNK比值(p-JNK/JNK)升高(均P<0.01);GXN处理后VSMC表型转换为收缩型,表现为与ox-LDL组相比COLIA1、COLIA2和α-SMA水平增高(均P<0.01),MMP2、MMP9、MMP13、OPN、TNFAIP3的表达水平、p-ASK1/ASK1以及p-JNK/JNK都降低(均P<0.01)。沉默TNFAIP3后,与ox-LDL+10%GXN+sh-NC组相比,COLIA1、COLIA2、TNFAIP3和α-SMA水平降低(均P<0.01),MMP2、MMP9、MMP13、OPN、p-ASK1/ASK1以及p-JNK/JNK都升高(均P<0.01)。过表达ASK1后,与ox-LDL+10%GXN+oe-NC组相比,COLIA1、COLIA2和α-SMA的表达水平降低(均P<0.01),MMP2、MMP9、MMP13、OPN、p-ASK1/ASK1以及p-JNK/JNK都升高(均P<0.01)。结论GXN可能通过TNFAIP3调控ASK1/JNK通路介导VSMC表型转换,从而起到稳定动脉硬化斑块的作用。 展开更多

血清IRS-1和CTRP3水平与妊娠糖尿病患者不良妊娠结局的相关性

目的分析血清胰岛素底物受体-1(IRS-1)、C1q肿瘤坏死因子相关蛋白-3(CTRP3)水平与妊娠糖尿病(GDM)患者不良妊娠结局的相关性。方法采用前瞻性研究,选取2021年1月至2023年12月在陕西中医药大学第二附属医院分娩的132例GDM孕妇为患病组[年... 目的分析血清胰岛素底物受体-1(IRS-1)、C1q肿瘤坏死因子相关蛋白-3(CTRP3)水平与妊娠糖尿病(GDM)患者不良妊娠结局的相关性。方法采用前瞻性研究,选取2021年1月至2023年12月在陕西中医药大学第二附属医院分娩的132例GDM孕妇为患病组[年龄(28.76±3.18)岁,体重指数(BMI)为(25.72±2.82)kg/m^(2),孕周(38.06±1.42)周,经产孕妇66例,阴道分娩82例],根据妊娠结局分为良好结局组(80例)与不良结局组(52例);另选同期分娩的健康孕妇132名为对照组[年龄(28.65±3.06)岁,BMI(25.31±2.67)kg/m^(2),孕周(38.21±1.33)周,经产孕妇60例,阴道分娩86例]。对不同妊娠结局GDM患者临床资料进行单因素分析,多因素logistic回归分析孕妇不良妊娠结局的影响因素,绘制受试者操作特征曲线(ROC)分析IRS-1、CTRP3对GDM患者不良妊娠结局的诊断价值,分析IRS-1、CTRP3与临床指标及孕妇妊娠结局的相关性。统计学方法采用χ^(2)检验、t检验、Pearson及Spearman相关性分析。结果患病组血清IRS-1、CTRP3水平均低于对照组[(1.82±0.21)μg/L比(2.33±0.28)μg/L、(353.51±38.14)ng/L比(425.95±45.21)ng/L],差异均有统计学意义(t=16.74、14.07,均P<0.05);不良结局组血清IRS-1、CTRP3水平均低于良好结局组,空腹血糖(FBG)、糖化血红蛋白(HbAlc)、胰岛素抵抗指数(HOMA-IR)水平均高于良好结局组,差异均有统计学意义(t=8.61、7.49、7.26、8.40、32.73,均P<0.05);FBG、HbAlc、HOMA-IR水平升高均为影响患者不良妊娠结局的危险因素[比值比(OR)=1.498,95%置信区间(CI)1.099~2.042;OR=1.698,95%CI 1.092~2.639;OR=2.014,95%CI 1.063~3.816;均P<0.05],IRS-1、CTRP3均为影响患者不良妊娠结局的保护因素[OR=0.774,95%CI 0.621~0.964;OR=0.731,95%CI 0.573~0.932;均P<0.05];ROC显示,IRS-1、CTRP3联合诊断患者不良妊娠结局的曲线下面积(AUC)为0.899,联合诊断的AUC优于IRS-1、CTRP3单独诊断(Z=2.094、2.056,均P<0.05);IRS-1、CTRP3与孕妇妊娠结局及FBG、HbAlc、HOMA-IR均呈负相关(均P<0.05)。结论不良妊娠结局GDM患者血清IRS-1、CTRP3水平降低,二者联合对患者不良妊娠结局有一定预测价值。 展开更多

