Preparation,characterization and antioxidant activity analysis of three Maillard glycosylated bone collagen hydrolysates from chicken,porcine and bovine

Bone collagen hydrolysates(peptides)derived from byproduct of animal product processing have been used to produce commercially valuable products due to their potential antioxidant activity.Maillard glycosylated reaction is considered as a promising method to enhance the antioxidant activity of peptides.Hence,this research aims at investigating the Maillard glycosylation activity and antioxidant activity of bone collagen hydrolysates from different sources.In this study,3 glycosylated bone collagen hydrolysates were prepared and characterized,and cytotoxicity and antioxidant activity were analyzed and evaluated.The free amino groups loss,browning intensity,and fluorescence intensity of G-Cbcp(glycosylated chicken bone collagen hydrolysates(peptides))were the heaviest,followed by G-Pbcp(glycosylated porcine bone collagen hydrolysates(peptides))and G-Bbcp(glycosylated bovine bone collagen hydrolysates(peptides)).The results of amino acid analysis showed that amino acid composition of different bone collagen hydrolysates was significantly different and the amino acid decreased to different degrees after Maillard glycosylated reaction,which may lead to differences in Maillard glycosylated reaction activity.Furthermore,the 3 glycosylated hydrolysates showed no significant cytotoxicity.The results showed that glycosylation process significantly increased the antioxidant activity of bone collagen hydrolysates,and G-Cbcp showed the strongest antioxidant activity,followed by G-Pbcp and G-Bbcp.Therefore,compared with the bone collagen hydrolysates,3 glycosylated hydrolysates showed significant characteristic and structural changes,and higher antioxidant activity. 展开更多

This study investigated the antioxidant activity and alpha-amylase inhibitory potential of the methanolic extract from Vangueria infausta fruits with the aim of exploring its therapeutic and nutritional applications. Bioactive compounds such as total phenolics (TPC) and total alkaloids (TAC) were analyzed. Antioxidant activity was assessed using the DPPH, ABTS, FRAP, Ferrozine, Phosphomolybdate, and Folin-Ciocalteu methods, while alpha-amylase inhibitory activity was determined using the 3,5-Dinitrosalicylic Acid (DNSA) and 2-chloro-p-nitrophenyl-α-D-maltotrioside (CNPG3) methods. The results revealed significant variations in antioxidant activity across the methods used, with IC50 values ranging from 318.42 to 1255.56 µg/mL. Alpha-amylase inhibition was also significant, with IC50 values ranging from 140.85 to 121.06 µg/mL. The TPC was found to be 95.6 mg of gallic acid equivalent per 100 g of dry powder, and the TAC was 0.188 mg of atropine equivalent per 100 g of dry powder. These findings suggest that Vangueria infausta possesses promising antioxidant and alpha-amylase inhibitory potential, indicating its potential application in antioxidant therapies and in the management of carbohydrate digestion. 展开更多

It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. Objective:To evaluate and compare the antioxidant potential and anti-inflammatory activity of ethanolic extract of flowers of Moringa oleifera(M.oleifera)grown in Oman.Methods:Flowers of M.oleiferawere collected in the month of December 2012 and identified by a botanist.Alcoholic extract of the dry pulverized flowers ofM.oleiferawere obtained by cold maceration method.The ethanolic flower extract was subjected to preliminary phytochemical screening as the reported methods.Folin-Ciocalteu reagent was used to estimate total phenolic content.DPPH was used to determinein-vitroantioxidant activity and anti-inflammatory activity of flowers was investigated by protein denaturation method.Results:Phytochemical analysis of extract showed presence of major classes of phytochemicals such as tannins,alkaloids,flavonoids,cardiac glycosidesetc.M.oleiferaflowers were found to contain 19.31 mg/g of gallic acid equivalent of total phenolics in dry extract but exhibited moderate antioxidant activity.The anti-inflammatory activity of plant extract was significant and comparable with the standard drug diclofenac sodium.Conclusions:The results of our study suggest that flowers ofM.oleiferapossess potent anti-inflammatory activity and are also a good source of natural antioxidants.Further study is needed to identify the chemical compounds responsible for their anti-inflammatory activity. 展开更多

