Phenylalanine ammonia lyase(PAL)is the rate-limiting and pivotal enzyme of the general phenylpropanoid path-way,but few reports have been found on PAL genes in Pinus yunnanensis.In the present study,three PAL genes we...Phenylalanine ammonia lyase(PAL)is the rate-limiting and pivotal enzyme of the general phenylpropanoid path-way,but few reports have been found on PAL genes in Pinus yunnanensis.In the present study,three PAL genes were cloned and identified from P.yunnanensis seedlings for thefirst time,namely,PyPAL-1,PyPAL-2,and PyPAL-3.Our results indicated that the open-reading frames of PyPAL genes were 2184,2157,and 2385 bp.Phylogenetic tree analysis revealed that PyPALs have high homology with other known PAL genes in other plants.In vitro enzymatic analysis showed that all three PyPAL recombinant proteins could catalyze the deamination of L-phenylalanine to form trans-cinnamic acid,but only PAL1 and PAL2 can catalyze the conversion of L-tyrosine toρ-coumaric acid.Three PyPAL genes were expressed in different tissues in 1-year-old P.yunnanensis,and such genes had different expression patterns.This study lays a foundation for further understanding of the biosynthesis of secondary metabolites in P.yunnanensis.展开更多
Plant-based fermentations provide an untapped source for novel biotechnological applications.In this study,a probiotic named Lactobacillus fermentum 21828 was introduced to ferment Lentinus edodes.Polysaccharides were...Plant-based fermentations provide an untapped source for novel biotechnological applications.In this study,a probiotic named Lactobacillus fermentum 21828 was introduced to ferment Lentinus edodes.Polysaccharides were extracted from fermented and non-fermented L.edodes and purified via DEAE-52 and Sephadex G-100.The components designated F-LEP-2a and NF-LEP-2a were analyzed by FT-IR,HPGPC,HPAEC,SEM,GC-MS and NMR.The results revealed that probiotic fermentation increased the molecular weight from 1.16×10^(4) Da to 1.87×10^(4) Da and altered the proportions of glucose,galactose and mannose,in which glucose increased from 45.94%to 48.16%.Methylation analysis and NMR spectra indicated that F-LEP-2a and NF-LEP-2a had similar linkage patterns.Furthermore,their immunomodulatory activities were evaluated with immunosuppressive mice.NF-LEP and F-LEP improved immune organ indices,immunoglobulin(Ig G and Ig M)and cytokines concentrations;restored the antioxidation capacity of liver;and maintained the balance of gut microbiota.F-LEP displayed better moderating effects on the spleen index,immunoglobulin,cytokines and the diversity of gut microbiota than NF-LEP(200,400 mg/kg).Our study provides an efficient and environment-friendly way for the structural modification of polysaccharides,which helps to enhance their biological activity and promote their wide application in food,medicine and other fields.展开更多

