Cultural relics line graphic serves as a crucial form of traditional artifact information documentation,which is a simple and intuitive product with low cost of displaying compared with 3D models.Dimensionality reduct...Cultural relics line graphic serves as a crucial form of traditional artifact information documentation,which is a simple and intuitive product with low cost of displaying compared with 3D models.Dimensionality reduction is undoubtedly necessary for line drawings.However,most existing methods for artifact drawing rely on the principles of orthographic projection that always cannot avoid angle occlusion and data overlapping while the surface of cultural relics is complex.Therefore,conformal mapping was introduced as a dimensionality reduction way to compensate for the limitation of orthographic projection.Based on the given criteria for assessing surface complexity,this paper proposed a three-dimensional feature guideline extraction method for complex cultural relic surfaces.A 2D and 3D combined factor that measured the importance of points on describing surface features,vertex weight,was designed.Then the selection threshold for feature guideline extraction was determined based on the differences between vertex weight and shape index distributions.The feasibility and stability were verified through experiments conducted on real cultural relic surface data.Results demonstrated the ability of the method to address the challenges associated with the automatic generation of line drawings for complex surfaces.The extraction method and the obtained results will be useful for line graphic drawing,displaying and propaganda of cultural relics.展开更多

The Drake Passage is located between the Antarctic Peninsula and Tierra del Fuego in the south of South America.Surface seawater samples were collected at seven sites in the Drake Passage during the austral summer of ...The Drake Passage is located between the Antarctic Peninsula and Tierra del Fuego in the south of South America.Surface seawater samples were collected at seven sites in the Drake Passage during the austral summer of 2012.The 16 S rRNA sequences were analyzed from 187 isolated bacterial strains.Three phyla,29 genera and 56 species were identified.The three phyla were Actinobacteria,Firmicutes and Proteobacteria;the Proteobacteria included a-Proteobacteria,P-Proteobacteria and y-Proteobacteria.y-Proteobacteria,Actinobacteria and Firmicutes were the dominant class or phyla in terms of quantity and species.Gram-positive bacteria(Actinobacteria and Firmicutes) accounted for 57.8% of all types identified.There were nine dominant genera,including Curtobacterium,Staphylococcus,and Halomonas,and 14 dominant species including Curtobacterium flaccumfaciens,Curtobacterium pusillum,and Staphylococcus sciuri.Of the strains identified,87.2% were catalase positive or weakly positive.展开更多
High enzymatic activity is required for laccase applications.Central composite design (CCD)-based response surface methodology (RSM) can effectively increase the enzymatic activity of Pleurotus ostreatus P40 in li...High enzymatic activity is required for laccase applications.Central composite design (CCD)-based response surface methodology (RSM) can effectively increase the enzymatic activity of Pleurotus ostreatus P40 in liquid substrate fermentation.Initial screening of the nutritional components was performed using a Plackett-Burman design.The variables,namely,bran,bagasse,Tween 80,and yeast extract,were found to have statistically significant effects on laccase activity.These variables were further optimized using CCD-based RSM.Optimal concentrations for the maximum laccase activity were 8.144 2 g/L bran,50 g/L bagasse,0.424 1 mL/L Tween 80,and 2.832 5 g/L yeast extract.Under optimized conditions,the maximum measured laccase activity reached 96 480 U/L,which was close to the predicted value (104 830 U/L) by RSM.Therefore,RSM can be used to optimize culture components for laccase activity from Pieurotus ostreatus P40.展开更多

The central composite desig...The factors selected to optimize the productivity of Pleurotus flabellatus biomass in 250ml working volume Erlenmeyer flask were agitation rate, initial pH value and incubation temperature. The central composite design was applied to study the significant factors and the interactions between the chosen factors, if present. The Design Expert software generated 20 runs. The optimized conditions obtained were as follows: the agitation rate of 129.8 rpm, incubation temperature at 27.8°C, and initial pH of 6.06. The optimized conditions tripled the productivity at the range of 980 - 1040 mg/litre/day compared to the initial rate productivity at 310 mg/litre/day. From the quadratic equation,the agitation rate, temperature and the interaction between agitation rate and temperature were found to be significant (p < 0.05). At optimum conditions, the experimental data supported the theoretical estimate.展开更多
Objective This study optimizes three-dimensional(3D) culture conditions of HepG2 using response surface methodology(RSM) based on the VitroGel system to facilitate the cell model in vitro for liver tissues.Method HepG...Objective This study optimizes three-dimensional(3D) culture conditions of HepG2 using response surface methodology(RSM) based on the VitroGel system to facilitate the cell model in vitro for liver tissues.Method HepG2 cell was 3D cultured on the VitroGel system.Cell viability was detected using Cell Counting Kit-8(CCK-8) assay of HepG2 lived cell numbers.The proliferation of HepG2 cell and clustering performance was measured via fluorescence staining test.Albumin concentration in the culture medium supernatant as an index of HepG2 cell biological function was measured with ELISA kit.Independent factor tests were conducted with three key factors:inoculated cell concentration,cultured time,and dilution degree of the hydrogel.The preliminary results of independent factor tests were used to determine the levels of factors for RSM.Result The selected optimal culture conditions are as follows:concentration of inoculated cells was4.44 × 10^(5)/mL,culture time was 4.86 days,and hydrogel dilution degree was 1:2.23.The result shows that under optimal conditions,the predicted optical density(OD) value of cell viability was 3.10 and measured 2.978 with a relative error of 3.94%.Conclusion This study serves as a reference for the 3D HepG2 culture and constructs liver tissues in vitro.Additionally,it provides the foundation for repeated dose high-throughput toxicity studies and other scientific research work.展开更多

