The results showed that the qualities and active substance content of mulberry leaves changed from increasing to decreasing with its development; crude protein and solu-ble sugar achieved the peak on August 20; total flavonoid and 1-deoxynojirimycin was the highest on May 15. Fertilizations with N, P and K fertilizers at different amounts had significant effects on quality of mulberry leaf and content of active substances. Specifically, as fertilizer amount increased, the content of active sub- stances grew dramatically and achieved the highest at level 2 (X^2X=X~_). Based on fertilizer effect functions of objective yield, the recommended amounts of N, P and K fertilizers based on crude protein, soluble sugar, flavonoid content and DNJ in test sites were 718.46, 220.11 and 305.23 kg/hm2, when the highest of crude protein in mulberry leaf was 1 813.83 kg/hm2. When N, P and K fertilizers were recommended at 666.54, 204.41 and 243.18 kg/hm2, soluble sugar in mulberry leaf achieved the peak at 1 042.65 kg/hm2. When N, P and K fertilizers were at 675.96, 326.49 and 462.90 kg/hm2, flavonoid content achieved the maximum at 147.90 kg/hm2. When N, P and K fertilizers were at 720.9, 225.11 and 323.63 kg/hm2, DNJ content was the highest at 13.55 kg/hm2.展开更多

In order to further study its control mechanism, the sin- gle-factor tests of temperature and pH value, the activated carbon adsorption test, the organic solvent extraction test and the crude extract of active substance test were carried out to explore the characteristics of active substances. The results showed that the algae-lysing active ingredient of A1 strain had strong thermal stabili- ty, which still had good algae-lysing effect after being treated at 121 ℃ ; when the pH values of fermentation liquid were adjusted to 2.0 and 4.0 respectively, the active substances lost their activities, but their control ability against moss would be enhanced under the alkaline conditions ; the active substances could not be ad- sorbed by activated carbon; the algae-lysing active substances showed strong hydrophilie ability when they were extracted by ethyl acetate, petroleum ether and chloroform. Therefore, it could be speculated that the active substances belonged to carbohydrates. The crude extract results of algae-lysing active substances showed that the active substances secreted by A1 strain were composed by a variety of algae-lysing active substances.展开更多

Kuntze, the isolation and structural identification were researched in this paper. The crude extract from the root ofF. bidentis (L.) Kuntze was extracted by petroleum ether, ethyl acetate, and water saturation of n-butyl alcohol, respectively, and the extraction fluid was separated by using the method of TLC, then the main fraction was separated by HPLC, and the structure of the herbicidal active substance was analyzed by LC-MS, elemental analysis and ~H-NMR. The results showed that the petroleum extraction had the strongest herbicidal activity, and the purple blue stripe separated by TLC had the strongest effect on Digitaria sanguinalis. The herbicidal active substance was identified as ct-terthienyl according to the data of LC-MS, elemental analysis and 1H-NMR.展开更多
In the present review, we first summarize the main benefits, limitations and pitfalls of conventional in vivo approaches to assessing male reproductive structures and functions in rodents in cases of endocrine active ...In the present review, we first summarize the main benefits, limitations and pitfalls of conventional in vivo approaches to assessing male reproductive structures and functions in rodents in cases of endocrine active substance (EAS) exposure from the postulate that they may provide data that can be extrapolated to humans. Then, we briefly present some integrated approaches in rodents we have recently developed at the organism level. We particularly focus on the possible effects and modes of action (MOA) of these substances at low doses and in mixtures, real-life conditions and at the organ level, deciphering the precise effects and MOA on the fetal testis. It can be considered that the in vivo experimental EAS exposure of rodents remains the first choice for studies and is a necessary tool (together with the epidemiological approach) for understanding the reproductive effects and MOA of EASs, provided the pitfalls and limitations of the rodent models are known and considered. We also provide some evidence that classical rodent models may be refined for studying the multiple consequences of EAS exposure, not only on the reproductive axis but also on various hormonally regulated organs and tissues, among which several are implicated in the complex process of mammalian reproduction. Such models constitute an interesting way of approaching human exposure conditions. Finally, we show that organotypic culture models are powerful complementary tools, especially when focusing on the MOA. All these approaches have contributed in a combinatorial manner to a better understanding of the impact of EAS exposure on human reproduction.展开更多

