Cancer is one of the most prevalent diseases worldwide,which causes significant morbidity and mortality.Designing and developing a potential anti-cancer drug is an active field of research worldwide.Microorganisms hav...Cancer is one of the most prevalent diseases worldwide,which causes significant morbidity and mortality.Designing and developing a potential anti-cancer drug is an active field of research worldwide.Microorganisms have been considered a potential source of anti-cancer drugs.One such microbe-derived compound is surfactin,which shows potential anti-cancer activities.In this study,we evaluated the binding potential of surfactin with several cancer cell ligands via an in-silico approach.Hence,molecular docking studies were performed to test the binding potential of surfactin against four targets.The analyses revealed that surfactin from Bacillus sp.can bind with the targeted ligands(coenzyme A,D-leucine,glycerol,and(R)-3-hydroxytetradecanal)with significant affinity.Surfactin showed the highest binding affinity(-7.7 kcal mol-1)to coenzyme A among the targeted ligands.These results may be useful for developing anti-cancer drugs.Nevertheless,further experimental studies are needed to investigate the ligand binding capacity and anti-cancer potential of such surfactin-like molecules.展开更多

These volatiles strongly inhibited the growth of the most tested pathogenic fungi with wide host plants, induced the mycelial morphological abnormalities, and decreased the sclerotoid production of Sclerotinia sclerotiorum in sealed plates. Spores of Botrytis cinerea exposed to these volatiles for 24-48 h in cavity slides cracked and the sporaceous inclusion became brown and effused to the suspension. An interesting phenomenon observed was that all the bacterial volatiles exhibited intense inhibitory activities against the pigment formation of tested pathogenic fungi, including Ascochyta citrullina, Alternaria solani, Alternaria brassicae, and so on. Interactions mediated by microbial volatiles could be widespread in soils, and volatiles may play an important role in reducing disease levels. A phylogenetic analysis based on 16S rDNA sequence placed the four bacteria in three species Paenibacillus polymyxa (BMP-11), Bacillus subtilis (BL02), and Bacillus pumilus (BSH-4 and ZB 13). Through headspace sampling and GC-MS analysis, a rich profile was found from B. subtilis and overlapping volatile patterns could be found among the different species. Studies are under the way to find the possible action mechanisms and to seek the effective application of bacterial volatiles in greenhouse.展开更多

[ Methods] Using flat isolation and culture method, the bacteria that could produce Bacillus spp. were isolated from rhizosphere soil, and carried out primary and secondary screening by confrontation culture. The lipopeptide substances of Y-3 strain with better antifungal effect were carried out I-IPLC analysis. [Result] Nine strains of bacteria had certain antagonistic effects against P. infestans in primary screening, Y-1 and Y-3 strains had significant inhibition effect against the growth of P. infestans in secondary screening, with inhibition rate higher than 80% ; the crude extract of lipepeptide produced by Y-3 strain had better antagonistic effect against P. infestans, with inhibition rate higher than 90%. HPLC analysis showed that the crude extract of lipepeptide mainly contained 8 components. Objective: To evaluate the ability of Bacillus spp. as direct-fed microbials(DFM) to biodegrade al atoxin B1(AFB1) by using an in vitro digestive model simulating in vivo conditions.Methods: Sixty-nine Bacillus isolat...Objective: To evaluate the ability of Bacillus spp. as direct-fed microbials(DFM) to biodegrade al atoxin B1(AFB1) by using an in vitro digestive model simulating in vivo conditions.Methods: Sixty-nine Bacillus isolates were obtained from intestines, and soil samples were screened by using a selective media method against 0.25 and 1.00 μg/m L of AFB1 in modii ed Czapek-Dox medium. Plates were incubated at 37 °C and observed every two days for two weeks. Physiological properties of the three Bacillus spp. candidates were characterized biochemically and by 16 S r RNA sequence analyzes for identii cation. Tolerance to acidic p H, osmotic concentrations of Na Cl, bile salts were tested, and antimicrobial sensitivity proi les were also determined. Bacillus candidates were individually sporulated by using a solid fermentation method and combined. Spores were incorporated into 1 of 3 experimental feed groups: 1) Negative control group, with unmedicated starter broiler feed without AFB1; 2) Positive control group, with negative control feed contaminated with 0.01% AFB1; 3) DFM treated group, with positive control feed supplemented with 109 spores/g. After digestion time(3:15 h), supernatants and digesta were collected for high-performance liquid chromatography l uorescence detection analysis by triplicate.Results: Three out of those sixty-nine DFM candidates