Free radicals scavenging activity of PAP was studied through 3 free radicals scavenging tests. We make the comparison of 11-year’s phase between relative sunspot numbers and sunspot group numbers. Objective:To elucidate its pharmacological activities and medicinal potential of extract of Etlingera elatior(E.elatior).Methods:Phytochemical screening of the flower extract was done to determine the phytochemical ...Objective:To elucidate its pharmacological activities and medicinal potential of extract of Etlingera elatior(E.elatior).Methods:Phytochemical screening of the flower extract was done to determine the phytochemical in the extract.The pharmacological study included the determination of antimicrobial activity and minimum inhibitory concentration(MIC) of metabolic flower extract.The antimicrobial activity of the extract was tested against medically important bacterial,yeast and fungal strains.Apart from that,the methanolic extract of E.elatior flower was further tested in vivo toxicity using the brine shrimp lethality test.Moreover,the flower extract was qualitatively screened for their free radical scavenging activity by 2,2-diphenyl-1- picrylhydrazyl radical(DPPH) assay.Results:The extract was effective on tested microorganisms and MIC values were in the range of 1.563-50.000 mg/mL.The brine shrimp lethality test exhibited no significant toxicity(LC<sub>50</sub>= 2.52 mg/mL) against Artemia salina.The E.elatior flower extract with high LC<sub>50</sub> value signified that this plant is not toxic to humans.While the phytochemical screening of the flower extract revealed the presence of the following compounds: flavonoids,terpenoids,saponin,tannins and carbohydrates whereas,alkaloids,anthraquinone and reducing sugars were absent.The concentration of the flower extract required for 50% inhibition of DPPH radical scavenging effect(IC<sub>50</sub>) were 9.14 mg/mL and 8.08 mg/mL for butylated hydroxytoluene 8.08 mg/mL.Conclusions:These findings indicate that the extract of E.elatior flower possesses pharmacological properties and potential to develop natural products based pharmaceuticals products.展开更多
Continued innovation in screening methodologies remains important for the discovery of high-quality multiactive fungi,which have been of great significance to the development of new drugs.Mangrove-derived fungi,which ...Continued innovation in screening methodologies remains important for the discovery of high-quality multiactive fungi,which have been of great significance to the development of new drugs.Mangrove-derived fungi,which are well recognized as prolific sources of natural products,are worth sustained attention and further study.In this study,118 fungi,which mainly included Aspergillus spp.(34.62%)and Penicillium spp.(15.38%),were isolated from the mangrove ecosystem of the Maowei Sea,and 83.1%of the cultured fungi showed at least one bioactivity in four antibacterial and three antioxidant assays.To accurately evaluate the fungal bioactivities,the fungi with multiple bioactivities were successfully evaluated and screened by principal component analysis(PCA),and this analysis provided a dataset for comparing and selecting multibioactive fungi.Among the 118 mangrove-derived fungi tested in this study,Aspergillus spp.showed the best comprehensive activity.Fungi such as A.clavatonanicus,A.flavipes and A.citrinoterreus,which exhibited high comprehensive bioactivity as determined by the PCA,have great potential in the exploitation of natural products and the development of new drugs.This study demonstrated the first use of PCA as a time-saving,scientific method with a strong ability to evaluate and screen multiactive fungi,which indicated that this method can affect the discovery and development of new drugs.展开更多

By using the NCEP, SST and JTWC data, the causes of the bimodal pattern storm activity of the...Storms that occur at the Bay of Bengal(Bo B) are of a bimodal pattern, which is different from that of the other sea areas. By using the NCEP, SST and JTWC data, the causes of the bimodal pattern storm activity of the Bo B are diagnosed and analyzed in this paper. The result shows that the seasonal variation of general atmosphere circulation in East Asia has a regulating and controlling impact on the Bo B storm activity, and the "bimodal period" of the storm activity corresponds exactly to the seasonal conversion period of atmospheric circulation. The minor wind speed of shear spring and autumn contributed to the storm, which was a crucial factor for the generation and occurrence of the"bimodal pattern"storm activity in the Bo B. The analysis on sea surface temperature(SST) shows that the SSTs of all the year around in the Bo B area meet the conditions required for the generation of tropical cyclones(TCs). However,the SSTs in the central area of the bay are higher than that of the surrounding areas in spring and autumn, which facilitates the occurrence of a "two-peak"storm activity pattern. The genesis potential index(GPI) quantifies and reflects the environmental conditions for the generation of the Bo B storms. For GPI, the intense low-level vortex disturbance in the troposphere and high-humidity atmosphere are the sufficient conditions for storms, while large maximum wind velocity of the ground vortex radius and small vertical wind shear are the necessary conditions of storms.展开更多
Background:Outdoor physical activity(PA)brings important health benefits,but exposure to polluted air increases health risks.This study aimed to quantify the tradeoff of PA under fine particulate matter(PM_(2.5))air p...Background:Outdoor physical activity(PA)brings important health benefits,but exposure to polluted air increases health risks.This study aimed to quantify the tradeoff of PA under fine particulate matter(PM_(2.5))air pollution by estimating the optimal PA duration under various pollution levels.Methods:A risk-benefit analysis was performed to estimate the optimal outdoor moderate-intensity PA(MPA)duration under varying PM_(2.5) concentrations.Results:An inverse nonlinear relationship was identified between optimal MPA duration and background PM_(2.5) concentration levels.When background PM_(2.5) concentration increased to 186 μg/m^(3),the optimal outdoor MPA duration decreased to 2.5 h/week,the minimum level recommended by current PA guidelines.When background PM_(2.5) concentration further increased to 235 μg/m^(3),the optimal outdoor MPA duration decreased to 1 h/week.The relationship between optimal MPA duration and background PM_(2.5) concentration levels was stronger when exercising at a location closer to a source of air pollution.Compared to the general adult population,adults aged 60 years and older had substantially steeper curves—the optimal outdoor MPA duration decreased to 2.5 h/week when background PM_(2.5) concentration reached 45 μg/m^(3).Conclusion:The health benefit of outdoor MPA by far outweighs the health risk of PM_(2.5) pollution for the global average urban background concentration(22 μg/m^(3),).This modeling study examined a single type of air pollutant and suffered from measurement errors and estimation uncertainties.Future research should examine other air pollutants and indoor PA,incorporate short-and mid-term health effects of MPA and air pollution into the risk-benefit analysis,and provide estimates specific for high-risk subgroups.展开更多

