Nine primer combinations were used and 385 fragments were detected. Among the 385 fragments, 80 bands (20.78%) were polymorphic. And it can be speculated that the genetic diversity of the three cultured populations of jade perch was very poor according to the gene genetic diversity among populations (Ht), the gene genetic diversity within populations (Hs) and Shannon-Weiner index (I). From gene differentiation (Gst), genetic distance (D), genetic similarity (5), and UPGMA analysis, it is found that the relationship among the three populations is very closed, and the difference in genetic diversity among the three populations is not significant. Further clustering relationships of the jade perch cultured populations also are correlated to historical-breeding observations and genetic relationships. The GZ population clustered together with the QD population first, then with FS population.展开更多

In the contrasts of immune-associated gene expression between areas and populations, significant differences were found, expression levels of these immune-associated genes were lower in the clear water area than in the poor water quantity area, and lower in May than in October. MY was more sensitive to environmental factors than DQ, which was coincident with the water quality in the culturing areas. Differential analyses of the expression levels of these immune-associated genes showed that significant up-regulation could be triggered by poor environmental factors. The expression patterns indicated that the expression levels of these genes were sensitive to ecological changes, thereby the immune-associated genes, especially Pgrn-a, Ifit2, β2m, Il25 and Hsp96, might serve as immediate and sensitive indicators of population immunologic vigor and ecosystem health. But the expression of immunity-associated genes at the level of gene transcription is highly influenced by multiple factors, and the exact causes or influencing factors of the up-regulation or down-regulation of these genes still need further thorough investigation.展开更多

The change in soil bacterial PLFAs was always accompanied by a change in the soil eulturable bacterial population in the first flush. Comparatively higher culturable bacterial population and bacterial PLFAs were found in the casing soil at the primordia formation stage of the first flush. There was a significant increase in the ratio of fungal to bacterial PLFAs during mushroom growth. Multivariate analysis of PLFA data demonstrated that the mushroom cropping stage could considerably affect the microbial community structure of the casing soil. The bacterial population increased significantly from casing soil application to the primordia formation stage of the first flush. Casing soil application resulted in an increase in the ratio of gram-negative bacterial PLFAs to gram-positive bacterial PLFAs, suggesting that some gram-negative bacteria might play an important role in mushroom sporophore initiation.展开更多
