Blood vessels constitute a closed pipe system distributed throughout the body,transporting blood from the heart to other organs and delivering metabolic waste products back to the lungs and kidneys.Changes in blood ve...Blood vessels constitute a closed pipe system distributed throughout the body,transporting blood from the heart to other organs and delivering metabolic waste products back to the lungs and kidneys.Changes in blood vessels are related to many disorders like stroke,myocardial infarction,aneurysm,and diabetes,which are important causes of death worldwide.Translational research for new appro-aches to disease modeling and effective treatment is needed due to the huge socio-economic burden on healthcare systems.Although mice or rats have been widely used,applying data from animal studies to human-specific vascular physiology and pathology is difficult.The rise of induced pluripotent stem cells(iPSCs)provides a reliable in vitro resource for disease modeling,regenerative medicine,and drug discovery because they carry all human genetic information and have the ability to directionally differentiate into any type of human cells.This review summarizes the latest progress from the establishment of iPSCs,the strategies for differentiating iPSCs into vascular cells,and the in vivo trans-plantation of these vascular derivatives.It also introduces the application of these technologies in disease modeling,drug screening,and regenerative medicine.Additionally,the application of high-tech tools,such as omics analysis and high-throughput sequencing,in this field is reviewed.展开更多
BACKGROUND Type 2 diabetes(T2 D) is characterized by insufficient insulin secretion caused by defective pancreatic β-cell function or insulin resistance,resulting in an increase in blood glucose.However,the mechanism...BACKGROUND Type 2 diabetes(T2 D) is characterized by insufficient insulin secretion caused by defective pancreatic β-cell function or insulin resistance,resulting in an increase in blood glucose.However,the mechanism involved in this lack of insulin secretion is unclear.The level of vascular endothelial growth factor B(VEGF-B) is significantly increased in T2 D patients.The inactivation of VEGF-B could restore insulin sensitivity in db/db mice by reducing fatty acid accumulation.It is speculated that VEGF-B is related to pancreatic β-cell dysfunction and is an important factor affecting β-cell secretion of insulin.As an in vitro model of normal pancreatic β-cells,the MIN6 cell line can be used to analyze the mechanism of insulin secretion and related biological effects.AIM To study the role of VEGF-B in the insulin secretion signaling pathway in MIN6 cells and explore the effect of VEGF-B on blood glucose regulation.METHODS The MIN6 mouse pancreatic islet β-cell line was used as the model system.By administering exogenous VEGF-B protein or knocking down VEGF-B expression in MIN6 cells,we examined the effects of VEGF-B on insulin secretion,Ca2+ and cyclic adenosine monophosphate(cAMP) levels,and the insulin secretion signaling pathway.RESULTS Exogenous VEGF-B inhibited the secretion of insulin and simultaneously reduced the levels of Ca2+ and cAMP in MIN6 cells.Exogenous VEGF-B also reduced the expression of phospholipase C gamma 1(PLCγ1),phosphatidylinositol 3-kinase(PI3 K),serine/threonine kinase(AKT),and other proteins in the insulin secretion pathway.Upon knockdown of VEGF-B,MIN6 cells exhibited increased insulin secretion and Ca2+ and cAMP levels and upregulated expression of PLCγ1,PI3 K,AKT,and other proteins.CONCLUSION VEGF-B can regulate insulin secretion by modulating the levels of Ca2+ and cAMP.VEGF-B involvement in insulin secretion is related to the expression of PLCγ1,PI3 K,AKT,and other signaling proteins.These results provide theoretical support and an experimental basis for the study of VEGF-B in the pathogenesis of T2 D.展开更多