TLC, UV Spectroscopy and differential spectrophotometry methods of analysis were employed in this study. Identified BAS included flavonoids, polysaccharides and amino acids. Following state pharmacopoeia of Ukraine (SPU) requirements to validate the developed techniques were employed in this study, and the following validation properties specificity, linearity, range, precision accuracy (convergence) and robustness was determined. These characteristics did not exceed the eligibility criteria as stated in (SPU) Appendix 2. Thus the technique can be made to the SPU monograph on fruits of <i>Tetrapleura tetraptera</i>.展开更多
Background: Psychoactive substance use was common among commercial motorcycle operators. Studies had shown that there were some socio-demographic factors which were commoner among commercial motorcycle operators who used psychoactive substances. However, this was not explored among commercial motorcycle operators in Kano, North Western Nigeria. Aim: The aim of the study was to determine the socio-demographic factors associated with psychoactive substance use among motorcycle operators in Kano, Northwestern Nigeria. Methods: It was a cross-sectional study. Instrument used for data collection included socio-demographic questionnaire and World Health Organisation Student Drug Use Questionnaire. Data analysis was done using Statistical Package for Social Sciences (SPSS) 16th edition. Results: Commercial motorcycle operators who used substances were found to be of younger age group (X<sup>2</sup> = 10.9, P < 0.001), single (X<sup>2</sup> = 4.3, P = 0.034) and had a father who used tobacco (X<sup>2</sup> = 4.28, P = 0.04). Participants who were single were more likely to use inhalants (X<sup>2</sup> = 21.06, P =0.001), opiates (X<sup>2</sup> = 11.82, P = 0.012), cannabis (X<sup>2</sup> = 5.12, P = 0.028) and stimulants (X<sup>2</sup> = 5.12, P = 0.028) compared to those who were married. Conclusion: This study found that being of younger age, being single, and having a father who smoked were significantly associated with psychoactive substance use. Participants who were singles were more likely to use inhalants, opiates, cannabis and stimulants. No relationship was observed between the age of the respondents and specific substance use. Legislations should be enacted to reduce the rate of psychoactive substance use among motorcycle operators and general population at large.展开更多

[Method] The efficiency of five methods (H2SO4, formaldehyde-NaOH, mixing, heating and NaOH) on the extraction of EPS was investigated comparatively. The optimal extraction conditions of the most suitable method were determined. [Result] NaOH method is most effective in extracting EPS with less DNA contamination and shortened extraction period. The optimal extraction condition was pH of 11, extraction time of 10 min and agitation speed of 80-120 r/min. [Conclusion] The determined optimal extraction condition provided theoretical basis for EPS study.展开更多

They play a crucial role in the flocculation, settling and dewat...Extracellular polymeric substances (EPS) are the predominant constituents of activated sludge and represent up to 80% of the mass of activated sludge. They play a crucial role in the flocculation, settling and dewatering of activated sludge. Furthermore, EPS also show great efficiency in binding heavy metals. So EPS are key factors influencing reduction in sludge volume and mass, as well as activity and utilization of sludge. EPS are of considerable environmental interest and hundreds of articles on EPS have been published abroad, while information on EPS in China is limited. In this paper, results of over 60 publications related to constituents and characteristics of EPS and their influences on flocculation, settling and dewatering of sludge are compiled and analyzed. Metal-binding ability of EPS is also discussed, together with a brief consideration of possible research interests in the future.展开更多
Flos Sophorae Immaturus(FSI)extract and its main active secondary metabolites have health benefits including antioxidant activity,but the drying process may negatively affect their quality and quantity.To investigate ...Flos Sophorae Immaturus(FSI)extract and its main active secondary metabolites have health benefits including antioxidant activity,but the drying process may negatively affect their quality and quantity.To investigate the effect of ultrasound-assisted freeze-drying(UAFD)on FSI,the drying rate,colour,and microstructure samples were analysed.Furthermore,the flavonoids and antioxidant activity of dried products were evaluated.The results showed that the drying rates were significantly enhanced with UAFD.At 600 W and 15 min,the drying time of UAFD was 40%shorter than that of freeze-drying(FD)alone.The micrographs showed that USAD created more porous structures that allowed water to migrate outward from inside the FSI tissue.Processing conditions of 600 W ultrasonic power and 5 min ultrasonication time produced better antioxidant activity in dried products than in other conditions.The highest characteristic properties were DPPH(75.51%),ABTS(95.14%),FRAP(83.05%),acceptable drying time(3 h),and low colour difference(1.09).Under these conditions,the contents of total flavonoids(298.34 mg/g),rutin(256.66 mg/g),quercetin(20.13 mg/g),kaempferol(19.64 mg/g),isorhamnetin(17.77 mg/g),and genistein(13.97 mg/g)were determined to be as indicated.The results confirmed the obvious benefits of incorporating the ultrasonic effect into the FD method.展开更多

