Chinese peony(Paeonia lactiflora Pall.)is both medicinally and aesthetically beneficial.Powdery mildew is a common fungal disease that seriously jeopardizes the value of numerous species,including peonies as a crop.In...Chinese peony(Paeonia lactiflora Pall.)is both medicinally and aesthetically beneficial.Powdery mildew is a common fungal disease that seriously jeopardizes the value of numerous species,including peonies as a crop.In order to provide a basis for the prevention and treatment of peony powdery mildew,we examined the microbial diversity,the malondialdehyde(MDA)concentrations and antioxidant enzyme activities of peony leaves infected with three levels of powdery mildew to determine any modifications to the leaf's antioxidant enzyme systems and microbial community structure following the onset of disease.The results show that the MDA content rose as the degree of infection became worse.Antioxidant enzyme activity rose and then declined.Following the initiation of powdery mildew,fungal community diversity decreased,whereas there was not any appreciable change in bacterial communities according to microbial diversity sequencing.The relative abundance of more than half of fungal species decreased,with the bacterial genera displaying both abundant and diminished communities with less pronounced alterations in their community structure after the disease spread.Significant different taxa that were critical to the organization of each microbiome were found.Correlation analysis showed that the relative abundance of powdery mildew pathogenic fungal genus Erysiphe was correlated with those of 11 fungal genera and one bacterial genus.Among them,Aureobasidium,Neosetophoma and Sclerostagonospora showed significant positive correlations with Erysiphe and MDA.展开更多

Seeds were exposed to 25, 50, and 100 μmol/L CdCl2 concentrations, for a period of 10, 15, 20 and 25 d. The extent of damage to chlorophyll, protein, proline, nitrate and nitrite reductase, antioxidant enzyme activity in leaves and roots were evaluated after 10 d of cadmium stress. The higher concentration of cadmium (100 μmol/L) resulted (leaves and roots) total chlorophyll 91.01%, protein 79.51%, 83.61%, nitrate reductase 79.39%, 80.72% and nitrite reductase 77.07%, 75.88% activity decreased with increase in cadmium concentrations and exposure periods. Cadmium caused significant changes in the activity of antioxidative enzymes. Contrastingly Cd treated plant tissues showed an increase in proline 159.87%, 239.6%, gluthion reductase (GR) 337.72%, 306.14%, superoxide disumutase (SOD) 688.56%, 381.72%, ascorbate peroxidase (APX) 226.47%, 252.14%, peroxidase (POD) 72.19%, 60.29% and catalase (CAT) 228.96%, 214.74% as compared to control. Cadmium stress caused a significant increase in the rate of SOD activity in leaves and roots of plant species. Results show the crop A. hypogaea is highly sensitive even at very low cadmium concentrations.展开更多

Methods: Twenty-six male rats were randomly divided into three groups as follows: group Ⅰ, control, non-diabetic rats (n = 9); group Ⅱ, STZ-induced, untreated diabetic rats (n = 8); group Ⅲ, STZ-induced, melatonin-treated (dose of 10 mg/kg·day) diabetic rats (n = 9). Following 8-week melatonin treatment, all rats were anaesthetized and then were killed to remove testes from the scrotum. Results: As compared to group Ⅰ, in rat testicular tissues of grouap Ⅱ, increased levels of malondialdehyde (MDA) (P 〈 0.01) and superoxide dismutase (SOD) (P 〈 0.01) as well as, decreased levels of catalase (CAT) (P 〈 0.01) and glutathione peroxidase (GSH-Px) (P 〉 0.05) were found. In contrast, as compared to group Ⅱ, in rat testicular tissues of group Ⅲ, levels of MDA decreased (but this decrease was not significant, P 〉 0.05) and SOD (P 〈 0.01) as well as CAT (P 〈 0.05) increased. GSH-Px was not influenced by any of the treatment. Melatonin did not significantly affect the elevated glucose concentration of diabetic group. At the end of the study, there was no significant difference between the melatonin-treated group and the untreated group by means of body and testicular weight. Conclusion: Diabetes mellitus increases oxidative stress and melatonin inhibits lipid peroxidation and might regulate the activities of antioxidant enzymes of diabetic rat testes.展开更多

The V. zizanioides uptake capacity of Zn and Cd also determined. The results showed that plant growth of V. zizanioides was inhibited by Zn and Cd. The shoot dry weight (SDW) and root dry weight (RDW) decrease by 14.2%, 14.1%, 17.0% and 17.3%, 32.5%, 35.7%, respectively, compared to the control without EDTA addition. After adding EDTA, shoot and root dry weight decreased over 10% and 15%, respectively. The toxicity from insoluble Zn and Cd in soil on SDW and RDW of V. zizanioides was in order: Zn+Cd 〉 Cd 〉 Zn. The activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), and contents of MDA and proline increased significantly, while the contents of GSH and water-soluble proteins decreased markedly with increasing Zn and Cd toxicity. With EDTA, shoot and root Zn concentrations increased in the Zn treatment by 7.3% and 37.4%, and Cd concentrations in the combined Zn and Cd treatment increased by 18.6% and 391.9% compared to the treatment without EDTA. However, Zn and Cd concentrations in shoot and roots decreased in the Cd treatment compared to the plants grown in absence of EDTA, with exception of root Cd concentration in the presence of EDTA.展开更多

