Expression of hepatocyte nuclear factor 4 alpha,wingless-related integration site,andβ-catenin in clinical gastric cancer

BACKGROUND Gastric cancer(GC)is the second most common cause of cancer-related deaths worldwide.Hepatocyte nuclear factor 4 alpha(HNF4α)that belongs to the nuclear hormone receptor superfamily,is overexpressed in GC tissues,and might be involved in the development of GC by regulating its downstream winglessrelated integration site(WNT)/β-catenin signaling.AIM To clarify the expression of HNF4α/WNT5a/β-catenin signaling proteins in clinical GC tissues.METHODS We immunohistochemically stained pathological blocks of GC and matched paracancerous tissues.The intensity of HNF4α,WNT5a andβ-catenin staining in the tumor cells was determined according to cell rates and staining intensity.The correlations between GC and HNF4α,WNT5a,andβ-catenin expression using chisquare and paired chi-square tests.Relationships between double-positive HNF4αand WNT5a expression and types of gastric tumor tissues were assessed using regression analysis.Correlations between HNF4αand WNT5a expression at the RNA level in GC tissues found in the TCGA database were analyzed using Pearson correlation coefficients.RESULTS We found more abundant HNF4αand WNT5a proteins in GC,especially in mucinous adenocarcinoma and mixed GC than in adjacent tissues(P<0.001).Low and high levels of cytoplasmicβ-catenin respectively expressed in GC and adjacent tissues(P<0.001)were not significantly associated with pathological parameters.CONCLUSION The expressions of HNF4αand WNT5a could serve as early diagnostic biomarkers for GC. 展开更多

A steam power plant can work as a dual purpose plant for simultaneous production of steam and elec-trical power. In this paper we seek the optimum integration of a steam power plant as a source and a site utility sys-tem as a sink of steam and power. Estimation for the cogeneration potential prior to the design of a central utility system for site utility systems is vital to the targets for site fuel demand as well as heat and power production. In this regard, a new cogeneration targeting procedure is proposed for integration of a steam power plant and a site utility consisting of a process plant. The new methodology seeks the optimal integration based on a new cogenera-tion targeting scheme. In addition, a modified site utility grand composite curve(SUGCC) diagram is proposed and compared to the original SUGCC. A gas fired steam power plant and a process site utility is considered in a case study. The applicability of the developed procedure is tested against other design methods(STAR? and Thermoflex software) through a case study. The proposed method gives comparable results, and the targeting method is used for optimal integration of steam levels. Identifying optimal conditions of steam levels for integration is important in the design of utility systems, as the selection of steam levels in a steam power plant and site utility for integration greatly influences the potential for cogeneration and energy recovery. The integration of steam levels of the steam power plant and the site utility system in the case study demonstrates the usefulness of the method for reducing the overall energy consumption for the site. 展开更多

Global Positioning System(GPS)measurements of integrated water vapor(IWV)for two years(2014 and 2015)are presented in this paper.Variation of IWV during active and break spells of Indian summer monsoon has been studied for a tropical station Hyderabad(17.4°N,78.46°E).The data is validated with ECMWF Re-Analysis(ERA)91 level data.Relationships of IWV with other atmospheric variables like surface temperature,rain,and precipitation efficiency have been established through cross-correlation studies.A positive correlation coefficient is observed between IWV and surface temperature over two years.But the coefficient becomes negative when only summer monsoon months(June,July,August,and September)are considered.Rainfall during these months cools down the surface and could be the reason for this change in the correlation coefficient.Correlation studies between IWV-precipitation,IWVprecipitation efficiency(P.E),and precipitation-P.E show that coefficients are-0.05,-0.10 and 0.983 with 95%confidence level respectively,which proves that the efficacy of rain does not depend only on the level of water vapor.A proper dynamic mechanism is necessary to convert water vapor into the rain.The diurnal variations of IWV during active and break spells have been analyzed.The amplitudes of diurnal oscillation and its harmonics of individual spell do not show clear trends but the mean amplitudes of the break spells are approximately double than those of the active spells.The amplitudes of diurnal,semidiurnal and ter-diurnal components during break spells are 1.08 kg/m^(2),0.52 kg/m;and 0.34 kg/m;respectively.The corresponding amplitudes during active spells are 0.68 kg/m^(2),0.41 kg/m;and 0.23 kg/m;. 展开更多