Sanya (China), then extracted the antibacterial substances using 10% glacial acetic acid with the help of antiprotease inhibitors, and tested the antibacterial activity by a terrestrial bacterium (Staphylococcus aurevs) and a marine bacterium (Vibrio anguillarum). The result showed that, there were antibacterial agents in the mucus, and their antibacterial activities were lost by treatment of the sample at 90 ℃ water for 10 min.展开更多

The strain SLYY- 3, which produced a bacteriocin-like substance (BLS), was characterized to be a strain of Baci...In the present research, the strain SLYY-3 was isolated from sediments of Jiaozhou Bay, Qingdao, China. The strain SLYY- 3, which produced a bacteriocin-like substance (BLS), was characterized to be a strain of Bacillus subtillis by biochemical profiling and 16S rDNA sequence analysis. It is the first time to report that Bacillus subtilis from Jiaozhou Bay sediments could produce a BLS. The BLS of B. subtillis SLYY-3 exhibited strong inhibitory activity against gram-positive bacteria (including Staphylococcus aureus and B. subtillis) and some fimgi (including Penicillium glaucum, Aspergillus niger and Aspergillus flavus). The antimicrobial activity was detected from culture in the exponential growth phase and reached its maximum when culture entered into stationary growth phase. It was thermo-tolerant even when being kept at 100~C for 60 min without losing any activity and stable over a wide pH range from 1.0 to 12.0 while being inactivated by proteolytic enzyme and trypsin, indicating the proteinaceous nature of the BLS. The BLS was purified by precipitation with hydrochloric acid (HC1) and gel filteration (Sephadex G-100). SDS-PAGE analysis of the extracellular peptides of SLYY-3 revealed a bacteriocin-like protein with a molecular mass of 66 kDa. Altogether, these characteristics indicate the potential of the BLS for food industry as a protection against pathogenic and spoilage microorganisms.展开更多

A bacterial strain with high flocculating activity was isolated from seawater samples. The strain was defined as Bacillus licheniformis dhs-40 by 16S rDNA identification and Biolog test. Ultrasonication test confirmed the flocculating activity of the strain was both in fermentation supernatant and cell. According to flocculating activity curve, the ideal fermentation time for collecting flocculating active substances was two days. The flocculating activity of the strain was sensitive to pH. The strain could only preserve flocculating activity while pH varied from 7 to 11. However, it could preserve flocculating activity while temperature varied from -20℃to 100 ℃ Saccharide, protein, lipid, nucleic acids qualitative test and RNase, Proteinase K treatment confirmed the flocculating active substances were proteins. Their flocculating activities were insensitive to Proteinase K.展开更多

Overcoming seed dormancy is an important component of efficient and cost-effective seedling production of Cyclocarya paliurus. Changes in biochemical composition and enzyme activity were investigated during dormancy release. The activities of all the studied enzymes in the stratified seeds increased significantly, compared to those in the control samples. Of the enzymes examined, the activities of protease increased the most (413.8%), followed by peroxidase (278.7%), lipase (161.0%), glucose-6-phosphate dehydrognase (149.1%) and amylase (60.6%) after 8 months of stratification. Crude fat and protein constituted the bulk of the storage reserves in mature seeds of C. pal# urus. Compared with the seeds before stratification, about 45% of the starch, 46% of the protein and 11% of the crude fat were depleted during dormancy release of C. paliurus seeds, while the soluble sugar content was enhanced by 101.5% in the germinating seeds. Correlation analysis showed, during dormancy release of C. paliurus seeds, a close positive relationship between POD and G6PDH activity as well as soluble sugar content and amylase activity, while there was a significant negative relationship between storage substances and their related enzyme activities.展开更多