However, physiological processes involved in alkali stress in oat seedlings are not well understood. In this study, physiological responses and yield of oat to alkali stress were studied using the alkali-tolerant oat genotype Vao-9 and the alkali-sensitive oat genotype Baiyan 5. The results were: (i) low concentrations of alkali stress (25 and 50 mmol L^-1) significantly reduced the yield and grain weight while increased the oat grain number per spike. A negative correlation between yield and malondialdehyde (MDA) content at the jointing and grain filling stages and positive correlations between yield on one hand and superoxide dismutase (SOD), and peroxidase (POD) activities on the other at the jointing stage were observed. There was a positive correlation between MDA and soluble sugar at the grain filling stage; (ii) soluble sugar content was increased at the jointing and grain filling stages and decreased at the heading stage by alkali stress; (iii) alkali stress increased the SOD activity during the heading and grain filling stages, and increased the POD activity at the heading stage. As compared to the control, the increase of MDA contents in alkali-treated oat was observed, during the jointing, heading and grain filling stages; (iv) under alkali stress, the oat genotype Vao-9 showed higher antioxidant enzyme activity and lower soluble sugar contents during the heading stage, and lower MDA contents than those in the oat genotype Baiyan 5 under alkali stress. Compared with the control, excess Mn significantly inhibited net photosynthetic rate (Pn), stomatal conductance, as well as the maximum yield of the photosystem II photochemical reactions (Fv/Fm) and the quantum yield of photosysytem II electron transport (Φ PSII), application of Si reversed the negative effects of excess Mn. In the further investigation, it was obtained that application of Si significantly increased the activities of enzymes related with ascorbate-glutathione cycle including ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) in cucumber chloroplast under excess Mn, this could be responsible for the lower accumulation of H2O2 and lower lipid peroxidation of chloroplast induced by Mn, and resulted in keeping higher photosynthesis.展开更多

The aim of the present study is to investigate the effects on growth, pigments, lipid peroxidation, and some antioxidant enzyme activities of marine microalga Pavlova viridis, in response to elevated concentrations of cobalt (Co) and manganese (Mn), especially with regard to the involvement of antioxidative defences against heavy metal-induced oxidative stress. In response to Co^2+, lipid peroxidation was enhanced compared to the control, as an indication of the oxidative damage caused by metal concentration assayed in the microalgal cells but not Mn^2+. Exposure of Pavlova viridis to the two metals caused changes in enzyme activities in a different manner, depending on the metal assayed: after Co^2+ treatments, total superoxide dismutase (SOD) activity was irregular, although it was not significantly affected by Mn^2+ exposure. Co^2+ and Mn^2+ stimulated the activities of catalase (CAT) and glutathione (GSH), whereas, glutathione peroxidase (GPX) showed a remarkable increase in activity in response to Co^2+ treatments and decreased gradually with Mn^2+ concentration, up to 50 μmol/L, and then rose very rapidly, reaching to about 38.98% at 200 μmol/L Mn^2+. These results suggest that an activation of some antioxidant enzymes was enhanced, to counteract the oxidative stress induced by the two metals at higher concentration.展开更多

Cucumis sativus cv. Jinchun 4 was hydroponically cultivated to the two-leaf stage, at which point seedlings were watered with different concentrations of silicon (0, 0.1 and 1 mmol L^-1) and separately exposed to normal (25/18℃) or chilling (15/8℃) temperatures for six days under low light (100μmol m^-2 s^-9. Data were collected from the second leaves on the percentage of withering and the levels of endogenous silicon, malondialdehyde (MDA), hydrogen peroxide (H202), superoxide radical (O2^.-), superoxide dismutase (SOD, EC 1.15.1.1), glutathione peroxidase (GSH-Px, EC 1.11.1.9), ascorbate peroxidase (APX, EC 1.11.1.11), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), glutathione reductase (GR, EC 1.6.4.2), reduced glutathione (GSH) and ascorbate (AsA). Compared to normal temperatures, chilling resulted in partially withered leaves and increased MDA content. When 0.1 or 1 mmol L^-1 exogenous silicon was combined with chilling, the withering of the cucumber leaves was reduced relative to the original chilling treatment, while the endogenous silicon content was increased, antioxidants such as SOD, GSH-Px, APX, MDHAR, GR, GSH, and AsA were more active, and the levels of H2O2, O2^.-, and MDA were lower. We propose that exogenous silicon leads to greater deposition of endogenous silicon and thereby increases antioxidant activities and reduces lipid peroxidation induced by chilling.展开更多

The wheat cultivars tested were Yangmai 9 with weak-gluten and Yangmai 12 with medium-gluten. Compared with 25℃, the higher the temperature was, the higher was the MDA content in flag leaves, while lower were the SOD, POD, and CAT activities. SOD and CAT activities in Yangmai 12 appeared to be more sensitive to high temperature than that in Yangmai 9. But POD activity in Yangmai 12 was less sensitive to high temperature. MDA content in Yangmai 12 was higher than that in Yangmai 9. The 1000-grain weight declined with increase in temperature.展开更多

For better understanding of chilling stress responses in tobacco (Nicotiana tabacum), growth rate and antioxidant enzymes of seedlings in 2 tobacco cultivars, viz., MSk326 (chilling sensitive variety) and Honghuadajinyuan (HHDJY, chilling tolerant variety) at chilling temperature (5℃) were studied. The results showed that the relative growth rate in chilling period to that in recovery period was significantly higher in roots than that in shoots for both cultivars, suggesting that shoots growth was more easily affected by chilling stress. Chilling stress increased peroxidase (POD) activity and reduced superoxide dismutase (SOD) activity in shoots of HHDJY, and catalase (CAT) activity was little affected. In the roots of HHDJY, chilling stress increased SOD and CAT activities, and had little effect on POD activity. For MSk326, chilling treatment increased SOD activity in shoots and declined CAT activity in roots. MDA concentration in both varieties was increased under the chilling stress, while it was decreased after seedlings were recovered growth for 4 d at normal temperature (25℃). It showed that tobacco seedlings might have the capacity of recovering from chilling injury for a short term, The relationship between the growth rate and antioxidant enzyme activity was analyzed by stepwise regression. It was found that there was a close relationship between relative growth rate of tobacco seedlings and CAT activity under chilling stress condition and regression equations containing CAT could be used in predicting seedling growth rate of tobacco under chilling stress condition.展开更多

