Selenium(Se)-enriched Brassica napus L.is a valuable organic Se supplement. In this study, the fermentation broth enriched with organic Se(FFS)was prepared using Lactobacillus plantarum to ferment the substrate of Se-enriched Brassica napus L.Significant increases were observed after fermentation in total sugars, reducing sugars, soluble proteins, total phenolic content(TPC),and total flavonoid content(TFC).The organic Se was retained at a concentration of 54.75 mg/g in the freeze-dried sample.Principal component analysis and cluster analysis showed good separation between the FFS and unfermented(FS)groups.Fragrant 2-ethyloxetane had the highest content among all volatiles, while sinapine had the highest content among all phenolic compounds. The fermentation process showed remarkable improvement in the abundance and concentration of volatile compounds and phenolic contents, making FFS exhibit strong antioxidant activity and inhibitory capacity againstα-glucosidase activity. The bioaccessibility of phenolic compounds was significantly greater in FFS compared to FS.ADMET analysis revealed that the majority of phenolic compounds contained in FFS did not exhibit mutagenicity toxicity, hepatotoxicity, skin sensitization,or blood-brain barrier penetration, indicating a favorable level of biosafety.Overall, our study provides a new insight into the further utilization of Se-enriched Brassica napus L.in foods. 展开更多

The aim of the present work is to assess the value of Detarium Senegalense by determining the content of total phenols, total flavonoids and total anthocyanins, and by evaluating the free radical scavenging activity of Detarium Senegalense extracts. For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). DPPH free radical scavenging activity was 1.674 ± 0.023 mg/mL for the acetone extract and 0.934 ± 0.24 mg/mL for the methanol extract of roots. . 展开更多

A novel alkali-soluble polysaccharide from Lepista sordida(LSAP)mycelia with antioxidative and hepatoprotective activities was characterized. The weight-average molecular weight and number-average molecular weight of LSAP were 1.442×10^(3) and 6.05×10^(2) kDa,respectively.LSAP was consisted of glucose(57.9%),xylose(31.8%),and small amounts of rhamnose, arabinose, galactose, glucuronic acid, and galacturonic acid(1.2%–3.1%).The FT-IR and 2D NMR confi rmed that LSAP was composed of Xylp,Araf,4-O-Me-α-D-GlcpA,(1→4)-linkedβ-D-Glcp,and(1→4)-α-D-GalA,andβ-glycosidic linkages between these sugar units.LSAP displayed notable effects on 1,1-dephenyl-2-picryhydrazyl(DPPH)radical scavenging, hydrogen peroxide scavenging, lipid peroxidation inhibitory ability, reducing power and Fe^(2+)chelating property. These biological effects were further verifi ed by suppressing CCl_(4)-induced oxidative liver damage in mice at doses of 100 and 200 mg/kg.Administration of LSAP in mice prior to CCl_(4) signifi cantly prevented the CCl_(4)-induced elevation in serum alanine aminotransferase, aspartate aminotransferase, and hepatic malondialdehyde.Mice treated with LSAP demonstrated to increase activities in superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the liver. We also found out that LSAP prevented CCl_(4)-induced oxidative liver histological alteration.LSAP may exert hepatoprotective effects against CCl_(4)-induced damage through antioxidant mechanisms in model mice. 展开更多

Objective:To investigate the relationship between triterpenoid saponin content and antioxidant, antimicrobial, and α-glucosidase inhibitory activities of 70%ethanolic,butanolic,aqueous,supernate and precipitate extracts of Juglans regia leaves. Methods:Triterpenoid saponins of different Juglans regia leaf extracts were measured by the vanillin method.Antioxidant activity was evaluated against DPPH and ABTS free radicals. We also assessed α-glucosidase inhibitory and antimicrobial activities of the leaf extracts.Pearson’s correlation coefficient was evaluated to determine the correlation between the saponin content and biological activities. Results:The butanolic extract was most effective against DPPH with an IC50of 6.63μg/mL,while the aqueous extract showed the highest scavenging activity against ABTS free radical with an IC50of 42.27μg/mL.Pearson’s correlation analysis indicated a strong negative correlation (r=-0.956) between DPPH radical scavenging activity (IC50) and the saponin content in the samples examined. In addition, the aqueous extract showed the best α-glucosidase inhibitory activity compared with other extracts. All the extracts had fair antibacterial activity against Bacillus subtilis,Escherichia coli, and Klebsiella pneumoniae except for the aqueous extract. Conclusions:Juglans regia extracts show potent antioxidant, antimicrobial, and α-glucosidase inhibitory activities. There is a correlation between saponin levels in Juglans regia leaf extracts and the studied activities. However, additional research is required to establish these relationships by identifying the specific saponin molecules responsible for these activities and elucidating their mechanisms of action. 展开更多