Four Gram positive bacterial cultures (HA-21, HA-68, HA- MS-105 and HA-MS-119) were isolated from the sponge Amphimedon ochracea, collected from the Red Sea coast of Egypt. Bacterial species were identified based on the phylogenetic analysis of the nucleotide sequences of their 16S rDNA genes. The Sequences similarity values of 98% - 100% to other strains in the NCBI database showed strong similarities with the 16S rDNA genes of firmicutes (Bacillus sp.). The four bacterial species were submitted to the GenBank database and had accession numbers of: HA-21 [JQ-768238];HA-68 [JQ751264];HA-MS-105 [JQ768239];HAMS-119 [JQ768240]. The cytotoxic activities of the bacterial isolates were tested against three established human cancer cell lines;HepG2 (hepatocellular carcinoma), HCT (colon carcinoma) and MCF-7 (breast carcinoma). The inhibitory effect on these cell lines, measured by MTT cell assay protocol, revealed promising cytotoxic activity of the four isolates (IC50 values (μg/mL) were: HA-21: 13.2, 9.3 and 12.2;HA-68: 10.42, 4.3 and 5.5;HA-MS-105: 46.9, 28.6 and 21.3;HAMS-119: 10.42, 6.3 and 22.1;respectively). The recovery of bacterial strains with cytotoxic activity suggests that marine invertebrates remain a rich source for the isolation of culturable isolates capable of producing novel bioactive secondary metabolites.展开更多

However, this method presents problems owing to tedious quantitation, radioisotopic handling. In order to alleviate these inconveniences, a novel telomerase DNA sequencing assay together with TRAP to detect human telomerase activity was developed. It was used to detect telomerase activity in Hela, HLF, MCF, K562, SMMC 7721 cells, Leukocytes and RNase pretreated or heat treated cells as control. Telomerase activity assayed by this method was positive when the number of K562 cells examined was 102,103, and 104. The telomerase activity depended on the number of K562 cells used in the assay. Telomerase activity of Rnase pretreated cells or heat treated cells, and human normal peripheral blood leukocyte(Leu) were negative. The result of this method was available within a few hours and was handled without radioisotope. Further studies should be taken to detect telomerase activity in quantitation.展开更多