CTRP3联合NT-proBNP对MVD患者发生HHcy的预测价值

目的探讨补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)联合N末端B型利钠肽前体(NT-proBNP)对冠状动脉(冠脉)多支病变(MVD)患者发生高同型半胱氨酸血症(HHcy)的预测价值。方法回顾性分析200例MVD患者的临床资料,根据血浆同型半胱氨酸(Hcy)水平... 目的探讨补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)联合N末端B型利钠肽前体(NT-proBNP)对冠状动脉(冠脉)多支病变(MVD)患者发生高同型半胱氨酸血症(HHcy)的预测价值。方法回顾性分析200例MVD患者的临床资料,根据血浆同型半胱氨酸(Hcy)水平是否>15μmol/L分为高Hcy组(111例)和正常Hcy组(89例)。比较两组基线资料[年龄、性别、冠心病病程、吸烟史、糖尿病病史、高血压病史、体质量指数(BMI)],生化指标(NT-proBNP、尿酸(UA)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、Hcy、CTRP3),采用二元Logistic回归分析法分析影响MVD患者发生HHcy危险因素,绘制受试者工作特征(ROC)曲线分析CTRP3联合NT-proBNP对MVD患者发生HHcy的预测效能。结果两组年龄、高血压病史、BMI、UA、TG、TC、HDL-C比较均无统计学差异(P>0.05);高Hcy组男性患者数量70例、冠心病病程(6.66±4.49)年、吸烟史55例、糖尿病病史60例,均大于正常Hcy组的36例、(3.67±2.31)年、28例、17例,NT-proBNP(5234.95±7476.76)pg/ml、LDL-C(3.55±0.95)mmol/L、Hcy(44.75±12.48)μmol/L均高于正常Hcy组的(1296.94±3864.78)pg/ml、(2.95±0.88)mmol/L、(10.58±2.80)μmol/L,CTRP3(65.20±13.61)μg/L低于正常Hcy组的(88.69±14.94)μg/L,两组间比较差异均具有统计学意义(P<0.05)。多因素Logistic回归分析显示:男性[OR=3.745,95%CI=(1.398,10.030)]、糖尿病病史[OR=3.262,95%CI=(1.264,8.417)]、冠心病病程[OR=1.194,95%CI=(1.022,1.394)]、LDL-C[OR=2.254,95%CI=(1.337,3.800)]为导致MVD患者发生HHcy的独立危险因素,而CTRP3[OR=0.902,95%CI=(0.873,0.933)]为其独立保护因素(P<0.05)。ROC曲线结果显示:CTRP3联合NT-proBNP预测MVD患者发生HHcy的AUC为0.901,敏感度为83.8%,特异度为86.5%,优于LDL-C、NT-proBNP或CTRP3单独预测(P<0.05)。结论性别、糖尿病病史、冠心病病程、LDL-C为导致MVD患者发生HHcy的独立危险因素,而CTRP3为其独立保护因素。血清CTRP3联合NT-proBNP可提高MVD患者发生HHcy的预测价值。 展开更多

