[Objectives]This study was conducted to systematically present the research and development status of Wuling Powder over the years, and comprehensively analyze the hot spots and trends of Wuling Powder in the treatment of diseases, so as to provide reference for scholars in this field in the future. [Methods] China National Knowledge Network(CNKI) database was used as the literature retrieval source to search the literature related to Wuling Powder in the treatment of nephropathy, and then the data visualization analysis was carried out by using the software citespace.5.7.R2. [Results] Through accurate retrieval, a total of 204 effective literatures were included. It was found that domestic research on the treatment of nephropathy by Wuling Powder was on the rise. From the analysis of knowledge graph, it was concluded that after long-term research and development, a relatively concentrated core research group had been formed in China, which mainly focused on "Wuling Powder", "chronic kidney disease" and "diabetic nephropathy". [Conclusions] The research on the treatment of nephropathy by Wuling Powder shows a good development trend, and the research content is constantly enriched, and this research is still the current and future research hotspot. 展开更多

Effects of Wuling Powder Mediating Notch Pathway on Mice with Nephrotic Syndrome

[Objectives]This study was conducted to investigate the renal protective effects of Wuling Powder on mice with nephrotic syndrome(NS)based on Notch pathway.[Methods]Sixty KM mice were randomly divided into normal group,model group,prednisone acetate positive group,high-dose Wuling Powder group,medium-dose Wuling Power group and low-dose Wuling Power group,with 10 mice in each group.Three days after prophylactic administration,a comprehensive nephropathy model was prepared by injecting 1 mg/ml doxorubicin hydrochloride solution(7.5 mg/kg)into the tail vein.After successful modeling,prednisone acetate and Wuling SAN were given high,medium and low doses for intervention for 28 d,respectively.After that,urinary protein and creatinine contents of mice in each group were detected,and pathological damage of renal tissue was observed by HE and Masson staining.The mRNA levels of Notch1,Jagged1 and Hes1 in mouse kidney tissues were detected by RT-PCR,and the expression levels of Notch1,Jagged1 and Hes1 proteins were detected by Western blot.[Results]Wuling Powder could effectively reduce the contents of urine protein(P<0.01)and Scr(P<0.01)in NS mice,and alleviate the pathological injury of kidney.Compared with the model group,the prednisone acetate group and various Wuling Powder groups could down-regulate the expressions of Notch1,Jagged1 and Hes1 mRNA in the kidney tissue of mice(P<0.01),and the expression of Notch1 protein in the renal tissue of mice decreased(P<0.01).The contents of Hes1 in the prednisone acetate group and the high-and medium-dose Wuling Powder groups significantly decreased(P<0.05).[Conclusions]Wuling Powder could protect the kidneys in mice with NS through Notch pathway. 展开更多

Efficacy and Safety of Wuling Powder and Western Medicine in Treatment of Hyperuricemia:A Meta-analysis of Clinical Trials

[Objectives]To compare the clinical efficacy and safety of Wuling powder and conventional Western medicine for lowering uric acid in the treatment of hyperuricemia.[Methods]Databases such as CNKI,Wanfang Data,VIP Database,PubMed,Cochrane library and Embase,were retrieved,and comparative experiments of efficacy(serum uric acid level)and safety(incidence of adverse reactions)between Wuling powder(used singly or combined with other prescriptions)and conventional Western medicine in the treatment of hyperuricemia were included,and effect size was pooled with Revman 5.3.[Results]A total of 11 articles,involving 817 patients,were included,and their research quality was evaluated to be low.In terms of overall effect,Wuling powder(used singly or combined with other prescriptions)was better than the control drug in reducing serum uric acid level,but the difference was insignificant[SMD=-0.37,95%CI(-0.90,0.16)].The heterogeneity between studies was high.The incidence of adverse reactions in the experimental group was significantly lower than that in the control group.Subgroup analysis shows that Wuling powder combined with other prescriptions was better than the control drug in reducing serum uric acid level[SMD=-0.37,95%CI(-0.90,0.16),P=0.17],and the heterogeneity was reduced.[Conclusions]Wuling powder combined with other prescriptions is better than conventional Western medicine in the treatment of hyperuricemia,and has better safety.Wuling powder alone is not as effective as conventional Western medicine.It is suggested that in clinical work,doctors may need to modify the prescription of Wuling powder.The low quality of the literature supporting this conclusion suggests that this evidence should be interpreted with caution. 展开更多

Wuling powder alleviates depressive-like behavior by attenuating endoplasmic reticulum stress in the mouse hippocampus

