Process parameters of microwave assisted extraction (MAE) of the polysaccharides from pumpkin viz. extraction temperature, time and liquid-solid ratio were studied by using single factor and response surface methodology method. The results showed that the liquid-solid ratio was the most important factor in polysaccharides yield, followed the extraction temperature was the least important factor. The optimum microwave assisted extraction co by ndi extraction time, and tions for the highest polysaccharides yield from pumpkin (16.76%-4-0.38%) were obtained by using the response surface methodology with extraction time of 29 min, an extraction temperature of 79 ℃ and a liquid-solid ratio of 22 mL·g^-1. In the present work,an enzyme assisted extraction method is used to isolate crude polysaccharides from Ganoderma lucidum. The isolating effect was optimized with orthographic graph statistic method with three levels and four independent variables. Complex enzyme,extraction temperature,extraction time and extraction pH were combined to obtain the best possible combination to get maximum amount of extract and crude polysaccharides yield. The optimum extraction conditions were:complex enzyme amount of 3 %(w/v),extraction temperature at 45 ℃,extraction time of 3 h and extraction pH at 7. Under these conditions,the experimental amount of extract is 8.9 % and the yield of crude polysaccharides is 1.1 %,which are in close agreement with the value predicted by the model. 展开更多

Orange pomace is the solid waste of the orange juice industry which accounts for approximately 50%of the quantity of the fruits processed into juice and is a good raw material for production of high added value products with diverse uses.Orange pomace is rich in polyphenols and flavonoids which can substitute the potentially hazardous or less desirable chemical antioxidants/antimicrobials used in agro-food and cosmetics industry.In this work,an eco-friendly aqueous microwave assisted extraction of orange pomace was investigated and optimized in order to produce aqueous bioactive antioxidant/antimicrobial extracts.A three factorial Response Surface Optimization methodology with centered Box&Behnken experimental design was used to obtain optimum values of total polyphenols and total flavonoids and build predictive models for their optimal extraction conditions.The three optimization factors in terms of applied process parameters were(a)water/solid ratio,(b)extraction temperature and(c)extraction time.The effectiveness and statistical soundness of the two corresponding models regarding optimal total polyphenols and flavonoids were verified by Analysis of Variance(ANOVA). 展开更多

In this paper,using to the comprehensive scoring method,pressure shear assisted extraction was used to prepare polysaccharide from Auricularia auricula(PS-AAP)by orthogonal test to optimize the extraction.The highest comprehensive score(90.70)was obtained under 1:70 g/mL solid-liquid ratio,110°C reaction temperature,60 min reaction time and 1.0 MPa reaction pressure.Compared to AAP by water bath(WB-AAP),PS-AAP led to a higher extraction efficiency and decreased the relative molecular weight.In addition,infrared spectrum(FT-IR)analysis showed that pressure shear assisted did not change the polysaccharide structure.Scanning electron microscope(SEM)showed that PS-AAP was smaller and more broken by pressure shear assisted.Furthermore,the ratio of glucose to mannose in PS-AAP was relatively higher than WB-AAP,which also proved that PS-AAP had certain antioxidant and hypoglycemic activities.Compared with WB-AAP,the inhibitionα-glucosidase andα-amylase activity of PS-AAP increased by 11.37%and 24.43%respectively.These results revealed that PS-AAP has the poten-tial value of being a antioxidant and hypoglycemic agent. 展开更多

Cimicifugae Rhizome (RC) is one of many commonly used traditional Chinese medicines. This study used Box-Behnken Design (BBD), a widely used form of response surface methodology (RSM), to optimize the conditions of the ultrasound-assisted extraction (UAE) for the extraction of phenolic compounds from CimicifugaeRhizome. The influence of three independent variables, including ethanol concentration (%), extraction time (min) and solvent-to-material ratio (mL/g), on the extraction yields of phenolic compounds were examined. The results demonstrated that the optimal ultrasound-assisted extraction (UAE) conditions were as follows: ethanol concentration of 68%, an extraction time of 19 min, and solvent-to-material ratio of 30 mL/g for the extraction of caffeic acid, cimicifugoside, ferulic acid, isoferulicacid, cimifugin, 4′-O-β-glucopyranosyl-5-O-methylvisamminol. These results demonstrated the suitability of UAE for the extraction of phenolic compounds from CimicifugaeRhizome. 展开更多