Background Tumor necrosis factor-induced protein 3 (TNFAIP3) gene has been shown important in cardiac remodeling. The aim of the present study was to investigate whether the variants of TNFAIP3 gene are associated with left ventricular hypertrophy (LVH) in hypertensive patients.Methods Four representatives of all the other single nucleotide polymorphisms (SNPs) in TNFAIP3 gene were tested for association with hypertrophy in two independent hypertensive populations (n=2120 and n=324).Results We found that only the tag SNP (rs5029939) was consistently lower in the hypertensives with cardiac hypertrophy than in those without cardiac hypertrophy in the two study populations, indicating a protective effect on LVH (odds ratio (OR) (95% confidence interval (CI))0.58 (0.358-0.863), P=0.035; OR (95% CI)=0.477 (0.225-0.815), P〈0.05,respectively). Multiple regression analyses confirmed that the patients with G allele of rs5029939 had less thickness in inter-ventricular septum, left ventricular posterior wall, relative wall thickness and left ventricular mass index than did those with CC allele in the hypertensive patients in both study populations (all P〈0.01).Conclusion These findings indicate that the SNP (rs5029939) in the TNFAIP3 gene may serve as a novel protective genetic marker for the development of LVH in patients with hypertension 展开更多

Background The microemboli produced during spontaneous plaque rupture and ulceration and during coronary intervention will reduce coronary reserve and cause cardiac dysfunction. It is though that inflammation caused by the microinfarction induced by the microembolization may play an essential role. It is known that the activation of p38 mitogen-activated protein kinases (MAPK) in both infected and non-infected inflammation in myocardium may cause a contractile dysfunction. But the relation between the activation of p38 MAPK and microembolization is still unknown. Methods Sprague-Dawley rats were randomly divided into three groups: Sham group, coronary microembolization (CME) group and SB203580 group (n=-10 per group). CME rats were produced by injection of 42 pm microspheres into the left ventricle with occlusion of the ascending aorta. SB203580, a p38 MAPK inhibitor, was injected into the femoral vein after the injection of microspheres to make the SB203580 group. Left ventricular ejection fraction (LVEF) was determined by echocardiography. The protein concentration of P38 MAPK in the myocardium was assessed by Western blotting. The relative expression of mRNA for tumor necrosis factor (TNF)-a was assessed by the technique of semi-quantitative polymerase chain reaction amplification. Results LVEF was depressed at three hours up to 12 hours in the CME group. Increased p38 MAPK activity and TNF-a mRNA expression were observed in the CME group. The administration of SB203580 partly inhibited p38 MAPK activity, but did not fully depress the TNF-α expression, and partly preserved cardiac contractile function. Conclusions p38 MAPK is significantly activated by CME and the inhibition of p38 MAPK can partly depress the TNF-a expression and preserve cardiac contractile function. 展开更多

目的研究血清半乳糖凝集素3(Gal-3)、肿瘤坏死因子样弱凋亡诱导因子(TWEAK)对精神分裂症的诊断价值及与精神症状严重程度的关系。方法选取2019年3月至2021年3月在该院诊治的96例精神分裂症患者作为病例组,以同期60例健康体检者为对照组... 目的研究血清半乳糖凝集素3(Gal-3)、肿瘤坏死因子样弱凋亡诱导因子(TWEAK)对精神分裂症的诊断价值及与精神症状严重程度的关系。方法选取2019年3月至2021年3月在该院诊治的96例精神分裂症患者作为病例组,以同期60例健康体检者为对照组,进行回顾性分析。采用酶联免疫吸附试验检测血清Gal-3、TWEAK水平。采用Pearson相关分析血清Gal-3、TWEAK水平与临床指标的相关性;采用多因素Logistic回归分析影响精神分裂症发生的因素;采用受试者工作特征(ROC)曲线分析血清Gal-3、TWEAK单项及联合检测对精神分裂症的诊断价值。结果观察组PANSS评分总分、阳性症状评分、阴性症状评分、一般精神病理学症状评分及血清Gal-3、TWEAK水平均高于对照组,差异均有统计学意义(P<0.001)。血清Gal-3、TWEAK水平与PANSS评分总分、阳性症状评分、阴性症状评分、一般精神病理学症状评分呈正相关(P<0.001)。PANSS评分总分、阳性症状评分、一般精神病理学症状评分升高及血清Gal-3、TWEAK水平升高均是精神分裂症发生的独立危险因素(P<0.001)。血清Gal-3、TWEAK联合检测诊断精神分裂症的曲线下面积为0.870(95%CI:0.831~0.919),明显高于血清Gal-3、TWEAK单项检测诊断精神分裂症的AUC[0.812(95%CI:0.769~0.847)、0.820(95%CI:0.771~0.852)],差异均有统计学意义(Z=4.345,P<0.001;Z=4.010,P=0.002)。结论精神分裂症患者血清Gal-3、TWEAK水平升高,二者与精神症状严重程度有关,血清Gal-3、TWEAK联合检测对精神分裂症具有较高的诊断价值。 展开更多