Objective: To investigate effect of aerobic exercise training combined with conventional drug on red blood cell parameters, vascular endothelial function and cardiac function in elderly patients with coronary heart di...Objective: To investigate effect of aerobic exercise training combined with conventional drug on red blood cell parameters, vascular endothelial function and cardiac function in elderly patients with coronary heart disease complicated with chronic heart failure. Methods: A total of 110 elderly patients with coronary heart disease and chronic heart failure according to random data table were divided into control group (n=54) and observation group (n=56). The patients in control group were treated with conventional drug, and patients in the observation group received conventional drug combined with aerobic exercise training. Before and after treatment, levels of red blood cell parameters, vascular endothelial function and cardiac function indexes were compared between the two groups. Results: Before treatment, the difference of HCT, RDW, RBC, NO, ET-1, LVEF, LVEDD and LVESD levels in the two groups were not significantly. After treatment, HCT and RBC levels in two groups were no statistically significant difference;The levels of RDW, ET-1, LVEDD and LVESD in observation group were significantly lower than those in this group before treatment, and significantly lower than those in the control group after treatment;The levels of NO and LVEF in two groups were significantly higher than those in the group before treatment, and levels of NO and LVEF in the observation group were significantly higher than those in control group after treatment. Conclusion: The clinical effect of aerobic exercise training combined with conventional drug in treatment of senile coronary heart disease with chronic heart failure was significant, which can effectively increase the RDW level of patients, improve vascular endothelial function and heart function, it has important clinical value.展开更多

Acupuncture at Conception and Governor vessels als...Mesenchymal stem cell transplantation is a novel means of treating cerebral ischemia/reper- fusion, and can promote angiogenesis and neurological functional recovery. Acupuncture at Conception and Governor vessels also has positive effects as a treatment for cerebral ischemia/ reperfusion. Therefore, we hypothesized that electro-acupuncture at Conception and Governor vessels plus mesenchymal stem cell transplantation may have better therapeutic effects on the promotion of angiogenesis and recovery of neurological function than either treatment alone. In the present study, human umbilical cord blood-derived mesenchymal stem cells were isolated, cultured, identified and intracranially transplanted into the striatum and subcortex of rats at 24 hours following cerebral ischemia/reperfusion. Subsequently, rats were electro-acupunctured at Conception and Governor vessels at 24 hours after transplantation. Modified neurological severity scores and immunohistochemistry findings revealed that the combined interventions of electro-acupuncture and mesenchymal stem cell transplantation clearly improved neurological impairment and up-regulated vascular endothelial growth factor expression around the isch- emic focus. The combined intervention provided a better outcome than mesenchymal stem cell transplantation alone. These findings demonstrate that electro-acupuncture at Conception and Governor vessels and mesenchymal stem cell transplantation have synergetic effects on promot- ing neurological function recovery and angiogenesis in rats after cerebral ischemia/reperfusion.展开更多

METHODS Esophagea...OBJECTIVE To examine the expression of vascular endothelial growth factor C(VEGF-C)in human esophageal squamous cell carcinoma(ESCC), and to clarify its role in lymphatic metastasis in ESCC patients. METHODS Esophageal carcinoma EC9706 cel s and samples from 49 patients with primary ESCC were investigated by using S-P immunohisto- chemistry(IHC),the semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR)and in situ hybridization(ISH)methods for VEGF-C expression. RESULTS VEGF-C positive expression was found in EC9706 cells through IHC,ISH and RT-PCR.Positive IHC for VEGF-C was observed in 36 of 49 cases of ESCC.There was a significant difference between the expres- sion of VEGF-C in a lymph-node-positive group compared to a node-nega- tive group(χ2=4.7,P<0.05).Positive ISH for VEGF-C mRNA was observed in 23 of 49 cases of ESCC.There was a significant difference between the expression of VEGF-C in the lymph-node-positive group and node-negative group(χ2=31.3,P<0.01).The expression of VEGF-C was significantly higher in the lymph-node-positive group compared to the node-negative group.Of 49 ESCC tissues,RT-PCR for VEGF-C mRNA was observed positively