The response to endoplasmic reticulum(ER)stress has been noted in both human depression cases and depression models in rodents.Wuling powder,derived from the mycelium of the esteemed fungus Xylaria Nigripes(Kl.)Sacc,has demonstrated efficacy in alleviating depressive symptoms.The purpose of this research was to explore the antidepressant properties of Wuling powder and its basic molecular effects,particularly regarding alterations in ER stress.A model of social defeat stress was created by introducing a mouse to the cage of an unfamiliar,hostile mouse for intervals of 5–10 min daily over a span of 10 d.Subsequently,the mice received oral doses of Wuling powder for 2 weeks.The social approach-avoidance assay was employed to evaluate signs of depression-like behaviors.Moreover,protein and gene expressions linked to ER stress triggered by social defeat were analyzed through Western blotting analysis and quantitative real-time PCR.The behavioral tests indicated that Wuling powder ameliorated behaviors associated with depression due to social defeat stress.Treatment with Wuling powder markedly reduced the increased levels of the 78-k Da glucose-regulated protein and protein disulfide isomerase caused by social defeat stress.It also diminished the expression of inositol-requiring enzyme 1α(IRE1α)and spliced X box-binding protein-1(s XBP1)at the protein and m RNA levels.Furthermore,Wuling treatment notably decreased the levels of phosphorylated eukaryotic initiation factor 2 alpha kinase(P-e IF2α),activating transcription factor 4(ATF4)and C/EBP homologous protein(CHOP),simultaneously enhancing the ratio of B-cell lymphoma 2(Bcl-2)to Bcl-2-associated X protein(Bax).These results suggested that Wuling powder could alleviate ER stress and inhibit cell apoptosis in the hippocampus by inhibiting protein translation and synthesis,thereby attenuating depressive-like behavior. 展开更多

Investigation on the mechanisms of Wuling Powder in treating myocardial ischemia-reperfusion injury based on network pharmacology and molecular docking with experiment verification

Background Myocardial ischemia-reperfusion injury is characterized by the inability of tissue and organ function to recover after the perfusion blood flow is restored followed by myocardial ischemia.Previous studies have shown that many Chinese herbal compounds can reduce myocardial ischemia-reperfusion injury,but the efficacy of Wuling Powder in treating myocardial ischemia-reperfusion injury has not been reported.The present study aimed to investigate the mechanism of Wuling Powder in treating myocardial ischemia-reperfusion injury by network pharmacology and molecular docking.Methods The active constituents of Wuling Powder were obtained by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the related targets of active constituents of Wuling Powder were screened by the Swiss Target Prediction database.Disease targets of myocardial ischemia-reperfusion injury were obtained by Gene Cards,Dis Ge NET and OMIM gene databases.The active compound-target network of Wuling Powder was constructed using Cytoscape software.The drug prediction targets were mapped to the disease target set,and the intersection targets were obtained.The Gene Ontology(GO)function and Kyoto Encyclopedia of Genes anf Genomes(KEGG)pathway enrichment analysis were performed on the intersection targets obtained by using String database.Auto Dock Vina was used for molecular docking of core targets and the top 3 key active ingredients with degree values.Results Through screening,56 active ingredients and 116 targets of Wuling Powder were obtained.There were 27 common targets of Wuling Powder and myocardial ischemia-reperfusion injury in Myocardial ischemia-reperfusion injury(MIRI).Protein-Protein Interaction(PPI)network analysis in this study showed that CASP3,PTGS2,CAT,JUN,ESR1,CASP8,CASP9 and RELA may be the key targets of Wuling Powder to exert the protective effect of MIRI.GO functional analysis and KEGG pathway enrichment analysis showed that biological processes such as apoptosis,lipopolysaccharide response,steroid hormone response and signal transduction pathways such as PTGS2,TNF-α,nuclear transcription factor-κB(NF-κB),MAPK,PI3K/AKT,NLRP3,and autophagy play an important role in the occurrence and development of MIRI.Molecular docking results showed thatβ-sitosterol and hederagenin had good binding activities with key targets such as PTGS2,CASP3,and CAT.Conclusions Wuling Powder may act on CASP3,PTGS,CAT,JUN,ESR1,CASP8,CASP9 and other targets throughβ-sitosterol,hederagenin,3β-acetoxyatractylone,taxifolin and other active ingredients.[S Chin J Cardiol 2024;25(3):179-192] 展开更多

Osteoporosis is a health problem to cause global concern.A lot of methods have been used to prevent and treat osteoporosis,but there is still a lack of effective treatment for osteoporosis owing to limited understanding of its mechanism.Therefore,the aim of this present study is to explore the underlying mechanism of Wuling Powder,a traditional Chinese medicine on treating osteoporosis.In this study,we firstly screened and identified the common targets between Wuling Powder and osteoporosis through the related databases,and then explored the relationships among these targets,Wuling Powder and osteoporosis by using Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)and network analyses.Subsequently,the molecular docking was performed by using systemsDock to evaluate the potential binding relationships between the active components of Wuling Powder and their related targets.The results showed that in total of 14 common targets including CREBBP,ADAM17,GOT1,GAPDH,USP8,ERBB2,EEF1 A1,MTOR,RAC1,ETS1,DDX58,GCK,EGF and S100 A8 were screened.EGF,ERBB2,MTOR and HIF-1 were the potential therapeutic targets for osteoporosis,and they were also the related targets for predicting active components in Wuling Powder.Taken together,we concluded that Wuling Powder might be used to treat osteoporosis through above these targets. 展开更多
