【背景】鞘脂质是第二大类膜脂,作为信号转导物质参与细胞生长发育、繁殖及凋亡等过程。鞘脂质代谢酶调控鞘脂质代谢以维持生物体内代谢的稳态。【目的】以重要水稻害虫褐飞虱(Nilaparvata lugens)为研究对象,通过RNA干扰(RNA interfere... 【背景】鞘脂质是第二大类膜脂,作为信号转导物质参与细胞生长发育、繁殖及凋亡等过程。鞘脂质代谢酶调控鞘脂质代谢以维持生物体内代谢的稳态。【目的】以重要水稻害虫褐飞虱(Nilaparvata lugens)为研究对象,通过RNA干扰(RNA interference,RNAi)技术检测微小RNA(microRNA,miRNA)生物合成通路核心组分NlAgo1、NlDicer1和NlDrosha沉默后褐飞虱鞘脂质代谢通路相关基因的相对转录水平,结合small RNA测序分析沉默丝氨酸棕榈酰转移酶1(serine palmitoyltransferase 1,SPT1)和鞘磷脂酶4(sphingomyelinase 4,SMase4)基因的差异miRNA,探究miRNA在褐飞虱鞘脂质代谢中的作用,为害虫防治提供新的分子靶标。【方法】利用RNAi技术,分别对羽化后第1天(1 PAE,post adult eclosion)的雌成虫NlAgo1、NlDicer1和NlDrosha进行dsRNA注射,以ds GFP为对照;分别解剖羽化后第5天的卵巢组织,以β-actin作为内参基因,采用实时荧光定量PCR(qRT-PCR)方法检测NlAgo1、NlDicer1和NlDrosha沉默后鞘脂质代谢通路相关基因的表达量变化;根据已有的小RNA文库联合miRNA-靶基因预测软件对可能调控NlSPT1和NlSMase4表达的miRNA进行预测;通过small RNA测序技术对沉默NlSPT1和NlSMase4的差异miRNA进行鉴定和靶基因富集性分析。【结果】与对照组相比,沉默NlAgo1、NlDicer1或NlDrosha显著上调卵巢中NlSPT1和NlSMase4等鞘脂质代谢通路相关基因的表达;靶基因预测结果显示,有6条miRNA能与NlSPT1结合,13条miRNA能与NlSMase4结合;沉默NlSPT1和NlSMase4的差异miRNA的靶基因显著富集在细胞核和蛋白质结合等生物学过程以及内吞作用、内质网加工、MAPK信号通路、TOR信号通路、凋亡、脂质代谢等代谢通路。【结论】NlAgo1、NlDicer1和NlDrosha依赖性的miRNA通过影响鞘脂质代谢相关基因的表达影响鞘脂质代谢。NlSPT1和NlSMase4沉默引起褐飞虱卵巢miRNA表达水平的改变。研究结果可为基于鞘脂质代谢基因为靶标的害虫防治提供理论依据。 展开更多

Small nucleolar RNAs(snoRNAs)represent a class of non-coding RNAs that play pivotal roles in post-transcriptional RNA processing and modification,thereby contributing significantly to the maintenance of cellular functions related to protein synthesis.SnoRNAs have been discovered to possess the ability to influence cell fate and alter disease progression,holding immense potential in controlling human diseases.It is suggested that the dysregulation of snoRNAs in cancer exhibits differential expression across various cancer types,stages,metastasis,treatment response and/or prognosis in patients.On the other hand,colorectal cancer(CRC),a prevalent malignancy of the digestive system,is characterized by high incidence and mortality rates,ranking as the third most common cancer type.Recent research indicates that snoRNA dysregulation is associated with CRC,as snoRNA expression significantly differs between normal and cancerous conditions.Consequently,assessing snoRNA expression level and function holds promise for the prognosis and diagnosis of CRC.Nevertheless,current comprehension of the potential roles of snoRNAs in CRC remains limited.This review offers a comprehensive survey of the aberrant regulation of snoRNAs in CRC,providing valuable insights into the discovery of novel biomarkers,therapeutic targets,and potential tools for the diagnosis and treatment of CRC and furnishing critical cues for advancing research into CRC and the judicious selection of therapeutic targets. 展开更多