血清CTRP3、DKK-1水平在绝经后骨质疏松患者骨折发生中的临床价值

目的 探讨血清C1q肿瘤坏死因子相关蛋白3(CTRP3)和Dickkopf相关蛋白1(DKK-1)水平与绝经后骨质疏松(PMOP)患者发生骨折的关系。方法 选取2020年6月-2022年12月武警江西总队医院PMOP患者126例(PMOP组)和健康体检者(无骨质疏松、自然绝经1... 目的 探讨血清C1q肿瘤坏死因子相关蛋白3(CTRP3)和Dickkopf相关蛋白1(DKK-1)水平与绝经后骨质疏松(PMOP)患者发生骨折的关系。方法 选取2020年6月-2022年12月武警江西总队医院PMOP患者126例(PMOP组)和健康体检者(无骨质疏松、自然绝经1年以上)118名(正常对照组)。根据PMOP患者是否发生骨折分为骨折组(41例)和无骨折组(85例)。收集所有研究对象一般资料和实验室生化项目检测结果,同时检测血清CTRP3、DKK-1水平和腰椎(L_(1)-L_(4))的骨密度。采用Spearman相关分析评估血清CTRP3和DKK-1水平与PMOP患者发生骨折的相关性。采用多因素Logistic回归分析评估PMOP患者发生骨折的影响因素。采用受试者工作特征(ROC)曲线评估血清CTRP3、DKK-1水平和骨密度判断PMOP患者发生骨折的效能。结果 与正常对照组比较,PMOP组25-羟基维生素D[25-(OH)D]、CTRP3水平和骨密度显著降低(P<0.001),DKK-1水平显著升高(P<0.001),其他指标2个组之间差异均无统计学意义(P>0.05)。与无骨折组比较,骨折组骨密度和血清CTRP3水平显著降低(P<0.001),DKK-1水平显著升高(P<0.001)。血清CTRP3水平与PMOP患者发生骨折呈负相关(r=-0.574,P<0.05),血清DKK-1水平与PMOP患者发生骨折呈正相关(r=0.658,P<0.05)。DKK-1升高、CTRP3降低、骨密度降低均是PMOP患者发生骨折的危险因素[比值比(OR)值分别为2.249、0.529、0.424],95%可信区间(CI)分别为1.199~4.219、0.299~0.936、0.188~0.958,P<0.05。血清CTRP3、DKK-1、骨密度单项检测和联合预测判断PMOP患者发生骨折的曲线下面积(AUC)分别为0.871、0.904、0.806和0.974。结论 PMOP患者血清CTRP3水平显著降低,DKK-1显著升高。2项指标与骨密度联合检测对判断PMOP患者发生骨折有较高的效能,有助于临床对PMOP患者中的骨折高风险人群及时干预。 展开更多