Skeletal muscles are essential for locomotion,posture,and metabolic regulation.To understand physiological processes,exercise adaptation,and muscle-related disorders,it is critical to understand the molecular pathways that underlie skeletal muscle function.The process of muscle contra ction,orchestrated by a complex interplay of molecular events,is at the core of skeletal muscle function.Muscle contraction is initiated by an action potential and neuromuscular transmission requiring a neuromuscular junction.Within muscle fibers,calcium ions play a critical role in mediating the interaction between actin and myosin filaments that generate force.Regulation of calcium release from the sarcoplasmic reticulum plays a key role in excitation-contraction coupling.The development and growth of skeletal muscle are regulated by a network of molecular pathways collectively known as myogenesis.Myogenic regulators coordinate the diffe rentiation of myoblasts into mature muscle fibers.Signaling pathways regulate muscle protein synthesis and hypertrophy in response to mechanical stimuli and nutrient availability.Seve ral muscle-related diseases,including congenital myasthenic disorders,sarcopenia,muscular dystrophies,and metabolic myopathies,are underpinned by dys regulated molecular pathways in skeletal muscle.Therapeutic interventions aimed at preserving muscle mass and function,enhancing regeneration,and improving metabolic health hold promise by targeting specific molecular pathways.Other molecular signaling pathways in skeletal muscle include the canonical Wnt signaling pathway,a critical regulator of myogenesis,muscle regeneration,and metabolic function,and the Hippo signaling pathway.In recent years,more details have been uncovered about the role of these two pathways during myogenesis and in developing and adult skeletal muscle fibers,and at the neuromuscular junction.In fact,research in the last few years now suggests that these two signaling pathways are interconnected and that they jointly control physiological and pathophysiological processes in muscle fibers.In this review,we will summarize and discuss the data on these two pathways,focusing on their concerted action next to their contribution to skeletal muscle biology.However,an in-depth discussion of the noncanonical Wnt pathway,the fibro/a dipogenic precursors,or the mechanosensory aspects of these pathways is not the focus of this review. 展开更多

According to the investigation of Taiyanggou Block and Lvshun Industrial School Site in Lvshun (China), this paper aims to discuss the understanding and conservation of modern heritage. Firstly, it interprets the opening of modern Lvshun and the formation of Taiyanggou Block to provide the fundamental knowledge. After a brief introduction of Lvshun Industrial School Site, the paper analyzes the problems in conservation practice and proposes countermeasures on heritage management and reuse strategy. Seismic response characteristics of nuclear island structure at generic soil and rock sites 被引量:1

The number of traditionally excellent coastal lithologic nuclear power plants is limited.It is a trend to develop nuclear power plants on soil sites in inland areas.Therefore,the seismic safety and adaptability of non-rock nuclear power plant(NPP)sites are the key concerns of nuclear safety researchers.Although the five site categories are clearly defined in the AP1000 design control documents,the effects of nuclear power five site conditions and soil nonlinearity on the seismic response characteristics of nuclear island buildings have not been systematically considered in previous related studies.In this study,targeting the AP1000 nuclear island structure as the research object,three-dimensional finite element models of a nuclear island structure at five types of sites(firm rock site(FR),soft rock site(SR),soft-to-medium soil site(SMS),upper bound soft-to-medium site(SMS-UB),and soft soil site(SS))are established.The partitioned analysis method of soil-structure interaction(PASSI)in the time-domain is used to investigate the effects of site hardness and nonlinearity on the acceleration,displacement,and acceleration response spectrum of the nuclear island structure under seismic excitation.The incremental equilibrium equation and explicit decoupling method are used to analyze the soil nonlinearity described by the Davidenkov model with simplified loading-reloading rules.The results show that,in the linear case,with the increase of site hardness,the peak ground acceleration(PGA)and the peak of acceleration response spectrum of the nuclear island structure increase except for the FR site,while the maximum displacement decreases.In nonlinear analysis,as the site hardness increases,the PGA,maximum displacement,and the peak of acceleration response spectrum of the nuclear island structure increase.The peak value of the acceleration response spectrum in the nonlinear case is greater than that in the linear case for FR,while smaller for SR and soil sites.The site nonlinearity reduces the peak values of the response spectrum for SR and soil sites much more as the site hardness decreases.The results of this study can provide a reference for design of nuclear island structures on soil and rock sites. 展开更多