Objective:To determine the genetic diversity of Plasmodium(P.)knowlesi isolates from Sabah,Malaysian Borneo and Peninsular Malaysia,targeting the S-type SSU rRNA gene and including aspects of natural selection and haplotype.Methods:Thirty-nine blood samples infected with P.knowlesi were collected in Sabah,Malaysian Borneo and Peninsular Malaysia.The S-type SSU rRNA gene was amplified using polymerase chain reaction,cloned into a vector,and sequenced.The natural selection and haplotype of the S-type SSU rRNA gene sequences were determined using DnaSP v6 and illustrated using NETWORK v10.This study's 39 S-type SSU rRNA sequences and eight sequences from the Genbank database were subjected to phylogenetic analysis using MEGA 11.Results:Overall,the phylogenetic analysis showed no evidence of a geographical cluster of P.knowlesi isolates from different areas in Malaysia based on the S-type SSU rRNA gene sequences.The S-type SSU rRNA gene sequences were relatively conserved and with a purifying effect.Haplotype sharing of the S-type SSU rRNA gene was observed between the P.knowlesi isolates in Sabah,Malaysian Borneo,but not between Sabah,Malaysian Borneo and Peninsular Malaysia.Conclusions:This study suggests that the S-type SSU rRNA gene of P.knowlesi isolates in Sabah,Malaysian Borneo,and Peninsular Malaysia has fewer polymorphic sites,representing the conservation of the gene.These features make the S-type SSU rRNA gene suitable for comparative studies,such as determining the evolutionary relationships and common ancestry among P.knowlesi species. 展开更多

Small RNAs are non-coding RNA molecules with 20-30 nucleotides (nt) in length that mainly play regulatory roles in gene expression at the post-transcription level by directly cutting target mRNA or inhibiting its translation. Small RNAs play regulatory roles in the growth and development process of plants at the core of gene regulatory networks, which has been widely studied and confirmed in sporophyte generation of plants. However, few researches have been conducted on small RNAs and gametophyte generation. It is reported that small RNAs play important roles in floral organ development, gametogenesis, fertilization, and early zygotic development of plants. In addition, various small RNAs also play roles in controlling genetic integrity, cell differentiation and functions during the sexual reproduction process of plants. However, most of the specific functions of small RNAs in the sexual reproduction process are unknown yet. This study mainly aimed to introduce small RNAs in plants, summarize the latest advances in researches of small RNAs and plant sexual reproduction, and make prospect on its future. 展开更多

小RNA(small RNA,sRNA)介导的RNA沉默或RNA干扰(RNA silencing or RNA interference,RNAi)是真核生物基因表达调控的保守机制。内源或外源双链RNA(double-stranded RNA,dsRNA)被加工成sRNA,sRNA通过碱基互补配对识别靶标基因mRNA或DNA,... 小RNA(small RNA,sRNA)介导的RNA沉默或RNA干扰(RNA silencing or RNA interference,RNAi)是真核生物基因表达调控的保守机制。内源或外源双链RNA(double-stranded RNA,dsRNA)被加工成sRNA,sRNA通过碱基互补配对识别靶标基因mRNA或DNA,通过降解mRNA、抑制翻译或者DNA甲基化在转录后或者转录水平调控基因表达。由于sRNA作用靶标特异性,RNAi技术被广泛地应用于基因功能研究、生物医药、作物分子设计育种以及开发新型农药等领域。自然界中sRNA能够在不同物种间传递并发挥作用,这一现象为RNAi应用技术的开发提供了理论基础。研究发现,dsRNA诱导RNAi的效率受多种因素影响,例如长度、剂量以及施用方式等。在细胞中,RNA结构的复杂性决定了其功能的多样性。本文概述了基于RNAi的作物病害防控技术的原理,包括宿主诱导的基因沉默(host-induced gene silencing,HIGS)技术、喷施诱导的基因沉默(spray-induced gene silencing,SIGS)技术和微生物诱导的基因沉默(microbe-induced gene silencing,MIGS)技术;总结了靶标RNA和sRNA结构影响RNAi效率的实验证据,以期加深对RNA结构影响RNAi效率地理解,为靶标筛选以及dsRNA设计提供经验,为开发高效RNAi技术提供参考;最后归纳了RNA结构检测和预测的代表性方法,为辅助设计高效诱导RNAi的dsRNA提供方法。 展开更多