The present investigation was aimed to study functional properties,antioxidant activity and in-vitro digestibility characteristics of brown and polished flours obtained from four rice cultivars(SR-4,K-39,Mushq Budij and Zhag)of Kashmir.Brown rice flours had higher total dietary fibre(3.08%-3.68%),oil absorption(116.0%-139.0%),emulsion capacity(4.78%-9.52%),emulsion stability(87.46%-99.93%)and resistant starch content(6.80%-9.00%)than polished flours.However,polished flours presented greater water absorption(102.0%-122.0%),foaming capacity(8.00%-13.63%),apparent amylose(19.16%-22.62%),peak(2260.0-2408.0 cP),trough(1372.0-1589.0 cP)and breakdown(714.0-978.0 cP)viscosities than their brown counterparts.Brown rice flours depicted highest total phenolic content(4.40-6.40 mg GAE/g)and inhibition of lipid peroxidation(19.50%-33.20%).However,equilibrium starch hydrolysis percentage(C∞)and predicted glycemic index of brown rice flours were lower than their polished counterparts.Among rice cultivars,brown Zhag flour had the highest total dietary fibre(3.68%),emulsion capacity(9.52%),emulsion stability(99.93%),resistant starch(9.00%),DPPH radical scavenging activity(85.45%)and inhibition of lipid peroxidation(33.20%),respectively.Emulsion capacity and emulsion stability were positively correlated with protein content of rice flours.However,peak,trough,breakdown and setback viscosities were negatively correlated with protein and fat contents of rice flour.The present investigation will be helpful in identifying nutritive role of rice flours from studied cultivars in human diet. 展开更多

The results showed that the antioxidant activity of polysaccharides in yam bulbils was signifi- cantly enhanced with the increase of concentration; they showed a strong scaveng- ing ability against DPPH. and .OH, and the scavenging ability was dose dependent to some extent; the scavenging rates reached 91.15% and 89.06% respectively when the dose reached 4.0 mg/ml; the polysaccharides in yam bulbils significantly educed the blood glucose in model rice, and the hypoglycemic effect of large-dose polysaccharides was more obvious. Flaxseed lignan macromolecules(FLM)are a class of important secondary metabolites in fl axseed,which have been widely concerned due to their biological and pharmacological properties,especially for their antioxidative activity.For the composition and structure of FLM,our results confirmed that ferulic acid glycoside(FerAG)was directly ester-linked with herbacetin diglucoside(HDG)or pinoresinol diglucoside(PDG),which might determine the beginning of FLM biosynthesis.Additionally,p-coumaric acid glycoside(CouAG)might determine the end of chain extension during FLM synthesis in fl axseed.FLM exhibited higher antioxidative activity in polar systems,as shown by its superior 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical scavenging capacity compared to the 2,2’-azinobis(3-ehtylbenzothiazolin-6-sulfnic acid)(ABTS)cation free radical scavenging capacity in non-polar systems.Moreover,the antioxidative activity of FLM was found to be highly dependent on its composition and structure.In particular,it was positively correlated with the number of phenolic hydroxyl groups(longer FLM chains)and inversely related to the steric hindrance at the ends(lower levels of FerAG and CouAG).These fi ndings verifi ed the potential application of FLM in nonpolar systems,particularly in functional food emulsions。 展开更多