Iron deficiency led to serious chlorosis in Chinese cabbage leaves, and resulted in significant decrease in plant growth, photosynthetic pigments, net photosynthetic rate, Fv/Fm, Ф ps Ⅱ and activities of antioxidant enzymes, and increase in lipid peroxidation. While treatment with SNP, a NO donor, it could revert the iron deficiency symptoms, increased photosynthetic rate as well as activities of antioxidant enzymes, and protected membrane from lipid peroxidation, as a result, the growth inhibition of Chinese cabbage by Fe deficiency was alleviated.展开更多

At three-leaf and one-heart stage of Luliangyou 996,15 μg/mL Ag-antibiotic 702 was sprayed,while Validamycin and distilled water were sprayed as positive control and negative control,respectively. The antioxidant enzyme( SOD,POD,PPO,CAT and PAL) activities and malondialdehyde( MDA) content in rice leaves were determined every 24 h within 168 h post spraying. Compared with CK,the MDA content and activities of the five antioxidant enzymes increased in Agantibiotic 702 treatment,Validamycin treatment and M. grisea inoculation treatment. The MDA content and SOD,CAT and POD activities in Ag-antibiotic 702 treatment were significantly higher than those in M. grisea inoculation treatment in early stage,and the APX and LOX activities increased more remarkably and rapidly. The induction effect of Ag-antibiotic 702 could reach or exceed that of Validamycin treatment. Ag-antibiotic 702 enhanced the resistance to rice blast because MDA content and antioxidant enzymes activities were improved in rice leaves. Therefore,the increase of rice blast resistance induced by Ag-antibiotic 702 in rice was positively correlated with the increase of MDA content and antioxidant enzymes activities in rice leaves.展开更多

Maxim. under drought stress. The free MDA content was enhanced during the experimental period, which may be an indicator of oxidative stress. Superoxide dismutase (SOD), peroxidase (POD), and ascorbate peroxidase (APX) activities in C. mongholica showed a significant increase during the experiment, but catalase (CAT) activity was slightly decreased. On the other hand, POD and APX activities showed a significant increase and SOD and CAT activity data had no significant changes in R. soongorica. APX, SOD, and CAT activities were higher in R. soongorica than in C. mongholica, but MDA content was lower, indicating that the lower values of MDA were attributed to higher activities of antioxidant enzyme in R. soongorica. Chlorophyll content decreased significantly in the two shrubs during the experiment, which indicated that there was a photoprotection mechanism through reducing light absorbance by decreasing pigments content. Caretonoids content increased in C. mongholica and decreased in R. soongorica. The ratio of Chla/Chlb decreased significantly but caretonoids/Chl revealed a significant increase in the two shrubs, which could be explained as no decrease of peripheral light-harvesting complexes and a higher tolerance to drought. Total flavonoid content and the activities of phenylalanine ammonialyase (PAL) and chalcone isomerase (CHI) showed different changes between C. mongholica and R. soongorica after treatment. These values decreased in R. soongorica and increased in C. mongholica except for PAL activity. However, anthocyanin content increased in the two shrubs, indicating that there was a different regulation response in the ftavonoid pathway in the two shrubs under drought stress, and anthocyanin should be an important antioxidant both in the shrubs. Our results demonstrated the different responses of antioxidant defense and drought tolerance ability between the two shrubs.展开更多

In order to investigate the biochemical process of cold acclimation in Ammopiptanthus mongolicus, seedlings were acclimated at 2℃ under 16-h photoperiod (150 μmol·m^-2·s^-1 photosynthetically active radiation) for 14 d. Freezing tolerance in seedlings increased after 14 d of cold-hardening. Contents of protein, proline and solute carbohydrate in cotyledon increased after cold acclimation. Patterns of isozymes of superoxide dismutase (SOD), peroxidase, catalase and polyphenol oxidase (PPO) were investigated. The activities of SOD, peroxidase and PPO in cold acclimated plants were increased during cold-hardening. We deduced that compatible solutes and antioxidant enzymes play important roles in development of freezing tolerance during cold acclimation in this evergreen woody plant.展开更多

Soil contamination by toxic trace metal elements,like barium(Ba),may stimulate various undesirable changes in the metabolic activity of plants.The plant responses are fast and with,direct or indirect,generation of rea...Soil contamination by toxic trace metal elements,like barium(Ba),may stimulate various undesirable changes in the metabolic activity of plants.The plant responses are fast and with,direct or indirect,generation of reactive oxygen species(ROS).To cope with the stress imposed by the ROS production,plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS,and their by-products,into stable nontoxic molecules.To assess the Ba stress response of two Brassicaceae species(Brassica juncea,a glycophyte,and Cakile maritime,a halophyte),plants were exposure to different Ba concentrations(0,100,200,300 and 500µM).The plants response was evaluated through their morphology and development,the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary metabolites.Results indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba(even at 500µM).The biomass production of C.maritima was slightly affected whereas an increase in biomass B.juncea was noticed.The stress imposed by Ba activated the antioxidant defense system in the two species,noticed by the changes in the leaves activity of catalase(CAT),ascorbate peroxidase(APX)and guaicol peroxidase(GPX),and of the secondary metabolites,through the production of total phenols and flavonoids.The enzymatic response was not similar within the two plant species:CAT and APX seem to have a more important role against the oxidative stress in C.maritima while in B.juncea is GPX.Overall,total phenols and flavonoids production was more significant in the plants aerial part than in the roots,of the both species.Although the two Brassicaceae species response was different,in both plants catalytic and non-catalytic transformation of ROS occurs,and both were able to overcome the Ba toxicity and prevent the cell damage.展开更多