胶质瘤组织lncRNA SNHG25、miR-497-5p表达与临床特征及预后的关系研究

目的探讨胶质瘤组织长链非编码RNA(lncRNA)小核仁RNA宿主基因(SNHG)25、微小RNA(miR)-497-5p表达与临床特征及预后的关系。方法选择2019年1月至2020年1月该院收治的157例胶质瘤患者为胶质瘤组,同期该院100例因颅脑损伤接受手术治疗的患... 目的探讨胶质瘤组织长链非编码RNA(lncRNA)小核仁RNA宿主基因(SNHG)25、微小RNA(miR)-497-5p表达与临床特征及预后的关系。方法选择2019年1月至2020年1月该院收治的157例胶质瘤患者为胶质瘤组,同期该院100例因颅脑损伤接受手术治疗的患者为对照组。分别取术中切除的胶质瘤组织和正常脑组织检测lncRNA SNHG25、miR-497-5p表达水平,术后随访3年。分析lncRNA SNHG25表达水平与miR-497-5p的相关性,lncRNA SNHG25、miR-497-5p表达水平与患者临床特征和预后的关系。结果与对照组比较,胶质瘤组lncRNA SNHG25表达水平升高(P<0.05),miR-497-5p表达水平降低(P<0.05)。与肿瘤最大径<4 cm、世界卫生组织(WHO)中枢神经系统肿瘤分级Ⅰ~Ⅱ级比较,肿瘤最大径≥4 cm、WHO中枢神经系统肿瘤分级Ⅲ~Ⅳ级的胶质瘤组织中lncRNA SNHG25表达水平升高,miR-497-5p表达水平降低(P<0.05)。胶质瘤患者的lncRNA SNHG25表达水平与miR-497-5p呈负相关(r=-0.370,P<0.05)。lncRNA SNHG25高表达组累积生存率低于lncRNA SNHG25低表达组(P<0.05),miR-497-5p低表达组累积生存率低于miR-497-5p高表达组(P<0.05)。WHO中枢神经系统肿瘤分级Ⅲ~Ⅳ级、lncRNA SNHG25高表达是胶质瘤患者预后不良的危险因素(P<0.05),miR-497-5p高表达则是保护因素(P<0.05)。结论胶质瘤组织中lncRNA SNHG25表达水平升高,miR-497-5p表达水平降低,且与肿瘤最大径增大、WHO中枢神经系统肿瘤分级高有关,可导致胶质瘤患者不良预后。 展开更多

目的探讨抑制线粒体RNA加工核糖核酸内切酶RNA组分(RMRP)表达对人肺癌细胞系A549增殖和侵袭的影响;检测非小细胞肺癌(NSCLC)患者全血和组织中RMRP表达,比较在不同临床指标表达差异性,以期为NSCLC机制研究提供参考资料。方法收集2016年3... 目的探讨抑制线粒体RNA加工核糖核酸内切酶RNA组分(RMRP)表达对人肺癌细胞系A549增殖和侵袭的影响;检测非小细胞肺癌(NSCLC)患者全血和组织中RMRP表达,比较在不同临床指标表达差异性,以期为NSCLC机制研究提供参考资料。方法收集2016年3月至2021年3年在焦作市人民医院行手术治疗的NSCLC患者122例,同期,在体检中心选取健康者50名作为对照组。RT-qPCR检测全血和组织中RMRP mRNA水平。培养A549细胞分为si-RMRP组、siRNA-NC组和空白组(blank组)。RT-qPCR、CCK-8法和Transwell小室法分别检测细胞中RMRP表达、增殖活性和侵袭细胞数。结果NSCLC患者全血中RMRP相对表达量显著高于对照组(P<0.001);NSCLC组织中RMRP相对表达量高于癌旁组织(P<0.001);低分化、淋巴结转移和TNM分期Ⅲ的NSCLC患者全血和组织中RMRP相对表达量较高分化、未发生淋巴结转化和TNM分期Ⅰ~Ⅱ明显升高(P<0.05);Si-RMRP组细胞中RMRP相对表达量低于空白组(blank组)和siRNA-NC组(P<0.001);相比与blank组和siRNA-NC组,24、48、72和96 h时si-RMRP组细胞吸光度(A)值均降低(P<0.05);Si-RMRP组细胞侵袭数低于blank组和siRNA-NC组(P<0.001)。结论NSCLC患者全血和组织中RMRP相对表达量升高,下调A549细胞中RMRP基因表达可抑制细胞增殖,减少细胞侵袭。 展开更多