The objective of this study was to investigate the structural and antioxidative properties of royal jelly protein(RJP)at different degrees of hydrolysis(DH)by partial enzymatic hydrolysis. RJP was hydrolyzed by alcalase for 0 min, 15 min, 1 h, 5 h and 8 h to obtain hydrolysates at DH of 5.34%, 11.65%, 15.19%, 21.38% and 23.91%, respectively. With the increased DH, the RJP hydrolysates showed elevated antioxidative activities. The molecular weight of RJP hydrolysates was significantly decreased but their primary backbone kept unchanged. Analysis of circular dichroism spectra revealed that the enzymolysis reduced the content of α-helix but increased the contents of β-sheet, β-turn and random coil. Meanwhile, the surface hydrophobicity and fluorescence intensity of RJP hydrolysates were decreased and a red shift occurred. As the enzymolysis continued, the surface morphology of RJP was gradually changed from a sheet-like structure into microparticles. Changes in antioxidative activities and structures generally followed a DH-dependent manner, however these changes became insignificant for samples at DH beyond 20%. Taking into consideration of both effectiveness and productivity, the optimum enzymatic duration was determined at 5 h. 展开更多

Cadmium(Cd)and Mercury(Hg)is among the heavy metals most hazardous for plant and human health.Known to induce oxidative stress in plants and disbalance equilibrium in the antioxidant defence system, these metals alter plant growth and cause damage at the cellular and molecular levels.Soybean is an important oilseed crop that is raised in soils often contaminated by Cd and Hg.The comparative studies on the deleterious effect of Cd and Hg and the defence system of antioxidants were not studied earlier in soybean plant. In this study, soybean plants were exposed to Cd(100μM CdCl_(2))and Hg(100μM HgCl_(2))and studied for physiological, biochemical and molecular responses. Both Cd and Hg treatment increased the magnitude of oxidative stress.Activities of antioxidant enzymes were significantly upregulated in response to Cd and Hg stress.Quantitative and qualitative assessment of isolated RNA showed significant differences in RNA under stress.Integrity values of RNA confirmed alterations.Transcript level of the Actin gene, involved in the morphogenesis of plants and also used as referenced gene in expression studies was analyzed using qRT-PCR just to check its stability and response under heavy metal stress. Results showed significant upregulation of the gene in the presence of Cd.It can be concluded that both Cd and Hg caused oxidative damage to plants, and adversely affected the quality of RNA.However, soybean tried to limit the adverse impacts of Cd and Hg stress by elevating the antioxidant system and upregulating Actin gene. 展开更多