in 29 cases.There was a significant difference between the expression of VEGF-C in the lymph-node-positive group and node-negative group(χ2=23.3, P<0.01).The expression of VEGF-C was significantly higher in the lymph- node-positive group compared to the node-negative group.Expressions of VEGF-C were not significantly associated with age,gender,and pathological grade.There was a relationship between VEGF-C mRNA expressions by RT-PCR and ISH(χ2=18.5,P<0.01)in ESCC cases,but with no significant difference between the two methods. CONCLUSION VEGF-C expression may induce lymphangiogenesis in human ESCC.There was a close correlation between VEGF-C expression and lymph node metastasis.VEGF-C can serve as a useful prognostic factor for ESCC patients.展开更多
Hyperglycemia reduces the number of circulating endothelial progenitor cells, accelerates their senescence and impairs their function.However, the relationship between blood glucose levels and endothelial progenitor c...Hyperglycemia reduces the number of circulating endothelial progenitor cells, accelerates their senescence and impairs their function.However, the relationship between blood glucose levels and endothelial progenitor cells in peripheral blood of patients with traumatic brain injury is unclear. In this study, 101 traumatic brain injury patients admitted to the Department of Neurosurgery, Tianjin Medical University General Hospital or the Department of Neurosurgery, Tianjin Huanhu Hospital, China, were enrolled from April 2005 to March 2007. The number of circulating endothelial progenitor cells and blood glucose levels were measured at 1, 4, 7, 14 and 21 days after traumatic brain injury by flow cytometry and automatic biochemical analysis, respectively. The number of circulating endothelial progenitor cells and blood sugar levels in 37 healthy control subjects were also examined. Compared with controls, the number of circulating endothelial progenitor cells in traumatic brain injury patients was decreased at 1 day after injury, and then increased at 4 days after injury,and reached a peak at 7 days after injury. Compared with controls, blood glucose levels in traumatic brain injury patients peaked at 1 day and then decreased until 7 days and then remained stable. At 1, 4, and 7 days after injury, the number of circulating endothelial progenitor cells was negatively correlated with blood sugar levels(r =-0.147, P < 0.05). Our results verify that hyperglycemia in patients with traumatic brain injury is associated with decreased numbers of circulating endothelial progenitor cells. Results showed that 5-bromodeoxyuridine-labeled cells aggregated arou...In the present study, human umbilical cord blood mesenchymal stem cells were injected into a rat model of traumatic brain injury via the tail vein. Results showed that 5-bromodeoxyuridine-labeled cells aggregated around the injury site, surviving up to 4 weeks post-transplantation. In addition, transplantation-related death did not occur, and neurological functions significantly improved. Histological detection revealed attenuated pathological injury in rat brain tissues following human umbilical cord blood mesenchymal stem cell transplantation. In addition, the number of apoptotic cells decreased. Immunohistochemistry and in situ hybridization showed increased expression of brain-derived neurotrophic factor, nerve growth factor, basic fibroblast growth factor, and vascular endothelial growth factor, along with increased microvessel density in surrounding areas of brain injury. Results demonstrated migration of transplanted human umbilical cord blood mesenchymal stem cells into the lesioned boundary zone of rats, as well as increased angiogenesis and expression of related neurotrophic factors in the lesioned boundary zone.展开更多