Objective: To study and compare the conventional extraction procedure with microwave assisted extraction (MAE) for some Ayurvedic Rasayana drugs and to evaluate their antioxidant potential and carry out the characterization of extracts by thin layer chromatography. Methods: Three Ayurvedic rasayana plants Allium sativum Linn., Bombax ceiba Linn. and Inula racemosa Hook. were evaluated for an improved MAE methodology by determining the effects of grinding degree, extraction solvent, effect of dielectric constant and duration of time on the extractive value. Antioxidant potential of all three drugs was evaluated with 2,2-diphenyl-l-picrylhydrazyl (DPPH) radical scavenging activity and reducing power was determined by using Gallic acid as standard. Further thin layer chromatographic (TLC) analysis was performed on pre-activated Silica Gel G plates and Rf value were compared with those reported for the important biomarkers. Results: The total extractive value for Allium sativum Linn. was 36.95% (w/w) and 49.95% (w/w) for ethanol extraction respectively. In case of Bombax ceiba Linn. the yield of aqueous extract by MAE was 50% (w/w) compared to 42% (w/w) in ethanol (50% v/v). Percent yield of Inula racernosa Hook. in aqueous extract was found to be 27.55% (w/w) which was better than ethanol extract (50%) where the yield was 25.95% (w/w). Upon antioxidant activity evaluation A. safivum extract showed an absorbance of 0.980 ± 0.92 at concentration of 500 μg with maximum reducing capacity. This was followed by B. ceiba Linn. 0.825 ± 0.98 and I. racemosa Hook. with 0.799 ± 2.01 at a concentration of 500 μ g. TLC based standardization of A. sativum Linn. extract shows single spot with Rf value of 0.38, B. ceiba Linn. extract shows Rf values were 0.23, 0.58, 0.77, 0.92 and I. racemosa Hook. extract spot had a Rf value of 0.72. Conclusions: A significant improvement in extractive values was observed as a factor of time and other advantages by using MAE technology. All three drugs have high antioxidant potential and a TLC profiling similar to reported ones. The presence of fructan type polysaccharide can be further utilized for bioactivity directed fracUonation and evaluation of immunomodulatory activity. 展开更多

In the present study, we aimed to investigate the effects of key extraction parameters including extraction time(10–20 min), extraction temperature(30–60 °C), ultrasonic power(60–90 W) and solvent-to-solid(S/S) ratio(10–30 m L/g) on yield of celastrol from Celastrus monospermus Roxb. To optimize the conditions, we investigated the effects of parameters on the ultrasound assisted extraction(UAE) with the Box-Behnken Design(BBD), one widely used form of Response Surface Methodology(RSM). In all tested solvents, ethanol was the most effective for celastrol extraction, followed by methanol, ethanol, ethyl acetate, n-butanol and water. A second order polynomial model was fitted well to the extraction experimental data with R2 of 0.9928. Extraction yield of 3.116 mg/g was obtained for celastrol under the optimized extraction conditions of extraction time(20 min), extraction temperature(46 °C), ultrasonic power(60 W) and S/S ratio(30 m L/g). Experimental validation was performed, and the experimental values agreed well with the predicted values. The results indicated that the UAE was good extraction material for celastrol from C. monospermus Roxb. 展开更多

Comparative study on yields and quality parameters of argan oils extracted by conventional and green extraction techniques

Argan oil is most frequently sold as pure oil,which can be directly applied topically due to its cosmetological proprieties or ingested in order to provide several health benefits.It's also commonly mixed into a number of cosmetic products like shampoos,soaps and conditioners.In this study we aimed to improve the argan oil extraction yield and quality parameters by comparing the effects of different extraction technologies.Argan kernel oils were extracted using four methods:mechanical cold press,Soxhlet extraction with n-hexane,supercritical fluid extraction(SFE),and enzyme assisted extraction with three different enzyme solutions cellulase(cellulast),pectinase(Pectinex)and a mixture of carbohydrase enzymes(Viscozyme®).The quality parameters was evaluated by determining the acid,peroxide and iodine values aswell as the extinction coefficients K_(232) and K_(270) as measures of conjugated dienes and trienes,respectively.The results showed that the highest yield(66.37%±3.3%)was obtained by enzyme assisted extraction using the carbohydrases enzymes mixture(Viscozyme®),followed by Soxhlet extraction(59.5%±3.1%)and pectinase extraction(52.03%±3.55%).All argan oils samples obtained by the different methods showed a good oxidation stability,with acid,peroxide and iodine values lower than 0.8 mg/g,15 meq/kg and 110 g/100 g according to the official argan oil norm,respectively.The results of argan oils quality parameters demonstrated that the enzyme extracted argan oils showed low oxidation,with the following quality parameters:acid values(0.4-0.6 mg/g),iodine values(95-100 g/100 g),dienes(K_(232)<2),trienes(K_(270)<0.35),and peroxide values(<1.5 meq/kg).The results proved that the enzyme assisted extraction method can be applied to improve the argan oil yield without affecting the oil quality.The enzyme extraction method may be a great alternative to solvent and cold press extractions for this eco-friendly processing approach. 展开更多

Polysaccharides are the most important pharmacologically active constituents of Ganoderma lucidum. In this work, polysaccharides were extracted from Ganoderma lucidum with boiling water method and enzyme assisted method. The human liver hepatocellular carcinoma cell line HepG2 was used to compare the an- titumor effect of the two kinds of extraction with 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) test. Both of these two kinds of Ganoderma lucidum polysaccharides reduced cell viability of cancer cell HepG2 in a dose and time-dependent manner. At low concentrations, there was no significant difference in the effectiveness of L 1 and L2; while at concentrations over 0.8 ug/mL, the difference in the effectiveness of L2 in comparison to L1 became significant. At the concentrations of 3.2 ug/mL, the cancer cells were almost killed in 2 d. 展开更多