目的探讨血清补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)、脂蛋白相关磷脂酶A2(Lp-PLA2)、可溶性髓样细胞触发受体-1(sTREM-1)水平在预测脑血管狭窄人群的颅脑动脉支架术后支架内再狭窄(ISR)方面的价值。方法选取2020年5月至2022年6月廊坊... 目的探讨血清补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)、脂蛋白相关磷脂酶A2(Lp-PLA2)、可溶性髓样细胞触发受体-1(sTREM-1)水平在预测脑血管狭窄人群的颅脑动脉支架术后支架内再狭窄(ISR)方面的价值。方法选取2020年5月至2022年6月廊坊市人民医院收治的107例接受颅脑动脉支架成形术的患者。根据患者术后6个月内是否发生ISR分为ISR组(n=26)与NISR组(n=81)。比较两组患者的低密度脂蛋白胆固醇(LDL-C)、支架数量、球囊扩张支架的占比、狭窄程度、残余狭窄程度,以及CTRP3、Lp-PLA2、sTREM-1水平。结果ISR组LDL-C、支架数量、球囊扩张支架的占比、狭窄程度、残余狭窄程度、Lp-PLA2水平、sTREM-1水平均高于NISR组,差异有统计学意义(P<0.05);ISR组CTRP3水平低于NISR组,差异有统计学意义(P<0.05)。支架数量多、狭窄程度及残余狭窄程度高、CTRP3水平降低、Lp-PLA2、LDL-C、sTREM-1水平上升为脑血管狭窄病患血管成形术后出现ISR的独立危险因素(P<0.05)。CTRP3、Lp-PLA2、sTREM-1三者联合在脑血管狭窄病患颅脑血管成形术后出现ISR的预测曲线下面积分别为0.840、0.784、0.809、0.899,三者联合预测价值显著高于单一指标。结论CTRP3水平降低,Lp-PLA2、sTREM-1水平升高与脑血管狭窄患者颅脑动脉支架术后发生ISR有关,联合指标检测对脑血管狭窄患者颅脑动脉支架术后发生ISR具有一定预测价值。 展开更多

连翘苷通过调控CTRP3表达对高糖诱导的人视网膜血管内皮细胞损伤的影响及其机制研究

目的比较不同剂量连翘苷(PHN)对高糖(HG)诱导的人视网膜血管内皮细胞损伤的影响,并分析其对补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)表达的调控作用及可能的作用机制。方法采用HG培养人视网膜血管内皮细胞并建立细胞损伤模型(HG组)。HG+PH... 目的比较不同剂量连翘苷(PHN)对高糖(HG)诱导的人视网膜血管内皮细胞损伤的影响,并分析其对补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)表达的调控作用及可能的作用机制。方法采用HG培养人视网膜血管内皮细胞并建立细胞损伤模型(HG组)。HG+PHN-L组、HG+PHN-M组、HG+PHN-H组人视网膜血管内皮细胞分别用1μmol·L^(-1)、10μmol·L^(-1)、100μmol·L^(-1)PHN处理后进行HG诱导。HG+pcDNA组、HG+pcDNA-CTRP3组分别用pcDNA、pcDNA-CTRP3转染至人视网膜血管内皮细胞后进行HG诱导。HG+PHN-H+sh-NC组、HG+PHN-H+sh-CTRP3组分别用sh-NC、sh-CTRP3转染后,用100μmol·L^(-1)PHN处理HG诱导的人视网膜血管内皮细胞。检测细胞丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平。采用流式细胞术检测细胞凋亡率。采用实时荧光定量聚合酶链反应(qRT-PCR)、Western blot分别检测B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关蛋白(Bax)、CTRP3蛋白表达水平。结果与Con组比较,HG组细胞凋亡率、MDA水平、Bax蛋白水平均升高,SOD、GSH-Px、Bcl-2蛋白、CTRP3 mRNA及蛋白水平均降低(均为P<0.05)。与HG组比较,HG+PHN-L组、HG+PHN-M组、HG+PHN-H组细胞凋亡率、MDA水平、Bax蛋白水平均降低,且HG+PHN-H组<HG+PHN-M组<HG+PHN-L组(均为P<0.05),SOD、GSH-Px、Bcl-2蛋白、CTRP3 mRNA及蛋白水平均升高,且HG+PHN-H组>HG+PHN-M组>HG+PHN-L组(均为P<0.05)。与HG+pcDNA组比较,HG+pcDNA-CTRP3组细胞凋亡率、MDA水平、Bax蛋白水平均降低,SOD、GSH-Px、CTRP3蛋白、Bcl-2蛋白水平均升高(均为P<0.05)。与HG+PHN-H+sh-NC组比较,HG+PHN-H+sh-CTRP3组细胞凋亡率、MDA水平、Bax蛋白水平均升高,SOD水平、GSH-Px水平、CTRP3蛋白、Bcl-2蛋白水平均降低(均为P<0.05)。结论PHN可减轻HG诱导的人视网膜血管内皮细胞损伤,其作用机制可能与上调CTRP3表达有关。 展开更多