Succulent level, inorganic ions (Na^+, K^+), organics such as betaine, proline, malondialdehyde, and antioxidant enzyme activities including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), betaine aldehyde dehydrogenase (BADH) were measured to reveal its salt tolerance mechanism. When the composite salt concentration reaches 5.4%, SOD activity level, and MDA content is five times the control group; when it reaches 2.7%, the succulent level of seedlings, and the content ofK+ in roots is nearly two times the NaCl treatment; the dry weight is more than three times the control group; with the NaCl treatment, MDA is three times the contrast; when the salt concentration is 2.7%, POD reaches the maximum. Results indicate that Si^4+, K^+, and Ca^20 from composite salt in the roots of H. strobilaceum improved the water-holding capacity. The activities of antioxidant enzyme were raised by the accumulation ofproline and betaine, which increased the salt tolerance. The absorption of K^+ promoted the high ratio of K^+/Na^+ and alleviated the damage of cell membranes of H. strobilaceum, which is associated with osmotic contents such as betaine and proline.展开更多
Paeonia ostii is an economically important oil crop,which has been widely cultivated in the middle and lower reaches of the Yangtze River in China in recent years.Although P.ostii is highly adaptable to the environmen...Paeonia ostii is an economically important oil crop,which has been widely cultivated in the middle and lower reaches of the Yangtze River in China in recent years.Although P.ostii is highly adaptable to the environment,the prolonged high summer temperature in this region severely inhibits its growth,which adversely affects seed yield and quality.In this study,P.ostii plants were subjected to 20℃/15℃(day/night)and 40℃/35℃(day/night)temperatures for 15 days.The changes in physiological and biochemical indicators of P.ostii under high-temperature stress were initially investigated.The results showed that with the deepening of leaf etiolation,chlorophyll a and chlorophyll b concentration,carotenoid concentration,Soil Plant Analysis Development(SPAD)values and leaf relative water content decreased significantly,while both relative electrical conductivity(REC)and free proline concentration showed an upward trend.Meanwhile,the continuous accumulation of reactive oxygen species(ROS)in P.ostii plants,led to an increased activity of antioxidant enzymes including superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)and ascorbate peroxidase(APX).Moreover,with the extension of the high-temperature treatment,the anatomical structures of P.ostii were destroyed,resulting in a decreased photochemical efficiency of the photosystem II(PSII)reaction center and photosynthesis was inhibited.Taken together,these results provide reference values for understanding the physiological response of P.ostii to hightemperature stress and establish a foundation for further research on the relevant underlying molecular mechanisms.展开更多
In order to provide reference for using Sigartus guttatus to control overgrowth of Ertteromorpha prolifra, the growth, serum biochemical and antioxidant enzyme indices of juvenile S. guttatus respectively fed with E. ...In order to provide reference for using Sigartus guttatus to control overgrowth of Ertteromorpha prolifra, the growth, serum biochemical and antioxidant enzyme indices of juvenile S. guttatus respectively fed with E. prolifra and artificial feed were studied. One hundred and eighty individuals were cultivated for 90 days in six 2.5 m × 1.5 m × 1.5 m cages (30 ind. per cage) which mesh size were 0.5 mm. The experimental animals were divided into two dietary groups ( three cages for each group) that were fed with E. prolifra and artificial feed respectively. During the culture period, temperatures ranged from 23.0 to 26.5℃, pH was between 7.8 and 8.2, dissolved oxygen was more than 5.0 mg/L. The weight gain rate, specific growth rate, relative growth rate, hepato-somatic index of juvenile S. guttatus fed with E. prolifra were significantly lowe than that fed with artificial feed (P 〈 0.05). The total protein (TP), urea nitrogen (UN) and alkaline phosphatase (AKP) of juvenile S. guttatus fed with E. Prolifra were significantly higher than that fed with artificial feed (P 〈0.05), while glutamic oxalacetie transaminase (AST/GOT) and glutamic-pyruvic transaminase (ALT/GPT) of juvenile S. guttatus fed with E. prolifra were significantly lower than that fed with artificial feed (P 〈 0.05 ). SOD, GPX and anti-superoxide anion radical activity in liver, kidney, gill and muscle of juvenile S. guttatus fed with E. prolifra were higher than that fed with artificial feed, and these antioxidant enzyme activities in kidney of juvenile S. gtatatus fed with E. prolifra was significantly higher than that fed with artificial feed (P 〈 0.05), while CAT and hydroxyl radical-inhibiting activity in liver of juvenile S. guttatus fed with arti- ficial feed were significantly higher than that fed with E. prolifra (P 〈 0.05 ). Juvenile S. guttatus fed with E. prolifra showed poor growth performance but better antioxidant defense system. S. guttatus is easily raised and has stronger ability of digestion and absorption of E. Prolifra. The technique that uses S. gtatatus to control E. prolifra deserves deeply study.展开更多

文摘Chinese peony(Paeonia lactiflora Pall.)is both medicinally and aesthetically beneficial.Powdery mildew is a common fungal disease that seriously jeopardizes the value of numerous species,including peonies as a crop.In order to provide a basis for the prevention and treatment of peony powdery mildew,we examined the microbial diversity,the malondialdehyde(MDA)concentrations and antioxidant enzyme activities of peony leaves infected with three levels of powdery mildew to determine any modifications to the leaf's antioxidant enzyme systems and microbial community structure following the onset of disease.The results show that the MDA content rose as the degree of infection became worse.Antioxidant enzyme activity rose and then declined.Following the initiation of powdery mildew,fungal community diversity decreased,whereas there was not any appreciable change in bacterial communities according to microbial diversity sequencing.The relative abundance of more than half of fungal species decreased,with the bacterial genera displaying both abundant and diminished communities with less pronounced alterations in their community structure after the disease spread.Significant different taxa that were critical to the organization of each microbiome were found.Correlation analysis showed that the relative abundance of powdery mildew pathogenic fungal genus Erysiphe was correlated with those of 11 fungal genera and one bacterial genus.Among them,Aureobasidium,Neosetophoma and Sclerostagonospora showed significant positive correlations with Erysiphe and MDA.