Inflammatory jaw bone diseases are common in stomatology,including periodontitis,peri-implantitis,medication-related osteonecrosis of the jaw,radiation osteomyelitis of the jaw,age-related osteoporosis,and other speci...Inflammatory jaw bone diseases are common in stomatology,including periodontitis,peri-implantitis,medication-related osteonecrosis of the jaw,radiation osteomyelitis of the jaw,age-related osteoporosis,and other specific infections.These diseases may lead to tooth loss and maxillofacial deformities,severely affecting patients'quality of life.Over the years,the reconstruction of jaw bone deficiency caused by inflammatory diseases has emerged as a medical and socioeconomic challenge.Therefore,exploring the pathogenesis of inflammatory diseases associated with jaw bones is crucial for improving prognosis and developing new targeted therapies.Accumulating evidence indicates that the integrated bone formation and dysfunction arise from complex interactions among a network of multiple cell types,including osteoblast-associated cells,immune cells,blood vessels,and lymphatic vessels.However,the role of these different cells in the inflammatory process and the'rules'with which they interact are still not fully understood.Although many investigations have focused on specific pathological processes and molecular events in inflammatory jaw diseases,few articles offer a perspective of integration.Here,we review the changes and mechanisms of various cell types in inflammatory jaw diseases,with the hope of providing insights to drive future research in this field.展开更多
目的:探究血管内皮生长因子(VEGF)基因修饰大鼠羊膜间充质干细胞对肾病综合征大鼠血液生化指标的影响。方法:选取60只雄性大鼠,随机分为4组,各15例,分别为对照组(健康)、模型组(肾病综合征)、治疗1组(羊膜间充质干细胞悬液)、治疗2组(V...目的:探究血管内皮生长因子(VEGF)基因修饰大鼠羊膜间充质干细胞对肾病综合征大鼠血液生化指标的影响。方法:选取60只雄性大鼠,随机分为4组,各15例,分别为对照组(健康)、模型组(肾病综合征)、治疗1组(羊膜间充质干细胞悬液)、治疗2组(VEGF基因修饰羊膜间充质干细胞悬液)。观察4组肾组织病理形态学及羊膜间充质干细胞存活、分布情况,并比较尿肌酐(SCr)、血尿素(BU)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、24 h尿蛋白定量(24 h UTP)、阻力指数(RI)、舒张期末期流速(EDV)、收缩期峰值流速(PSV)及肾组织VEGF相关表达。结果:对照组大鼠肾组织具有完整清晰结构,模型组大鼠肾组肾小球表现为肿胀,肾小球内存在较多细胞,系膜区明显增宽,系膜细胞及基质中到重度增生;相较于模型组,各治疗组病理改变均较轻,各治疗组病理改变程度均较模型组轻,其中治疗1组病理改变较治疗2组病理改变明显;对照组及模型组大鼠肾组织均未见荧光细胞;治疗1组、治疗2组大鼠肾组织均可见红色荧光的阳性细胞,且仅可见于肾间质及小管内,在肾小球中未见其表达。与治疗1组比较,治疗2组阳性细胞较多;与对照组比较,模型组SCr、BU、24 h UTP、TC、TG、LDL、IL-1β、TNF-α水平均明显升高(P<0.05),治疗1组、治疗2组SCr、BU、24 h UTP、TC、TG、LDL、IL-1β、TNF-α水平均较模型组明显降低,且治疗2组最低(P<0.05);与对照组比较,模型组RI明显升高,PSV、EDV明显下降(P<0.05),治疗1组、治疗2组PSV、EDV均较模型组显著升高,RI水平较模型组显著降低,且治疗2组RI最低,PSV、EDV最高(P<0.05);与对照组比较,模型组肾组织中VEGF蛋白及mRNA表达均明显下降(P<0.05),治疗1组、治疗2组肾组织中VEGF蛋白及mRNA表达均较模型组显著升高,且治疗2组最高(P<0.05)。结论:经VEGF基因修饰的羊膜间充质干细胞移植治疗,可显著改善肾病综合征大鼠血脂水平、微炎症状态、肾功能及肾动脉血流,效果更为显著,可为肾病综合征细胞移植治疗提供有力依据。展开更多
Steadily increasing evidence supports the idea that genetic diversities in the vascular bed are, in addition to hemodynamic influences, a major contributing factor in determining region-specific cardiovascular disease...Steadily increasing evidence supports the idea that genetic diversities in the vascular bed are, in addition to hemodynamic influences, a major contributing factor in determining region-specific cardiovascular disease susceptibility. Members of the phylogenetically highly conserved Hox gene family of developmental regulators have to be viewed as prime candidates for determining these regional genetic differences in the vasculature. During embryonic patterning, the regionally distinct and precisely choreographed expression patterns of HOX transcription factors are essential for the correct specification of positional identities. Apparently, these topographic patterns are to some degree retained in certain adult tissues, including the circulatory system. While an understanding of the functional significance of these localized Hox activities in adult blood vessels is only beginning to emerge, an argument can be made for a role of Hox genes in the maintenance of vessel wall homeostasis and functional integrity on the one hand, and in regulating the development and progression of regionally restricted vascular pathologies, on the other. Initial functional studies in animal models, as well as data from clinical studies provide some level of support for this view. The data suggest that putative genetic regulatory networks of Hox-dependent cardiovascular disease processes include genes of diverse functional categories(extracellular matrix remodeling, transmembrane signaling, cell cycle control, inflammatory response, transcriptional control, etc.), as potential targets in both vascular smooth muscle and endothelial cells, as well as cell populations residing in the adventitia.展开更多