Piptoporellus baudonii, previously known as Laetiporus baudonii, is an African species that was considered to be a sister species to Laetiporus sulphureus, another European species known for its medicinal value. While much is known about the edibility and antimicrobial properties of L. sulphureus, African species like P. baudonii remain understudied. This study investigated the antimicrobial and antioxidant properties of P. baudonii extracts (powder maceration) prepared using ethanol, methanol and water with fractions obtained via differential solubility in hexane, ethyl acetate and n-butanol. Before the antimicrobial analysis, the study material was accurately identified using both morphology and molecular techniques. Antimicrobial activity was tested against fungi, gram-positive, and gram-negative bacteria using a broth serial microdilution method, while antioxidant activity was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Ferric Reducing Antioxidant Power FRAP methods. Phylogenetic analysis confirmed the specimen as P. baudonii, with genetic material from Benin grouping it with other P. baudonii from Tanzania and other unknown regions, forming a well-supported clade (100/100). The ethanol (1.71), methanol (2.41) extracts, along with ethyl acetate (1.36), n-butanol (1.18), and hexane (12.91) fractions showed significant antioxidant activity with EC50 values below 20 µg∙mL−1. The highest antimicrobial inhibition was seen in the n-butanol (58%) and ethyl acetate (54%) fractions, followed by ethanol (49%) and hexane (48%). Methanol exhibited the lowest inhibition (46.10%). These values were compared to the standard (Vitamin C). The examined extracts demonstrated high bactericidal properties, with an MBC/MIC ratio (R) of 1 to 4, particularly effective ethyl acetate against Escherichia coli (R = 2) and ethanol extract with strong activity against Enterococcus faecalis (R = 4). Further chemical and cytotoxicity studies are warranted to fully explore the pharmaceutical potential of P. baudonii. 展开更多

The aim of the present work is to assess the value of Detarium Senegalense by determining the content of total phenols, total flavonoids and total anthocyanins, and by evaluating the free radical scavenging activity of Detarium Senegalense extracts. For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). Changes in Polyphenols and Antioxidant Activities of Yingshan Yunwu Tea during Digestion in Vitro

The antibacterial study was carried out with the help of agar disc dif usion method, MIC, MBC and viable count. The mode of action was determined with help of potassium ion l ux, cellular material release and scanning electron microscopy. The antioxidant activity was determined with the help of 2, 3-diphenyl-2-picrylhydrazyl method, nitric oxide scavenging activity and superoxide anion radical scavenging assay. Results: Total ten compounds were identii ed as trans-phytol(42.73%), cis-3-hexenol(12.95%), 毬-linalool(10.68%), trans-2-hexenal(8.86%), trans-2-hexenol(8.86%), myrcenol(3.86%), 4-vinylphenyl acetate(3.86%),(4Z)-4,6-heptadien-1-ol(3.18%), lemonol(2.73%) and benzeneacetaldehyde(2.27%) by gas chromatography coupled with mass spectrometry. The antibacterial study was demonstrated that leaf essential oil of F. koreana act against foodborne and other pathogenic bacteria. The mode of action revealed that this essential oil acted on the cytoplasmic membrane, resulting in loss of integrity and increased permeability. Objective:To examine the antioxidant activity and total phenolic content of different solvent fractions of Pedulium murex(P.murex)Linn fruits(Family:Pedaliaceae)as well as the correlation between the total antioxidant capacity and total phenolic content.Methods:In the present study,the antioxidant activities of P.murex were evaluated using six in-vitro assays,namely total antioxidant assay,DPPH assay,reducing power,nitric oxide scavenging,hydrogen peroxide scavenging and deoxyribose scavenging assays,and total phenol contents were also investigated.Results:The ethyl acetate(EA)fraction was found to have high levels of phenolic content(298.72±2.09 mg GAE/g).The EA fraction exhibit higher total antioxidant capacity,higher percentage of DPPH radical scavenging activity(135.11±2.95μg/mL),nitric oxide(200.57±4.5lμg/mL),hydrogen peroxide(2I7.91±6.12μg/mL),deoxyribose(250.01±4.68μg/mL)and higher reducing power.Correlation coefficient(r^2=0.914)was found to be significant between total phenolic content and total antioxidant activity.Conclusions:In general,the results indicate that the EA fractions are rich in phenolic antioxidants with potent free radical scavenging activity implying their importance to human health. 展开更多