This paper proposed the explicit generalized-a time scheme and periodic boundary conditions in the material point method(MPM)for the simulation of coseismic site response.The proposed boundary condition uses an intuitive particle-relocation algorithm ensuring material points always remain within the computational mesh.The explicit generalized-a time scheme was implemented in MPM to enable the damping of spurious high frequency oscillations.Firstly,the MPM was verified against finite element method(FEM).Secondly,ability of the MPM in capturing the analytical transfer function was investigated.Thirdly,a symmetric embankment was adopted to investigate the effects of ground motion arias intensity(I_(a)),geometry dimensions,and constitutive models.The results show that the larger the model size,the higher the crest runout and settlement for the same ground motion.When using a Mohr-Coulomb model,the crest runout increases with increasing I_(a).However,if the strain-softening law is activated,the results are less influenced by the ground motion.Finally,the MPM results were compared with the Newmark sliding block solution.The simplified analysis herein highlights the capabilities of MPM to capture the full deformation process for earthquake engineering applications,the importance of geometry characterization,and the selection of appropriate constitutive models when simulating coseismic site response and subsequent large deformations. 展开更多

为保障国家天然气安全平稳供应,2019年国家能源局启动了川渝天然气千亿立方米级“气大庆”的建设项目,将四川盆地天然气储气调峰和战略应急保供能力建设提升至国家战略安全高度。此外,中国石油在“十四五”期间确立了打造“西南储气中... 为保障国家天然气安全平稳供应,2019年国家能源局启动了川渝天然气千亿立方米级“气大庆”的建设项目,将四川盆地天然气储气调峰和战略应急保供能力建设提升至国家战略安全高度。此外,中国石油在“十四五”期间确立了打造“西南储气中心”的战略目标,积极攻关各项地下储气库(以下简称储气库)建库技术难题,已初步形成“西南储气中心”建设格局。为此,在分析四川盆地建设储气库面临的挑战基础上,系统梳理总结了中国石油近17年在四川盆地的发展规划、建设现状、技术进展等方面的成果,并对打造“中国西南储气能力”提出了具体的攻关方向。研究结果表明:(1)四川盆地天然气资源丰富且拥有与全国天然气管网互连互通的完善管网设施,能够为盆地储气库建设提供坚实基础;(2)提出了复杂盆地气藏的“十指标”选址体系,建立了完善的储气库库址优选技术;(3)形成了储气库建库运行七大特色技术,健全了碳酸盐岩气藏建库主体技术系列;(4)构建了国内首个储气库地质体、井、地面完整性管理评价技术体系,保障了储气库安全稳定运行。结论认为,为支撑国家天然气千亿立方米级“气大庆”建设,提出了“中国西南储气能力”建设的概念,其核心是以中国石油初步建成的企业级“西南储气中心”为基础,走出单一油公司储气库建设模式,建立政府主导协调、跨企业的储气库协同共建共享共营机制,全面推动四川盆地整体储气能力建设,保障国家天然气能源安全。 展开更多

