The structure, surface static electricity and hydrophobicity of scFv were investigated. Computer graphic modelling indicated that all regions of scFv including the linker, variable regions of the heavy (VH) and light (VL) chains were suitable. The VH region and the VL region were involved in composing the 'hydrophobic pocket'. The linker was drifted away VH and VL regions. The complementarity determining regions (CDRs) of VH and VL regions surrounded the 'hydrophobic pocket'. This study provides a theory basis for improving antibody affinity, investigating antibody structure and analyzing the functions of VH and VL regions in antibody activity.展开更多

The nonlinear consti- tutive relations of the ME material are taken into account, and the influences of the nonlinear material properties on the ME effect are investigated for the static and dynamic cases, respectively. The influences of different constraint conditions on the ME effect are discussed. In the dynamic case considering nonlinear material properties, the double frequency ME response （The response frequency is twice the applied magnetic frequency） is obtained and discussed, which can be used to explain the experiment phenomenon in which the input signal with frequency f is converted to the output signal with 2f in ME laminated structures. Some calculations on nonlinear ME effect are conducted. The obtained results indicate that the nonlinear material properties affect not only the magnitude of the ME effect in the static case but also the ME response frequency in the dynamic case展开更多
One of the important issues in the system identification and the spectrum analysis is the frequency resolution, i.e., the capability of distinguishing between two or more closely spaced frequency components. In the mo...One of the important issues in the system identification and the spectrum analysis is the frequency resolution, i.e., the capability of distinguishing between two or more closely spaced frequency components. In the modal identification by the empirical mode decomposition （EMD） method, because of the separating capability of the method, it is still a challenge to consistently and reliably identify the parameters of structures of which modes are not well separated. A new method is introduced to generate the intrin- sic mode functions （IMFs） through the filtering algorithm based on the wavelet packet decomposition （GIFWPD）. In this paper, it is demonstrated that the CIFWPD method alone has a good capability of separating close modes, even under the severe condition beyond the critical frequency ratio limit which makes it impossible to separate two closely spaced harmonics by the EMD method. However, the GIFWPD-only based method is impelled to use a very fine sampling frequency with consequent prohibitive computational costs. Therefore, in order to decrease the computational load by reducing the amount of samples and improve the effectiveness of separation by increasing the frequency ratio, the present paper uses a combination of the complex envelope displacement analysis （CEDA） and the GIFWPD method. For the validation, two examples from the previous works are taken to show the results obtained by the GIFWPD-only based method and by combining the CEDA with the GIFWPD method.展开更多
Epidemiological evidence points to increased dairy and meat consumption,staples of the Western diet,as major risk factors for the development of type 2 diabetes(T2D).This paper presents a new concept and comprehensive...Epidemiological evidence points to increased dairy and meat consumption,staples of the Western diet,as major risk factors for the development of type 2 diabetes(T2D).This paper presents a new concept and comprehensive review of leucine-mediated cell signaling explaining the pathogenesis of T2D and obesity by leucine-induced over-stimulation of mammalian target of rapamycin complex 1(mTORC1).mTORC1,a pivotal nutrient-sensitive kinase,promotes growth and cell proliferation in response to glucose,energy,growth factors and amino acids.Dairy proteins and meat stimulate insulin/insulin-like growth factor 1 signaling and provide high amounts of leucine,a primary and independent stimulator for mTORC1 activation.The downstream target of mTORC1,the kinase S6K1,induces insulin resistance by phosphorylation of insulin receptor substrate-1,thereby increasing the metabolic burden of β-cells.Moreover,leucine-mediated mTORC1-S6K1-signaling plays an important role in adipogenesis,thus increasing the risk of obesity-mediated insulin resistance. High consumption of leucine-rich proteins explains exaggerated mTORC1-dependent insulin secretion, increased β-cell growth and β-cell proliferation promoting an early onset of replicative β-cell senescence with subsequent β-cell apoptosis.Disturbances of β-cell mass regulation with increased β-cell proliferation and apoptosis as well as insulin resistance are hallmarks of T2D,which are all associated with hyperactivation of mTORC1.In contrast,the anti-diabetic drug metformin antagonizes leucine-mediated mTORC1 signaling.Plant-derived polyphenols and flavonoids are identified as natural inhibitors of mTORC1 and exert anti-diabetic and anti-obesity effects.Furthermore,bariatric surgery in obesity reduces increased plasma levels of leucine and other branched-chain amino acids.Attenuation of leucine-mediated mTORC1 signaling by defining appropriate upper limits of the daily intake of leucine-rich animal and dairy proteins may offer a great chance for the prevention of T2D and obesity,as well as other epidemic diseases of civilization with increased mTORC1 signaling,especially cancer and neurodegenerative diseases,which are frequently associated with T2D.展开更多

The average hardness and elastic...Fe40Ni40P14B6 bulk metallic glass rods have been prepared by water quenching the fluxed alloy. The deformation behavior was investigated by nanoindentation tests and compressing tests. The average hardness and elastic modulus of the as-prepared Fe40Ni40P14B6 BMG (bulk metallic glass) measured by nanoindentation tests are 8.347 and 176.61 GPa respectively. The displace- ment-load curve shows “pop-in” characteristics which correspond to the loading rate bursts. Many shear bands around the indent were observed. The as-prepared Fe-based BMG exhibits a compressive plastic strain of 5.21%, which is much larger than that of other Fe-based glassy alloys and most of other BMGs.展开更多

Numerical solutions are obtained in an anisotropic sphere, whose material is transversely isotropic in the radial direction. It is shown that the bifur- cation may occur either to the right or to the left, depending on the degree of material anisotropy. The deformation and stress contribution in the sphere before cavitation are di?erent from those after cavitation. The stability of solutions is discussed through a comparison of energy.展开更多

A suitable flexible functional form for the radial distribution function G（R） is assumed for the mixture, with R as a variable. The function G（R） has an arbitrary parameter m and a different equation of state can be obtained with a suitable choice of m. For m = 0.75 and m = 0.83 results are close to molecular dynamics （MD） result for pure HS and LJ fluid respectively.展开更多

Methods: Based on the principal component analysis of element contents in Chinese herbs, the effects of 25 chemical elements in the 11 kinds of Chinese herbs in the caine kidney mixture have been analyzed. Results: The traditional Chinese medicines of rehmannia, astragalus, mulberry, salvia miltiorrhiza, rhubarb in the caine kidney mixture play a major role in the treatment of chronic renal failure. Conclusion: The principal component analysis is very practical in the compatibility of medicines and concentrated prescriptions.展开更多
