Objective: To evaluate the scientific basis for the use of Kang 601 heji(K-601) as an anti-in flammatory and antipyretic agent using appropriate animal models. Methods: Carrageenan-induced rat paw and xylene-induced ear oedemas were models used to investigate anti-in flammatory actions of K-601. Lipopolysaccharide-induced pyrexia model was used to evaluate antipyretic activity in Wistar rats. The antiin flammatory and antipyretic mechanisms were evaluated by detecting prostaglandins E2, nitric oxide, interleukin-1b and tumour necrosis factora levels using appropriate reagents and ELISA kits. Results: The results revealed that K-601 reduced the level of in flammations in both antiin flammatory models in a dose-dependent manner. The same was true for the antipyretic model. The possible mechanisms of actions were through the inhibition of prostaglandins E2, interleukin-1b, tumour necrosis factora and nitric oxide. Conclusions: K-601 has proven anti-in flammatory and antipyretic actions. The findings provide a scientific basis for the use of K-601 as anti-in flammatory and antipyretic agent in traditional Chinese medicinal practice. 展开更多

Objective: To evaluate the other pharmacological actions of silymarin in Albino rats and mice such as antipyretic, anti-inflammatory, antinociceptive and antihyperlipidemic effects. Methods: Rats were injected intramuscularly with pyrogenic dose of brewer's yeast for the antipyretic test of silymarin. Another group of rats injected with 0.1 mL of 1% carrageenan solution in saline at the subplanter area of the right hind paw for the anti-inflammatory test of silymarin. Another group of mice tested by hot plate method for determination of antinociceptive effect of silymarin. Hyperlipidemia was induced using high fat diet for 2 months to estimate the antihyperlipidemic activity of silymarin. Results: Silymarin showed a significant antipyretic effect of both doses(50 and 100 mg/kg) compared with control untreated group. Moreover, silymarin elucidated a significant anti-inflammatory effect of both doses reflected on the decrease of the rat paw edema every hour interval for 4 h after administration in comparison with control positive group. By the same taken, both doses of silymarine revealed a significant antinociceptive action in hot plate method at 30 and 60 min post administration. Besides, it lowered significantly the serum levels of prostaglandin E2, tumor necrosis factor alpha and interleukin 1 beta after 2 h of silymarin administration in carrageenan induced rat paw edema besides the significant decrease of total cholesterol, triglycerides, low density lipoprotein and significantly elevated high density lipoprotein after 2 weeks of silymarin administration. Conclusions: These outcomes delivered a new vision into the possible pharmacological mechanisms by which silymarin advances antipyretic, anti-inflammatory, antinociceptive and antihyperlipidemic effects. 展开更多

Objective:To explore the anti-inflammatory and antipyretic effects of methanolic leaf extract from Arbutus andrachne and its mechanism of action. Methods:Paw edema was induced by intraplantar(i.pl.)injection ofλ-carrageenan(1%w/v,100μL/paw)while pyrexia was evoked by intraperitoneal(i.p.)injection of 20% baker’s yeast(20 mL/kg body wt)in male Wistar rats. The anti-inflammatory and antipyretic effects of Arbutus andrachne methanolic leaf extract were explored by injecting rats with different doses of the plant extract(150,300,and 600 mg/kg body wt,i.p.).Selective antagonists for transient receptor potential vanilloid-1(TRPV1),cannabinoid receptor 1(CB1),and alpha-2 adrenergic receptor(α2-AR)were used to unravel the extracts’mechanism of action.Blood samples were collected from the heart of rats to measure the levels of interleukin-6(IL-6)and prostaglandin E2(PGE2)by enzyme-linked immunosorbent assay. Results:The extract exhibited potent anti-inflammatory activity by decreasing paw thickness and IL-6 levels. In addition,yeast-evoked pyrexia was attenuated by the extract treatment via TRPV1 and CB1 receptors and a reduction in PGE2 levels. No significant effects were found for α2-AR.Moreover,the rats that received the plant extract demonstrated similar responses to the positive control group. Conclusions:Arbutus andrachne can be a good candidate for treating inflammation and pyrexia and should be further investigated. 展开更多

