To study the resistant mechanism and clinical significance of pseudomonas aeruginosa toβ- lactam antibiotics,the outer mem brane permeability rate of30 P.aeruginosa strains to5 β- lactam antibiotics was m easured and their production ofβ- lactamase and theβ- lactam ase genes they carried detected. Furthermore,the relationship between the perm eability,β- lactam ase and the clinical effects ofβ- lactam antibiotics was observed. By using 1 4C- penicillin and liquid- scintillant isotope assay,the affinity of penicillin binding proteins(PBPs) was m easured and their roles in the resistant m echanism studied.Itwas revealed thatthe perm eability rate was higher in sensitive strains than in resistantones(P<0 .0 5 ) .All strains harbored1- 4 β- lactamase genes and produced β- lactam ase.Higher permeability rate and higher degree of stability toβ- lactamase indicated better clinical therapeutic effects. The affinity of PBPs changed little without regard to the perm eability andβ- lactam ase. These results suggested that the permeability of outer mem brane andβ- lacta- mase,but not PBPs,played im portant roles in the resistant mechanism of P. aeruginosa toβ- lac- tam antibiotics and affected the clinical therapeutic effectiveness of som e patients. 展开更多

The effect of protective genital care protocol on preventing diaper dermatit development in 0 - 18 month old children using antibiotics

Diaper dermatit is among the widely seen inflamatory skin diseases in neonatals and small 0 - 18 month old children. Control of causative agents as well as providing good genital care may prevent or reduce the risk of the disease. The aim of this study is find out the effect of protective genital care on reducing the diaper dermatit development in 0 - 18 months old inpatient 0 - 18 month old children who are using antibiotic. The study was conducted as prospective half-experimetal using control-experimental groups. Our samples comprise of 0 - 18 month old children admitted at Dokuz Eylul University Practical and Research Hospital, pediatric clinic between January and May 2009, 41 0 - 18 month old children as a control group were not under protective genital care, and 42 0 - 18 month old children as an experimental group were under the protecion and using vaseline. The mothers of the experimental group 0 - 18 month old children were given education. All 0 - 18 month old children were followed-up 3 - 7 days and the collected data were evaluated in computer using Pearson’s chisquare, Fisher’s exact chi-square and Yatest-corrected chi-square tests. K You 48-2 is a new combination of early hybrid rice with good grain quality and high yield, developed by combining male sterile line K-Qing A having japonica cytoplasm and indica genome and restorer line Ce 48-2. In the regional trial of Hubei Province in 1991, its yield was 4.4% higher than that of the CK Weiyou 48-2, being significant at 0.05 level. In the same year, in another regional trial of Sichuan Province, its yield was 8.9% higher than that of the CK Weiyou 48-2, being significant at 0.01 level. It has the highest yield among all tested varieties in both provincial trials. It has shown good grain quality with 60% head rice recovery and moderate resistance to rice blast. 展开更多

云对全球的能量收支及大气循环系统具有不可替代的调节作用,是影响天气及气候变化的重要因子。云顶高度作为云的重要参数,有助于分析云在大气中的物理机制,对局部地区监测和预报具有实质性作用。基于多通道扫描辐射成像仪(AGRI)数据反... 云对全球的能量收支及大气循环系统具有不可替代的调节作用,是影响天气及气候变化的重要因子。云顶高度作为云的重要参数,有助于分析云在大气中的物理机制,对局部地区监测和预报具有实质性作用。基于多通道扫描辐射成像仪(AGRI)数据反演东北地区云顶高度,采用红外分裂窗查算表方法,并对不同季节和不同云类型建立查算表,探讨基于静止卫星反演中高纬度地区云顶高度的可行性。同时,引入主动式高精度仪器正交偏振云-气溶胶偏振雷达(CALIOP),结合11、12μm通道亮温数据搭建云顶高度反演模型。分析结果表明:1)AGRI遥感的云顶高度与CALIPSO搭载的CALIOP云顶高度变化趋势一致,算法计算效率较高;2)从4个季节的统计结果来看,均方根误差(RMSE)上都小于2.3 km,平均偏差在500 m左右,受季节的影响较小,反演结果与CALIOP探测结果具有较高的一致性;3)利用该方法基于AGRI结果精度不亚于官方采用的CO_(2)薄片法,该方法估算的RMSE和相对误差较小,并且该算法的反演模型只提取红外分裂窗区通道数据,无需大气廓线资料进行辅助计算,节省计算资源,提高计算效率。 展开更多