目的探究环状RNA hsa_circ_0085576对口腔鳞状细胞癌(OSCC)细胞迁移和侵袭的影响及其分子机制。方法使用实时荧光定量聚合酶链反应(qRT-PCR)和蛋白印迹法检测OSCC细胞中hsa_circ_0085576、微小RNA-498(miR-498)以及B细胞特异性莫洛尼鼠... 目的探究环状RNA hsa_circ_0085576对口腔鳞状细胞癌(OSCC)细胞迁移和侵袭的影响及其分子机制。方法使用实时荧光定量聚合酶链反应(qRT-PCR)和蛋白印迹法检测OSCC细胞中hsa_circ_0085576、微小RNA-498(miR-498)以及B细胞特异性莫洛尼鼠白血病病毒整合位点1(BMI-1)的表达水平。使用CCK-8、划痕实验、Transwell实验以及qRT-PCR、蛋白印迹法分别检测SCC-15细胞增殖活力、迁移及侵袭能力以及相关基因和蛋白的相对表达量。结果OSCC细胞中hsa_circ_0085576与BMI-1表达上调,miR-498表达下调(P<0.05)。下调hsa_circ_0085576表达或过表达miR-498后SCC-15细胞的增殖活性、划痕愈合率、侵袭细胞数目以及细胞周期蛋白D1、波形蛋白表达水平下调,miR-498和E-钙黏蛋白表达水平上调(P<0.05);抑制miR-498表达可减弱下调hsa_circ_0085576表达对OSCC细胞增殖、迁移及侵袭的抑制作用;上调BMI-1表达可减弱过表达miR-498对OSCC细胞增殖、迁移及侵袭的抑制作用。结论下调hsa_circ_0085576表达可通过激活miR-498/BMI-1轴抑制OSCC细胞增殖、迁移及侵袭。 展开更多

山区铁路传统工程建设运输物流基地选址模型较少将未来可能需求纳入考量范畴,忽视了此类临时工程在社会效益方面的可持续发展潜力。针对此问题,在传统工程建设运输物流基地选址模型考虑影响因素及假设基础上,加入永临结合决策变量,构建... 山区铁路传统工程建设运输物流基地选址模型较少将未来可能需求纳入考量范畴,忽视了此类临时工程在社会效益方面的可持续发展潜力。针对此问题,在传统工程建设运输物流基地选址模型考虑影响因素及假设基础上,加入永临结合决策变量,构建同时考虑未来社会需求分布的不确定性的工程选址模型。以工程地质条件、建设施工成本、道路情况等多个因素作为评价指标,采用预测未来社会需求并引入波动变量构建非精确机会约束的方式,提出适用于永临结合模式设施选址的“多面体-DRO”二阶段分布鲁棒模型。研究结果表明:服务水平是选址决策的主要影响因素;对于高海拔施工点,选择附近的高海拔设施更加可靠;为尽可能服务高海拔地区,永久设施点更倾向于在高海拔地区修建。建立的模型能够有效抵抗不确定性带来的代价,从而避免决策失误,在山区铁路工程永临结合设施修建方面有着重要理论价值和实际指导意义。 展开更多