文摘Tree peony(Paeonia suffruticosa Andr.)is a traditional Chinese flower,which prefers cool weather.However,high temperature in summer in the middle and lower reaches of the Yangtze River restricts its growth and development.In this study,osmotic regulation,antioxidant enzyme activities,and photosynthetic characteristics of tree peony in response to high-temperature stress were investigated.The results showed that high-temperature stress had destroyed the cell membrane,manifested as the increased relative electrical conductivity and malondialdehyde content.Moreover,high-temperature stress led to excessive accumulation of reactive oxygen species,thereby,activating antioxidant enzyme activities.Also,photosynthetic parameters and chlorophyll fluorescence parameters directly reflected the damage to the photosystem II reflection center under high-temperature stress.In addition,high-temperature stress led to stomatal closure and chloroplast damage.This study revealed the physiological responses of tree peony to high-temperature stress,laying a foundation for the promotion of tree peony in high-temperature areas and the improvement of high-temperature resistance.
文摘Phytotoxicity of cadmium on growing Arachis hypogaea L. seedlings was studied. Seeds were exposed to 25, 50, and 100 μmol/L CdCl2 concentrations, for a period of 10, 15, 20 and 25 d. The extent of damage to chlorophyll, protein, proline, nitrate and nitrite reductase, antioxidant enzyme activity in leaves and roots were evaluated after 10 d of cadmium stress. The higher concentration of cadmium (100 μmol/L) resulted (leaves and roots) total chlorophyll 91.01%, protein 79.51%, 83.61%, nitrate reductase 79.39%, 80.72% and nitrite reductase 77.07%, 75.88% activity decreased with increase in cadmium concentrations and exposure periods. Cadmium caused significant changes in the activity of antioxidative enzymes. Contrastingly Cd treated plant tissues showed an increase in proline 159.87%, 239.6%, gluthion reductase (GR) 337.72%, 306.14%, superoxide disumutase (SOD) 688.56%, 381.72%, ascorbate peroxidase (APX) 226.47%, 252.14%, peroxidase (POD) 72.19%, 60.29% and catalase (CAT) 228.96%, 214.74% as compared to control. Cadmium stress caused a significant increase in the rate of SOD activity in leaves and roots of plant species. Results show the crop A. hypogaea is highly sensitive even at very low cadmium concentrations.
文摘Aim: To examine the effects of melatonin treatment on lipid peroxidation (LPO) and the activities of antioxidant enzymes in the testicular tissue of streptozotocin (STZ)-induced diabetic rats. Methods: Twenty-six male rats were randomly divided into three groups as follows: group Ⅰ, control, non-diabetic rats (n = 9); group Ⅱ, STZ-induced, untreated diabetic rats (n = 8); group Ⅲ, STZ-induced, melatonin-treated (dose of 10 mg/kg·day) diabetic rats (n = 9). Following 8-week melatonin treatment, all rats were anaesthetized and then were killed to remove testes from the scrotum. Results: As compared to group Ⅰ, in rat testicular tissues of grouap Ⅱ, increased levels of malondialdehyde (MDA) (P 〈 0.01) and superoxide dismutase (SOD) (P 〈 0.01) as well as, decreased levels of catalase (CAT) (P 〈 0.01) and glutathione peroxidase (GSH-Px) (P 〉 0.05) were found. In contrast, as compared to group Ⅱ, in rat testicular tissues of group Ⅲ, levels of MDA decreased (but this decrease was not significant, P 〉 0.05) and SOD (P 〈 0.01) as well as CAT (P 〈 0.05) increased. GSH-Px was not influenced by any of the treatment. Melatonin did not significantly affect the elevated glucose concentration of diabetic group. At the end of the study, there was no significant difference between the melatonin-treated group and the untreated group by means of body and testicular weight. Conclusion: Diabetes mellitus increases oxidative stress and melatonin inhibits lipid peroxidation and might regulate the activities of antioxidant enzymes of diabetic rat testes.

文摘A root-bag experiment was conducted to study the effects of insoluble Zn, Cd, and ethylenediaminetetraacetic acid (EDTA) on the plant growth, activities of antioxidant enzymes, proline, glutathione (GSH), water-soluble proteins and malondialdehyde (MDA) of Vetiveria zizanioides. The V. zizanioides uptake capacity of Zn and Cd also determined. The results showed that plant growth of V. zizanioides was inhibited by Zn and Cd. The shoot dry weight (SDW) and root dry weight (RDW) decrease by 14.2%, 14.1%, 17.0% and 17.3%, 32.5%, 35.7%, respectively, compared to the control without EDTA addition. After adding EDTA, shoot and root dry weight decreased over 10% and 15%, respectively. The toxicity from insoluble Zn and Cd in soil on SDW and RDW of V. zizanioides was in order: Zn+Cd 〉 Cd 〉 Zn. The activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), and contents of MDA and proline increased significantly, while the contents of GSH and water-soluble proteins decreased markedly with increasing Zn and Cd toxicity. With EDTA, shoot and root Zn concentrations increased in the Zn treatment by 7.3% and 37.4%, and Cd concentrations in the combined Zn and Cd treatment increased by 18.6% and 391.9% compared to the treatment without EDTA. However, Zn and Cd concentrations in shoot and roots decreased in the Cd treatment compared to the plants grown in absence of EDTA, with exception of root Cd concentration in the presence of EDTA.

文摘Alkali stress can cause severe crop damage and reduce production. However, physiological processes involved in alkali stress in oat seedlings are not well understood. In this study, physiological responses and yield of oat to alkali stress were studied using the alkali-tolerant oat genotype Vao-9 and the alkali-sensitive oat genotype Baiyan 5. The results were: (i) low concentrations of alkali stress (25 and 50 mmol L^-1) significantly reduced the yield and grain weight while increased the oat grain number per spike. A negative correlation between yield and malondialdehyde (MDA) content at the jointing and grain filling stages and positive correlations between yield on one hand and superoxide dismutase (SOD), and peroxidase (POD) activities on the other at the jointing stage were observed. There was a positive correlation between MDA and soluble sugar at the grain filling stage; (ii) soluble sugar content was increased at the jointing and grain filling stages and decreased at the heading stage by alkali stress; (iii) alkali stress increased the SOD activity during the heading and grain filling stages, and increased the POD activity at the heading stage. As compared to the control, the increase of MDA contents in alkali-treated oat was observed, during the jointing, heading and grain filling stages; (iv) under alkali stress, the oat genotype Vao-9 showed higher antioxidant enzyme activity and lower soluble sugar contents during the heading stage, and lower MDA contents than those in the oat genotype Baiyan 5 under alkali stress. The result suggested that the high ROS scavenging capacity and soluble sugar levels might play roles in oat response to alkali stress,