EGFR siRNA序列的筛选及其对HepG2细胞活性的影响

目的构建EGFR shRNA重组真核表达载体,并筛选抑制效果最好的EGFR shRNA序列。方法应用在线工具设计人EGFR siRNA序列,采用限制性内切酶BamHI和HindIII切割真核表达质粒pcDNA3.1(+),构建三种人EGFR的siRNA干扰序列载体(psilencer 4.1-CMV... 目的构建EGFR shRNA重组真核表达载体,并筛选抑制效果最好的EGFR shRNA序列。方法应用在线工具设计人EGFR siRNA序列,采用限制性内切酶BamHI和HindIII切割真核表达质粒pcDNA3.1(+),构建三种人EGFR的siRNA干扰序列载体(psilencer 4.1-CMV neo-EGFR siRNA)。将重组质粒载体转化大肠杆菌DH5α感受态并筛选阳性克隆,通过DNA测序鉴定重组质粒。应用脂质体lipo2000将三种人EGFR siRNA干扰序列载体转染到HepG2细胞,通过荧光显微镜观察转染效率,实时定量PCR检测EGFR mRNA表达水平,MTT法检测细胞活性。结果Psilencer 4.1-CMV neo-EGFR siRNA重组质粒被成功克隆。EGFR shRNA-1、EGFR shRNA-2和EGFR shRNA-3敲低EGFR mRNA的效率分别是80%、60%和70%以上。shRNA-2和shRNA-3使细胞活性分别下降50%(P<0.05),但shRNA-1对细胞活性无明显影响(P>0.05)。结论重组psilencer 4.1-CMV neo-EGFR siRNA质粒可下调肝癌细胞株EGFR表达水平和细胞活性。EGFR shRNA-3较EGFR shRNA-1和shRNA-2对HepG2细胞的抑制作用更显著。 展开更多

鹅绒藤(Cynanchum chinense)是一种传统中药,为明确造成鹅绒藤叶片花叶症状的原因,本研究采用小RNA深度测序技术结合RT-PCR的方法鉴定引起山西太谷鹅绒藤病毒花叶症状的病原,并通过生物信息学的手段对病毒序列进行分析。结果表明,发病... 鹅绒藤(Cynanchum chinense)是一种传统中药,为明确造成鹅绒藤叶片花叶症状的原因,本研究采用小RNA深度测序技术结合RT-PCR的方法鉴定引起山西太谷鹅绒藤病毒花叶症状的病原,并通过生物信息学的手段对病毒序列进行分析。结果表明,发病鹅绒藤样品经小RNA深度测序技术共获得15039334个原始序列,将拼接contigs与NCBI中的病毒数据库进行比对注释,结果显示病原为苜蓿花叶病毒(AMV)和黄瓜花叶病毒(CMV)。利用特异引物克隆了AMV和CMV的外壳蛋白(CP)和移动蛋白(MP)基因全序列,分别命名为AMV-BR和CMV-BR。通过序列比对分析,发现AMV-BR CP氨基酸序列和核苷酸序列与AMV分离物AMV-Gyn(MH332899)、Dich-rep(MW835989)和VIC-320(MF075254)的相似性均达到100%;MP氨基酸序列和核苷酸序列与AMV分离物AMV-Gyn(MH332899)的相似性最高,均为99.3%。CMV-BR CP氨基酸序列和核苷酸序列与CMV亚组ⅠA中CMV分离物YA17(MH119159)的相似性最高,分别为100%和99.1%;MP氨基酸序列和核苷酸序列与CMV亚组Ⅰ分离物PV-0185(ON013887)的相似性最高,分别为98.2%和96.4%。MP和CP氨基酸序列系统进化分析表明,CMV-BR属于CMV亚组Ⅰ中的成员。这是首次在山西鹅绒藤上检测到AMV和CMV,该研究有助于深入了解AMV、CMV的分子进化,也为鹅绒藤病毒病的防治提供一定的理论依据。 展开更多