目的探究焦虑抑郁情绪对高血压病患者血压变异性及血管内皮细胞功能的影响。方法入选2015年1月至2018年12月于解放军总医院第七医学中心就诊的高血压病患者155例,根据患者是否伴有焦虑抑郁情绪分为观察组(57例,合并焦虑抑郁情绪)和对照...目的探究焦虑抑郁情绪对高血压病患者血压变异性及血管内皮细胞功能的影响。方法入选2015年1月至2018年12月于解放军总医院第七医学中心就诊的高血压病患者155例,根据患者是否伴有焦虑抑郁情绪分为观察组(57例,合并焦虑抑郁情绪)和对照组(98例,未合并焦虑抑郁情绪)。比较2组24 h动态血压监测(ABPM)参数[24 h平均收缩压(24 h SBP)、24 h平均舒张压(24 h DBP)、白天平均收缩压(dSBP)、白天平均舒张压(dDBP)、夜间平均收缩压(nSBP)、夜间平均舒张压(nDBP)]、血压变异性指标[24 h收缩压变异系数(24 h SBPCV)、24 h舒张压变异系数(24 h DBPCV)、白天收缩压变异系数(dSBPCV)、白天舒张压变异系数(dDBPCV)、夜间收缩压变异系数(nSBPCV)、夜间舒张压变异系数(nDBPCV)]、血压节律类型和血管内皮功能指数(RHI),并评价RHI与ABPM参数和血压变异性指标的相关性。结果观察组ABPM参数24 h SBP、24 h DBP、dSBP、dDBP、nSBP、nDBP均高于对照组,血压异常节律比例高于对照组,血压变异性指标24 h SBPCV、24 h DBPCV、dSBPCV、dDBPCV、nSBPCV、nDBPCV均高于对照组,RHI明显低于对照组[(1.17±0.39)比(2.12±0.18)],2种或多种降压药物联合使用率高于对照组(均P<0.01)。RHI与所有ABPM参数、血压变异性指标均呈负相关(均P<0.05)。结论高血压病合并焦虑抑郁患者血压变异性大,血管内皮功能障碍加重,焦虑抑郁情绪、血压变异性增加可能会造成血管内皮细胞功能损伤加重。展开更多

文摘BACKGROUND Type 2 diabetes(T2 D) is characterized by insufficient insulin secretion caused by defective pancreatic β-cell function or insulin resistance,resulting in an increase in blood glucose.However,the mechanism involved in this lack of insulin secretion is unclear.The level of vascular endothelial growth factor B(VEGF-B) is significantly increased in T2 D patients.The inactivation of VEGF-B could restore insulin sensitivity in db/db mice by reducing fatty acid accumulation.It is speculated that VEGF-B is related to pancreatic β-cell dysfunction and is an important factor affecting β-cell secretion of insulin.As an in vitro model of normal pancreatic β-cells,the MIN6 cell line can be used to analyze the mechanism of insulin secretion and related biological effects.AIM To study the role of VEGF-B in the insulin secretion signaling pathway in MIN6 cells and explore the effect of VEGF-B on blood glucose regulation.METHODS The MIN6 mouse pancreatic islet β-cell line was used as the model system.By administering exogenous VEGF-B protein or knocking down VEGF-B expression in MIN6 cells,we examined the effects of VEGF-B on insulin secretion,Ca2+ and cyclic adenosine monophosphate(cAMP) levels,and the insulin secretion signaling pathway.RESULTS Exogenous VEGF-B inhibited the secretion of insulin and simultaneously reduced the levels of Ca2+ and cAMP in MIN6 cells.Exogenous VEGF-B also reduced the expression of phospholipase C gamma 1(PLCγ1),phosphatidylinositol 3-kinase(PI3 K),serine/threonine kinase(AKT),and other proteins in the insulin secretion pathway.Upon knockdown of VEGF-B,MIN6 cells exhibited increased insulin secretion and Ca2+ and cAMP levels and upregulated expression of PLCγ1,PI3 K,AKT,and other proteins.CONCLUSION VEGF-B can regulate insulin secretion by modulating the levels of Ca2+ and cAMP.VEGF-B involvement in insulin secretion is related to the expression of PLCγ1,PI3 K,AKT,and other signaling proteins.These results provide theoretical support and an experimental basis for the study of VEGF-B in the pathogenesis of T2 D.
文摘Objective: To investigate effect of aerobic exercise training combined with conventional drug on red blood cell parameters, vascular endothelial function and cardiac function in elderly patients with coronary heart disease complicated with chronic heart failure. Methods: A total of 110 elderly patients with coronary heart disease and chronic heart failure according to random data table were divided into control group (n=54) and observation group (n=56). The patients in control group were treated with conventional drug, and patients in the observation group received conventional drug combined with aerobic exercise training. Before and after treatment, levels of red blood cell parameters, vascular endothelial function and cardiac function indexes were compared between the two groups. Results: Before treatment, the difference of HCT, RDW, RBC, NO, ET-1, LVEF, LVEDD and LVESD levels in the two groups were not significantly. After treatment, HCT and RBC levels in two groups were no statistically significant difference;The levels of RDW, ET-1, LVEDD and LVESD in observation group were significantly lower than those in this group before treatment, and significantly lower than those in the control group after treatment;The levels of NO and LVEF in two groups were significantly higher than those in the group before treatment, and levels of NO and LVEF in the observation group were significantly higher than those in control group after treatment. Conclusion: The clinical effect of aerobic exercise training combined with conventional drug in treatment of senile coronary heart disease with chronic heart failure was significant, which can effectively increase the RDW level of patients, improve vascular endothelial function and heart function, it has important clinical value.