Con...Correlations between estrogenic activity and DOC/UV260 ratio in wastewater treatment processes were investigated to propose a simple, reliable and comprehensive indicator for the presence of estrogenic substances. Contrary to this, when short-term bioassays such as the E-SCREEN, receptor binding and reporter gene expression assays are used for detecting estrogenic activity in the wastewater sample, they require a long time, at least a few days. The major factors contributing to the estrogenic activity were found to be 17β-estradiol (E2) and estrone (El). A good relationship between the DOC/ UV260 ratio and the concentration of estrogens (El and E2) in the effluent of the activated sludge process was found: the E2 concentration increased as the DOC/UV260 ratio increased while the E1 concentration decreased. The relative estrogenic activity and DOC/UV260 ratio showed a good correlation (R^2 = 0.84) for all sewage samples except the ozonized samples in the sewage treatment plants. This study shows that the estrogenic compounds are hard to be mineralized by the conventional biological processes. Advanced oxidation processes are required to further remove estrogenic substances in the secondary effluent. By analysis of DOC and UV260, the estrogenic activity in the wastewater can be rapidly estimated.展开更多

Aims: The representativeness of the mandatory bacterial strains specified in European standards for in vitro assay of the bactericidal activity of antiseptics was evaluated by testing the activity of an antiseptic com...Aims: The representativeness of the mandatory bacterial strains specified in European standards for in vitro assay of the bactericidal activity of antiseptics was evaluated by testing the activity of an antiseptic combining chlorhexidine digluconate 0.2% and benzalkonium chloride 0.5% against 21 additional bacterial strains, and the positive interaction between these two biocidal agents was assessed. Methods and Results: The bactericidal activity of the antiseptic solution used pure or diluted was assessed according to the European standards EN 1040 and EN 13727. The contact time was 1 min at 20°C. Interfering substances used in the EN 13727 assay were bovine serum albumin and sheep erythrocytes, simulating “dirty” conditions, and hard water. A reduction of colony-forming units by ≥5 log10 was deemed to meet the requirements to conclude bactericidal activity. Under “basic” conditions, the bactericidal activity of the antiseptic was observed against all four mandatory strains specified in the standards as well as against nearly all the additional strains tested, including most of those with acquired antibiotic-resistance. The positive interaction between the two biocidal agents was also confirmed. Under “dirty” conditions, the bactericidal activity of the antiseptic solution was maintained against all the mandatory strains and was reduced against only four of the additional strains tested. Conclusions: With regard to the antiseptic tested and under the experimental conditions described, bactericidal activity evidenced against the mandatory strains appeared to be representative of that manifested against a wide range of the main pathogenic bacteria. Reduced bacterial activity against some of the additional strains tested (e.g. Enterobacteriaceae) was observed under “dirty” conditions. Significance and Impact of the Study: EN 13727 with some experimental adjustments represents an additional appropriate standard that needs to be considered for mucocutaneous antiseptic assessment. However, it may be worth including other specific bacterial strains to those specified in the standard, when evaluating antiseptics intended for use in certain clinical situations.展开更多

Bearberry(Arctostaphylos uva-ursi L.)is a perennial plant of the heather family(Ericaceae).The leaves are dominated by arbutin,phenol carbonic acids flavonoids,saponins,etc.It was previously shown that the bearberry l...Bearberry(Arctostaphylos uva-ursi L.)is a perennial plant of the heather family(Ericaceae).The leaves are dominated by arbutin,phenol carbonic acids flavonoids,saponins,etc.It was previously shown that the bearberry leaves extract reduced blood glucose level in healthy animals under glucose overload,so it need to be studied more detail.The aim of the study was to investigate the chemical composition and the effect of dry alcohol extract from bearberry leaves,which enriched with cysteine,on the rats pancreas under experimental dexamethasone-induced insulin resistance(IR).Arctostaphylos uva-ursi L.leaves extract and its modified cysteine preparation were obtained according to the developed method with 50%ethanol solution.Their phytochemical profile,hypoglycaemic and pancreatic protective effect were investigated.Phenologlycoside(arbutin),phenolic carboxylic acid(gallic acid),5 flavonoids and 4 hydroxy-cinnamic acids were identified and quantified in the extracts by HPLC.Present data revealed that bearberry leaves alcoholic dry extract enriched with cysteine has a hypoglycaemic and pancreatic protective effect in treated animals under dexamethasone-induced IR model.Treatment improved hyperglycaemia,insulin resistance and beta cell reduc-tion induced by dexamethasone injections.展开更多