目的 探讨脑小血管病(CSVD)患者血清小核仁RNA宿主基因8(SNHG8)表达水平变化与疾病进展及认知功能障碍(CI)的相关性。方法 选取2021年5月至2023年5月在承德医学院附属医院进行诊治的100例CSVD患者为研究对象,根据患者脑动脉指数(PI),将C... 目的 探讨脑小血管病(CSVD)患者血清小核仁RNA宿主基因8(SNHG8)表达水平变化与疾病进展及认知功能障碍(CI)的相关性。方法 选取2021年5月至2023年5月在承德医学院附属医院进行诊治的100例CSVD患者为研究对象,根据患者脑动脉指数(PI),将CSVD患者分为轻度组(n=31)、中度组(n=45)和重度组(n=24);另根据蒙特利尔评估量表(MoCA)评分,将CSVD患者分为CI组(n=51)和非CI组(n=49);同时,选取同期在我院体检的健康人群100例为对照组。实时荧光定量PCR法测定血清SNHG8水平;比较对照组、CI组与非CI组一般资料及血清SNHG8水平;比较不同病情程度CSVD患者血清SNHG8水平;Spearman相关性分析血清SNHG8水平与病情程度、MoCA评分之间的关系;受试者工作特征(ROC)曲线分析血清SNHG8水平对CSVD患者并发CI的诊断价值;Logistic回归分析CSVD患者并发CI的影响因素。结果 对照组、CI组与非CI组在性别、年龄、基础病史、TC、TG、HDL-C、LDL-C上差异无统计学意义(P>0.05),在受教育程度、IL-33及IL-18水平上差异有统计学意义(P<0.05);不同病情程度CSVD患者血清SNHG8水平差异有统计学意义(P<0.05),且随CSVD疾病进展,血清SNHG8水平逐渐降低;Spearman相关性分析结果显示,血清SNHG8水平与病情程度呈负相关(r=-0.561,P<0.05),与MoCA评分呈正相关(r=0.583,P<0.05);ROC曲线结果显示,血清SNHG8诊断CSVD患者并发CI的AUC为0.860,敏感度为86.3%,特异度为72.0%;多因素Logistic回归分析结果显示,受教育程度、血清IL-18、IL-33、SNHG8均是CSVD患者发生CI的影响因素。结论 随着CSVD患者病情进展,血清SNHG8水平逐渐降低,且血清SNHG8水平与CSVD患者并发CI密切相关。 展开更多

目的探讨中国汉族人群中小核RNA激活复合体多肽3(SNAPC3)基因rs4741506多态性与缺血性脑卒中(IS)及其中医证候的关系。方法选取2016年1月至2019年12月在广西中医药大学第一附属医院脑病科住院治疗的774例IS患者作为观察组,同期本院体检... 目的探讨中国汉族人群中小核RNA激活复合体多肽3(SNAPC3)基因rs4741506多态性与缺血性脑卒中(IS)及其中医证候的关系。方法选取2016年1月至2019年12月在广西中医药大学第一附属医院脑病科住院治疗的774例IS患者作为观察组,同期本院体检中心的健康体检者以及医院骨科轻症外伤患者793例作为对照组。对IS患者进行中医证候辨证,对SNAPC3基因多态性位点rs4741506进行基因分型检测。建立4种遗传模型,应用PLINK软件和SPSS 21.0软件进行遗传关联及基因位点多态性与IS及其中医证候和患者临床指标的相关性分析。结果观察组与对照组rs4741506位点的基因型频率分布差异有统计学意义(χ^(2)=6.366,P=0.041)。在加性模型、显性模型、隐性模型中,SNAPC3基因rs4741506多态性与IS的发生风险均无显著相关性(均P>0.05)。多因素Logistic回归模型分析结果显示,校正年龄和性别后SNAPC3基因rs4741506多态性与IS风证(隐性模型:比值比=0.45,95%置信区间:0.22~0.92,P=0.029)、痰证(隐性模型:比值比=0.39,95%置信区间:0.19~0.81,P=0.011)发生风险相关,而与血瘀证的发生风险无明显相关性(显性模型:比值比=1.42,95%置信区间:1.00~2.01,P=0.051)。在隐性模型下,校正年龄和性别后SNAPC3基因rs4741506多态性与IS痰证患者舒张压水平相关(β=-10.93,95%置信区间:-20.64~-1.22,P=0.028)。一般线性回归分析结果显示,校正年龄和性别后SNAPC3基因rs4741506多态性与IS痰证患者血清载脂蛋白A1(加性模型、显性模型)、载脂蛋白B(隐性模型)、血小板计数(加性模型、显性模型)水平、凝血酶时间(显性模型)显著相关(均P<0.05)。结论SNAPC3基因rs4741506多态性可能影响IS风证及痰证的发生发展。 展开更多