[Method] By using lightning location data, basic characteristics of lightning activities in Lianyungang and Yancheng both located at the eastem coast of Jiangsu were analyzed and contrasted. [Result] CG characteristics in Uanyungang and Yancheng were same, and negative CG was dominant. Monthly change activity rule of CG basically presented single-peak distribution, and peak value appeared in August. Monthly change of the lightning was drastic, and high- occurrence period was both in summer. Daily change trend of CG in the two sites was consistent. High-occurrence period of the lightning was in af- temoon and early morning, and occurrence frequency of the lightning in the morning was less. Spatial distribution of CG in Lianyungang and Yancheng was very uneven. Lightning activity in Lianyungang mainly concentrated in northwest and southeast areas of Donghai County and western area of Guannan County. Especially in Donghai County, it was the area where lightning strike density was the maximum. Lightning strike activity in Yanchang was more concentrated, and mainly concentrated in the west, namely near Jiangying Town and Louwang Town. Distribution characteris- tics of CG intensities in the two sites were same. Waveform changes of the positive CG in the two sites were slower than that of the negative CG, and current intensity concentrated from + 20 to ~ 50 kA. [ Conclusion ] By contrasting and analyzing lightning activity rules in Lianyungang and Yanchang, we could share and complement data in the two cities better, and serve for routine lightning prevention and early-warning better.展开更多

The plant is aromatic, stimulant and used for allaying nausea, headache and vomiting. Its oil is one of the most popular widely used essential oils in food products, cosmetics, pharmaceuticals, dental preparations, mouthwashes, soaps and alcoholic liquors. The antibacterial potential of six extracts from leaf, stem and root of Mentha piperita against pathogenic bacteria such as Bacillus subtilis, Streptococcus pneumonia, Staphylococcus aureus, Escherichia coli, Proteus vulgaris and Klebsiella pneumonia were evaluated by agar well diffusion method. The organic (ethanol, methanol, ethyl acetate, chloroform, hexane and petroleum ether) extracts of the leaves were found to possess strong antibacterial activity against a range of pathogenic bacteria. The ethyl acetate leaf extract of Mentha piperita showed pronounced inhibition than chloroform, petroleum ether and hexane. The leaf extract activity being more on Bacillus subtilis, Staphylococcus aureus and Proteus vulgaris than Escherichia coli, Streptococcus pneumonia and Klebsiella pneumonia. In the present study, we also evaluated the phytochemical analysis for the presence of various secondary metabolites. The analysis revealed the presence of alkaloids, flavonoids, steroids, tannins, and phenols.展开更多

Temporal activity pattern is an in...Temporal activity patterns in animals emerge from complex interactions between choices made by organisms as responses to biotic interactions and challenges posed by external factors. Temporal activity pattern is an inherently continuous process, even being recorded as a time series. The discreteness of the data set is clearly due to data-acquisition limitations rather than a true underlying discrete nature of the phenomenon itself. Therefore, curves are a natural representation for high-frequency data. Here, we fully model temporal activity data as curves integrating wavelets and functional data analysis, allowing for testing hypotheses based on curves rather than on scalar and vector-valued data. Temporal activity data were obtained experimentally for males and females of a small-bodied marsupial and modelled as wavelets with independent and identically distributed errors and dependent errors. The null hypothesis of no difference in temporal activity pattern between male and female curves was tested with functional analysis of variance (FANOVA). The null hypothesis was rejected by FANOVA and we discussed the differences in temporal activity pattern curves between males and females in terms of ecological and life-history attributes of the reference species. We also performed numerical analysis that shed light on the regularity properties of the wavelet bases used and the thresholding parameters.展开更多

文摘Plant-based fermentations provide an untapped source for novel biotechnological applications.In this study,a probiotic named Lactobacillus fermentum 21828 was introduced to ferment Lentinus edodes.Polysaccharides were extracted from fermented and non-fermented L.edodes and purified via DEAE-52 and Sephadex G-100.The components designated F-LEP-2a and NF-LEP-2a were analyzed by FT-IR,HPGPC,HPAEC,SEM,GC-MS and NMR.The results revealed that probiotic fermentation increased the molecular weight from 1.16×10^(4) Da to 1.87×10^(4) Da and altered the proportions of glucose,galactose and mannose,in which glucose increased from 45.94%to 48.16%.Methylation analysis and NMR spectra indicated that F-LEP-2a and NF-LEP-2a had similar linkage patterns.Furthermore,their immunomodulatory activities were evaluated with immunosuppressive mice.NF-LEP and F-LEP improved immune organ indices,immunoglobulin(Ig G and Ig M)and cytokines concentrations;restored the antioxidation capacity of liver;and maintained the balance of gut microbiota.F-LEP displayed better moderating effects on the spleen index,immunoglobulin,cytokines and the diversity of gut microbiota than NF-LEP(200,400 mg/kg).Our study provides an efficient and environment-friendly way for the structural modification of polysaccharides,which helps to enhance their biological activity and promote their wide application in food,medicine and other fields.
文摘The issue of the extremely imbalanced gender ratio in preschool teachers has received widespread attention,and there are few studies on teacher-child verbal interaction behavior based on gender differences in preschool teachers.This article takes the“Little Light Bulb Is On”,a scientific exploration activity done by 5-6 years old kindergarten students as an example,and uses the improved Flanders Interaction Analysis System(iFIAS)as a tool to analyze the speech interaction behavior of male and female preschool teachers.The research results indicate that there are gender differences in teacher child language interaction between male and female teachers in terms of the atmosphere,the teaching structure,the teaching tendency,the way of raising questions,and the overall trend of interaction.