文摘Effects of silicon on photosynthetic parameters and antioxidant enzymes of chloroplast in cucumber seedlings under excess Mn were studied. Compared with the control, excess Mn significantly inhibited net photosynthetic rate (Pn), stomatal conductance, as well as the maximum yield of the photosystem II photochemical reactions (Fv/Fm) and the quantum yield of photosysytem II electron transport (Φ PSII), application of Si reversed the negative effects of excess Mn. In the further investigation, it was obtained that application of Si significantly increased the activities of enzymes related with ascorbate-glutathione cycle including ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) in cucumber chloroplast under excess Mn, this could be responsible for the lower accumulation of H2O2 and lower lipid peroxidation of chloroplast induced by Mn, and resulted in keeping higher photosynthesis.

文摘Pollution of marine environment has become an issue of major concern in recent years. Serious environmental pollution by heavy metals results from their increasing utilization in industrial processes and because most heavy metals are transported into the marine environment and accumulated without decomposition. The aim of the present study is to investigate the effects on growth, pigments, lipid peroxidation, and some antioxidant enzyme activities of marine microalga Pavlova viridis, in response to elevated concentrations of cobalt (Co) and manganese (Mn), especially with regard to the involvement of antioxidative defences against heavy metal-induced oxidative stress. In response to Co^2+, lipid peroxidation was enhanced compared to the control, as an indication of the oxidative damage caused by metal concentration assayed in the microalgal cells but not Mn^2+. Exposure of Pavlova viridis to the two metals caused changes in enzyme activities in a different manner, depending on the metal assayed: after Co^2+ treatments, total superoxide dismutase (SOD) activity was irregular, although it was not significantly affected by Mn^2+ exposure. Co^2+ and Mn^2+ stimulated the activities of catalase (CAT) and glutathione (GSH), whereas, glutathione peroxidase (GPX) showed a remarkable increase in activity in response to Co^2+ treatments and decreased gradually with Mn^2+ concentration, up to 50 μmol/L, and then rose very rapidly, reaching to about 38.98% at 200 μmol/L Mn^2+. These results suggest that an activation of some antioxidant enzymes was enhanced, to counteract the oxidative stress induced by the two metals at higher concentration.

文摘In order to increase vegetable productivity by improving environmental conditions, this article investigates the effects of exogenous silicon on the activities of major antioxidant enzymes and on lipid peroxidation under chilling stress, and it examines whether silicon-induced chilling tolerance is mediated by an increase in antioxidant activity. Cucumis sativus cv. Jinchun 4 was hydroponically cultivated to the two-leaf stage, at which point seedlings were watered with different concentrations of silicon (0, 0.1 and 1 mmol L^-1) and separately exposed to normal (25/18℃) or chilling (15/8℃) temperatures for six days under low light (100μmol m^-2 s^-9. Data were collected from the second leaves on the percentage of withering and the levels of endogenous silicon, malondialdehyde (MDA), hydrogen peroxide (H202), superoxide radical (O2^.-), superoxide dismutase (SOD, EC 1.15.1.1), glutathione peroxidase (GSH-Px, EC 1.11.1.9), ascorbate peroxidase (APX, EC 1.11.1.11), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), glutathione reductase (GR, EC 1.6.4.2), reduced glutathione (GSH) and ascorbate (AsA). Compared to normal temperatures, chilling resulted in partially withered leaves and increased MDA content. When 0.1 or 1 mmol L^-1 exogenous silicon was combined with chilling, the withering of the cucumber leaves was reduced relative to the original chilling treatment, while the endogenous silicon content was increased, antioxidants such as SOD, GSH-Px, APX, MDHAR, GR, GSH, and AsA were more active, and the levels of H2O2, O2^.-, and MDA were lower. We propose that exogenous silicon leads to greater deposition of endogenous silicon and thereby increases antioxidant activities and reduces lipid peroxidation induced by chilling.

文摘On the basis of the phytotron, the effects of high temperature (daily average temperature 25, 30, 35 and 40℃, respectively) on antioxidant enzymes and lipid peroxidation in flag leaves of wheat at 50% relative air moisture during grain fastest filling stage [19-21 days after anthesis (DAA)] were studied. The wheat cultivars tested were Yangmai 9 with weak-gluten and Yangmai 12 with medium-gluten. Compared with 25℃, the higher the temperature was, the higher was the MDA content in flag leaves, while lower were the SOD, POD, and CAT activities. SOD and CAT activities in Yangmai 12 appeared to be more sensitive to high temperature than that in Yangmai 9. But POD activity in Yangmai 12 was less sensitive to high temperature. MDA content in Yangmai 12 was higher than that in Yangmai 9. The 1000-grain weight declined with increase in temperature.