Correlation of the abnormal expression of HK-II and TNFAIP3 in diffuse large B-cell lymphoma with the malignant features of tumor cells

Objective: To study the correlation of the abnormal expression of hexokinase-Ⅱ (HK-Ⅱ) and tumor necrosis factor alpha-induced protein 3 (TNFAIP3) in diffuse large B-cell lymphoma (DLBCL) with the malignant features of tumor cells. Methods: DLBCL patients who underwent surgical resection in our hospital between March 2016 and March 2018 were selected as the DLBCL group, and the patients who underwent surgical resection during the same period and were pathologically confirmed as reactive hyperplasia of lymph nodes were selected as the control group. The lesions were collected to measure the expression levels of HK-Ⅱ, TNFAIP3, proliferation genes and invasion genes. Results: HK-Ⅱ mRNA expression level in the lesions of the DLBCL group was significantly higher than that of the control group while TNFAIP3 mRNA expression level was significantly lower than that of the control group;HK-Ⅱ mRNA expression levels in the lesions of DLBCL group of patients with lymphoma stage ⅡI-IV and lymphoma group B were significantly higher than those of the patients with lymphoma stage I-Ⅱ and group A while TNFAIP3 mRNA expression levels were significantly lower than those of the patients with lymphoma stage I-Ⅱ and lymphoma group A;CyclinD2, PDE4B, BCL2, XIAP, CCL5, CXCR4 and MMP26 mRNA expression levels in the lesions of the DLBCL group were significantly higher than those of the control group, positively correlated with HK-Ⅱ and negatively correlated with TNFAIP3 while Caspase-3 and TIMP4 mRNA expression levels were significantly lower than those of the control group, negatively correlated with HK-Ⅱ and positively correlated with TNFAIP3. Conclusion: The high expression of HK-Ⅱ and the low expression of TNFAIP3 in DLBCL are closely related to the pathological process of tumor as well as the proliferation and invasion of tumor cells. 展开更多