文摘A crude polysaccharide was extracted from Physalis alkekengi L. fruit. HPLC was used for the component analysis of the polysaccharide. The results indicate that Physalis alkekengi L. polysaccharide(PAP) was composed of rhamnose, xylose, arabinose, galactose, and glucose. Free radicals scavenging activity of PAP was studied through 3 free radicals scavenging tests. PAP exhibited high scavenging effects on OH and DPPH radicals, and both the scavenging rates were about 80%. The scavenging rate of O2^- radical was about 22%.

文摘Properties of the Schwabe cycles in solar activity are investigated by using wavelet transform. We study the main range of the Schwabe cycles of the solar activity recorded by relative sunspot numbers, and find that the main range of the Schwabe cycles is the periodic span from 8-year to 14-year. We make the comparison of 11-year’s phase between relative sunspot numbers and sunspot group numbers. The results show that there is some difference between two phases for the interval from 1710 to 1810, while the two phases are almost the same for the interval from 1810 to 1990.

文摘Objective:To elucidate its pharmacological activities and medicinal potential of extract of Etlingera elatior(E.elatior).Methods:Phytochemical screening of the flower extract was done to determine the phytochemical in the extract.The pharmacological study included the determination of antimicrobial activity and minimum inhibitory concentration(MIC) of metabolic flower extract.The antimicrobial activity of the extract was tested against medically important bacterial,yeast and fungal strains.Apart from that,the methanolic extract of E.elatior flower was further tested in vivo toxicity using the brine shrimp lethality test.Moreover,the flower extract was qualitatively screened for their free radical scavenging activity by 2,2-diphenyl-1- picrylhydrazyl radical(DPPH) assay.Results:The extract was effective on tested microorganisms and MIC values were in the range of 1.563-50.000 mg/mL.The brine shrimp lethality test exhibited no significant toxicity(LC<sub>50</sub>= 2.52 mg/mL) against Artemia salina.The E.elatior flower extract with high LC<sub>50</sub> value signified that this plant is not toxic to humans.While the phytochemical screening of the flower extract revealed the presence of the following compounds: flavonoids,terpenoids,saponin,tannins and carbohydrates whereas,alkaloids,anthraquinone and reducing sugars were absent.The concentration of the flower extract required for 50% inhibition of DPPH radical scavenging effect(IC<sub>50</sub>) were 9.14 mg/mL and 8.08 mg/mL for butylated hydroxytoluene 8.08 mg/mL.Conclusions:These findings indicate that the extract of E.elatior flower possesses pharmacological properties and potential to develop natural products based pharmaceuticals products.