The aim of our work is to present extraction technics of the bioactive substances from gleba (ascoms) of Tirmania pinoyi (Maire) Malen?on and from the culture filtrate of mycelium of Terfezia claveryi Chatin. Also, we present antifungal and antibacterial activity of these substances in vitro and their identification. There is antifungal activity of different extracts on solid medium, in vitro, against species of Aspergillus, Fusarium, Gliocladium, Candida. Also, there is antibacterial activity against species of Staphylococus, Streptococcus, Bacillus, Enterococcus, Escherichia, and Salmonella. Results show that extracts inhibition varies according to microbial tested species. Bioactive substances of the different extracts were analyzed by GC (gas chromatography)-mass, RMN (Nuclear Magnetic Resonance) and infrared in order to determine the chemical identity. The results showed that the algae-lysing active ingredient of A1 strain had strong thermal stabili- ty, which still had good algae-lysing effect after being treated at 121 ℃ ; when the pH values of fermentation liquid were adjusted to 2.0 and 4.0 respectively, the active substances lost their activities, but their control ability against moss would be enhanced under the alkaline conditions ; the active substances could not be ad- sorbed by activated carbon; the algae-lysing active substances showed strong hydrophilie ability when they were extracted by ethyl acetate, petroleum ether and chloroform. Therefore, it could be speculated that the active substances belonged to carbohydrates. The crude extract results of algae-lysing active substances showed that the active substances secreted by A1 strain were composed by a variety of algae-lysing active substances.

文摘In order to understand the composition and structure of herbicidal active substance from the root of Flaveria bidentis (L.) Kuntze, the isolation and structural identification were researched in this paper. The crude extract from the root ofF. bidentis (L.) Kuntze was extracted by petroleum ether, ethyl acetate, and water saturation of n-butyl alcohol, respectively, and the extraction fluid was separated by using the method of TLC, then the main fraction was separated by HPLC, and the structure of the herbicidal active substance was analyzed by LC-MS, elemental analysis and ~H-NMR. The results showed that the petroleum extraction had the strongest herbicidal activity, and the purple blue stripe separated by TLC had the strongest effect on Digitaria sanguinalis. The herbicidal active substance was identified as ct-terthienyl according to the data of LC-MS, elemental analysis and 1H-NMR.
文摘In the present review, we first summarize the main benefits, limitations and pitfalls of conventional in vivo approaches to assessing male reproductive structures and functions in rodents in cases of endocrine active substance (EAS) exposure from the postulate that they may provide data that can be extrapolated to humans. Then, we briefly present some integrated approaches in rodents we have recently developed at the organism level. We particularly focus on the possible effects and modes of action (MOA) of these substances at low doses and in mixtures, real-life conditions and at the organ level, deciphering the precise effects and MOA on the fetal testis. It can be considered that the in vivo experimental EAS exposure of rodents remains the first choice for studies and is a necessary tool (together with the epidemiological approach) for understanding the reproductive effects and MOA of EASs, provided the pitfalls and limitations of the rodent models are known and considered. We also provide some evidence that classical rodent models may be refined for studying the multiple consequences of EAS exposure, not only on the reproductive axis but also on various hormonally regulated organs and tissues, among which several are implicated in the complex process of mammalian reproduction. Such models constitute an interesting way of approaching human exposure conditions. Finally, we show that organotypic culture models are powerful complementary tools, especially when focusing on the MOA. All these approaches have contributed in a combinatorial manner to a better understanding of the impact of EAS exposure on human reproduction.
文摘This study was conducted to identify the biological active substances (BAS) from the fruit of <i>Tetrapleura tetraptera</i> obtained from Ghana. TLC, UV Spectroscopy and differential spectrophotometry methods of analysis were employed in this study. Identified BAS included flavonoids, polysaccharides and amino acids. Following state pharmacopoeia of Ukraine (SPU) requirements to validate the developed techniques were employed in this study, and the following validation properties specificity, linearity, range, precision accuracy (convergence) and robustness was determined. These characteristics did not exceed the eligibility criteria as stated in (SPU) Appendix 2. Thus the technique can be made to the SPU monograph on fruits of <i>Tetrapleura tetraptera</i>.
文摘Background: Psychoactive substance use was common among commercial motorcycle operators. Studies had shown that there were some socio-demographic factors which were commoner among commercial motorcycle operators who used psychoactive substances. However, this was not explored among commercial motorcycle operators in Kano, North Western Nigeria. Aim: The aim of the study was to determine the socio-demographic factors associated with psychoactive substance use among motorcycle operators in Kano, Northwestern Nigeria. Methods: It was a cross-sectional study. Instrument used for data collection included socio-demographic questionnaire and World Health Organisation Student Drug Use Questionnaire. Data analysis was done using Statistical Package for Social Sciences (SPSS) 16th edition. Results: Commercial motorcycle operators who used substances were found to be of younger age group (X<sup>2</sup> = 10.9, P < 0.001), single (X<sup>2</sup> = 4.3, P = 0.034) and had a father who used tobacco (X<sup>2</sup> = 4.28, P = 0.04). Participants who were single were more likely to use inhalants (X<sup>2</sup> = 21.06, P =0.001), opiates (X<sup>2</sup> = 11.82, P = 0.012), cannabis (X<sup>2</sup> = 5.12, P = 0.028) and stimulants (X<sup>2</sup> = 5.12, P = 0.028) compared to those who were married. Conclusion: This study found that being of younger age, being single, and having a father who smoked were significantly associated with psychoactive substance use. Participants who were singles were more likely to use inhalants, opiates, cannabis and stimulants. No relationship was observed between the age of the respondents and specific substance use. Legislations should be enacted to reduce the rate of psychoactive substance use among motorcycle operators and general population at large.

