Natural killer (NK) cell is a type of immune cell and is known to be particularly responsible for innate immunity such as anti-cancer immunity, defense mechanisms against infections, and secretion of various cytokines and chemokines for increasing recruitment of other immune cells. In this study, we investigated the potentials of NK-enriched lymphocytes (NKEL) conditioned media (CM) on skin care for cosmeceutical compositions. Various cytokines of NKEL CM can improve wound healing through epithelial-mesenchymal transition (EMT) by increasing KLKs (kallikreins) and reduce metalloproteinase (MMP)-1 and MMP-2 to inhibit wrinkle formation. Our results suggest that NKEL CM which has various cytokines promotes up-regulation of cell migration and KLKs and down-regulation of MMP-1 and MMP-2 by stimulating HaCaT keratinocytes migration. Therefore, NKEL CM can be used as a cosmetic composition that can play a role in skin regeneration and anti-aging. 展开更多

Background and Aims: Androgenetic alopecia (AGA) is a common form of hair loss in both men and women. Despite its high prevalence and associated patient morbidity, the approved therapeutic options are limited to finasteride and minoxidil. The present study is aimed at assessing the efficacy of hair serum formulation, Trichosera<sup>®</sup>containing Bone marrow-derived mesenchymal stromal cells conditioned media as an active ingredient, for hair fall control and hair regrowth in healthy Indian human volunteers. Methods: The product was made using a 20% concentration of 10X Conditioned Media along with excipients. The final product was tested for physicochemical parameters, biomarkers, total protein content and microbial limits as per our in-house specifications. Results: The primary irritation patch test showed that the product is non-irritant and dermatologically safe. A clinical study on 40 subjects was conducted to evaluate the effectiveness of the bioactive formulation in hair fall control and hair regrowth in healthy volunteers. Phototrichogram measurement showed hair density and hair growth rate increased significantly by 11.54% and 18.66% at week 24. Hair tensile strength also increased significantly by 41.10% at 12 weeks follow-up. Hair pull test, to see a reduction in pulled hair and comb’s test to show a decrease in hair fall significantly improved from week 4 onwards. There were no significant adverse events in response to the product application. Conclusion: It is concluded that the hair serum product is completely safe on direct application to the scalp and showed significant improvement in the hair growth rate, hair density, scalp condition and reduction in hair fall. . 展开更多

Statistic analyses of sectional focal depth frequency relation for 1739 M S≥4.0 events in North China, 541 M S≥4.0 events in Xingtai seismic region and 999 M S≥4.0 events in Tangshan seismic region have been carried out for the first step of the study. Then the sectional focal depth frequency diagrams (H N diagram) for each region were obtained. The study shows that: (1) The intraplate earthquakes in North China occur within the continental crust, and are mainly concentrated within a certain depth range of the middle and upper crust, making up a seismic source layer; (2) The seismic source layer consists of a main layer and an auxiliary layer adjacent to it. The tops and bottoms of the main and auxiliary layers correspond to the G and C, as well as to B and G geologic boundaries respectively; (3) The H N diagrams for Xingtai and Tangshan seismic regions are significantly different from each other, indicating the regionality and localization of the seismic source layers; (4) DSS and MTS observations show that the regionality and localization of the seismic source layer depend mainly on the environment of the deep structures of the region; (5) The seismic source layer is a substance of medium layer, which can be related to the brittle ductile transition zone within the crust. The seismic source layer is also a stress conducting layer. As the media above and below this layer are subjected to failure or flowage, then the stress will concentrate in the stress conducting layer, in which the earthquake occurs. 展开更多

Experimental Investigation on Media Condition of Leaching Scandium from Silt of Yangtze

The minerals of the silt of the end of Sanxia reservoir of Yangtze River were studied. Through the spectral analysis of the samples, the analysis of chemistry components and the content mensuration of main minerals, chemistry components and composition of the main minerals were tested. The results show that: (1) The silt of Sanxia of the Yangtze in Chongqing is mainly composed of silicolites and quartz, containing Fe and Ti in small quantity and Yt, Yb and Sc in minim quantity. The content of Yt, Yb and Sc is 25.8, 2.06, and 6.60 g·t -1 respectively. (2) Thereinto, the content of Sc is 101 g·t -1 in ilmenite and 121 g·t -1 in titanium-picrite. On the basis of these results, Sc leaching conditions and Sc leaching media are investigated by leaching media of HCl, H_2SO_4 and HNO_3. The results show that HCl can be used as the leaching media and its best leaching conditions are as follows: The leaching concentration is 2280%; the leaching ration on liquid-solid is 2∶1; the leaching temperature is 90 ℃; the leaching time is 10 h and the leaching granularity of the test sample is -0.040 mm. The best condition of HCl adding assiting solvent is that assisting solvent is No.7 and its dosage is 40 g·t -1, which leaching ratio is 81.31%. 展开更多