文摘Mesenchymal stem cell transplantation is a novel means of treating cerebral ischemia/reper- fusion, and can promote angiogenesis and neurological functional recovery. Acupuncture at Conception and Governor vessels also has positive effects as a treatment for cerebral ischemia/ reperfusion. Therefore, we hypothesized that electro-acupuncture at Conception and Governor vessels plus mesenchymal stem cell transplantation may have better therapeutic effects on the promotion of angiogenesis and recovery of neurological function than either treatment alone. In the present study, human umbilical cord blood-derived mesenchymal stem cells were isolated, cultured, identified and intracranially transplanted into the striatum and subcortex of rats at 24 hours following cerebral ischemia/reperfusion. Subsequently, rats were electro-acupunctured at Conception and Governor vessels at 24 hours after transplantation. Modified neurological severity scores and immunohistochemistry findings revealed that the combined interventions of electro-acupuncture and mesenchymal stem cell transplantation clearly improved neurological impairment and up-regulated vascular endothelial growth factor expression around the isch- emic focus. The combined intervention provided a better outcome than mesenchymal stem cell transplantation alone. These findings demonstrate that electro-acupuncture at Conception and Governor vessels and mesenchymal stem cell transplantation have synergetic effects on promot- ing neurological function recovery and angiogenesis in rats after cerebral ischemia/reperfusion.

文摘OBJECTIVE To examine the expression of vascular endothelial growth factor C(VEGF-C)in human esophageal squamous cell carcinoma(ESCC), and to clarify its role in lymphatic metastasis in ESCC patients. METHODS Esophageal carcinoma EC9706 cel s and samples from 49 patients with primary ESCC were investigated by using S-P immunohisto- chemistry(IHC),the semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR)and in situ hybridization(ISH)methods for VEGF-C expression. RESULTS VEGF-C positive expression was found in EC9706 cells through IHC,ISH and RT-PCR.Positive IHC for VEGF-C was observed in 36 of 49 cases of ESCC.There was a significant difference between the expres- sion of VEGF-C in a lymph-node-positive group compared to a node-nega- tive group(χ2=4.7,P<0.05).Positive ISH for VEGF-C mRNA was observed in 23 of 49 cases of ESCC.There was a significant difference between the expression of VEGF-C in the lymph-node-positive group and node-negative group(χ2=31.3,P<0.01).The expression of VEGF-C was significantly higher in the lymph-node-positive group compared to the node-negative group.Of 49 ESCC tissues,RT-PCR for VEGF-C mRNA was observed positively in 29 cases.There was a significant difference between the expression of VEGF-C in the lymph-node-positive group and node-negative group(χ2=23.3, P<0.01).The expression of VEGF-C was significantly higher in the lymph- node-positive group compared to the node-negative group.Expressions of VEGF-C were not significantly associated with age,gender,and pathological grade.There was a relationship between VEGF-C mRNA expressions by RT-PCR and ISH(χ2=18.5,P<0.01)in ESCC cases,but with no significant difference between the two methods. CONCLUSION VEGF-C expression may induce lymphangiogenesis in human ESCC.There was a close correlation between VEGF-C expression and lymph node metastasis.VEGF-C can serve as a useful prognostic factor for ESCC patients.

文摘Hyperglycemia reduces the number of circulating endothelial progenitor cells, accelerates their senescence and impairs their function.However, the relationship between blood glucose levels and endothelial progenitor cells in peripheral blood of patients with traumatic brain injury is unclear. In this study, 101 traumatic brain injury patients admitted to the Department of Neurosurgery, Tianjin Medical University General Hospital or the Department of Neurosurgery, Tianjin Huanhu Hospital, China, were enrolled from April 2005 to March 2007. The number of circulating endothelial progenitor cells and blood glucose levels were measured at 1, 4, 7, 14 and 21 days after traumatic brain injury by flow cytometry and automatic biochemical analysis, respectively. The number of circulating endothelial progenitor cells and blood sugar levels in 37 healthy control subjects were also examined. Compared with controls, the number of circulating endothelial progenitor cells in traumatic brain injury patients was decreased at 1 day after injury, and then increased at 4 days after injury,and reached a peak at 7 days after injury. Compared with controls, blood glucose levels in traumatic brain injury patients peaked at 1 day and then decreased until 7 days and then remained stable. At 1, 4, and 7 days after injury, the number of circulating endothelial progenitor cells was negatively correlated with blood sugar levels(r =-0.147, P < 0.05). Our results verify that hyperglycemia in patients with traumatic brain injury is associated with decreased numbers of circulating endothelial progenitor cells. 文摘In the present study, human umbilical cord blood mesenchymal stem cells were injected into a rat model of traumatic brain injury via the tail vein. Results showed that 5-bromodeoxyuridine-labeled cells aggregated around the injury site, surviving up to 4 weeks post-transplantation. In addition, transplantation-related death did not occur, and neurological functions significantly improved. Histological detection revealed attenuated pathological injury in rat brain tissues following human umbilical cord blood mesenchymal stem cell transplantation. In addition, the number of apoptotic cells decreased. Immunohistochemistry and in situ hybridization showed increased expression of brain-derived neurotrophic factor, nerve growth factor, basic fibroblast growth factor, and vascular endothelial growth factor, along with increased microvessel density in surrounding areas of brain injury. Results demonstrated migration of transplanted human umbilical cord blood mesenchymal stem cells into the lesioned boundary zone of rats, as well as increased angiogenesis and expression of related neurotrophic factors in the lesioned boundary zone.