文摘Continued innovation in screening methodologies remains important for the discovery of high-quality multiactive fungi,which have been of great significance to the development of new drugs.Mangrove-derived fungi,which are well recognized as prolific sources of natural products,are worth sustained attention and further study.In this study,118 fungi,which mainly included Aspergillus spp.(34.62%)and Penicillium spp.(15.38%),were isolated from the mangrove ecosystem of the Maowei Sea,and 83.1%of the cultured fungi showed at least one bioactivity in four antibacterial and three antioxidant assays.To accurately evaluate the fungal bioactivities,the fungi with multiple bioactivities were successfully evaluated and screened by principal component analysis(PCA),and this analysis provided a dataset for comparing and selecting multibioactive fungi.Among the 118 mangrove-derived fungi tested in this study,Aspergillus spp.showed the best comprehensive activity.Fungi such as A.clavatonanicus,A.flavipes and A.citrinoterreus,which exhibited high comprehensive bioactivity as determined by the PCA,have great potential in the exploitation of natural products and the development of new drugs.This study demonstrated the first use of PCA as a time-saving,scientific method with a strong ability to evaluate and screen multiactive fungi,which indicated that this method can affect the discovery and development of new drugs.

文摘Storms that occur at the Bay of Bengal(Bo B) are of a bimodal pattern, which is different from that of the other sea areas. By using the NCEP, SST and JTWC data, the causes of the bimodal pattern storm activity of the Bo B are diagnosed and analyzed in this paper. The result shows that the seasonal variation of general atmosphere circulation in East Asia has a regulating and controlling impact on the Bo B storm activity, and the "bimodal period" of the storm activity corresponds exactly to the seasonal conversion period of atmospheric circulation. The minor wind speed of shear spring and autumn contributed to the storm, which was a crucial factor for the generation and occurrence of the"bimodal pattern"storm activity in the Bo B. The analysis on sea surface temperature(SST) shows that the SSTs of all the year around in the Bo B area meet the conditions required for the generation of tropical cyclones(TCs). However,the SSTs in the central area of the bay are higher than that of the surrounding areas in spring and autumn, which facilitates the occurrence of a "two-peak"storm activity pattern. The genesis potential index(GPI) quantifies and reflects the environmental conditions for the generation of the Bo B storms. For GPI, the intense low-level vortex disturbance in the troposphere and high-humidity atmosphere are the sufficient conditions for storms, while large maximum wind velocity of the ground vortex radius and small vertical wind shear are the necessary conditions of storms.
文摘Background:Outdoor physical activity(PA)brings important health benefits,but exposure to polluted air increases health risks.This study aimed to quantify the tradeoff of PA under fine particulate matter(PM_(2.5))air pollution by estimating the optimal PA duration under various pollution levels.Methods:A risk-benefit analysis was performed to estimate the optimal outdoor moderate-intensity PA(MPA)duration under varying PM_(2.5) concentrations.Results:An inverse nonlinear relationship was identified between optimal MPA duration and background PM_(2.5) concentration levels.When background PM_(2.5) concentration increased to 186 μg/m^(3),the optimal outdoor MPA duration decreased to 2.5 h/week,the minimum level recommended by current PA guidelines.When background PM_(2.5) concentration further increased to 235 μg/m^(3),the optimal outdoor MPA duration decreased to 1 h/week.The relationship between optimal MPA duration and background PM_(2.5) concentration levels was stronger when exercising at a location closer to a source of air pollution.Compared to the general adult population,adults aged 60 years and older had substantially steeper curves—the optimal outdoor MPA duration decreased to 2.5 h/week when background PM_(2.5) concentration reached 45 μg/m^(3).Conclusion:The health benefit of outdoor MPA by far outweighs the health risk of PM_(2.5) pollution for the global average urban background concentration(22 μg/m^(3),).This modeling study examined a single type of air pollutant and suffered from measurement errors and estimation uncertainties.Future research should examine other air pollutants and indoor PA,incorporate short-and mid-term health effects of MPA and air pollution into the risk-benefit analysis,and provide estimates specific for high-risk subgroups.
文摘Most of marine sponges harbor dense and diverse microbial communities of bioactivity importance. Four Gram positive bacterial cultures (HA-21, HA-68, HA- MS-105 and HA-MS-119) were isolated from the sponge Amphimedon ochracea, collected from the Red Sea coast of Egypt. Bacterial species were identified based on the phylogenetic analysis of the nucleotide sequences of their 16S rDNA genes. The Sequences similarity values of 98% - 100% to other strains in the NCBI database showed strong similarities with the 16S rDNA genes of firmicutes (Bacillus sp.). The four bacterial species were submitted to the GenBank database and had accession numbers of: HA-21 [JQ-768238];HA-68 [JQ751264];HA-MS-105 [JQ768239];HAMS-119 [JQ768240]. The cytotoxic activities of the bacterial isolates were tested against three established human cancer cell lines;HepG2 (hepatocellular carcinoma), HCT (colon carcinoma) and MCF-7 (breast carcinoma). The inhibitory effect on these cell lines, measured by MTT cell assay protocol, revealed promising cytotoxic activity of the four isolates (IC50 values (μg/mL) were: HA-21: 13.2, 9.3 and 12.2;HA-68: 10.42, 4.3 and 5.5;HA-MS-105: 46.9, 28.6 and 21.3;HAMS-119: 10.42, 6.3 and 22.1;respectively). The recovery of bacterial strains with cytotoxic activity suggests that marine invertebrates remain a rich source for the isolation of culturable isolates capable of producing novel bioactive secondary metabolites.
文摘OBJECTIVE To investigate the hpyerglycemic activity and elucidate the active composition of the fruit of Phyllanthus emblica L.,an edible and medicinal plant used for traditional Chinese medicine and Tibetan medicine to cure the blood disease,bacon disease,liver disease,heart disease and hypertension.METHODS The LO2 normal liver cell lines were used to investigate the hpyerglycemic activity,The LO2 cells were treated with 100,200 and 400μg·mL-1 extracts of P.emblica L.;real-Time PCR was used to detect the expression of glucose transporter type 2(GLUT-2)and peroxisome proliferator-activated receptor-γ(PPARγ)mRNA.The PPRE(peroxisome proliferator response element)and NF-κB activities were detected by luciferase method.The chemical compositions were analyzed by HPLC-MS.RESULTS The expression of GLUT-2 and PPARγmRNA were significantly increased treated with 400μg·mL-1 extracts of P.emblica L.(P<0.05)and in a dose-dependant manner;PPRE activity was significantly increased in a dosedependant manner(P<0.05);however,NF-κB activity induced by LPS was inhibited also in a dose-dependant manner(P<0.05).The chemical composition analyzed by HPLC-MS showed that the gallic acid was the major component in P.emblica L.CONCLUSION P.emblica L.showed promising hpyerglycemic activity and the gallic acid was the main constituent.

