In this pilot study, we conducted a transcriptomic analysis of a resistant (R) and susceptible (S) variety of alfalfa, with (+A) or without (-A) aphids (totally four treatments). We used the resistant cultivar Zhongmu t and the susceptible cultivar Soca. A total of 3 549 mRNAs were differentially expressed, of which 1 738 up-regulated and 1 307 down-regulated genes were identified in S+A/S-A plants, while 543 up-regulated and 331 down-regulated genes were identified in the R+A/R-A plants. KEGG analysis mapped 112 and 546 differentially expressed genes to 8 and 17 substantially enriched pathways for Zhongmu 1 and Soca, respectively. Six shared pathways were linked to plant resistance traits, including phenylpropanoid biosynthesis associated with salicylic acid synthesis, and linoleic acid metabolism associated with both jasmonic acid and flavonoid biosynthesis. Ultimately, we proposed a preliminary regulatory mechanism of alfalfa cultivar resistance response to aphids feeding based on transcriptome analyses and published documents.展开更多

AQR is based on the correlation between aphid populations and their host plants and is an important tool that has been utilized in evaluating Medicago sativa(alfalfa) cultivar resistance to aphids. However, assessment of alfalfa resistance to aphids can be confused by the presence of aphid predators, causing the assessment of plant resistance to aphids to be based on incorrect aphid population data. To refine the AQR and account for the effect of predators on aphid population assessments, we introduced a parameter ‘α', corresponding to the predator quantity ratio, and used αAQR as the ratio to quantify aphid populations. Populations of both aphids(4 species) and their predators(12 species) occurring in 28 M. sativa cultivars were sampled over two years at a research station near Cangzhou, Hebei Province, China. Results showed that the most suitable evaluation period was from May to June, as the aphid population was stable during this period. Compared with the AQR method, the predator population numbers based on the αAQR had a significant inverse relationship with aphid population numbers and the 28 cultivars were clustered into three classes: the resistant class, tolerant class, and susceptible class. In addition, 17 cultivars were reassigned when evaluated using αAQR. All numerical values calculated by αAQR were displayed as a Gaussian distribution, which showed that the 28 cultivars could be clustered into nine groups using a median value(±SE) of 1±0.1. Hence, ongoing alfalfa breeding trials will be assessed using the αAQR to establish a robust system that includes agronomic performance parameters in order to generalize the new method for further studies.展开更多

Unfortunately, it is inaccurate, and the results appear significantly inconsistent when analysing the same cultivars in the same field study. To improve this model, we first studied the resistance of 28 alfalfa cultivars to thrips in Cangzhou, Hebei Province, north China. The results showed that the most suitable evaluation period was from May to June, as the thrips population was stable during this period. Second, we found that the natural enemy population was significantly positively correlated with the thrips population density(R=0.7275, P〈0.0001), which might influence resistance estimation. Hence, we introduced a parameter ‘α', corresponding to the natural enemy quantity ratio, to eliminate the effect of the natural enemy using "αTQR". Using the improved method, 28 cultivars were clustered into three classes: the resistant class, sensitive class, and median class. All numerical values were calculated for αTQR displayed as a Gaussian distribution. This information showed that all data should be divided into nine groups using a median value of 1±0.1 with an equal difference of 0.1. Defence response in olive fruits injured both by pathogens and by mechanical damages has been associated with the enzyme β-glucosidase, which specifically hydrolyses oleuropein, producing highly reactive aldehyde molecules. In situ detection of ^-glucosidase activity in olive fruit tissues following injury, which simulates Bactrocera oleae punctures, is reported. The assay was performed in two cultivars showing different degrees of susceptibilities to fly infestation. In both cultivars, the histochemical assay for β-glucosidase showed that within 20 min after the injury, a strong ^-glucosidase activity could be observed in the damaged tissues. Thereafter a progressive enzyme inactivation occurred starting from tissues around the boundary of the injury with decrease of the enzyme activity and stopped after 3 h. Whereas the mass of active cells reached a distance of (300±50) μm from the edge of the injury. Biochemical analyses showed that in extracts of the injured fruit, β-glucosidase activity rapidly increased within 20 min from injury, thereafter decreasing and reaching values comparable with those in intact fruits. Following puncture, the oleuropein contents did not change significantly in the high susceptibility cultivar, whereas it rapidly decreased in the cultivar showing low susceptibility. The results strongly suggest that olive fruits susceptible towards fly infestation could be related to the ability of the oleuropein-degrading-β-glucosidase to produce the highly reactive molecules in the damaged tissues. As a consequence of puncture, high level of peroxidase activity was detected. This feature also suggested that this enzyme could play a key role in the defence response against insect injuries.展开更多
Lodging in maize is one of the major problems in maize production worldwide,which causes serious yield and economic losses annually.By evaluating cultivar lodging resistance performance in target growing environments ...Lodging in maize is one of the major problems in maize production worldwide,which causes serious yield and economic losses annually.By evaluating cultivar lodging resistance performance in target growing environments before cultivar extension and application,the risks and losses can be significantly reduced.In this study,a GIS-based quantitative method for evaluating maize cultivar lodging resistance performance in target growing environments was established based on full cognition of environment stress,cultivar resistance,and the interaction between them.At first,comprehensive environment lodging stress is measured by three factors:1)extreme wind event in maize vegetative stage which is the direct factor,2)soil potassium content in target growing environment which is an indirect factor affecting corn stem sturdiness,and 3)planting density which is a human influence factor.Quantification methods of extreme probability analysis,spatial interpolation,normalization,and so on were used.Then,maize cultivar lodging resistance was determined using cumulative frequency distribution analysis of tested lodging data.At last,an evaluation matrix was established combining environment lodging stress and cultivar lodging resistance together,which was very simple and easy to understand method and the result is promising providing good direct support in practical cultivar application.The method used in this study,at county-level,cultivar-level and stress-level with GIS,can facilitate the identification of better-adapted growing environments for a specific maize cultivar,and provide direct support for maize cultivar recommendation and extension,so as to reduce the risk and loss of lodging in maize.It is more easy-operational and feasible than traditional surveying approach,especially for large-scale spatial trend analysis.So it is of both academic significance in accelerating precision agriculture development and practical significance in improving maize cultivar application.展开更多

Nodal cuttings of the tolerant cultivar, Kennebec, and the sens...To understand the response of potato to salt stress, antioxidant enzyme activities and ion content were analyzed for a sensitive and a tolerant cultivar. Nodal cuttings of the tolerant cultivar, Kennebec, and the sensitive cultivar, Concord, were exposed to media without or with 30, 60, 90 or 120 mmol/L NaCI for 4 weeks. On exposure to NaCI, the length and fresh and dry weight of both shoots and roots of Concord showed greater decrease than those of Kennebec. The decrease in shoot growth was more severe than that of the root for both cultivars. The K^+ content of shoots and roots of both cultivars was reduced in a dose-dependent manner by exposure to NaCl; the Na^+ content increased. Activities of ascorbate peroxidase, catalase and glutathione reductase were increased in NaCl-exposed shoots of Kennebec; the corresponding activities in NaCI-exposed shoots of Concord were decreased. Roots of both cultivars showed similar changes in the activities of these enzymes on exposure to NaCI. These studies established that enzyme activities in Concord shoots are inversely related to the NaCI concentration, whereas those in Kennebec do not show a dose dependency, which is also the case for the roots of both cultivars. Our findings suggest that an increase in activity of antioxidant enzymes, such as ascorbate peroxidase, catalase and glutathione reductase, can contribute to salt tolerance in Kennebec, a salt resistant cultivar of potato.展开更多