C. ambrosioides is a Congolese medicinal plant used in the treatment of several pathologies. This work was initiated to evaluate the analgesic and antipyretic effect of the aqueous extract of leaves of C. ambrosioides. Analgesic effect was evaluated by using the acetic acid-induced writhing, the Tilk Flick test as well as the pain induced by formaldehyde. The results obtained showed that the aqueous extract significantly reduces the number of abdominal writhes (p C. ambrosioides (400 and 600 mg/kg) compared to control group (saline water). This result suggests an antipyretic effect. Chemical screening revealed the presence of alkaloids, flavones and mucilage. The presence of alkaloids and flavones could explain the analgesic and antipyretic effects attributed to this aqueous extract. 展开更多

The effects of isatin (2,3-dioxo-indole) and isatin analogues (5-methylisatin, 6-hydroxyisatin, 7-ethylisatin, N-acety- lisatin) were tested on prostaglandin E2 (PGE2)-induced fever in mice and rats. Two modes of administration were tested. Isatin or an analogue was injected simultaneously with PGE2 and the development of fever was tested, or the test compound was given 30 min following PGE2 administration and the effects on the already existing fever were measured, in mice and in rats. Isatin in a dose of 3.12 mg/kg ip was found to block the development of PGE2-induced fever in mice, while in a dose of 12.5 mg/kg ip it attenuated the existing fever. In rats isatin in a dose of 12.5 mg/kg ip blocked fever initiation, and at 25.0 mg/kg ip attenuated existing PGE2-induced fever. In mice, 5-methylisatin in a dose of 0.21 mg/kg ip blocked the initiation of fever, and at 6.72 mg/kg ip attenuated the existing fever. In rats in a dose of 3.36 mg/kg ip it blocked the development of fever, and at 13.44 mg/kg ip attenuated existing PGE2-induced fever. In mice 5,6-dimetylisatin in a dose of 0.02 mg/kg ip both blocked fever initiation and attenuated the existing fever in mice, in rats in a dose of 0.42 mg/kg ip it blocked the initiation of fever, and at 0.21 mg/kg ip attenuated the existing PGE2-induced fever. In mice 6-hydroxyisatin in a dose of 5.2 mg/kg blocked the development of fever, and at 10.4 mg/kg attenuated the existing fever. In rats in a dose of 10.40 mg/kg ip it blocked fever development and also attenuated the existing fever. In mice, 7-ethylisatin in a dose of 0.02 mg/kg ip both blocked fever initiation and also attenuated the existing fever. In rats, a dose of 0.11 mg/kg both blocked fever initiation and also attenuated the existing fever. In mice, N-acethylisatin in the dose of 0.005 mg/kg blocked fever initiation, while at 1.024 mg/kg it attenuated existing fever, in rats, in a dose of 0.096 mg/kg it blocked fever initiation, and at 0.384 mg/kg attenuated the existing fever. The results demonstrate that 7-ethyl- and N-acetylisatin are the most effective of these compounds both in blocking the development of PGE2-induced fever and also in attenuating existing the PGE 2-induced fever. 展开更多

Objective: Mahuang-Shigao herb-pair is a famous formula composed of Ephedra and Gypsum. The herb-pair is frequently used for treating cold symptoms and bronchial asthma in the clinical practice of Chinese medicine (CM). In the present study, we evaluated evidence for the benefit of combined use of Ephedra and Gypsum by analyzing the antipyretic and anti-asthmatic activities of Ephedra-Gypsum. Methods: The antipyretic effects of Ephedra-Gypsum were evaluated in yeast-induced hyperthermia test. Thirty male Wistar rats were randomly divided into 5 groups, including control group, standard aspirin group, and 3 Ephedra- Gypsum groups of different doses (6, 12, 24 g/kg). Ephedra-Gypsum extract and asprin were administered orally 6 h after the injection of yeast solution and body temperature was measured every 1 h for 8 h. The anti- asthmatic effects of Ephedra-Gypsum were evaluated using an ovalbumin (OVA)-induced asthmatic rat model. Thirty-six male SD rats were randomly divided into 6 groups. Rats were alternately sensitized and OVA+Al(OH)3 challenged by exposure to mists of ovalbumin. Ephedra-Gypsum extracts (6, 12, 24 g/kg) or dexamethasone were administered 45 min prior to the allergen challenge for 8 days. Latent period and the weight of wet to dry ratio of lung were determined. In addition, the eosinophils in blood and white blood cell (WBC) were counted by an YZ-Hemavet Analyzer. Results: The Ephedra-Gypsum extracts at test dose (6, 12, 24 g/kg) significantly and dose-dependently attenuated yeast-induced fever in rats. The Ephedra-Gypsum extracts also prolonged the latent period, reduced OVA-induced increases in eosinophils and WBC, and decreased the wet and dry weight ratio of the lungs in the anti-asthmatic test. Conclusions: These findings indicate that the Ephedra-Gypsum extract has antipyretic and anti-asthmatic properties. Hence, the results support additional scientific evidence in prescriptions. 展开更多

