To observe the effect of Gardenia extract ZG on the adsorption quantity of herpes simplex virus type 1 (HSV-1) so as to explore the mechanism of its antiviral activity, fluorescein isothiocyanate (FITC) was used as the fluorescent probe to label viruses and heparin sodium was used as control. Meanwhile, the effect of Gardenia extract ZG on the adsorption quantity on the surface of Hep-2 cells was determined by flow cytometry. It was demonstrated that adsorption of HSV-1 on the surface of Hep-2 cells exhibited the character of saturation and specificity and heparin sodium could prevent attachment of viruses on these ceils. These results are in accord with those reported previously. It was also proved that the manner of drug-use prior to adsorption or simultaneous use of drug and adsorption was better than adsorption prior to drug-use, and the inhibition rates of the former and latter manner were 84.76% and 82.92% respectively. Three manners of drug-use with Gardenia extract ZG were all effective to reduce the adsorption quantity of viruses, especially the manner of simultaneous use of drug and adsorption with an adsorption inhibition rate of 68.46%. From the above observation, it is apparent that the mechanism of anti-viral activity of Gardenia extract ZG may be via several steps involved in the HSV-1 adsorption. 展开更多

Given the problem of the long time required for testing gas pressure, we propose a fast-test method in which we used a technique of fast borehole sealing and air replenishing. Based on the characteristics of gas emission from boreholes to be tested, we built a theoretical model for calculating parameters during the process of increasing natural pressure and aeration. Using this model, we investigated the effect of different aeration conditions on velocity of pressure tests. The result shows that: 1) aerating air into boreholes can speed up gas pressure tests and 2) the more similar the pressure of the aerated air to the original gas pressure, the smaller the gas volume absorbed by coal and the shorter the time needed in pressure test. A case study in the Lu’an mining area shows that the time needed for gas pressure test is only 4 h using our method of aeration and 29 h under conditions of increasing natural pressure, saving time by 86.2%. This case study also indicates that, by using the aeration method, only one hour is needed for gas pressure to reach a stable state, which breaks the record of the shortest time needed for gas pressure tests in China. 展开更多

Microorganisms being used in Cd-contaminated soil remediation have broad prospects in application.In order to obtain more Cd-tolerant microbial resources,paddy soils around lead-zinc tailings were collected to extract Cdtolerant bacterial strains by using the methods of enrichment culture,purification and gradient screening.The strains were identified by morphological and biochemical characteristics and 16S rDNA sequence analysis.The Cd-tolerance and Cdadsorption abilities were then analyzed.The results showed that 6 Cd-tolerant bacterial strains were isolated,3 strains could grow in solid medium which had a cadmium concentration of 700 mg/L.These three strains(i.e.KGJ,KDT,and TU)were members of the Pseudomonas genus(Pseudomonas sp.)identified by morphological and biochemical characteristics and 16S rDNA sequence analysis.The growth curve indicated that the growth rates of the strains were obviously inhibited under 250 mg/L cadmium treatment.The three strains were tolerant of cadmium concentrations over 250 mg/L for long periods of time up to the stationary phase,and TU had the strongest Cd-tolerance.The order of metal tolerance of TU to different heavy metals was Cd^2+ >Zn^2+ >Pb^2+respectively.Chemical analysis showed that the adsorption quantity of the 3 bacterial strains of KGJ,KDT,and TU were 7.596 mg/g,12.237 mg/g and 36.926 mg/g respectively by dry-weight,when the concentration of cadmium was 150 mg/L.All 3 strains had good tolerance and ability to adsorb cadmium,and therefore are considered important potential microbial resources for the remediation of Cd pollution in soil. 展开更多