文摘[Objective] To explore the optimal extraction conditions of extracellular polymeric substances (EPS) from activated sludge. [Method] The efficiency of five methods (H2SO4, formaldehyde-NaOH, mixing, heating and NaOH) on the extraction of EPS was investigated comparatively. The optimal extraction conditions of the most suitable method were determined. [Result] NaOH method is most effective in extracting EPS with less DNA contamination and shortened extraction period. The optimal extraction condition was pH of 11, extraction time of 10 min and agitation speed of 80-120 r/min. [Conclusion] The determined optimal extraction condition provided theoretical basis for EPS study.

文摘Extracellular polymeric substances (EPS) are the predominant constituents of activated sludge and represent up to 80% of the mass of activated sludge. They play a crucial role in the flocculation, settling and dewatering of activated sludge. Furthermore, EPS also show great efficiency in binding heavy metals. So EPS are key factors influencing reduction in sludge volume and mass, as well as activity and utilization of sludge. EPS are of considerable environmental interest and hundreds of articles on EPS have been published abroad, while information on EPS in China is limited. In this paper, results of over 60 publications related to constituents and characteristics of EPS and their influences on flocculation, settling and dewatering of sludge are compiled and analyzed. Metal-binding ability of EPS is also discussed, together with a brief consideration of possible research interests in the future.

文摘Flos Sophorae Immaturus(FSI)extract and its main active secondary metabolites have health benefits including antioxidant activity,but the drying process may negatively affect their quality and quantity.To investigate the effect of ultrasound-assisted freeze-drying(UAFD)on FSI,the drying rate,colour,and microstructure samples were analysed.Furthermore,the flavonoids and antioxidant activity of dried products were evaluated.The results showed that the drying rates were significantly enhanced with UAFD.At 600 W and 15 min,the drying time of UAFD was 40%shorter than that of freeze-drying(FD)alone.The micrographs showed that USAD created more porous structures that allowed water to migrate outward from inside the FSI tissue.Processing conditions of 600 W ultrasonic power and 5 min ultrasonication time produced better antioxidant activity in dried products than in other conditions.The highest characteristic properties were DPPH(75.51%),ABTS(95.14%),FRAP(83.05%),acceptable drying time(3 h),and low colour difference(1.09).Under these conditions,the contents of total flavonoids(298.34 mg/g),rutin(256.66 mg/g),quercetin(20.13 mg/g),kaempferol(19.64 mg/g),isorhamnetin(17.77 mg/g),and genistein(13.97 mg/g)were determined to be as indicated.The results confirmed the obvious benefits of incorporating the ultrasonic effect into the FD method.

文摘Coral mucus covers the surface of coral and contains antibacterial substances as a first line of defense. Coral mucus not only enables the coral itself to resist disease, but also provides antibacterial agents for people. We collected mucus from a scleractinian coral (Symphyllia gigantea) at Sta. Sanya (China), then extracted the antibacterial substances using 10% glacial acetic acid with the help of antiprotease inhibitors, and tested the antibacterial activity by a terrestrial bacterium (Staphylococcus aurevs) and a marine bacterium (Vibrio anguillarum). The result showed that, there were antibacterial agents in the mucus, and their antibacterial activities were lost by treatment of the sample at 90 ℃ water for 10 min.