Hepatocellular carcinoma(HCC) is the 5th most common malignancy which is responsible for more than half million annual mortalities; also, it is the third leading cause of cancer related death. Unfavorablesystemic side-effects of chemotherapeutic agents and susceptibility to the degradation of small interfering RNAs(si RNAs), which can knock down a specific gene involved in the disease, have hampered their clinical application. So, it could be beneficial to develop an efficient carrier for the stabilization and specific delivery of drugs and si RNA to cells. Targeted nanoparticles have gained considerable attention as an efficient drug and gene delivery system, which is due to their capability in achieving the highest accumulation of cytotoxic agents in tumor tissue, modifiable drug pharmacokinetic- and bio-distribution, improved effectiveness of treatment, and limited sideeffects. Recent studies have shed more light on the advantages of novel drug loaded carrier systems vs free drugs. Most of the animal studies have reported improvement in treatment efficacy and survival rate using novel carrier systems. Targeted delivery may be achieved passively or actively. In passive targeting, no ligand as homing device is used, while targeting is achieved by incorporating the therapeutic agent into a macromolecule or nanoparticle that passively reaches the target organ. However, in active targeting, the therapeutic agent or carrier system is conjugated to a tissue or cell-specific receptor which is overexpressed in a special malignancy using a ligand called a homing device. This review covers a broad spectrum of targeted nanoparticles as therapeutic and nonviral si RNA delivery systems, which are developed for enhanced cellular uptake and targeted gene silencing in vitro and in vivo and their characteristics and opportunities for the clinical applications of drugs and therapeutic si RNA are discussed in this article. Asialoglycoprotein receptors, low-density lipoprotein, ganglioside GM1 cell surface ligand, epidermal growth factor receptor receptors, monoclonal antibodies, retinoic acid receptors, integrin receptors targeted by Arg-Gly-Asp peptide, folate, and transferrin receptors are the most widely studied cell surface receptors which are used for the site specific delivery of drugs and si RNA-based therapeutics in HCC and discussed in detail in this article. 展开更多

RNA interfering(RNAi), mediated by small interfering RNAs and microRNAs, is currently one of the most promising tools of gene therapy. Small RNAs are capable of inducing specific post-transcriptional gene silencing, providing a potentially effective platform for the treatment of a wide array of diseases. However, similar to other nucleic acid-based drugs,the major hurdle of RNAi therapy is lack of efficient and non-immunogenic delivery vehicles. Currently, viruses, synthetic polymers, and lipid-based carriers are among the most widely studied vehicles for small RNA delivery. However, many drawbacks are reported to be associated with these delivery vehicles. There is a pressing need to replace them with more efficient and better-tolerated approaches. Exosomes secreted from the endocytic compartment of live cells, are a subtype of endogenous extracellular vesicles that transfer genetic and biochemical information among different cells, thus playing an important role in cellcell communication. Recently, accumulating attention has been focused on harnessing exosomes as nanaocarriers for small RNAs delivery. Due to their natural role in shuttling endogenous nucleic acid in our body, exosomes may exhibit higher delivery efficiency, lower immunogenicity, and better compatibility than existing foreign RNA carriers. Importantly,exosomes own intrinsic homing capacity that can guide small RNAs across natural membranous barriers. Moreover, such a capacity can be further improved by adding appropriate targeting moieties. In this manuscript, we briefly review the progress and challenges of RNAi therapy, and discuss the potential of exosomes' applications in small RNA delivery with focus on the most recent advances in exosome-based small RNA delivery for disease therapy. 展开更多

Transfer RNAs(tRNAs), a type of ncRNA, are the second most abundant type of RNA in cells. Recent studies have shown that tRNAs can be cleaved into a heterogeneous population of ncRNAs with lengths of 18–40 nucleotides, known as tRNA-derived small RNAs(tsRNAs). There are two main types of tsRNA, based on their length and the number of cleavage sites that they contain: tRNA-derived fragments and tRNA-derived stress-induced RNAs. These RNA species were first considered to be byproducts of tRNA random cleavage. However, mounting evidence has demonstrated their critical functional roles as regulatory factors in the pathophysiological processes of various diseases, including neurological diseases. However, the underlying mechanisms by which tsRNAs affect specific cellular processes are largely unknown. Therefore, this study comprehensively summarizes the following points:(1) The biogenetics of tsRNA, including their discovery, classification, formation, and the roles of key enzymes.(2) The main biological functions of tsRNA, including its miRNA-like roles in gene expression regulation, protein translation regulation, regulation of various cellular activities, immune mediation, and response to stress.(3) The potential mechanisms of pathophysiological changes in neurological diseases that are regulated by tsRNA, including neurodegeneration and neurotrauma.(4) The identification of the functional diversity of tsRNA may provide valuable information regarding the physiological and pathophysiological mechanisms of neurological disorders, thus providing a new reference for the clinical treatment of neurological diseases. Research into tsRNAs in neurological diseases also has the following challenges: potential function and mechanism studies, how to accurately quantify expression, and the exact relationship between tsRNA and miRNA. The practical use of these molecules is however limited by the identification of optimal molecular targets and especially by the lack of effective and targeted HCC delivery systems.