文摘In this study, a novel phenoxyethylamine derivative 1-(bis(4-methoxy phenyl)-methyl)-4-(2-(2-methylphenoxy)ethyl)piperazine(C28 H35 N2O3, Mr = 454.58) has been synthesized and its structure was characterized by 1 H NMR, 13 C NMR, HRMS and single-crystal X-ray diffraction. The compound crystallizes in triclinic system, space group P1 with a = 6.1980(12),b = 12.212(2), c = 19.527(4)A,α= 93.08(3)°,β= 98.62(3)°,γ= 90.72(3)°, V = 1458.9(5)?3, Z =2, Dc = 1.035 g/cm^3, F(000)= 489,μ= 0.067 mm-1, Mo Kα radiation(λ= 0.71073 ?), the final R= 0.0693 and wR = 0.1759 for 3030 observed reflections with I > 2σ(I). The bioassay results indicated that the title compound 5 displayed effective activities against glutamine-induced neurotoxicity in PC12 cells.

文摘Telomeric repeat amplification protocol (TRAP) is now a conventional assay for detecting telomerase activity. However, this method presents problems owing to tedious quantitation, radioisotopic handling. In order to alleviate these inconveniences, a novel telomerase DNA sequencing assay together with TRAP to detect human telomerase activity was developed. It was used to detect telomerase activity in Hela, HLF, MCF, K562, SMMC 7721 cells, Leukocytes and RNase pretreated or heat treated cells as control. Telomerase activity assayed by this method was positive when the number of K562 cells examined was 102,103, and 104. The telomerase activity depended on the number of K562 cells used in the assay. Telomerase activity of Rnase pretreated cells or heat treated cells, and human normal peripheral blood leukocyte(Leu) were negative. The result of this method was available within a few hours and was handled without radioisotope. Further studies should be taken to detect telomerase activity in quantitation.