文摘Chilling is one of the major abiotic stresses limiting yield and quality of many important crops. For better understanding of chilling stress responses in tobacco (Nicotiana tabacum), growth rate and antioxidant enzymes of seedlings in 2 tobacco cultivars, viz., MSk326 (chilling sensitive variety) and Honghuadajinyuan (HHDJY, chilling tolerant variety) at chilling temperature (5℃) were studied. The results showed that the relative growth rate in chilling period to that in recovery period was significantly higher in roots than that in shoots for both cultivars, suggesting that shoots growth was more easily affected by chilling stress. Chilling stress increased peroxidase (POD) activity and reduced superoxide dismutase (SOD) activity in shoots of HHDJY, and catalase (CAT) activity was little affected. In the roots of HHDJY, chilling stress increased SOD and CAT activities, and had little effect on POD activity. For MSk326, chilling treatment increased SOD activity in shoots and declined CAT activity in roots. MDA concentration in both varieties was increased under the chilling stress, while it was decreased after seedlings were recovered growth for 4 d at normal temperature (25℃). It showed that tobacco seedlings might have the capacity of recovering from chilling injury for a short term, The relationship between the growth rate and antioxidant enzyme activity was analyzed by stepwise regression. It was found that there was a close relationship between relative growth rate of tobacco seedlings and CAT activity under chilling stress condition and regression equations containing CAT could be used in predicting seedling growth rate of tobacco under chilling stress condition.
文摘The effects of exogenous nitric oxide (NO) on plant growth, chlorophyll contents, photosynthetic and chlorophyll fluorescence parameters as well as lipid peroxidation and activities of antioxidant enzymes were investigated in Chinese cabbage plants exposed to iron (Fe) deficiency. Iron deficiency led to serious chlorosis in Chinese cabbage leaves, and resulted in significant decrease in plant growth, photosynthetic pigments, net photosynthetic rate, Fv/Fm, Ф ps Ⅱ and activities of antioxidant enzymes, and increase in lipid peroxidation. While treatment with SNP, a NO donor, it could revert the iron deficiency symptoms, increased photosynthetic rate as well as activities of antioxidant enzymes, and protected membrane from lipid peroxidation, as a result, the growth inhibition of Chinese cabbage by Fe deficiency was alleviated.

文摘The paper was to explore the effect of rice blast resistance induced by Ag-antibiotic 702 on antioxidant enzyme activities and malondialdehyde( MDA)content. At three-leaf and one-heart stage of Luliangyou 996,15 μg/mL Ag-antibiotic 702 was sprayed,while Validamycin and distilled water were sprayed as positive control and negative control,respectively. The antioxidant enzyme( SOD,POD,PPO,CAT and PAL) activities and malondialdehyde( MDA) content in rice leaves were determined every 24 h within 168 h post spraying. Compared with CK,the MDA content and activities of the five antioxidant enzymes increased in Agantibiotic 702 treatment,Validamycin treatment and M. grisea inoculation treatment. The MDA content and SOD,CAT and POD activities in Ag-antibiotic 702 treatment were significantly higher than those in M. grisea inoculation treatment in early stage,and the APX and LOX activities increased more remarkably and rapidly. The induction effect of Ag-antibiotic 702 could reach or exceed that of Validamycin treatment. Ag-antibiotic 702 enhanced the resistance to rice blast because MDA content and antioxidant enzymes activities were improved in rice leaves. Therefore,the increase of rice blast resistance induced by Ag-antibiotic 702 in rice was positively correlated with the increase of MDA content and antioxidant enzymes activities in rice leaves.

文摘Antioxidant enzyme activity, photosynthetic pigment content, and free malondialdehyde (MDA), as well as flavonoid content and the key enzyme activity in the flavonoid pathway were determined in two desert shrubs, Caryopteris mongolica Bunge and Reaumuria soongorica (Pall.) Maxim. under drought stress. The free MDA content was enhanced during the experimental period, which may be an indicator of oxidative stress. Superoxide dismutase (SOD), peroxidase (POD), and ascorbate peroxidase (APX) activities in C. mongholica showed a significant increase during the experiment, but catalase (CAT) activity was slightly decreased. On the other hand, POD and APX activities showed a significant increase and SOD and CAT activity data had no significant changes in R. soongorica. APX, SOD, and CAT activities were higher in R. soongorica than in C. mongholica, but MDA content was lower, indicating that the lower values of MDA were attributed to higher activities of antioxidant enzyme in R. soongorica. Chlorophyll content decreased significantly in the two shrubs during the experiment, which indicated that there was a photoprotection mechanism through reducing light absorbance by decreasing pigments content. Caretonoids content increased in C. mongholica and decreased in R. soongorica. The ratio of Chla/Chlb decreased significantly but caretonoids/Chl revealed a significant increase in the two shrubs, which could be explained as no decrease of peripheral light-harvesting complexes and a higher tolerance to drought. Total flavonoid content and the activities of phenylalanine ammonialyase (PAL) and chalcone isomerase (CHI) showed different changes between C. mongholica and R. soongorica after treatment. These values decreased in R. soongorica and increased in C. mongholica except for PAL activity. However, anthocyanin content increased in the two shrubs, indicating that there was a different regulation response in the ftavonoid pathway in the two shrubs under drought stress, and anthocyanin should be an important antioxidant both in the shrubs. Our results demonstrated the different responses of antioxidant defense and drought tolerance ability between the two shrubs.

In order to investigate the biochemical process of cold acclimation in Ammopiptanthus mongolicus, seedlings were acclimated at 2℃ under 16-h photoperiod (150 μmol·m^-2·s^-1 photosynthetically active radiation) for 14 d. Freezing tolerance in seedlings increased after 14 d of cold-hardening. Contents of protein, proline and solute carbohydrate in cotyledon increased after cold acclimation. Patterns of isozymes of superoxide dismutase (SOD), peroxidase, catalase and polyphenol oxidase (PPO) were investigated. The activities of SOD, peroxidase and PPO in cold acclimated plants were increased during cold-hardening. We deduced that compatible solutes and antioxidant enzymes play important roles in development of freezing tolerance during cold acclimation in this evergreen woody plant.

文摘The effects of excess Mn on oxidative stress and antioxidant enzymes in cucumber roots under different pH (pH 4.5 and 6.5) were studied. The results indicated that lipid peroxidation was increasingly serious with the increased concentration of Mn, especially under low pH. As a result, the growth of cucumber roots and shoots was significantly inhibited. Excess Mn led to an elevation in activities of Mn-SOD and Fe-SOD, which played an important role in removing O2^-. CAT was sensitive to excess Mn, and its activity significantly decreased under excess Mn and low pH, while activities of GPX, APX, DHAR and GR increased under low pH and to some extent under excess Mn, which indicated their important roles in scavenging reactive oxygen in tolerance to low pH and excess Mn of cucumber roots.