文摘In the present research, the strain SLYY-3 was isolated from sediments of Jiaozhou Bay, Qingdao, China. The strain SLYY- 3, which produced a bacteriocin-like substance (BLS), was characterized to be a strain of Bacillus subtillis by biochemical profiling and 16S rDNA sequence analysis. It is the first time to report that Bacillus subtilis from Jiaozhou Bay sediments could produce a BLS. The BLS of B. subtillis SLYY-3 exhibited strong inhibitory activity against gram-positive bacteria (including Staphylococcus aureus and B. subtillis) and some fimgi (including Penicillium glaucum, Aspergillus niger and Aspergillus flavus). The antimicrobial activity was detected from culture in the exponential growth phase and reached its maximum when culture entered into stationary growth phase. It was thermo-tolerant even when being kept at 100~C for 60 min without losing any activity and stable over a wide pH range from 1.0 to 12.0 while being inactivated by proteolytic enzyme and trypsin, indicating the proteinaceous nature of the BLS. The BLS was purified by precipitation with hydrochloric acid (HC1) and gel filteration (Sephadex G-100). SDS-PAGE analysis of the extracellular peptides of SLYY-3 revealed a bacteriocin-like protein with a molecular mass of 66 kDa. Altogether, these characteristics indicate the potential of the BLS for food industry as a protection against pathogenic and spoilage microorganisms.
文摘Microbial flocculants have become a hot spot in recent years. A bacterial strain with high flocculating activity was isolated from seawater samples. The strain was defined as Bacillus licheniformis dhs-40 by 16S rDNA identification and Biolog test. Ultrasonication test confirmed the flocculating activity of the strain was both in fermentation supernatant and cell. According to flocculating activity curve, the ideal fermentation time for collecting flocculating active substances was two days. The flocculating activity of the strain was sensitive to pH. The strain could only preserve flocculating activity while pH varied from 7 to 11. However, it could preserve flocculating activity while temperature varied from -20℃to 100 ℃ Saccharide, protein, lipid, nucleic acids qualitative test and RNase, Proteinase K treatment confirmed the flocculating active substances were proteins. Their flocculating activities were insensitive to Proteinase K.

文摘Cyclocarya paliurus is only propagated from seeds which have pronounced dormancy. Overcoming seed dormancy is an important component of efficient and cost-effective seedling production of Cyclocarya paliurus. Changes in biochemical composition and enzyme activity were investigated during dormancy release. The activities of all the studied enzymes in the stratified seeds increased significantly, compared to those in the control samples. Of the enzymes examined, the activities of protease increased the most (413.8%), followed by peroxidase (278.7%), lipase (161.0%), glucose-6-phosphate dehydrognase (149.1%) and amylase (60.6%) after 8 months of stratification. Crude fat and protein constituted the bulk of the storage reserves in mature seeds of C. pal# urus. Compared with the seeds before stratification, about 45% of the starch, 46% of the protein and 11% of the crude fat were depleted during dormancy release of C. paliurus seeds, while the soluble sugar content was enhanced by 101.5% in the germinating seeds. Correlation analysis showed, during dormancy release of C. paliurus seeds, a close positive relationship between POD and G6PDH activity as well as soluble sugar content and amylase activity, while there was a significant negative relationship between storage substances and their related enzyme activities.
文摘Correlations between estrogenic activity and DOC/UV260 ratio in wastewater treatment processes were investigated to propose a simple, reliable and comprehensive indicator for the presence of estrogenic substances. Contrary to this, when short-term bioassays such as the E-SCREEN, receptor binding and reporter gene expression assays are used for detecting estrogenic activity in the wastewater sample, they require a long time, at least a few days. The major factors contributing to the estrogenic activity were found to be 17β-estradiol (E2) and estrone (El). A good relationship between the DOC/ UV260 ratio and the concentration of estrogens (El and E2) in the effluent of the activated sludge process was found: the E2 concentration increased as the DOC/UV260 ratio increased while the E1 concentration decreased. The relative estrogenic activity and DOC/UV260 ratio showed a good correlation (R^2 = 0.84) for all sewage samples except the ozonized samples in the sewage treatment plants. This study shows that the estrogenic compounds are hard to be mineralized by the conventional biological processes. Advanced oxidation processes are required to further remove estrogenic substances in the secondary effluent. By analysis of DOC and UV260, the estrogenic activity in the wastewater can be rapidly estimated.