showed ability to biodegrade AFB1 in vitro based on growth as well as reduction of l uorescence and area of clearance around each colony in modii ed Czapek-Dox medium which was clearly visible under day light after 48 h of evaluation. Analysis of 16S-DNA identii ed the strains as Bacillus amyloliquefaciens, Bacillus megaterium and Bacillus subtilis. The three Bacillus strains were tolerant to acidic conditions(p H 2.0), tolerant to a high osmotic pressure(Na Cl at 6.5%), and were able to tolerate 0.037% bile salts after 24 h of incubation. No signii cant dif erences(P > 0.05) were observed in the concentrations of AFB1 in neither the supernatants nor digesta samples evaluated by highperformance liquid chromatography with l uorescence detection between positive control or DFM treated groups. Conclusions: In vitro digestion time was not enough to confirm biodegradation of AFB1. Further studies to evaluate the possible biodegradation ef ects of the BacillusDFM when continuously administered in experimentally contaminated feed with AFB1, are in progress.展开更多

The results indicated that the optimized medium was glucoses 20 g/L, soybean meal 30.0 g/L, K2HPO4 1.0 g/L;fermentation temperature is 37℃, the inoculum age 12 h, initial pH 7.0, 2% inoculation amount, loading volume of liquid 20 mL/250 mL. Under the optimized conditions of culture medium and fermentation for compound Bacillus spp., spore yield was 10.24 times more than the initial medium, and the spore formation rate reached more than 90%.展开更多
The present study was taken up with a view to ascertain the possibility of introduction of alcohol resistant bacteria in vitro through the aseptically raised watermelon (Citrullus lanatus) seedlings in the backdrop of...The present study was taken up with a view to ascertain the possibility of introduction of alcohol resistant bacteria in vitro through the aseptically raised watermelon (Citrullus lanatus) seedlings in the backdrop of isolating such organisms from micropropagated watermelon stocks. Watermelon cv. Arka Manik seedlings grown in vitro from surface-sterilized seeds with the intact seed coat on MS medium appeared visibly clean largely, but upon subjecting them to tissue-indexing, the segments from the collar or root tissue showed bacterial colony growth on Nutrient Agar (NA) from 72% of such healthy seedlings and the cotyledon and hypocotyl tissue of 44% seedlings. The pooled colony growth from NA upon challenge with 90% alcohol yielded 10 distinct colony types, identified as B. pumilus (4×), B. subtilis (4×), B. cereus (1×) or B. safensis (1×) based on partial 16S rRNA sequence analysis. The shoot-tip tissue from the healthy index-negative seedlings cultured on watermelon proliferation medium partly turned index-positive within 2 - 4 sub-culture cycles while being apparently clean. On the other hand, those from the previously index-positive cultures tended to show obvious bacterial growth during subsequent in vitro culturing. The observations suggested the possibility of introduction of spore-forming Bacillus spp. through surface-sterilized seeds, their gradual emergence in vitro in visibly clean seedlings, possible transmittal of spores to the alcohol through tissue-culturing tools and the survival therein with the chances of unsuspected lateral spread. Seed coat removal followed by surface sterilization with sodium hypochlorite facilitated the raising of clean seedlings with no detectable bacterial association.展开更多
Microbes that are beneficial to plants are used to enhance the crop growth,yield and are alternatives to chemical fertilizers.Trichoderma and Bacillus are the predominant plant growth-promoting fungi and bacteria.The ...Microbes that are beneficial to plants are used to enhance the crop growth,yield and are alternatives to chemical fertilizers.Trichoderma and Bacillus are the predominant plant growth-promoting fungi and bacteria.The objective of this study was select,characterize,and evaluate isolates of Trichoderma spp.and Bacillus spp.native from the northern region of Sinaloa,Mexico,and assess their effect on growth promotion in maize(Zea mays L.).In greenhouse conditions,four Trichoderma isolates and twenty Bacillus isolates,as well as two controls,were tested in a completely randomized design with three replicates.We selected the two best strains of Trichoderma and Bacillus:TB=Trichoderma asperellum,TF=Trichoderma virens,B14=Bacillus cereus sensu lato and B17=Bacillus cereus,which were evaluated in the field in a completely randomized blocks in factorial arrangement design with three replicates applying different rates of nitrogen fertilizer(0,150 kg N/ha,and 300 kg N/ha).Treatments 5(B17=B.cereus)and 11(TF=T.virens)both fertilized with 150 kg N/ha showed similar yields and they did not reveal significant differences from the treatments fertilized with 300 kg N/ha.This indicated that treatment 5(B17=B.cereus with 150 kg N/ha)and treatment 11(TF=T.virens with 150 kg N/ha)were efficient as growth promoters,by not showing significant differences in root volume and dry weight of foliage.The results indicated a reduction of 50% in the rate of nitrogen to fertilizer required for maize(Zea mays L.)crops.These microorganisms Trichoderma and Bacillus could be an alternative to reduce the use of chemical fertilizers in maize.展开更多
