Background:miRNAs are closely related to bone metabolism.Studies have shown that Erxian decoction can improve bone metabolism,possibly achieving this regulatory effect through miRNA targets.Netinfer was used to predic...Background:miRNAs are closely related to bone metabolism.Studies have shown that Erxian decoction can improve bone metabolism,possibly achieving this regulatory effect through miRNA targets.Netinfer was used to predict the miRNA targets of Erxian decoction for the treatment of postmenopausal osteoporosis,and the results were validated by clinical trials.Methods:In this study,we identified possible targets of Erxian decoction in osteoporosis by means of network pharmacological analysis and bioinformatic prediction.Fifteen cases of postmenopausal osteoporosis with kidney Yin and Yang deficiency(In traditional Chinese medicine,kidney Yin nourishes and moistens the tissues of the internal organs of the body,while kidney Yang promotes and warms the tissues of the internal organs of the body.)were treated with Erxian decoction for four weeks,and serum bone metabolism indices(P1NP,osteocalcin,andβ-CTX)and miRNA-335-5p expression were measured before and after treatment.Results:The constructed miRNA postmenopausal osteoporosis related gene network of the effective compound of the Erxian decoction has 296 points and 981 edges.The 39 postmenopausal osteoporosis related genes regulated by miRNA-335-5p were enriched in ossification,while the signaling pathways were enriched in rheumatoid arthritis,the Toll signaling pathway,the HIF-1 signaling pathway,and the MAPK signaling pathway.After taking Erxian decoction,the expression of the serum bone formation index(P1NP,osteocalcin)and miRNA-335-5p gene expression levels increased significantly.The alterations in P1NP and osteocalcin were correlated with the changes in miRNA-335-5p.Conclusion:Circulating miRNA-335-5p may serve as an important target of Erxian decoction in the treatment of postmenopausal women.The effect of Erxian decoction on bone formation is significant,but the underlying mechanism requires further investigation.展开更多

This study investiga...Background: Alpinia officinarum Hance is valued as an edible medicinal plant. The rhizome is widely reported to have anticancer activity whereas little information is available on the aerial part. This study investigates chemical composition, proapoptotic and anti-osteoporosis activities of essential oil from aerial parts of A. officinarum (APEO). Methods: In this study, APEO was extracted by hydrodistillation and analyzed using GC-MS. The inhibitive activity of 0 - 2.5 μL/mL. APEO was investigated using MTT assay, while in vivo effect was evaluated in nude mice. The cell cycle, apoptosis, Δψm and expression of proteins analyses influenced by 0 - 0.313 μL/mL APEO were detected by PI, Annexin V/PI, JC-1, and Western blot, respectively. Alkaline phosphatase activity and mineralized nodules formation of rat osteoblasts with 0 - 0.156 μL/mL APEO were assayed using colorimetric method and alizarin red staining, respectively. Results: Total 45 constituents were identified accounting for 91.1% of APEO (sesquiterpene hydrocarbons for 44.4%). APEO significantly inhibited cancer cells growth in a dose-dependent manner. APEO also inhibited cancer growth in vivo. The percentage of S phase cells is up to 64.846% with 0.313 μL/mL APEO. The proportion of total apoptotic cells significantly increased to 79.6% at 0.313 μL/mL concentration. APEO treated cells accompanied with Bcl-2 and Δψm decrease, and caspase-3 and p53 upregulation. Furthermore, addition of APEO in rat osteoblasts led to a dose-dependent increase in ALP activity and formation of mineralized bone nodules. Conclusions: Our data suggest that APEO could be developed as an agent against human lung cancer and osteoporosis, especially cancer-induced bone loss.展开更多

Under axial rubbing action, the failure mode of the core yarn is the stripping of the sheath from the core. This paper introduces a method to test the anti - stripping property of the core yarn. With a modified Universal Testing Machine, the stripping resistance of friction spun core yarn can be continuously measured. Some factors Influencing the measurements are discussed in detail. The testing results are compared with those from a Y731 Yarn Abrasion Tester and fur - ther confirmed by weaving practice.展开更多
目的采用地塞米松建立斑马鱼骨质疏松模型,筛选先导物G39及其结构类似物的抗骨质疏松活性。方法斑马鱼 AB 系胚胎受精后6~72 h 浸于地塞米松溶液建模,受精后3 d分组给药直至受精后6 d,收集固定斑马鱼幼体;采用茜素红染色法显示成骨...目的采用地塞米松建立斑马鱼骨质疏松模型,筛选先导物G39及其结构类似物的抗骨质疏松活性。方法斑马鱼 AB 系胚胎受精后6~72 h 浸于地塞米松溶液建模,受精后3 d分组给药直至受精后6 d,收集固定斑马鱼幼体;采用茜素红染色法显示成骨发育状况,显微拍照后以 G39为对照,进行斑马鱼头面骨成骨面积和骨矿化密度的半定量分析,比较六种2-乙酰苯并五元杂环类化合物潜在的抗骨质疏松活性。结果 G395μg/ml 组对地塞米松导致的斑马鱼骨质疏松具有显著的改善作用,且实验结果与地塞米松大鼠模型具有一致性。以此筛选 G39衍生物体内活性,发现苯并呋喃衍生物410活性优于 G39。结论地塞米松斑马鱼骨质疏松模型评价周期缩短、成本低,使抗骨质疏松药物体内活性批量筛选成为可能,为更快更准地发现抗骨质疏松候选药物打下坚实基础。通过本研究筛选得到了全新结构类型的抗骨质疏松化合物410,其活性较 G39更强,值得进一步研究。展开更多

An edible fruit from traditional medicinal plant Capparis zeylanica (CZ) was studied for its anti diabetic, insulin secretagogue activities and mechanisms involved in it. In Streptozotocin induced diabetes rats, oral administration of Capparis zeylanica methanolic extract (CZME) (200 mg/kg body weight) for 28 days showed a significant reduction in blood glucose levels by 35.53% and enhanced circulating insulin levels by 81.82% than the diabetic control rats. The insulin secretagogue activity mechanisms of the extract were evaluated by using mouse insulinoma beta cell line (MIN6-β). The extract stimulated insulin release in dependent manner of glucose concentration (3 - 16.7 mM) and extract dose (5 - 500 μg/mL). The insulin releasing effect of the extract was significantly enhanced by 3-isobutyl-1-methyl xanthine, glibenclamide, elevated extracellular calcium and K+ depolarized media. This insulin release was significantly reduced in calcium blocking conditions (by nifedipine and EGTA), in the presence of potassium channel opener (diazoxide). Hence, anti diabetic activity of CZME might be a result of its stimulatory effect on insulin release from pancreatic beta cells via KATP channel dependent and independent ways. These results indicate that CZ fruits have the potential to use in diabetes therapy.展开更多