Fungal strain PA-2 was isolated from infected poplar leaves from the Ping’an District, Haidong City of Qinghai Province, China. Based on the culture characteristics and the internal transcribed spacer(ITS) sequence of its 16S r DNA, the strain was identified as Aureobasidium pullulans. The culture and metabolites of strain PA-2 showed high herbicidal potential to five tested weeds Galium aparine var. tenerum, Chenopodium album, Malva crispa, Polygonum lapathifolium and Avena fatua. For the in vitro test, 5 days after the detached leaves were inoculated with PA-2 culture, all leaves infected by the hyphae and became black and rotten. For the in vivo test, a metabolite filtrate of PA-2 culture was sprayed over the living weed plants, and five days after inoculation, the weed plants became withered and necrotic. Seven days after inoculation, the fresh weight reductions of G. aparine var. tenerum, C. album, M. crispa, P. lapathifolium and A. fatua were 87.25, 78.46, 82.25, 62.11, and 80.27%, respectively. Galium aparine var. tenerum and M. crispa exhibited significant reductions in fresh weight. The bio-safety test on the five crops showed no significant plant height reductions, which was also observed for wheat(Triticum aestivum), faba bean(Vicia faba), and barley(Hordeum vulgare). By contrast, oilseed rape(Brassica napus) and pea(Pisum sativum) exhibited light spots but no significant reductions in plant height. These results indicated that A. pullulans could be a potential microbial herbicide for the control of the target weeds in crops. Optimization of the carbon and nitrogen sources for cultural media and substances for solid-state fermentation indicated that PA-2 had better colony growth and spore production with the optimal carbon source of glucose(C4), nitrogen source of soybean flour(N2), and the optimal substance was wheat bran. The results in this study provide useful information for the development ofA. pulluans PA-2 as an herbicide for bio-control of the weed. 展开更多

BACKGROUND: The supernatant of interferon-gamma (IFNγ) co-cultured with neonatal rat cortical glia can promote the cells in embryonic basal forebrain/septal nuclei to differentiate into cholinergic neurons, but the mechanism is still unclear. OBJECTIVE: To analyze the pathways for IFNγ to promote the differentiation of primarily cultured cholinergic neurons in rat embryonic basal forebrain/septal nuclei through culture in different conditioned medium. DESIGN: A controlled experiment taking cells as the observational target. SETTINGS: Department of Biochemistry and Molecular Biology, Youjiang Medical College for Nationalities; Department of Cell Biology, Beijing University Health Science Center. MATERIALS: Sixty-four pregnant Wistar rats for 16 days (250-350 g) and 84 Wistar rats (either male or female, 5-7 g) of 0-1 day after birth were provided by the experimental animal department of Beijing University Health Science Center. Rat IFNγ were provided by Gibco Company; Glial fibrillary acidic protein by Huamei Company. METHODS: The experiments were carried out in the Department of Cell Biology, Beijing University Health Science Center and Daheng Image Company of Chinese Academy of Science from July 1995 to December 2002. ① Interventions: The nerve cells in the basal forebrain/septal nuclei of the pregnant Wistar rats for 16 days were primarily cultured, and then divided into four groups: Blank control group (not any supernatant and medium was added); Control group (added by mixed glial cell or astrocyte conditioned medium); IFNγ group (added by mixed glial cell or astrocyte conditioned medium+IFNγ). Antibody group (added by mixed glial cell or astrocyte conditioned medium+IFNγ+Ab-IFNγ). Mixed glial cell or astrocyte conditioned medium was prepared using cerebral cortex of Wistar rats of 0-1 day after birth. ② Evaluation: The immunohistochemical method was used to perform the choline acetyltransferase (ChAT) staining of cholinergic neurons. The ChAT positive cells were counted. MAIN OUTCOME MEASURES: Comparison of ChAT positive cells in rat basal forebrain and septal nuclei in different conditioned medium. RESULTS: ① ChAT positive cells in mixed glial cell conditioned medium: The ChAT positive cells in the IFNγ group and antibody group were significantly more than those in the control group (P 〈 0.01). ② ChAT positive cells in astrocyte conditioned medium: The ChAT positive cells in the IFNγ group were significantly more than those in the control group, but there was no significant difference between the antibody group and control group (P 〉 0.05). CONCLUSION: IFNγ cannot directly promote the differentiation of cholinergic neurons, but plays a role through activating glial cells (except astrocytes) to produce IFNγ like molecules. 展开更多

Tissue regeneration requires exogenous and endogenous signals,and there is increasing evidence that the exogenous microenvironment may play an even more dominant role in the complex process of coordinated multiple cells.The short-distance peripheral nerve showed a spontaneous regenerative phenomenon,which was initiated by the guiding role of macrophages.However,it cannot sufficiently restore long-distance nerve injury by itself.Based on this principle,we firstly constructed a proinflammatory model to prove that abnormal M2 expression reduce the guidance and repair effect of long-distance nerves.Furthermore,a bionic peptide hydrogel scaffold based on self-assembly was developed to envelop M2-derived regenerative cytokines and extracellular vesicles(EVs).The cytokines and EVs were quantified to mimic the guidance and regenerative microenvironment in a direct and mild manner.The bionic scaffold promoted M2 transformation in situ and led to proliferation and migration of Schwann cells,neuron growth and motor function recovery.Meanwhile,the peptide scaffold combined with CX3CL1 recruited more blood-derived M2 macrophages to promote long-distance nerve reconstruction.Overall,we systematically confirmed the important role of M2 in regulating and restoring the injury peripheral nerve.This bionic peptide hydrogel scaffold mimicked and remodeled the local environment for M2 transformation and recruitment,favoring long-distance peripheral nerve regeneration.It can help to explicate regulative effect of M2 may be a cause not just a consequence in nerve repair and tissue integration,which facilitating the development of pro-regenerative biomaterials. 展开更多