Microfluidic device with patterned microstructures on the substrate surface was used to regulate cell adhesion,morphology,and functions in tissue engineering.We developed a microfluidic device which employing microsca...Microfluidic device with patterned microstructures on the substrate surface was used to regulate cell adhesion,morphology,and functions in tissue engineering.We developed a microfluidic device which employing microscale patterned microstructures to achieve enhanced cell adhesion and migration.Biocompatible hydrogel substrates with micro-wavy and lattice-patterned microstructures were fabricated using standing surface acoustic waves and ultraviolet solidification.After seeding the L929 mouse fibroblast cells onto the patterned substrate of the microfluidic device,we determined that the viability and proliferation rate of cell migration can be greatly enhanced.Furthermore,L929 cells showed two types of gathering modes after 48 h of culturing.Cell growth was guided by the patterned substrate used in the microfluidic device and showed differences in the location distribution.Therefore,the developed microfluidic device with patterned microstructures can extend the application of in vitro cell culturing for future drug development and disease diagnosis.展开更多

Three different materials including platinum, gold, and pyrolyzed photoresist derived carbon material, in the forms of plasma-treated and non-treated ones, were tested. Surface properties of the microelectrodes in terms of surface morphology and wettability were examined; then their biocompatibility was tested by co-culturing with SK-N-SH neuroblastoma cells. Results of experiments demonstrated that, compared with platinum and gold, carbon could be a better substrate for cell adhesion and growth,especially for the plasma-treated carbon surface. The high wetting property of plasma-treated carbon accounted for the preferable adhesion of cell on its surface. Therefore, plasma-treated carbon can potentially be employed for fabrication of biocompatible and stable neural electrodes, which is beneficial for neural engineering research, such as regeneration from injury or disease therapy of neural system.展开更多

In this study, surface response models were developed to...Kinetin is an important growth hormone used for in vitro propagation, but its dynamic and temporal effects on Dioscoreaalata have not been thoroughly evaluated. In this study, surface response models were developed to better elucidate the effects ofkinetin on D. alata propagated in vitro. Nodal segments were obtained from Akaaba, an important D. alata cultivar in Ghana, andpropagated in vitro under five kinetin rates (0, 2.5, 5, 7.5 and 10 μM). The models were developed using segmented multipleregression with time and kinetin as the predictors. The effects on plant height, the number of leaves, shoots and roots were assessedwith three-dimensional figures for better observation of temporal trends. The model fit was very good with normalized root meansquared error (NRMSE) = 0.1, R-squared = 0.83 and adjusted R-squared = 0.82, averaged across the different growth parameters.Different kinetin levels elicited the maximum shoot, leaf and root formation, as well as the growth rates over time. Moderate kinetinlevels (2-4 μM) provided better growth at early culturing period. Higher kinetin levels (5-10 μM) suppressed the growth of theplantlets at early stages, but the plantlets recovered from the stress and resumed normal growth thereafter. After 4-5 weeks, thegrowth rates of the moderate kinetin levels (2-4 μM) declined much faster and were lower compared to the higher kinetin levels,except plant height and the number of roots which were still higher at the moderate kinetin level even after eight weeks of culturing.Thus, kinetin requirements vary depending on the growth parameters of interest.展开更多

This paper makes a general analysis of the negative transfer of surface-structure culture in language forms and that of deep-structure culture in values, thought patterns, religious beliefs and ethics. It holds that failure in intercultural communication will occur if inadequate attention is paid to cultural differences in the process of language and culture learning.展开更多