Considerable research has been conducted into the efficacy of individual probiotics in broiler production,however information on the most effective combinations of synergistic Bacillus probiotic is lacking.This study ...Considerable research has been conducted into the efficacy of individual probiotics in broiler production,however information on the most effective combinations of synergistic Bacillus probiotic is lacking.This study investigated the impact of different Bacillus strain combinations in broiler chickens,as well as in vitro enzyme production.In experiment one,a total of 576 Ross 308 broilers at 1 d old were grown for 21 d across 6 treatments of maize-soybean diets(n=12 pens per treatment)to compare three different strain combinations(formulation 1[F1]:3 strains Bacillus amyloliquefaciens;F2:Bacillus coagulans and 2 strains B.amyloliquefaciens;F3:B.coagulans,Bacillus licheniformis and 2 strains B.amyloliquefaciens;F5:Bacillus subtilis,B.licheniformis and 2 strains B.amyloliquefaciens),positive control(PC),and a negative control antibiotic treatment group(NC).In Exp.2,a total of 360 one-day-old ROSS308 broilers were used to test five treatments(n=9)including PC,NC,F1 and F5(selected from Exp.1),and F4(Bacillus pumilis and 2 strains B.amyloliquefaciens)in a maize-soybean diet.B.amyloliquefaciens F1 demonstrated a significant improvement in feed conversion ratio(FCR)compared to F2 at d 14(1.49 vs 2.10;P=0.038)and the body weight(BW)at d 21(847.0 g vs 787.4 g)compared to other combinations(P=0.027).The FCR at d 21 tended to be lower in birds fed F1(1.46 vs 1.66)compared to the control(P=0.068).Probiotic treatments had significantly improved nutrient digestibility compared to the PC and NC.Also,probiotic treatments supported the growth of Streptococcus,a common commensal genus and reduced the abundance of genera that correlated with low weight gain such as Akkermansia.Experiment two revealed that F4 improved FCR(P<0.001)and BW at 28 d(P=0.014).In vitro testing showed a high production of protease and amylase by Bacillus.Thus,the addition of Bacillus probiotics,particularly containing B.amyloliquefaciens strains and Bacillus pumilus,into the diet of broiler chickens improves production performance,nutrient digestibility,and allows the proliferation of beneficial gut microbiota.展开更多

Spores were incorporated into 1 of 3 experimental feed groups: 1) Negative control group, with unmedicated starter broiler feed without AFB1; 2) Positive control group, with negative control feed contaminated with 0.01% AFB1; 3) DFM treated group, with positive control feed supplemented with 109 spores/g. After digestion time(3:15 h), supernatants and digesta were collected for high-performance liquid chromatography l uorescence detection analysis by triplicate.Results: Three out of those sixty-nine DFM candidates showed ability to biodegrade AFB1 in vitro based on growth as well as reduction of l uorescence and area of clearance around each colony in modii ed Czapek-Dox medium which was clearly visible under day light after 48 h of evaluation. Analysis of 16S-DNA identii ed the strains as Bacillus amyloliquefaciens, Bacillus megaterium and Bacillus subtilis. The three Bacillus strains were tolerant to acidic conditions(p H 2.0), tolerant to a high osmotic pressure(Na Cl at 6.5%), and were able to tolerate 0.037% bile salts after 24 h of incubation. No signii cant dif erences(P > 0.05) were observed in the concentrations of AFB1 in neither the supernatants nor digesta samples evaluated by highperformance liquid chromatography with l uorescence detection between positive control or DFM treated groups. Conclusions: In vitro digestion time was not enough to confirm biodegradation of AFB1. Further studies to evaluate the possible biodegradation ef ects of the BacillusDFM when continuously administered in experimentally contaminated feed with AFB1, are in progress.
文摘In order to improve the spore yield of compound Bacillus spp. (B. amyloliquefaciens, B. laterosporus and B. megaterium), the effects of nutrient conditions including carbon source, nitrogen source, mineral salt and fermentation conditions including the inoculum age, inoculation amount, loading volume of liquid and initial pH on the spore yield were studied. The results indicated that the optimized medium was glucoses 20 g/L, soybean meal 30.0 g/L, K2HPO4 1.0 g/L;fermentation temperature is 37℃, the inoculum age 12 h, initial pH 7.0, 2% inoculation amount, loading volume of liquid 20 mL/250 mL. Under the optimized conditions of culture medium and fermentation for compound Bacillus spp., spore yield was 10.24 times more than the initial medium, and the spore formation rate reached more than 90%.