This study has been conducted to analyze the chemical composition and antimicrobial, antioxidant and antiplasmodial properties of ethanol extracts of Pycnostachys erici-rosenii R.E.Fr and Leucas martinicensis (Jack.) R.Br that are not yet well known. The HPLC-DAD-MS-ESI+ method was used to investigate the chemical profile, the disc diffusion and microdilution method for the antimicrobial activities, the DPPH and FRAP assays for antioxidant property and SYBR Green I-based growth inhibition assay on Pf 3D7 (CQ-sensitive) strain for antiplasmodial properties. The results indicated significant amounts of hydroxybenzoic acid, flavonol and hydroxycinnamic acid in the two species. The total polyphenols analysis gave 209.0 ± 16.1 mg GAE/100g DW for P. erici-rosenii and 175.3 ± 1.6 mg GAE/100g DW for L. martinicensis while for total flavonoids analysis 33.4 ± 1.8 mg TE/100g DW was found for P. erici-rosenii and 39.6 ± 0.6 mg TE/100g DW for L. martinicensis. Moreover, antimicrobial tests revealed 0.125 mg/mL on E. faecalis ATCC25922 for ethanol extracts P. erici-rosenii and L. martinicensis. The antioxidant assays revealed IC50 56.17 μg/mL and 78.43 μg/mL for DPPH assay, and 112.03 μM TE/100mL and 90.46 μM TE/100mL) for FRAP assay for ethanol extracts of P. erici-rosenii and L. martinicensis respectively. Finally, for Antiplasmodial activity, IC50 was 4.07 μg/mL for P. erici-rosenii and 12.29 μg/mL for L. martinicensis. Therefore, the antimicrobial, the antioxidant, and the antiplasmodial results suggest that the ethanolic extracts of the aerial parts of P. erici-rosenii and L. martinicensis contain compounds with antimicrobial, antioxidant, and antiplasmodial properties, which can be searched in new drug discovery for treatment of infectious diseases. 展开更多

In this study, the structural characters, antioxidant activities and bile acid-binding ability of sea buckthorn polysaccharides(HRPs)obtained by the commonly used hot water(HRP-W),pressurized hot water(HRP-H),ultrasonic(HRP-U),acid(HRP-C)and alkali(HRP-A)assisted extraction methods were investigated. The results demonstrated that extraction methods had significant effects on extraction yield, monosaccharide composition, molecular weight, particle size,triple-helical structure, and surface morphology of HRPs except for the major linkage bands.Thermogravimetric analysis showed that HRP-U with filamentous reticular microstructure exhibited better thermal stability. The HRP-A with the lowest molecular weight and highest arabinose content possessed the best antioxidant activities. Moreover, the rheological analysis indicated that HRPs with higher galacturonic acid content and molecular weight showed higher viscosity and stronger crosslinking network(HRP-C,HRP-W and HRP-U),which exhibited stronger bile acid binding capacity. The present findings provide scientific evidence in the preparation technology of sea buckthorn polysaccharides with good antioxidant and bile acid binding capacity which are related to the structure affected by the extraction methods. 展开更多

Background Milk synthesis in lactating animals demands high energy metabolism, which results in an increased production of reactive oxygen metabolites(ROM)causing an imbalance between oxidants and antioxidants thereby inducing oxidative stress(OS)on the animals. To mitigate OS and postpartum disorders in dairy goats and gain insight into the impact of dietary choices on redox status during lactation,a feeding trial was conducted using alfalfa silage inoculated with a high-antioxidant strain of Lactiplantibacillus plantarum. Methods Twenty-four Guanzhong dairy goats(38.1±1.20 kg)were randomly assigned to two dietary treatments:one containing silage inoculated with L.plantarum MTD/1(RSMTD-1),and the other containing silage inoculated with high antioxidant activity L.plantarum 24-7(ES24-7).Results ES24-7-inoculated silage exhibited better fermentation quality and antioxidant activity compared to RSMTD-1.The ES24-7 diet elevated the total antioxidant capacity(T-AOC),superoxide dismutase(SOD),glutathione peroxi-dase(GSH-Px),and catalase(CAT)activities in milk, serum, and feces of lactating goats(with the exception of T-AOC in milk).Additionally, the diet containing ES24-7 inoculated silage enhanced casein yield, milk free fatty acid(FFA)content, and vitamin A level in the goats’milk.Furthermore,an increase of immunoglobulin(Ig)A,IgG,IgM,inter-leukin(IL)-4,and IL-10 concentrations were observed, coupled with a reduction in IL-1β,IL-2,IL-6,interferon(IFN)-γ,and tumor necrosis factor(TNF)-αconcentrations in the serum of lactating goats fed ES24-7.Higher concentrations of total volatile fatty acid(VFA),acetate, and propionate were observed in the rumen fluid of dairy goats fed ES24-7 inoculated silage. Moreover, the diet containing ES24-7 inoculated silage significantly upregulated the expression of nuclear factor erythroid 2 like 2(NFE2L2),beta-carotene oxygenase 1(BCO1),SOD1,SOD2,SOD3,GPX2,CAT,glu-tathione-disulfide reductase(GSR),and heme oxygenase 1(HMOX1)genes in the mammary gland, while decreased the levels of NADPH oxidase 4(NOX4),TNF,and interferon gamma(IFNG).Conclusions These findings indicated that feeding L.plantarum 24-7 inoculated alfalfa silage not only improved rumen fermentation and milk quality in lactating dairy goats but also boosted their immunity and antioxidant status by modulating the expression of several genes related to antioxidant and inflammation in the mammary gland. 展开更多