Objective: To investigate the change in total phenolic compounds, antioxidant activity,and resveratrol content of five different germinated peanut cultivars.Methods: The germinated sprouts of five peanut cultivars(Kalasin1, Kalasin2, Konkaen,Konkaen4, and Tainan9) were extracted with 80% ethanol and collected as crude extract.The antioxidant capacities were determined with 2,2-diphenyl-1-picrylhydrazyl and ferric ion reducing antioxidant power method.The total phenolic compound was measured using the Folin–Ciocalteau assay.The qualification and quantification of resveratrol was performed by high performance liquid chromatography method.Results: Among the five cultivars, a three-day germination of Kalasin1 showed the highest phenolic content [(40.67 ± 2.62) mg gallic acid/g dry weight], expressed the highest 2,2-diphenyl-1-picrylhydrazyl antioxidant value [(80.51 ± 1.47) mmol/L Trolox/g dry weight], and ferric ion reducing antioxidant power antioxidant value [(171.33 ± 8.59)mmol/L ascorbic acid/g dry weight].However, the high performance liquid chromatography result of Kalasin2 significantly increased to the highest resveratrol content of(6.44 ± 1.26) mg/g dry weight on the second day of germination.Conclusions: The variation of phytochemical content in the peanut sprout is due to the effect of the peanut cultivar and the germination period. 展开更多

Mature-green tomato fruit (Solanum lycopersicum cv. Zhenfen 202) were exposed to different UV-C irradiation at 2, 4, 8, and 16 kJ m-2 and then stored under the dark at 14°C and 95% relative humidity (RH) for 35 d. Of these four doses, UV-C irradiation at 4 and 8 kJ m-2 significantly increased total phenolic contents in present tomato fruit by 21.2 and 20.2%, respectively. Furthermore, UV-C irradiation at 4 or 8 kJ m-2 promoted the accumulation of total flavonoids and increased the antioxidant activity. 2 or 16 kJ m-2 UV-C irradiation also enhanced antioxidant activity, but to a lesser extent. Seven phenolic compounds, viz., gallic acid, (+)-catechin, chlorogenic acid, cafferic acid, syringic acid, p-coumaric acid, and quercetin in tomato fruit were identified and quantified by HPLC. Gallic acid was the major phenolic compound in tomato fruit and significantly correlated with antioxidant activity. 4 or 8 kJ m-2 UV-C irradiation significantly increased the contents of gallic acid, chlorogenic acid, syringic acid, p-coumaric acid, and quercetin. The optimum dose of UV-C irradiation in terms of increased phenolic compound content and enhanced Antioxidant activity was determined to be 4 or 8 kJ m-2. 展开更多

Miq.(Rubiaceae) and study the tentative identification of their active constituents.Methods:The extract/fractions were screened for antioxidant activity using various in vitro assays viz.DPPH assay,ABTS assay,superoxide anion radical scavenging assay, reducing power assay and plasmid DNA nicking assay.Total phenolic content of extract/fractions was determined by colorimetric method.An ultra-performance LC-electrospray-quadrupoletime of flight mass spectrometry method was used to analyse the active constituents of extract/ fractions of A.cadamba.Results:The ethyl acetate fraction was found to be most active fraction in all the assays as compared to other extract/fractions.The IC<sub>50</sub> value of ethyl acetate fraction (ETAC fraction) was 21.24μg/mL,1.12μg/mL,9.68μg/mL and S7.81μg/mL in DPPH assay, ABTS assay,reducing power assay and superoxide scavenging assay respectively.All the extract/ fractions also showed the potential to protect the plasmid DNA(pBR322) against the attack of hydroxyl radicals generated by Fenton’s reagent.The bioactive compounds were identified by UPLC-ESI-QTOF-MS,by comparing the mass andλ<sub>max</sub> with literature values.Conclusions:The potential of the extract/fractions to scavenge different free radicals in different systems indicated that they may be useful therapeutic agents for treating radical-related pathologic damage. 展开更多