文摘The present study was taken up with a view to ascertain the possibility of introduction of alcohol resistant bacteria in vitro through the aseptically raised watermelon (Citrullus lanatus) seedlings in the backdrop of isolating such organisms from micropropagated watermelon stocks. Watermelon cv. Arka Manik seedlings grown in vitro from surface-sterilized seeds with the intact seed coat on MS medium appeared visibly clean largely, but upon subjecting them to tissue-indexing, the segments from the collar or root tissue showed bacterial colony growth on Nutrient Agar (NA) from 72% of such healthy seedlings and the cotyledon and hypocotyl tissue of 44% seedlings. The pooled colony growth from NA upon challenge with 90% alcohol yielded 10 distinct colony types, identified as B. pumilus (4×), B. subtilis (4×), B. cereus (1×) or B. safensis (1×) based on partial 16S rRNA sequence analysis. The shoot-tip tissue from the healthy index-negative seedlings cultured on watermelon proliferation medium partly turned index-positive within 2 - 4 sub-culture cycles while being apparently clean. On the other hand, those from the previously index-positive cultures tended to show obvious bacterial growth during subsequent in vitro culturing. The observations suggested the possibility of introduction of spore-forming Bacillus spp. through surface-sterilized seeds, their gradual emergence in vitro in visibly clean seedlings, possible transmittal of spores to the alcohol through tissue-culturing tools and the survival therein with the chances of unsuspected lateral spread. Seed coat removal followed by surface sterilization with sodium hypochlorite facilitated the raising of clean seedlings with no detectable bacterial association.
文摘Microbes that are beneficial to plants are used to enhance the crop growth,yield and are alternatives to chemical fertilizers.Trichoderma and Bacillus are the predominant plant growth-promoting fungi and bacteria.The objective of this study was select,characterize,and evaluate isolates of Trichoderma spp.and Bacillus spp.native from the northern region of Sinaloa,Mexico,and assess their effect on growth promotion in maize(Zea mays L.).In greenhouse conditions,four Trichoderma isolates and twenty Bacillus isolates,as well as two controls,were tested in a completely randomized design with three replicates.We selected the two best strains of Trichoderma and Bacillus:TB=Trichoderma asperellum,TF=Trichoderma virens,B14=Bacillus cereus sensu lato and B17=Bacillus cereus,which were evaluated in the field in a completely randomized blocks in factorial arrangement design with three replicates applying different rates of nitrogen fertilizer(0,150 kg N/ha,and 300 kg N/ha).Treatments 5(B17=B.cereus)and 11(TF=T.virens)both fertilized with 150 kg N/ha showed similar yields and they did not reveal significant differences from the treatments fertilized with 300 kg N/ha.This indicated that treatment 5(B17=B.cereus with 150 kg N/ha)and treatment 11(TF=T.virens with 150 kg N/ha)were efficient as growth promoters,by not showing significant differences in root volume and dry weight of foliage.The results indicated a reduction of 50% in the rate of nitrogen to fertilizer required for maize(Zea mays L.)crops.These microorganisms Trichoderma and Bacillus could be an alternative to reduce the use of chemical fertilizers in maize.

文摘Considerable research has been conducted into the efficacy of individual probiotics in broiler production,however information on the most effective combinations of synergistic Bacillus probiotic is lacking.This study investigated the impact of different Bacillus strain combinations in broiler chickens,as well as in vitro enzyme production.In experiment one,a total of 576 Ross 308 broilers at 1 d old were grown for 21 d across 6 treatments of maize-soybean diets(n=12 pens per treatment)to compare three different strain combinations(formulation 1[F1]:3 strains Bacillus amyloliquefaciens;F2:Bacillus coagulans and 2 strains B.amyloliquefaciens;F3:B.coagulans,Bacillus licheniformis and 2 strains B.amyloliquefaciens;F5:Bacillus subtilis,B.licheniformis and 2 strains B.amyloliquefaciens),positive control(PC),and a negative control antibiotic treatment group(NC).In Exp.2,a total of 360 one-day-old ROSS308 broilers were used to test five treatments(n=9)including PC,NC,F1 and F5(selected from Exp.1),and F4(Bacillus pumilis and 2 strains B.amyloliquefaciens)in a maize-soybean diet.B.amyloliquefaciens F1 demonstrated a significant improvement in feed conversion ratio(FCR)compared to F2 at d 14(1.49 vs 2.10;P=0.038)and the body weight(BW)at d 21(847.0 g vs 787.4 g)compared to other combinations(P=0.027).The FCR at d 21 tended to be lower in birds fed F1(1.46 vs 1.66)compared to the control(P=0.068).Probiotic treatments had significantly improved nutrient digestibility compared to the PC and NC.Also,probiotic treatments supported the growth of Streptococcus,a common commensal genus and reduced the abundance of genera that correlated with low weight gain such as Akkermansia.Experiment two revealed that F4 improved FCR(P<0.001)and BW at 28 d(P=0.014).In vitro testing showed a high production of protease and amylase by Bacillus.Thus,the addition of Bacillus probiotics,particularly containing B.amyloliquefaciens strains and Bacillus pumilus,into the diet of broiler chickens improves production performance,nutrient digestibility,and allows the proliferation of beneficial gut microbiota.