文摘Ideally,packaging materials should ensure the safety and quality of foods,without contributing to environmental degradation.Consequently,there is interest in the development of biodegradable films assembled from natural materials,such as polysaccharides,proteins,lipids,and their mixtures.In this review,the physicochemical attributes and functional characteristics of biodegradable films prepared from these food-grade natural substances are summarized.Recent advances in the production of active and intelligent packaging materials are also discussed.Active packaging is designed to improve the shelf life of packaged foods by including antimicrobials or antioxidants,such as essential oils.Intelligent packaging is designed to provide real-time information about the quality,freshness,or safety of packaged foods by including indicators in the film that are responsive to changes in storage conditions,gas levels,pH,etc.Potential applications of intelligent and active packaging materials to fruit,vegetable,meat,seafood,and dairy products are discussed.
文摘Aims: The representativeness of the mandatory bacterial strains specified in European standards for in vitro assay of the bactericidal activity of antiseptics was evaluated by testing the activity of an antiseptic combining chlorhexidine digluconate 0.2% and benzalkonium chloride 0.5% against 21 additional bacterial strains, and the positive interaction between these two biocidal agents was assessed. Methods and Results: The bactericidal activity of the antiseptic solution used pure or diluted was assessed according to the European standards EN 1040 and EN 13727. The contact time was 1 min at 20°C. Interfering substances used in the EN 13727 assay were bovine serum albumin and sheep erythrocytes, simulating “dirty” conditions, and hard water. A reduction of colony-forming units by ≥5 log10 was deemed to meet the requirements to conclude bactericidal activity. Under “basic” conditions, the bactericidal activity of the antiseptic was observed against all four mandatory strains specified in the standards as well as against nearly all the additional strains tested, including most of those with acquired antibiotic-resistance. The positive interaction between the two biocidal agents was also confirmed. Under “dirty” conditions, the bactericidal activity of the antiseptic solution was maintained against all the mandatory strains and was reduced against only four of the additional strains tested. Conclusions: With regard to the antiseptic tested and under the experimental conditions described, bactericidal activity evidenced against the mandatory strains appeared to be representative of that manifested against a wide range of the main pathogenic bacteria. Reduced bacterial activity against some of the additional strains tested (e.g. Enterobacteriaceae) was observed under “dirty” conditions. Significance and Impact of the Study: EN 13727 with some experimental adjustments represents an additional appropriate standard that needs to be considered for mucocutaneous antiseptic assessment. However, it may be worth including other specific bacterial strains to those specified in the standard, when evaluating antiseptics intended for use in certain clinical situations.

文摘Bearberry(Arctostaphylos uva-ursi L.)is a perennial plant of the heather family(Ericaceae).The leaves are dominated by arbutin,phenol carbonic acids flavonoids,saponins,etc.It was previously shown that the bearberry leaves extract reduced blood glucose level in healthy animals under glucose overload,so it need to be studied more detail.The aim of the study was to investigate the chemical composition and the effect of dry alcohol extract from bearberry leaves,which enriched with cysteine,on the rats pancreas under experimental dexamethasone-induced insulin resistance(IR).Arctostaphylos uva-ursi L.leaves extract and its modified cysteine preparation were obtained according to the developed method with 50%ethanol solution.Their phytochemical profile,hypoglycaemic and pancreatic protective effect were investigated.Phenologlycoside(arbutin),phenolic carboxylic acid(gallic acid),5 flavonoids and 4 hydroxy-cinnamic acids were identified and quantified in the extracts by HPLC.Present data revealed that bearberry leaves alcoholic dry extract enriched with cysteine has a hypoglycaemic and pancreatic protective effect in treated animals under dexamethasone-induced IR model.Treatment improved hyperglycaemia,insulin resistance and beta cell reduc-tion induced by dexamethasone injections.
文摘Desert truffles are edible mushrooms and they belong to groupe of ascomycetes. In Algeria, we find them in arid and semiarid area. They are represented by two most genuses: Tirmania (white terfez) and Terfezia (red terfez). The aim of our work is to present extraction technics of the bioactive substances from gleba (ascoms) of Tirmania pinoyi (Maire) Malen?on and from the culture filtrate of mycelium of Terfezia claveryi Chatin. Also, we present antifungal and antibacterial activity of these substances in vitro and their identification. There is antifungal activity of different extracts on solid medium, in vitro, against species of Aspergillus, Fusarium, Gliocladium, Candida. Also, there is antibacterial activity against species of Staphylococus, Streptococcus, Bacillus, Enterococcus, Escherichia, and Salmonella. Results show that extracts inhibition varies according to microbial tested species. Bioactive substances of the different extracts were analyzed by GC (gas chromatography)-mass, RMN (Nuclear Magnetic Resonance) and infrared in order to determine the chemical identity. Results show that these substances contain pyrazines and cyclic compounds like quinones.
