The results of PCR, restriction enzyme analysis and sequencing indicated that the Rxo1 gene had been cloned into pMNDRBBin6. This double right-border binary vector, named as pMNDRBBin6-Rxol, will play a role in breeding marker-free plants resistant to bacterial leaf streak of rice by genetic transformation.展开更多

For this purpose the wind field at a level under consideration is taken in the ratio form with that of 850 hPa level and the coefficients of the double Fouricr series are computed. These coefficients are subsequently used to computethe winds which are compared with the actual winds. The results of the double Fourier series technique are comparedwith those of the polynomial surface fitting method developed by Bavadekar and Khaladkar (1 992). The technique isalso applied for the daily wind data of 11. June, 1979 and the validation of the technique is tested for a few radiosondestations of india. The computed winds for these radiosonde stations arc quite close to observed winds.展开更多

According to the analysis of the received light spot size and its motion with the changes of the defocusing amount of detector photosensitive surface and the detector position attitude in the optical path, a mathematic expression of photoelectrical conversion is given, which can be applicable to random setting position of the detector at any time. Based on least square support vector machine (LS SVM), the mapping relationship among the output signal linear characteristic parameters (zero neighborhood gradient and intercept), the defocusing amount of the detector and the installation position attitude angle is established. Thus, the multiple dimensional high accuracy measuring and adjusting control system can be left out, and adaptive measurement of the detector parameters can be realized. Compared with existed measurement model and method, the presented model has the advantages of more clear physical meaning, closer to work mechanism of detector, acquiring more complete sample data and wiping out the dead spots or bad spots in measurement. And the accuracy of displacement measurement is increased to 3?μm. At the same time, this measurement mode provides a technical shortcut for three-dimensional small angle measurement.展开更多

A binary vector strategy was designed in 1983 to separate the T-DNA region in a small plasmid from the virulence genes in avirulent T-DNA-less Ti plasmid. The small plant vectors with the T-DNA region have been simply now called binary Ti vectors. A binary Ti vector consist of a broad host-range replicon for propagation in A. tumeraciens, an antibiotic resistance gene for bacterial selection and the T-DNA region that would be transferred to the plant genome via the bacterial virulence machinery. The T-DNA region delimited by the right and left border sequences contains an antibiotic resistance gene for plant selection, reporter gene, and/or any genes of interest. The ColEI replicon was also added to the plasmid backbone to enhance the propagation in Escherichia coli. A general trend in the binary vector development has been to increase the plasmid stability during a long co-cultivation period of A. tumefaciens with the target host plant tissues. A second trend is to understand the molecular mechanism of broad host-range replication, and to use it to reduce the size of plasmid for ease in cloning and for higher plasmid yield in E. coli. The broad host-range replicon of VS1 was shown to be a choice of replicon over those of pRK2, pRi and pSA because of the superior stability and of small well-defined replicon. Newly developed plant binary vectors pLSU has the small size of plasmid backbone (4566 bp) consisting of VS1 replicon (2654 bp), ColE1 replicon (715 bp), a bacterial kanamycin (999 bp) or tetracycline resistance gene, and the T-DNA region (152 bp).展开更多

Among the different change detection techniques, change vector analysis(CVA) have an exceptional advantage of discriminating change in terms of change magnitude and vector direction from multispectral bands. The estimation of precise threshold is one of the most crucial task in CVA to separate the change pixels from unchanged pixels because overall assessment of change detection method is highly dependent on selected threshold value. In recent years, integration of fuzzy clustering and remotely sensed data have become appropriate and realistic choice for change detection applications. The novelty of the proposed model lies within use of fuzzy maximum likelihood classification(FMLC) as fuzzy clustering in CVA. The FMLC based CVA is implemented using diverse threshold determination algorithms such as double-window flexible pace search(DFPS), interactive trial and error(T&E), and 3×3-pixel kernel window(PKW). Unlike existing CVA techniques, addition of fuzzy clustering in CVA permits each pixel to have multiple class categories and offers ease in threshold determination process. In present work, the comparative analysis has highlighted the performance of FMLC based CVA overimproved SCVA both in terms of accuracy assessment and operational complexity. Among all the examined threshold searching algorithms, FMLC based CVA using DFPS algorithm is found to be the most efficient method.展开更多