Salmonella enterica serovar 1,4,[5],12:i:-(S.1,4,[5],12:i:-)has been recognized as an emerging foodborne pathogen in recent years.It can cause human salmonellosis predominated by the contamination of animal-derived foods such as raw poultry and pork.This study aimed to characterize the genetic diversity,plasmid replicon types,and antibiotic resistance of 15 S.1,4,[5],12:i:-environmental isolates collected from two poultry farms using pulsed-field gel electrophoresis(PFGE),multilocus sequence typing(MLST),polymerase chain reaction-based replicon typing,and minimum inhibitory concentration approach.Ten different PFGE genotypes were detected,indicating a high diversity among these S.1,4,[5],12:i:-isolates.Three sequence types(ST19,ST1544,ST34)were identified by MLST.Among them,ST1544 was first detected in S.1,4,[5],12:i:-environmental isolates from poultry farms.All isolates were resistant to cefazolin,cefotetan,tobramycin,amikacin,and gentamicin,but susceptible to piperacillin-tazobactam,aztreonam,ceftazidime,cefepime,and ertapenem.Five incompatibility groups(Inc)of plasmids were identified,including IncFIIs(66.7%),IncHI2(20%),IncI1(6.7%),IncN(6.7%),and IncQ(6.7%).Among these isolates,80%carried at least one plasmid replicon type,and 20%carried multiple plasmid replicon types.Interestingly,the multidrug-resistant isolate 263 carried numerous resistance genes(i.e.qnrS,aac(6ʹ)-Ib-cr,bla_(TEM),bla_(CTX-M-9),bla_(OXA-1),sul1,sul2,sul3,floR,and mcr-1)and class I integronase gene intI1,which possessed both IncHI2 and IncQ plasmids,suggesting that resistance genes may be horizontally transferred by the combination of IncHI2 and IncQ plasmids.Collectively,antibiotic-resistant S.1,4,[5],12:i:-isolates were first found in poultry farm environments in China,and surveillance should be strengthened to prevent their further spread from poultry farms to foods. 展开更多

Objective: To evaluate the antimicrobial activity of total alkaloids extracted from Sophorea alopecuroides L. (TASA) against clinical isolated extended-spectrum beta-lactamases (ESBLs) producing Escherichia coil (E.. coil) strains. Methods: The antibacterial activity of TASA either itself or in combination with cefotaxime (CTX) or ceftazidime (CAZ) was investigated by using the microbroth dilution method and phenotypic confirmatory disk diffusion test against three clinical isolated ESBLs-producing E. coil strains; the interactions of TASA and C'I'X or CAZ were ascertained by evaluating the fractional inhibitory concentration index (FICI). Results: The antibacterial activity of either TASA itself or in combination with C'IX or CAZ was found. The minimum inhibitory concentration (MICs) of TASA against the ESBLs producing isolates was 12.5 mg/mL. In the combinations with a sub-inhibitory concentration of TASA, a synergistic effect on CTX and CAZ against the ESBLs producing isolates was observed. Similarly, the isolates exposed to lower dose of TASA yielded an increased susceptibility to CTX and CAZ by 8-16 folds determined by microdilution assay. Moreover, enzymatic detection of ESBLs demonstrated that TASA induced reversal resistance to CTX and CAZ partially by a mechanism of inhibition of ESBLs activity in these isolates. Additionally, in the tested isolates following the exposure of TASA, molecular analysis verified the SHV-type beta-lactamase encoding ESBL gene in these isolates, and no mutation was introduced into the ESBL gene. Conclusions: These results suggest that TASA could be used as a source of natural compound with pharmacological activity of reversal resistance to antimicrobial agent. These findings also indicated that the application of the TASA in combination with antibiotics might prove useful in the control and treatment of infectious diseases caused by the ESBLs producing enterobacteriaceae. 展开更多

Theβ-lactam antibiotic resistance caused by NDM-1 has become a major crisis of global public health.We have previously screened out(-)-epicatechin gallate(ECG)as a potent NDM-1 inhibitor.We further discussed its inhibitory effect and action mode in the present study.According to our results,ECG reversibly inactivated NDM-1 in a non-competitive mode,with an IC50 value of 4.48μM.ECG effectively recovered the activity of severalβ-lactam antibiotics against resistant strain harboring blaNDM-1.Especially,the effects on carbapenems were worth mentioning.The zinc supplement assay indicated a zinc-related mechanism of ECG.Different from traditional chelating agents,it showed low toxicity both in vivo and in vitro.In a word,our findings provided a promising NDM-1 inhibitor,ECG,which was able to assist carbapenems against NDM-1-producing strain. 展开更多