Quercetin compounds have antioxidant,anti-inflammatory and anticancer pharmacological functions.Longterm exposure to acrylamide(AA)can cause liver injury and endanger human health. However, whether quercetin compounds can attenuate AA-induced liver injury and the specific mechanism are not clear. Here,we studied the mechanism and structure-activity relationship of quercetin compounds in reducing AA-induced hepatotoxicity in vivo and in vitro. In vivo studies found that quercetin-like compounds protect against AAinduced liver injury by reducing oxidative stress levels, activating the Akt/m TOR signaling pathway to attenuate autophagy, and improving mitochondrial apoptosis and endoplasmic reticulum stress-mediated apoptosis. In vitro studies found that quercetin compounds protected Hep G2 cells from AA by attenuating the activation of AA-induced autophagy, lowering reactive oxygen species(ROS)levels by exerting antioxidant effects and thus attenuating oxidative stress, increasing mitochondrial membrane potential(MMP),and improving apoptosis-related proteins, thus attenuating AA-induced apoptosis.Furthermore, the conformational differences between quercetin compounds correlated with their protective capacity against AA-induced hepatotoxicity, with quercetin showing the best protective capacity due to its strongest antioxidant activity. In conclusion, quercetin compounds can protect against AA-induced liver injury through multiple pathways of oxidative stress, autophagy and apoptosis, and their protective capacity correlates with antioxidant activity. 展开更多

Wheat(Triticum aestivum L.)exhibits a greater capacity for cadmium(Cd)absorption compared to other cereal crops, leading to elevated daily Cd intake, and posing a significant threat to public health. For the mitigation of Cd stress in sustainable and environmentally friendly way,a pot study was designed by using exogenous application of various biostimulants,i.e.,Nigella sativa and Ocimum sanctum extracts:0%,10%,and 20%in combination with the chelating agent ethylenediaminetetraacetic acid(EDTA)using 0 and 5 mg kg^(-1) under various levels of Cd stress(i.e.,0,5,10,and 15 mg kg^(-1) soil). Results revealed that Cd stress significantly reduced the seed emergence, growth, root, and allometric characters and yield-related parameters of wheat crops. The most observable reduc-tion was documented in wheat plants exposed to a higher Cd concentration(15 mg kg^(-1)),followed by the lower Cd level(control). The combined application of bio-stimulants and EDTA minimized the negative impacts of Cd stress. The highest increase in seedling emergence(5.44%),leaf area(50.60%),number of tillers(31.02%),grain yield per plant(24.28%),biological yield(13.97%),and decrease in Cd levels in grains(40%)was noticed where 20%foliar application of N.sativa and 10%of O.sanctum biostimulants were done using 5 mg kg^(-1) of soil-applied EDTA.This intervention demonstrated a notable reduction in Cd-induced negative effects, highlighting the potential of these substances in promoting sustainable wheat cultivation in contaminated environments. Moreover,it is an eco-friendly and approachable method at thefield level able to ensure food safety. 展开更多