文摘Purpose:To investigate the spatial and temporal regulation effect of vascular endothelial growth factor(VEGF)on human fetal choroid vascularization.Methods:The eyeballs of 54human fetuses from the 9th week to the40th week due to accidental abortion were studied by immunohistochemically staining for the expression of VEGF and proliferation cell nuclear antigen(PCNA).Results:(1)The distribution of VEGF expression in the retinal pigment epithelium(RPE)decreased with the increase of age,the peak of which was between the 9th and 14th week,(2)PCNAimmunoreactivity was localized within choriocapillaris endothe-lium,THe expression level decreased alone with fetus age,In this period the chori-ocapillaris endothelium kept proliferation,differentiation.canalization and remodelled to form the choroid vessels.(3)Statistically significant correlations were shown between the expression of VEGF in the PRE and that of PCNAin choriocapillaris endothelium(r=0.933,P<0.01).Conclusion:VEGF expression in PRE was positively involved in modulating human fetal choroid vascularization.Eye Science2000;16:11-14.

文摘Inflammatory jaw bone diseases are common in stomatology,including periodontitis,peri-implantitis,medication-related osteonecrosis of the jaw,radiation osteomyelitis of the jaw,age-related osteoporosis,and other specific infections.These diseases may lead to tooth loss and maxillofacial deformities,severely affecting patients'quality of life.Over the years,the reconstruction of jaw bone deficiency caused by inflammatory diseases has emerged as a medical and socioeconomic challenge.Therefore,exploring the pathogenesis of inflammatory diseases associated with jaw bones is crucial for improving prognosis and developing new targeted therapies.Accumulating evidence indicates that the integrated bone formation and dysfunction arise from complex interactions among a network of multiple cell types,including osteoblast-associated cells,immune cells,blood vessels,and lymphatic vessels.However,the role of these different cells in the inflammatory process and the'rules'with which they interact are still not fully understood.Although many investigations have focused on specific pathological processes and molecular events in inflammatory jaw diseases,few articles offer a perspective of integration.Here,we review the changes and mechanisms of various cell types in inflammatory jaw diseases,with the hope of providing insights to drive future research in this field.