OBJECTIVE: The main objectives of this study were to qualitatively evaluate the profile of phytochemical constituents present in methanolic extract of Microcos paniculata bark(BME) and fruit(FME), as well as to evaluate their anti-inflammatory, analgesic and antipyretic activities.METHODS: Phytochemical constituents of BME and FME were determined by different qualitative tests such as Molisch’s test, Fehling’s test, alkaloid test, frothing test, Fe Cl3 test, alkali test, Salkowski’s test and Baljet test. The anti-inflammatory, analgesic and antipyretic activities of the extracts were evaluated through proteinase-inhibitory assay, xylene-induced ear edema test, cotton pellet-induced granuloma formation in mice, formalin test, acetic acid-induced writhing test, tail immersion test and Brewer’s yeastinduced pyrexia in mice.RESULTS: M. paniculata extracts revealed the presence of carbohydrates, alkaloids, saponins, tannins, flavonoids and triterpenoids. All of the extracts showed significant(P〈0.05, vs aspirin group) proteinaseinhibitory activity, whereas the highest effect elicited by plant extracts was exhibited by the BME(75.94% proteinase inhibition activity) with a half-maximal inhibitory concentration(IC50) of 61.31 μg/m L. Each extract at the doses of 200 and 400 mg/kg body weight showed significant(P〈0.05, vs control) percentage inhibition of ear edema and granuloma formation. These extracts significantly(P〈0.05, vs control) reduced the paw licking and abdominal writhing of mice. In addition, BME 400 mg/kg, and FME at 200 and 400 mg/kg showed significant(P〈0.05, vs control) analgesic activities at 60 min in the tail immersion test. Again, the significant(P〈0.05, vs control) post-treatment antipyretic activities were found by BME 200 and 400 mg/kg and FME 400 mg/kg respectively.CONCLUSION: Study results indicate that M. paniculata may provide a source of plant compounds with anti-inflammatory, analgesic and antipyretic activities. 展开更多

Objective To clarify the antipyretic effect of the Chinese materia medica, Violae Herba (Viola yedoensis), and its active fractions by examining the effects of V. yedoensis extracts with differing polarities on body temperature, total white blood cell (WBC) count, WBC differential count, and total serum complement of rabbits with lipopolysaccharide (LPS)-induced fever. Methods The rabbits were treated with water and ethanolic extracts of V. yedoensis, as well as petroleum ether, ethyl acetate, and n-butanol fractions of the ethanolic extract at low-, mid- and high- doses. The LPS was injected via the ear vein of rabbits in model and treatment groups 30 min post-gavage. Their body temperature was measured at 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5, 4.0, 4.5, 5.0, 5.5, and 6.0 h after the LPS challenge to calculate the temperature changes and thermal response index. After the last temperature measurement, blood samples were collected to determine the blood cell counts and total serum complement (CHs0) level. Results Compared with the model group, body temperature was significantly lower in the low-dose ethanolic extract group, low- and mid-dose petroleum ether fraction groups, and all three ethyl acetate fraction groups. Serum CHs0 levels were lower in all treatment groups, except the ethanolic extract groups, than that in the model group, with no significant difference. V. yedoensis had no significant effect on the blood cells of febrile rabbits challenged with LPS for 6 h. Conclusion The antipyretic effects of V. yedoensis are strong, and its active fractions are the petroleum ether and ethyl acetate fractions of ethanolic extract. 展开更多