Objective:To evaluate antioxidant,antimicrobial and cytotoxic activity of different parts(root, flower,leaf and stem)of Leucas aspera(L.aspera)(Labiatae).Methods:Different parts of L.aspera were extracted with 80%(v/v)methanol.The methanol extracts were subjected to antioxidant, antimicrobial and brine shrimp lethality assay.Results:All the extracts showed moderate to potent antioxidant activity,among which the root extract demonstrated the strongest antioxidant activity with the IC_(50)value of 6.552μg/mL.Methanol extract of root possessed antioxidant activity near the range of vitamin E and thus could be a potential rich source of natural antioxidant.In case of antimicrobial screening,crude extracts of root,flower,leaf and stem showed notable antibacterial activity against tested microorganisms.The root extract showed the highest mean zone of inhibition ranging from 9.0-11.0 mm against tested microorganisms,at a concentration of 100 mg/mL.In the brine shrimp lethality bioassay,it was evident that the methanol root extract did not show significant toxicity.The LC_(50)value for 12 h and 24 h observation was 2.890 mg/mL and 1.417 mg/mL,respectively.Conclusions:The present finding suggests that the methanol root extract of L.aspera could be developed as pharmaceutical products. 展开更多

Group 2 served as hepatotoxin(CCL_4 treated) group.Group 3 served as positive control(Silyntarin) group,and Group 4.S and ft served as(73,150 and 300 nig/kg bw p.o.)L.racemosa leaf extract treated groups.Moreover,in vitro antioxidant DPPH,hydroxyl radical scavenging activity(HRSA),NO,ferric reducing antioxidant power(FRAP),lipid hydroperoxide(LPO) and super oxide dismutase(SOD) were also analyzed for the leaf extract.Results:The levels of the serum parameters such as serum glutamic oxaloacetic transaminase(SGOT).serum glutamic pyruvic transaminase(SGPT).alkaline phosphatase(ALP),bilirubin,cholesterol(CHL).sugar and lactate dehydrogenase(LDH) were significantly increased in COL_4 treated rats when compared with the control group(P<0.05).But the L.racemosa leaf extract treated rats showed maximum reduction of SGOT[(210.16±19.63)IU/L].SGPT[(82.37±13.87) IU/L].ALP[(197.63±23.4.3)IU/L],bilurubilt[(2.13 ±0.84) mg/dL].cholesterol[(163.83± 13.63) mg/dL].sugar[(93.00±7.63) mg/dL]and LDH[(1134.00) ±285.00)IU/L]were observed with the high dose(300 mg/kg bw) of leaf extract treated rats. Histopathological scores showed that,no visible changes were observed with high dose(300 mg/ kgbw) of leaf extract treated rats except few mild necrosis.The IC_(50) values were observed as(56.37 ±4.87)μg/mL,(57.68±1.98) μg/mL,(64.15±2.90)μg/mL,(61.94±3.98)μg/mL,(94.53± 1.68) μg/mL and(69.7±2.65)μg/mL for DPPH,HRSA,NO,FRAP,LPO and SOL) radical scavenging activities, respectively.Conclusions:In conclusion,the hepatoprotective effect of the L.racemosa leaf extract might be due to the presence of phenolic groups,terpenoids and alkaloids and in vitro antioxidant properties. 展开更多

The objective of this study was to evaluate the effects of wheat variety, food processing, and milling method on antioxidant properties. Black wheat variety Heibaoshi 1 had the highest total phenolic content(659.8 μg gallic acid equivalents g-1), total flavonoid content(319.3 μg rutin equivalents g-1), and antioxidant activity, whereas light purple wheat variety Shandongzimai 1 had the lowest total flavonoid content(236.2 μg rutin equivalents g-1) and antioxidant activity. Whole wheat flour and partially debranned grain flour had significantly higher total phenolic contents, total flavonoid contents, and antioxidant activity than refined flour(P < 0.05). Compared with flour, total phenolic contents, total flavonoid contents and antioxidant activity decreased in noodles and steamed bread, whereas noodles had slightly higher total phenolic and flavonoid content than steamed bread. Antioxidant activities(by ferric reducing ability of plasma assay) of steamed bread made from whole wheat flour, partially debranned grain flour, and refined flour were 23.5%, 21.1%, and 31.6% lower, respectively, than the corresponding values of flour. These results suggested that black whole wheat flour and partially debranned grain flour are beneficial to human health. 展开更多