文摘目的:探究血管内皮生长因子(VEGF)基因修饰大鼠羊膜间充质干细胞对肾病综合征大鼠血液生化指标的影响。方法:选取60只雄性大鼠,随机分为4组,各15例,分别为对照组(健康)、模型组(肾病综合征)、治疗1组(羊膜间充质干细胞悬液)、治疗2组(VEGF基因修饰羊膜间充质干细胞悬液)。观察4组肾组织病理形态学及羊膜间充质干细胞存活、分布情况,并比较尿肌酐(SCr)、血尿素(BU)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、24 h尿蛋白定量(24 h UTP)、阻力指数(RI)、舒张期末期流速(EDV)、收缩期峰值流速(PSV)及肾组织VEGF相关表达。结果:对照组大鼠肾组织具有完整清晰结构,模型组大鼠肾组肾小球表现为肿胀,肾小球内存在较多细胞,系膜区明显增宽,系膜细胞及基质中到重度增生;相较于模型组,各治疗组病理改变均较轻,各治疗组病理改变程度均较模型组轻,其中治疗1组病理改变较治疗2组病理改变明显;对照组及模型组大鼠肾组织均未见荧光细胞;治疗1组、治疗2组大鼠肾组织均可见红色荧光的阳性细胞,且仅可见于肾间质及小管内,在肾小球中未见其表达。与治疗1组比较,治疗2组阳性细胞较多;与对照组比较,模型组SCr、BU、24 h UTP、TC、TG、LDL、IL-1β、TNF-α水平均明显升高(P<0.05),治疗1组、治疗2组SCr、BU、24 h UTP、TC、TG、LDL、IL-1β、TNF-α水平均较模型组明显降低,且治疗2组最低(P<0.05);与对照组比较,模型组RI明显升高,PSV、EDV明显下降(P<0.05),治疗1组、治疗2组PSV、EDV均较模型组显著升高,RI水平较模型组显著降低,且治疗2组RI最低,PSV、EDV最高(P<0.05);与对照组比较,模型组肾组织中VEGF蛋白及mRNA表达均明显下降(P<0.05),治疗1组、治疗2组肾组织中VEGF蛋白及mRNA表达均较模型组显著升高,且治疗2组最高(P<0.05)。结论:经VEGF基因修饰的羊膜间充质干细胞移植治疗,可显著改善肾病综合征大鼠血脂水平、微炎症状态、肾功能及肾动脉血流,效果更为显著,可为肾病综合征细胞移植治疗提供有力依据。

文摘Steadily increasing evidence supports the idea that genetic diversities in the vascular bed are, in addition to hemodynamic influences, a major contributing factor in determining region-specific cardiovascular disease susceptibility. Members of the phylogenetically highly conserved Hox gene family of developmental regulators have to be viewed as prime candidates for determining these regional genetic differences in the vasculature. During embryonic patterning, the regionally distinct and precisely choreographed expression patterns of HOX transcription factors are essential for the correct specification of positional identities. Apparently, these topographic patterns are to some degree retained in certain adult tissues, including the circulatory system. While an understanding of the functional significance of these localized Hox activities in adult blood vessels is only beginning to emerge, an argument can be made for a role of Hox genes in the maintenance of vessel wall homeostasis and functional integrity on the one hand, and in regulating the development and progression of regionally restricted vascular pathologies, on the other. Initial functional studies in animal models, as well as data from clinical studies provide some level of support for this view. The data suggest that putative genetic regulatory networks of Hox-dependent cardiovascular disease processes include genes of diverse functional categories(extracellular matrix remodeling, transmembrane signaling, cell cycle control, inflammatory response, transcriptional control, etc.), as potential targets in both vascular smooth muscle and endothelial cells, as well as cell populations residing in the adventitia.
文摘目的探究焦虑抑郁情绪对高血压病患者血压变异性及血管内皮细胞功能的影响。方法入选2015年1月至2018年12月于解放军总医院第七医学中心就诊的高血压病患者155例,根据患者是否伴有焦虑抑郁情绪分为观察组(57例,合并焦虑抑郁情绪)和对照组(98例,未合并焦虑抑郁情绪)。比较2组24 h动态血压监测(ABPM)参数[24 h平均收缩压(24 h SBP)、24 h平均舒张压(24 h DBP)、白天平均收缩压(dSBP)、白天平均舒张压(dDBP)、夜间平均收缩压(nSBP)、夜间平均舒张压(nDBP)]、血压变异性指标[24 h收缩压变异系数(24 h SBPCV)、24 h舒张压变异系数(24 h DBPCV)、白天收缩压变异系数(dSBPCV)、白天舒张压变异系数(dDBPCV)、夜间收缩压变异系数(nSBPCV)、夜间舒张压变异系数(nDBPCV)]、血压节律类型和血管内皮功能指数(RHI),并评价RHI与ABPM参数和血压变异性指标的相关性。结果观察组ABPM参数24 h SBP、24 h DBP、dSBP、dDBP、nSBP、nDBP均高于对照组,血压异常节律比例高于对照组,血压变异性指标24 h SBPCV、24 h DBPCV、dSBPCV、dDBPCV、nSBPCV、nDBPCV均高于对照组,RHI明显低于对照组[(1.17±0.39)比(2.12±0.18)],2种或多种降压药物联合使用率高于对照组(均P<0.01)。RHI与所有ABPM参数、血压变异性指标均呈负相关(均P<0.05)。结论高血压病合并焦虑抑郁患者血压变异性大,血管内皮功能障碍加重,焦虑抑郁情绪、血压变异性增加可能会造成血管内皮细胞功能损伤加重。