目的 探讨血清补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)、波形蛋白(VTN)水平与扩张型心肌病慢性心力衰竭患者预后的关系。方法 选取2021年1月至2022年8月河北省邢台市中心医院收治的扩张型心肌病慢性心力衰竭患者179例,根据1年预后分为预... 目的 探讨血清补体C1q/肿瘤坏死因子相关蛋白3(CTRP3)、波形蛋白(VTN)水平与扩张型心肌病慢性心力衰竭患者预后的关系。方法 选取2021年1月至2022年8月河北省邢台市中心医院收治的扩张型心肌病慢性心力衰竭患者179例,根据1年预后分为预后不良组(63例)和预后良好组(116例)。采用酶联免疫吸附试验检测血清CTRP3、VTN水平。单因素和多因素logistic回归分析影响扩张型心肌病慢性心力衰竭患者预后的因素,受试者操作特征曲线分析血清CTRP3、VTN水平对扩张型心肌病慢性心力衰竭患者预后不良的预测价值。结果 随访1年,179例扩张型心肌病慢性心力衰竭患者预后不良发生率为35.20%(63/179)。两组年龄、慢性心力衰竭病程、心功能分级、左室射血分数(LVEF)、CTRP3、VTN比较,差异有统计学意义(P<0.05)。两组性别、吸烟史、饮酒史、合并疾病、收缩压、舒张压、治疗药物、器械辅助治疗比较,差异无统计学意义(P>0.05)。年龄增加[OR(95%CI):1.090(1.007~1.179)]、心功能分级增加[OR(95%CI):3.868(1.405~10.652)]、VTN升高[OR(95%CI):1.101(1.039~1.167)],LVEF增加[OR(95%CI):0.680(0.543~0.850)]和CTRP3升高[OR(95%CI):0.946(0.916~0.978)]为扩张型心肌病慢性心力衰竭患者预后不良的影响因素(P<0.05)。血清CTRP3联合VTN水平预测扩张型心肌病慢性心力衰竭患者预后不良的曲线下面积(0.878)大于血清CTRP3、VTN水平单独预测的0.782、0.787(Z=3.779、3.546,P<0.05)。结论 扩张型心肌病慢性心力衰竭患者血清CTRP3水平降低、VTN水平升高与预后不良密切相关,二者联合预测扩张型心肌病慢性心力衰竭患者预后不良的价值较高。 展开更多

肠病药方调控PI3K/AKT/NF-κB信号通路改善溃疡性结肠炎的研究

目的探讨肠病药方调控磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)/核因子kappa B(NF-κB)信号通路对抗溃疡性结肠炎(UC)的作用机制。方法42只C57BL/6小鼠随机分为正常对照组(n=6,Control组,自由饮去离子水)、模型组(n=12,DSS组)、美沙拉嗪... 目的探讨肠病药方调控磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)/核因子kappa B(NF-κB)信号通路对抗溃疡性结肠炎(UC)的作用机制。方法42只C57BL/6小鼠随机分为正常对照组(n=6,Control组,自由饮去离子水)、模型组(n=12,DSS组)、美沙拉嗪组(n=12,MES组)及肠病药方组(n=12,CBD组),后3组小鼠自由饮用3%葡聚糖硫酸钠(DSS)溶液7 d诱导UC模型,后两组同期灌胃美沙拉嗪或肠病药方,前两组灌胃等体积去离子水;观察实验期间小鼠体质量改变,评测小鼠疾病活动指数(DAI);造模结束后麻醉处死小鼠,测量结肠长度、观察结肠内容物及黏膜,采用苏木素-伊红(HE)染色观察各组小鼠结肠病理组织学变化,酶联免疫吸附试验(ELISA)检测各组小鼠结肠组织匀浆白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,蛋白免疫印迹法(WB)检测各组小鼠结肠组织中磷酸化PI3K(p-PI3K)-p85、磷酸化AKT1(p-AKT1)、磷酸化NF-κB(p-NF-κB)-p65蛋白表达,实时荧光定量PCR技术(qRT-PCT)检测各组小鼠结肠组织PI3K、AKT、NF-κB信使RNA(mRNA)表达水平。结果与DSS组比较,CBD组治疗后,有效缓解UC小鼠症状,粪便性状及便血情况改善,结肠黏膜炎症浸润减少,体质量下降有所缓解,DAI评分降低及结肠长度接近正常,结肠组织匀浆中IL-6、TNF-α表达降低(P<0.05),p-PI3K-p85、p-AKT1、p-NF-κB-p65蛋白及mRNA表达明显减少(P<0.05)。结论肠病药方可缓解UC小鼠的症状、减轻结肠黏膜损伤,其机制可能与调解结肠组织中PI3K/AKT/NF-κB信号通路中相关蛋白质及mRNA表达、降低结肠组织炎症因子表达有关。 展开更多