Chinese herbal flavonoids play a role in improving the health of aquatic animals and optimizing the quality of aquatic products.The efficient extraction of flavonoids is beneficial to deepening the application of flavonoids in aquatic products.To explore the optimal way of flavonoid extraction has become a hot research topic.In this study,based on temperature,ultrasonic power and extraction time,the best ways to extract flavonoids from four Chinese herbal medicines,purslane,Verbena,Fructus Amomi and Alpinia oxyphylla,were explored.Through single-factor and multi-factor optimization extraction,the most appropriate conditions for flavonoid extraction were obtained.The results showed that the best way of ultrasonic assisted extraction of flavonoids was purslane at 40℃,300 W(400 W),Verbena at 70℃,400W,Fructus Amomi at 50℃,200 W,and A.oxyphylla at 40℃,400W,and 40 min.The results of orthogonal design showed that purslane could get 13.20 mg/g at 40℃,400 W and 50 min,Verbena could get 10.23 mg/g at 60℃,300 W and 30 min,Fructus Amomi could get 8.18 mg/g at 50℃,300 W and 60 min,and A.oxyphylla could get 7.31 mg/g at 30℃,300 W and 50 min. 展开更多

In the present experiment, the total phenolic content and antioxidant activity of the various extracts w</span><span style="font-family:"">ere</span><span style="font-family:""> measured. Thiobarbituric acid reactive substances (TBARS) assay was used to evaluate the ability of the BMR to protect lipid peroxidation in corn oil at 185</span><span style="font-family:"">&deg;</span><span style="font-family:"">C frying temperature. Lotus(Nelumbo nucifera Gaertn),a kind of perennial aquatic plant,is widely cultivated and consumed by people in Asian countries.Lotus plumule flavonoids(LPF)have been recognized as a hypoglycemic agent.LPF was optimally obtained using novel ultrasonic-microwave assisted synergistic extraction(UMSE)method by response surface methodology(RSM)on the basis of the results of single-factor experiments.Furthermore,the hypoglycemic activity of LPF was investigated by measuring the body weight,fasting blood glucose(FBG)level,and oral glucose tolerance test(OGTT)and analyzing the physiological indexes in streptozotocin-diabetic mice model.The optimum extraction conditions consisted of microwave power 355 W,ultrasonic power 423 W,extraction time 15 min,solid-liquid ratio 1:40,ultrasound/interval time 1/0,and ethanol concentration 70%with the maximum LPF yield of 2.62%.LPF supplementation significantly decreased the body weight,FBG,OGTT,serum total cholesterol(TC),serum total triglycerides(TG),and insulin levels,indicating the antidiabetic activity of LPF.This research verified that the UMSE technique was highly efficient to extract LPF to the maximum extent and the flavonoids from L.plumule exhibited hypoglycemic activity,which showed broad development and application prospects. 展开更多

A process for extraction of artemisinin(Ars)from Artemisia annua L.based on aqueous solution of cyclodextrin derivative under ultrasonic irradiation was proposed.Among the test additives,the system containing edible hydroxypropyl-β-cyclodextrin(HPBCD)was shown to be efficient for this extraction.The optimal extraction conditions were predicted by artificial neural network,under which the extraction amount of Ars could reach 8.66 mg per gram of the leaves,much higher than the value in water(1.70 mg g-1).Kinetics study indicated that HPBCD enhanced the equilibrium amount of Ars in water,and ultrasonic irradiation accelerated extraction rate and reduced the activation energy.Mechanism study showed that the formation of Ars-HPBCD complex during extraction enhanced the water solubility of Ars and facilitated the extraction,and the configuration of Ars-HPBCD complex was modeled by molecular docking,which was further verified by characterizations.This work provides a convenient way to fabricate the traditional tea-based medicament of natural drugs to make production of this famous antimalarial medicine cheaper. 展开更多

Soybean oil, rapeseed oil, palm oil and corn oil are primary feedstock for biodiesel production. However, biodiesel production from these traditional oil-rich crops is limited by land availability, climate, and environmental and social issues regarding the use of feed and food crops for fuel. Oleaginous microorganisms, including microalgae, bacteria, yeast and fungi can be cultivated with high lipid contents and used as promising feedstock for biodiesel production. However, the high cost of biodiesel production using oil microorganisms has been the biggest obstacle for its industrialization. The process of biodiesel production from microorganisms involves many steps, of which the lipids extraction is the most important and costly. Therefore, searching for an effective and economical extraction system is critical. Various approaches of lipids extraction are discussed in this review, including traditional extraction procedures such as solvent extraction, pressing and solvent integrated extraction, as well as some new procedures. 展开更多