文摘[Objective] The reseamh aimed to contrast and analyze CG activity rules in Lianyungang and Yancheng. [Method] By using lightning location data, basic characteristics of lightning activities in Lianyungang and Yancheng both located at the eastem coast of Jiangsu were analyzed and contrasted. [Result] CG characteristics in Uanyungang and Yancheng were same, and negative CG was dominant. Monthly change activity rule of CG basically presented single-peak distribution, and peak value appeared in August. Monthly change of the lightning was drastic, and high- occurrence period was both in summer. Daily change trend of CG in the two sites was consistent. High-occurrence period of the lightning was in af- temoon and early morning, and occurrence frequency of the lightning in the morning was less. Spatial distribution of CG in Lianyungang and Yancheng was very uneven. Lightning activity in Lianyungang mainly concentrated in northwest and southeast areas of Donghai County and western area of Guannan County. Especially in Donghai County, it was the area where lightning strike density was the maximum. Lightning strike activity in Yanchang was more concentrated, and mainly concentrated in the west, namely near Jiangying Town and Louwang Town. Distribution characteris- tics of CG intensities in the two sites were same. Waveform changes of the positive CG in the two sites were slower than that of the negative CG, and current intensity concentrated from + 20 to ~ 50 kA. [ Conclusion ] By contrasting and analyzing lightning activity rules in Lianyungang and Yanchang, we could share and complement data in the two cities better, and serve for routine lightning prevention and early-warning better.
文摘Mentha piperita L. (Peppermint) is a perennial glabrous and strongly scented herb belonging to family Lamiaceae. The plant is aromatic, stimulant and used for allaying nausea, headache and vomiting. Its oil is one of the most popular widely used essential oils in food products, cosmetics, pharmaceuticals, dental preparations, mouthwashes, soaps and alcoholic liquors. The antibacterial potential of six extracts from leaf, stem and root of Mentha piperita against pathogenic bacteria such as Bacillus subtilis, Streptococcus pneumonia, Staphylococcus aureus, Escherichia coli, Proteus vulgaris and Klebsiella pneumonia were evaluated by agar well diffusion method. The organic (ethanol, methanol, ethyl acetate, chloroform, hexane and petroleum ether) extracts of the leaves were found to possess strong antibacterial activity against a range of pathogenic bacteria. The ethyl acetate leaf extract of Mentha piperita showed pronounced inhibition than chloroform, petroleum ether and hexane. The leaf extract activity being more on Bacillus subtilis, Staphylococcus aureus and Proteus vulgaris than Escherichia coli, Streptococcus pneumonia and Klebsiella pneumonia. In the present study, we also evaluated the phytochemical analysis for the presence of various secondary metabolites. The analysis revealed the presence of alkaloids, flavonoids, steroids, tannins, and phenols.
文摘Temporal activity patterns in animals emerge from complex interactions between choices made by organisms as responses to biotic interactions and challenges posed by external factors. Temporal activity pattern is an inherently continuous process, even being recorded as a time series. The discreteness of the data set is clearly due to data-acquisition limitations rather than a true underlying discrete nature of the phenomenon itself. Therefore, curves are a natural representation for high-frequency data. Here, we fully model temporal activity data as curves integrating wavelets and functional data analysis, allowing for testing hypotheses based on curves rather than on scalar and vector-valued data. Temporal activity data were obtained experimentally for males and females of a small-bodied marsupial and modelled as wavelets with independent and identically distributed errors and dependent errors. The null hypothesis of no difference in temporal activity pattern between male and female curves was tested with functional analysis of variance (FANOVA). The null hypothesis was rejected by FANOVA and we discussed the differences in temporal activity pattern curves between males and females in terms of ecological and life-history attributes of the reference species. We also performed numerical analysis that shed light on the regularity properties of the wavelet bases used and the thresholding parameters.