目的研究右美托咪定对七氟烷诱发认知功能损伤的保护作用及可能机制。方法40只大鼠随机分为空白组、模型组、右美托咪定组及联合组,每组各10只。模型组、右美托咪定组、联合组建立七氟烷认知损伤模型。右美托咪定组、联合组大鼠建模前30... 目的研究右美托咪定对七氟烷诱发认知功能损伤的保护作用及可能机制。方法40只大鼠随机分为空白组、模型组、右美托咪定组及联合组,每组各10只。模型组、右美托咪定组、联合组建立七氟烷认知损伤模型。右美托咪定组、联合组大鼠建模前30 min腹腔注射右美托咪定50μg/kg,联合组另腹腔注射舒林酸5 mg/kg,空白组、模型组静脉注射等量生理盐水。Morris水迷宫测试检测认知功能;酶联免疫吸附实验(ELISA)检测血清同型半胱氨酸(Hcy)、单核细胞趋化蛋白-1(MCP-1)水平;高效液相色谱仪检测海马组织谷氨酸(Glu)、γ-氨基丁酸(GABA)含量;免疫印迹法检测海马组织糖原合成酶激酶3β(GSK-3β)、β-连环蛋白(β-catenin)蛋白表达量。结果右美托咪定组大鼠逃避潜伏期短于模型组(P<0.05),穿越原平台次数多于模型组(P<0.05),原平台象限停留时间长于模型组(P<0.05);联合组大鼠逃避潜伏期长于右美托咪定组(P<0.05),穿越原平台次数少于右美托咪定组(P<0.05),原平台象限停留时间短于右美托咪定组(P<0.05)。模型组大鼠血清Hcy、MCP-1水平高于空白组(P<0.05),右美托咪定组低于模型组(P<0.05),联合组高于右美托咪定组(P<0.05)。模型组海马组织GLu含量高于空白组(P<0.05),GABA含量低于空白组(P<0.05);右美托咪定组海马组织GLu含量低于模型组(P<0.05),GABA含量高于模型组(P<0.05);联合组海马组织GLu含量高于右美托咪定组(P<0.05),GABA含量低于右美托咪定组(P<0.05)。模型组海马组织GSK-3β蛋白表达量高于空白组(P<0.05),β-catenin蛋白表达量低于空白组(P<0.05);右美托咪定组海马组织GSK-3β蛋白表达量低于模型组(P<0.05),β-catenin蛋白表达量高于模型组(P<0.05);联合组海马组织GSK-3β蛋白表达量高于右美托咪定组(P<0.05),β-catenin蛋白表达量低于右美托咪定组(P<0.05)。结论右美托咪定可能通过激活Wnt/β-catenin信号通路改善七氟烷诱发认知功能损伤大鼠的认知功能,减轻炎症反应,增强神经递质活性。 展开更多

基于3D还原模型的驾驶舱眼位点反推

为获得飞机驾驶舱眼位点数据,本文研究了驾驶舱眼位点的反推。首先,通过建立某综合程序训练器的3D还原模型和简易飞机驾驶员模型,反推实际眼位点;其次,对实际眼位点和标准要求的参考眼位点进行对比分析,得到预测误差;最后将真实驾驶舱... 为获得飞机驾驶舱眼位点数据,本文研究了驾驶舱眼位点的反推。首先,通过建立某综合程序训练器的3D还原模型和简易飞机驾驶员模型,反推实际眼位点;其次,对实际眼位点和标准要求的参考眼位点进行对比分析,得到预测误差;最后将真实驾驶舱实测数据与该训练器数据进行对比分析,得到实际误差进行验证。结果表明:预测误差与实际误差相符,在纵向上预测误差为105.2 mm,实际误差为80 mm,在垂直和水平方向上存在细微误差。通过3D还原模型反推眼位点的方法能够反应综合程序训练器与真实驾驶舱之间存在的误差,可作为综合程序训练器鉴定等工作的参考依据。 展开更多

为准确测度“双碳”(即碳达峰、碳中和的合称)背景下具体工业门类选址与环境体系的结合发展实效,通过研究钢铁新产线的选址取向及环境关切转变,提出新产线选址的环境结合度评价框架。通过数据包络分析(Data Envelopment Analysis,DEA)... 为准确测度“双碳”(即碳达峰、碳中和的合称)背景下具体工业门类选址与环境体系的结合发展实效,通过研究钢铁新产线的选址取向及环境关切转变,提出新产线选址的环境结合度评价框架。通过数据包络分析(Data Envelopment Analysis,DEA)模型计算环境-经济效率,通过动态综合评价模型揭示时间序列上效率的变化特征,以构建一套钢铁新产线选址的环境结合度评价方法,进而对中国宝武钢铁集团部分生产基地开展实证分析。评价结果为上海宝山基地的环境结合度最高。研究表明:环境结合度是“双碳”背景下钢铁新产线向既有基地选址时主要的环境关切,而环境结合度的评价框架和评价方法可有效表征连续时间序列上既有基地与所在城市环境体系的互促共进态势,对大型钢企未来选址新产线具有一定指导作用。 展开更多