Bone collagen hydrolysates(peptides)derived from byproduct of animal product processing have been used to produce commercially valuable products due to their potential antioxidant activity.Maillard glycosylated reaction is considered as a promising method to enhance the antioxidant activity of peptides.Hence, this research aims at investigating the Maillard glycosylation activity and antioxidant activity of bone collagen hydrolysates from different sources. In this study,3 glycosylated bone collagen hydrolysates were prepared and characterized, and cytotoxicity and antioxidant activity were analyzed and evaluated. The free amino groups loss, browning intensity, and fluorescence intensity of G-Cbcp(glycosylated chicken bone collagen hydrolysates(peptides))were the heaviest, followed by G-Pbcp(glycosylated porcine bone collagen hydrolysates(peptides))and G-Bbcp(glycosylated bovine bone collagen hydrolysates(peptides)).The results of amino acid analysis showed that amino acid composition of different bone collagen hydrolysates was significantly different and the amino acid decreased to different degrees after Maillard glycosylated reaction, which may lead to differences in Maillard glycosylated reaction activity.Furthermore, the 3 glycosylated hydrolysates showed no significant cytotoxicity. The results showed that glycosylation process significantly increased the antioxidant activity of bone collagen hydrolysates, and G-Cbcp showed the strongest antioxidant activity, followed by G-Pbcp and G-Bbcp.Therefore,compared with the bone collagen hydrolysates,3 glycosylated hydrolysates showed significant characteristic and structural changes, and higher antioxidant activity. 展开更多

This study is the first to evaluate the phytochemical content and biological properties of Tunisian T.platyphyllos Scop.A total of 23 compounds of essential oils were identified by gas chromatography-mass spectrometry(GC-MS)analysis of bracts and fruit extracts. The results show that oxygenated monoterpenes were the dominant class of essential oils. The phenolic composition was investigated by matrix-assisted laser desorption/ionizationtime of flight(MALDI-TOF).The analysis showed that the chemical profiles of the ethanolic extracts of bracts and fruits are substantially similar. The highest polyphenol content was found in the ethanolic extracts of the fruits(7.65 mg gallic acid equivalents(GAE).g−1 on dry weight(DW).As for the antioxidant capacity,it was recorded in the essential oils of the fruits(IC_(50)=0.91 mg.mL^(−1)).The results obtained showed that the antioxidant activity of the fruit essential oil was higher than that of the ethanol extract. The fruit essential oil was also found to have the highest reducing power(IC_(50)=0.67 mg.mL^(−1)).In terms of iron-chelating power, fruit essential oil has the highest chelating power(IC_(50)=2.03 mg.mL^(−1)).Compared to the ethanolic extract, the essential oil had the maximum antioxidant capacity. The enzymatic activity of acetylcholinesterase(AChE)inhibition of the essential oil from T.platyphyllos bracts had the most potent inhibitory effect(IC_(50)=0.77 mg.mL^(−1)),followed by the essential oil from the fruits(IC_(50)=0.95 mg.mL^(−1)).The results suggest that T.platyphyllos can be used as a potential source of naturally occurring bioactive compounds and antioxidants. 展开更多

It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. DPPH scavenging activities of fractions 4 (55.59%) and 5 (55.64%) were significantly higher than the other fractions (p A. rigidula extracts contain gallic, caffeic, vanillic, p-coumaric, salicylic acids and vanillin. 展开更多

[Objectives]To study the optimum conditions of solid fermentation of natto with antioxidant function as an index.[Methods]Single factor experiment and orthogonal experiment were designed to study the effects of temperature, time, initial pH and inoculum amount on the antioxidant activity of natto solid fermentation. The optimum conditions of natto solid fermentation were determined and the antioxidant ac-tivity of natto extract was compared.[Results]The optimal fermentation conditions were as follows:temperature 32℃,initial pH 7.0,inocu-lation amount 8%,fermentation time 32 h.The hydroxyl radical scavenging rate of natto solid fermentation crude extract was the highest, which was 82.7%.The optimized nato fermentation extract showed stronger scavenging ability for-OH and O,:,and showed obvious dose-effect relationship.ICso was 3.63 and 4.24 mg/mL,respectively,and the scavenging efficiency was 1.3 and 1.9 times higher than that of the unoptimized fermentation extract, respectively.[Conclusions]Natto is rich in nattokinase and other functional factors, and its antioxidant ac-tivity can be improved by optimizing fermentation technology,so that natto products can be widely used, including cosmetic raw materials,nat-to skin care soap, health food and medicine, etc.,and have a broader development prospect. 展开更多