文摘Soil contamination by toxic trace metal elements,like barium(Ba),may stimulate various undesirable changes in the metabolic activity of plants.The plant responses are fast and with,direct or indirect,generation of reactive oxygen species(ROS).To cope with the stress imposed by the ROS production,plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS,and their by-products,into stable nontoxic molecules.To assess the Ba stress response of two Brassicaceae species(Brassica juncea,a glycophyte,and Cakile maritime,a halophyte),plants were exposure to different Ba concentrations(0,100,200,300 and 500µM).The plants response was evaluated through their morphology and development,the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary metabolites.Results indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba(even at 500µM).The biomass production of C.maritima was slightly affected whereas an increase in biomass B.juncea was noticed.The stress imposed by Ba activated the antioxidant defense system in the two species,noticed by the changes in the leaves activity of catalase(CAT),ascorbate peroxidase(APX)and guaicol peroxidase(GPX),and of the secondary metabolites,through the production of total phenols and flavonoids.The enzymatic response was not similar within the two plant species:CAT and APX seem to have a more important role against the oxidative stress in C.maritima while in B.juncea is GPX.Overall,total phenols and flavonoids production was more significant in the plants aerial part than in the roots,of the both species.Although the two Brassicaceae species response was different,in both plants catalytic and non-catalytic transformation of ROS occurs,and both were able to overcome the Ba toxicity and prevent the cell damage.

文摘The seedlings of Halocnermum strobilaceum were cultivated in 0.5% hoagland nutrient solution containing 0.0%, 0.9%, 2.7% and 5.4% of NaC1 as well as composite salt (Na^+, Ca^2+, K^+, Si^4+) for 20 days; all the contents are in weight ratio. Succulent level, inorganic ions (Na^+, K^+), organics such as betaine, proline, malondialdehyde, and antioxidant enzyme activities including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), betaine aldehyde dehydrogenase (BADH) were measured to reveal its salt tolerance mechanism. When the composite salt concentration reaches 5.4%, SOD activity level, and MDA content is five times the control group; when it reaches 2.7%, the succulent level of seedlings, and the content ofK+ in roots is nearly two times the NaCl treatment; the dry weight is more than three times the control group; with the NaCl treatment, MDA is three times the contrast; when the salt concentration is 2.7%, POD reaches the maximum. Results indicate that Si^4+, K^+, and Ca^20 from composite salt in the roots of H. strobilaceum improved the water-holding capacity. The activities of antioxidant enzyme were raised by the accumulation ofproline and betaine, which increased the salt tolerance. The absorption of K^+ promoted the high ratio of K^+/Na^+ and alleviated the damage of cell membranes of H. strobilaceum, which is associated with osmotic contents such as betaine and proline.

文摘Paeonia ostii is an economically important oil crop,which has been widely cultivated in the middle and lower reaches of the Yangtze River in China in recent years.Although P.ostii is highly adaptable to the environment,the prolonged high summer temperature in this region severely inhibits its growth,which adversely affects seed yield and quality.In this study,P.ostii plants were subjected to 20℃/15℃(day/night)and 40℃/35℃(day/night)temperatures for 15 days.The changes in physiological and biochemical indicators of P.ostii under high-temperature stress were initially investigated.The results showed that with the deepening of leaf etiolation,chlorophyll a and chlorophyll b concentration,carotenoid concentration,Soil Plant Analysis Development(SPAD)values and leaf relative water content decreased significantly,while both relative electrical conductivity(REC)and free proline concentration showed an upward trend.Meanwhile,the continuous accumulation of reactive oxygen species(ROS)in P.ostii plants,led to an increased activity of antioxidant enzymes including superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)and ascorbate peroxidase(APX).Moreover,with the extension of the high-temperature treatment,the anatomical structures of P.ostii were destroyed,resulting in a decreased photochemical efficiency of the photosystem II(PSII)reaction center and photosynthesis was inhibited.Taken together,these results provide reference values for understanding the physiological response of P.ostii to hightemperature stress and establish a foundation for further research on the relevant underlying molecular mechanisms.
文摘In order to provide reference for using Sigartus guttatus to control overgrowth of Ertteromorpha prolifra, the growth, serum biochemical and antioxidant enzyme indices of juvenile S. guttatus respectively fed with E. prolifra and artificial feed were studied. One hundred and eighty individuals were cultivated for 90 days in six 2.5 m × 1.5 m × 1.5 m cages (30 ind. per cage) which mesh size were 0.5 mm. The experimental animals were divided into two dietary groups ( three cages for each group) that were fed with E. prolifra and artificial feed respectively. During the culture period, temperatures ranged from 23.0 to 26.5℃, pH was between 7.8 and 8.2, dissolved oxygen was more than 5.0 mg/L. The weight gain rate, specific growth rate, relative growth rate, hepato-somatic index of juvenile S. guttatus fed with E. prolifra were significantly lowe than that fed with artificial feed (P 〈 0.05). The total protein (TP), urea nitrogen (UN) and alkaline phosphatase (AKP) of juvenile S. guttatus fed with E. Prolifra were significantly higher than that fed with artificial feed (P 〈0.05), while glutamic oxalacetie transaminase (AST/GOT) and glutamic-pyruvic transaminase (ALT/GPT) of juvenile S. guttatus fed with E. prolifra were significantly lower than that fed with artificial feed (P 〈 0.05 ). SOD, GPX and anti-superoxide anion radical activity in liver, kidney, gill and muscle of juvenile S. guttatus fed with E. prolifra were higher than that fed with artificial feed, and these antioxidant enzyme activities in kidney of juvenile S. gtatatus fed with E. prolifra was significantly higher than that fed with artificial feed (P 〈 0.05), while CAT and hydroxyl radical-inhibiting activity in liver of juvenile S. guttatus fed with arti- ficial feed were significantly higher than that fed with E. prolifra (P 〈 0.05 ). Juvenile S. guttatus fed with E. prolifra showed poor growth performance but better antioxidant defense system. S. guttatus is easily raised and has stronger ability of digestion and absorption of E. Prolifra. The technique that uses S. gtatatus to control E. prolifra deserves deeply study.
