Ilex cornuta (1.cornuta)is a traditional Chinese medicine (TCM)that has been used in clinical practice for hundreds of years.In order to provide more information about the chemical basis of its pharmacological effects...Ilex cornuta (1.cornuta)is a traditional Chinese medicine (TCM)that has been used in clinical practice for hundreds of years.In order to provide more information about the chemical basis of its pharmacological effects,phytochemical investigation on the roots of L cornuta was conducted in this study.The roots of the plant were firstly extracted with 95% EtOH,and then the crude was partitioned with petroleum ether,EtOAc and n-butyl alcohol.Different chromatographies were employed to isolate the crude step by step and the crude was further purified by semipreparative high performance liquid chromatography (HPLC).As a result,two new triterpenoid saponins (1,2),together with 12 known compounds (3-14),were isolated from the roots of I. cornuta.Their structures were determined based on nuclear magnetic resonance (NMR),mass spectrum (MS)technologies,chemical reactions as well as gas chromatography (GC).Compounds 4,6,8,11,12 and 13 were isolated from this genus for the first time.The structures of compounds 1 and 2 were determined as 3β-O-α-D-xylopyranosyl-(1→3)-α-L-2-O-acetylarabinopyranosyl-(1→2)-β-D glucopyranosyl-23-hydroxyl-20α(H)-urs-12-en-28-oic acid 28-O-β-D-glucopyranosyl ester (1)and 3β-O-α-D-xylopyranosly-(1→3)-α-L-2-O-acetylarabinopyranosyl-(1→2)-β-D-glucopyranosyl-19α,23-dihydroxyl-20α(H)-urs-12-en-28-oic acid 28-O-β-D-glucopyranosyl ester(2).展开更多
As the energizing and therapeutic effects of infusions of mate(Ilex paraguariensis A.St.-Hil)leaves are increasingly recognized,rising global consumption is also increasing the demand for plantlets with high genetic a...As the energizing and therapeutic effects of infusions of mate(Ilex paraguariensis A.St.-Hil)leaves are increasingly recognized,rising global consumption is also increasing the demand for plantlets with high genetic and morphological qualities.Mini-cuttings are an alternative for clonal plantlets;however,more details are needed on factors that affect the success of this technique.Here,toward maximizing production,we evaluated adventitious rooting competence of mini-cuttings of different sizes and the morphophysiological quality of the resultant plantlets.Small(one bud;length up to 2.5 cm),medium(multiple buds;length 2.6–5.0 cm),and large(multiple buds;length 5.1–10.0 cm)mini-cuttings were planted in a mixture of commercial substrate,vermiculite,and coarse sand(1:1:1 v:v:v),grown in a humidity chamber,then evaluated after 45,60,75,and 90 days for survival,percentage of callus and shoot formation,number and length of shoots,percentage of rooted cuttings,and number and length of roots.Any rooted mini-cuttings were then cultivated in commercial substrate and subsurface soil(2:1 v:v)and evaluated after 30,60,90 and 120 days of cultivation for morphophysiology of the plantlets,percentage of survival,number of leaves,shoot height,stem diameter,and ratio of shoot height to stem diameter.After 120 days,the plantlets were evaluated for the ratio of shoot and root dry mass,total length,surface area,total volume,and mean diameter of roots and Dickson quality index.Adventitious rooting of the mini-cuttings and the morphophysiological quality of the plantlets were not influenced by the size of the vegetative propagule.Therefore,single-bud mini-cuttings have similar adventitious rooting competence,and result in plantlets with adequate morphophysiological quality,based upon leaf development,shoot height,stem diameter and length,and surface area,volume and diameter of roots.Single-bud mini-cuttings up to 2.5 cm long can thus be used to maximize production of mate plantlets by mini-cuttings.展开更多

A reversed-phase high performance liquid chromatography (RP-HPLC) method with diode array detector (DAD) has been established to simultaneously determine six bioactive compounds in the roots ofllex pubescens, name...A reversed-phase high performance liquid chromatography (RP-HPLC) method with diode array detector (DAD) has been established to simultaneously determine six bioactive compounds in the roots ofllex pubescens, namely tortoside A, (+)-syringaresinol, ilexsaponin B3, ilexsaponin A1, ilexsaponin B1, ilexgenin A. The RP-HPLC assay was performed on a reversed-phase C18 column with a gradient elution. The mobile phase consisted of acetonitrile and water containing 0.1% (v/v) phosphoric acid. The flow rate was 1.0 mL/min and the detection wavelength was set at 210 nm. The six marker constituents were separated well with good linearity (r2〉0.9996), precision, stability and repeatability. The overall recoveries were in the range of 99.00%-104.52%. Cluster analysis was employed to analyze 15 batches of samples. The result indicated this method provide an efficient way to perform quality control as well as a scientific rationale for the Geo-authentication of I. pubescens.展开更多

There are 600 species in genus IlexL. containing various compounds such as terpenoids, saponins, glycosides, etc. Three species, L cornuta, I. chinensis, and L rotunda have been admitted in Chinese Pharmacopoeia 2015 to treat dyspepsia, stomatitis, and hyperactivity cough and protect the liver and kidney. Recent studies showed that several species have been daily drunk to promote human health and prevent cardiovascular diseases in the folk.
