In this study, mobile phase conditions suitable for chiral analyses with electrospray ionization LC-MS were systematically investigated using vancomycin as a representative CSP. The performance of a 2.7 μm SPP based vancomycin CSP (SPP-V) 10 cm ×0.21 cm column was compared to that of a corresponding 5 μm fully porous particles based analogue column. The results demonstrated that the SPP-V column provides higher efficiencies, 2-5 time greater sensitivity and shorter analysis time for a set of 22 basic pharma- ceutical drugs. The SPP-V was successfully applied for the analysis of the degradation products of racemic citalopram whose enantiomers could be selectively identified by MS.展开更多
Detection of drugs in hair has become popular in recent years. The significantly long drug detection window (months) in hair has allowed the retrospective investigation and measurement of past consumption of drug. As ...Detection of drugs in hair has become popular in recent years. The significantly long drug detection window (months) in hair has allowed the retrospective investigation and measurement of past consumption of drug. As the majority of drugs are basic, an extraction method was developed based on a methanolic solution for detection of basic/weak basic drugs in hair. It was compared with alkaline digestion (NaOH) followed by LLE. A filtration step with filtration vials was added and their materials were compared. After filtration, extracts were injected directly onto a C18 column coupled to Sciex ABI 2000 MSMS. The mobile phase was 50% methanol, 0.1% formic acid and 2 mM ammonium acetate (isocratic). Both methods were compared by applying them to real samples. Results showed that calibration was linear with r2 of 0.991-0.999 for 20 tested analytes. The matrix effect was assessed to be between 91.4%- 110.2% for 18 analytes. PTFE filter material showed better recoveries over the GMF and PVDF based filters. Stability of analytes during extraction in general was better with methanolic incubation rather than alkaline digestion. With regard to real sample recovery, 6 out of 10 analytes recovered better with alkaline digestion. In conclusion, the methanolic method is capable of extracting most basic drugs in hair samples but only part of the total incorporated drug. Therefore, these results suggest that a combination of both methods (methanolic and alkaline extractions) in hair sample processing for general detection of basic and weak basic drugs may produce better results. However, not all basic drugs are stable under alkaline digestion.展开更多

The average degree of substi-tution (Ds) of HP-g-CD were determined by fast atom bombardment mass spectrometry(FAB-MS). The effects of the Ds of HP-β-CD on CE res0lution of four basic drugs werestudied. The results show that Ds has a different effects on the separation of various drugsbecause of different molecular structures. At D.=0(β-CD), no separation was found for isoproterenol, ofloxacin and propranolol. The resolution of both isoproterenol and ofloxacin in-creased with the increase of Ds, but decreased when Ds was 14. The best reso1uti0n was atDs= 10. 2. The resolution of propranolol increased with increasing Ds. In contrast, the resolution of chlorophiramine decreased with the increase of Ds. The effects of the position ofsubstitution of HP-β-CD on separations were also investigated- The results demonstrate thatthe substitution at the second position of CD plays an important role in the separation ofenantiomers- ln general, the more substitution at the second position of CD, the better theseparation was obtained because the substitution of the second position made the wide open-ing become wider- This probably makes it easier for some molecular to enter the cyclodextrincavity, which c0uld facilitate enantiomeric separation.展开更多
